To investigate the correlation and clinical significance between programmed cell death factor 4 (PDCD4) and epidermal growth factor receptor 2 (Her-2) expressions and clinicopathological parameters in patients with gastric cancer, a total of 65 cases of gastric cancer and the corresponding normal mucosa with PDCD4 and Her-2 protein expressions were detected by SP immunohistochemical staining, and 50 cases of gastric cancer and the corresponding normal mucosa with PDCD4 and Her-2 protein expression quantities were detected by Western blot, in order to analyze the relationship between the positive expressions of PDCD4 and Her-2 protein and the clinicopathological features of patients with gastric cancer. The results showed that the positive rate of PDCD4 protein expression in gastric cancer tissues was 7.7%, which was significantly lower than that in the corresponding normal tissues, that is, 77.5% ( P < 0.05); the positive rate of Her-2 expression was 41.5%, which was significantly higher than that of the corresponding normal tissues, which is 2.5% ( P < 0.05). The Western blot test showed that the expression of PDCD4 protein in gastric cancer was 0.3105 ± 0.0073, which was significantly lower than that in the corresponding normal tissues, that is, 0.9428 ± 0.0127 ( P < 0.05); the expression level of Her-2 protein in gastric cancer tissues was 0.9428 ± 0.0127, which was significantly higher than that of the corresponding normal mucosa, which is 0.2054 ± 0.0264 ( P < 0.05). The positive expressions of PDCD4 (5/65) and Her-2 (27/65) were significantly correlated with the differentiation degrees and TNM stages of gastric cancer ( P < 0.05). However, no significant correlation can be observed from Table 2 ( P > 0.05), regarding sex, age, tumor size, and lymph node metastasis. Our research claimed that PDCD4 and Her-2 may play an important role in the invasion and metastasis of gastric cancer, which has a negative correlation with biological behaviors of gastric cancer. The low expression of PDCD4 and the high expression of Her-2 in gastric cancer may promote the occurrence and progression of cancer. The PDCD4 and Her-2 test can be used as an index to evaluate the malignant biological behaviors of gastric cancer and prognosis, and provide a theoretical basis for targeted therapy.

The aim of this study is to investigate the expression levels and clinical significance of ILF2 in gastric cancer. The mRNA and protein expression levels of ILF2 were, respectively, examined by quantitative real-time PCR (qRT-PCR) and Western blot from 21 paired fresh frozen GC tissues and corresponding normal gastric tissues. In order to analyze the expression pattern of ILF2 in GC, 60 paired paraffin-embedded GC slides and corresponding normal gastric slides were detected by immunohistochemistry (IHC) assay. The correlation between ILF2 protein expression levels and clinicopathological parameters, overall survival (OS), disease-free survival (DFS), and clinical prognosis were analyzed by statistical methods. Significantly higher levels of ILF2 were detected in GC tissues compared with normal controls at both mRNA and protein level. High expression of ILF2 was tightly correlated with depth of invasion, lymph node metastasis, pathological stage, and histological differentiation. Log-rank test showed that high expression of ILF2 was positively associated with poor clinical prognosis. Multivariate analysis identified that ILF2 was an independent prognostic factor for OS and DFS. Our findings suggest that ILF2 may be a valuable biomarker and a novel potential prognosis predictor for GC patients.

Abstract Background and objective: Human epidermal growth factor receptor 2 (HER2) is a key pathological characteristic in gastric cancer patients. However, the clinical significance of HER2 protein expression in gastric carcinoma remains controversial. The purpose of the study is to analyze the clinicopathological characteristics of HER2 protein expression, Lauren classification and P53 expression and evaluate the clinical significance of the HER2 protein expression. Methods: A total of 176 consecutive patients were recruited prospectively between January 2014 and December 2016 in The Second Affiliated hospital of Zhejiang University School of Medicine. Histological analysis was performed on resected tissue for HER2 protein expression by immunohistochemistry (IHC). The patients with IHC grade 2+ were analyzed by fluorescence in situ hybridization (FISH) to assess the expression status of HER2 protein. Moreover, standardized criteria of HER2 protein expression in gastric cancer was used in this study. Additionally, the expression status of HER2 protein and clinicopathological features were analyzed by Chi-square (c2) test. All statistical analyses were conducted using the SPSS 22.0 statistical software program (IBM Corp., SPSS statistics, Chicago, IL).Results: A total of 176 gastric cancer patients were enrolled in this study. Intratumorally heterogeneity of HER2 protein overexpression was 42 of 176 cases with IHC grade 2+ accompanied with FISH positivity and IHC grade 3+. HER2 protein expression correlated with tumor differentiation (p < 0.001), Lauren classification (p = 0.001), Borrmann type (p = 0.003) and P53 expression (p < 0.001). Overall survival (OS) was not analyzed because the follow-up duration was too short and the high rate of missed interview.Conclusions: The overexpression of HER2 protein was determined in 23.9% of the cases and significantly related to Lauren intestinal subtype and P53 expression.
