Methyl mercury induced visual deficits in rainbow trout

Scotopic spectral sensitivity of 15 rainbow trout was determined using a two-choice, operant-conditioning task. Maximum sensitivity occurred at 525 nm and generally declined at longer and shorter wavelengths with evidence of a "shoulder" at 600–650 nm. Fifteen days after injection with either saline control (n = 4) or 4.6–6.2 mg of methyl mercury chloride per kilogram of body weight (n = 9); measurements of response latency and trials to criterion revealed that methyl mercury (MEHG) did not affect memory or motor performance. MEHG did produce significant spectrally uniform decrements in visual sensitivity. These results suggest that MEHG impaired both the scotopic and photopic mechanisms of MEHG-treated rainbow trout.

The median tolerance limit (MTL) for methyl mercury chloride at 24, 48, and 96 h was 0.084, 0.045, and 0.024 mg/liter as mercury, respectively, for fry; and 0.125, 0.066, and 0.042 mg/liter as mercury, respectively, for fingerlings. The MTL (24 h) for mercuric chloride for fingerlings was 0.90 mg/liter as mercury. Fingerlings exposed to methyl mercury chloride concentrated mercury in their tissues much more rapidly than did those exposed to mercuric chloride. The acute toxic action of both compounds was exerted on the gills. Mercuric chloride caused severe epithelial necrosis. Poisoning by methyl mercuric chloride was characterized by epithelial cell swelling and hyperplasia, a marked increase in the number of epithelial cells in mitosis, and terminal epithelial desquamation.

Rainbow trout (Salmo gairdneri) fingerlings were fed rations containing 4, 8, 16, and 24 ppm mercury as methyl mercury chloride over a 105-day period. Fish receiving the 16- and 24-ppm rations had significantly higher blood packed cell volumes than did controls. Hyperplasia of the gill epithelium was the only morphologic alteration detected in these fish. Individual fish accumulated up to 30 ppm mercury in muscle tissue, but no mortality that could be attributed to mercury occurred.The results suggest that trout can tolerate a large body burden of mercury, if this mercury is acquired over a period of time.

Quantitative data are presented for three laboratory experiments concerning rate of gastric evacuation of pellets (fed in gelatin capsules) and pellet components. Rainbow trout, Salmo gairdneri, acclimated to a constant water temperature of 15 C were killed 12 hr after consuming an experimental meal. Subtraction of amount of dry matter remaining at autopsy from dry matter consumed yielded amount of food digested or evacuated or both, from the stomach per unit time.Meals of encapsulated pellet were evacuated from the stomach more rapidly (65.8% decrease in 12 hr) than encapsulated corn oil (42.6%), gelatin (50.8%), a combination of corn oil and gelatin (47.3%), saturated fat (28.8%), or methyl cellulose (50.3%).Groups of fish consuming five capsules equal to approximately 0.65 % of their body weight evacuated 35.9, 45.1, 64.2, 95.5, and 100% at intervals after killing from 6 to 36 hr, respectively. Similar groups consuming seven capsules equal to approximately 1.0% of their body weight evacuated 23.7, 57.9, 70.5, and 86.6% at intervals after killing from 4 to 20 hr, respectively.Ten groups of trout consuming amounts of dry matter equal to 0.24, 0.40, 0.74, 1.11, 1.31, 1.19, 1.59, 1.56, 1.91, and 2.26% of their body weight evacuated 90.7, 81.3, 64.2, 57.9, 56.6, 52.5, 53.4, 51.3, 58.7, and 50.0% in 12 hr, respectively. Gastric motility remains relatively constant once food has entered the stomach. However, when larger meals are fed a greater quantity is evacuated per unit time. This could only be accomplished by changes in volume of gastric contents pumped per peristaltic stroke or number of strokes per unit time.

The aim of this study was to determine oxidative stress caused by mercury chloride (HgCl2) in rainbow trout (Oncorhynchus mykiss) liver tissue. For this purpose, the LD50 value of HgCl2 on rainbow trout was determined as 551 μg/L. In the study, 40 fish in four groups were exposed to 25% and 50% (138 and 276 µg/L) of the two subletal doses of HgCl2 for 2 and 7 days, with 10 fish (n=10) in each group. To determine oxidative stress; peroxynitrite (ONOO−), total oxidant level (TOS), total antioxidant level (TAS), oxidative stress index (OSI) and malondialdehyde (MDA) were analyzed. In the study, it was observed that the differences between the groups in terms of ONOO−, TOS, TAS and OSI levels in the liver tissues was significant (P<0.05), however, this difference was not significant (P>0.05) in terms of MDA values. As a result, it can be concluded that HgCl2 increases ONOO−, TOS, TAS, OSI and MDA levels in liver tissue and even small doses of mercury are toxic to fish.

Objective To investigate whether the Notch–Hif-1α signaling pathway is involved in liver regeneration. Methods Rats were divided into two groups and treated with daily intraperitoneal injections of saline (control) or the gamma-secretase inhibitor, Fli-06, for 2 days. Two-thirds of the rat livers were resected and rats were later euthanized at specific time points post-resection to analyze the remnant livers. Each group's liver/body weight ratio was calculated, and immunostaining and western blotting were used to determine the cell proliferation marker, PCNA and Ki-67 expression. Real-time PCR and western blotting were used to compare the mRNA expression of Notch homolog-1 ( Notch1), hairy and enhancer of split-1 ( Hes1), and vascular endothelial growth factor ( Vegf), and the protein expression of NICD and HIF-1α, respectively. Results The liver/body weight ratios and number of Ki-67- and PCNA-positive cells were significantly lower in the experimental group than the control group, indicating lower levels of liver regeneration following the disruption of Notch signaling by Fli-06. The Hes1 and Vegf mRNA levels and NICD and HIF-1α protein expression levels were all down-regulated by Fli-06 treatment. Conclusion Notch–Hif-α signaling pathway activation plays an important role in liver regeneration, where it may contribute toward liver cell proliferation.
