Long non-coding RNAs (lncRNAs) play important roles during the initiation and progression of cancer. We identified DiGeorge Syndrome Critical Region Gene 5 (DGCR5) as a clear cell renal cell carcinoma (ccRCC) cancer- and lineage-specific lncRNA. Agarose gel electrophoresis analysis and sanger sequencing verified two main isoforms of DGCR5 in ccRCC patient tissues and cell lines. Quantitative polymerase chain reaction further demonstrated that the expression level of DGCR5 major isoform (isoform-1) was higher in ccRCC tissues than that in papillary/chromophobe RCC and other multiple solid malignant tumors. We investigate the biological functions of DGCR5 isoform-1 in ccRCC and show that DGCR5 isoform-1 exerts a tumor-promoting effect in ccRCC. DGCR5 isoform-1 is localized in cytoplasm and shares the same binding sequence to the tumor-suppressive miR-211-5p with the epithelial-to-mesenchymal transition key component SNAI. Furthermore, cellular and molecular experiments demonstrate that DGCR5 isoform-1 could sequester miR-211-5p, leading to the elevation of Snail protein and downregulation of its downstream targets and further promoting ccRCC cell proliferation and migration. Thus, our study indicates that DGCR5 isoform-1 could contribute to ccRCC progression by sponging miR-211-5p through regulating the expression of Snail protein and could serve as a reliable diagnostic biomarker in ccRCC.

Abstract The authors reported a potential candidate methylated mud snail protein (MeMsp) as an effective and eco-friendly flocculant to treat the high turbidity wastewater. MeMsp was obtained by extraction of mud snail protein (Msp) through isoelectric precipitation (PSC-IP) and then methylated via the esterification with side-chain carboxyl. Structural characterization of FT-IR, Zeta potential and elemental analysis were carried out and further confirmed the successful of the methylation Flocculation experiments with kaolin suspension simulated wastewater indicated that MeMsp-24 displayed more excellent flocculation efficiency at a low dosage. At the optimum dosage 27 mg/L, the maximum clarification efficiency of MeMsp-24 was 97.46% under pH 7.0. Furthermore, MeMsp-24 exhibited a wide flocculation window in the pH range of 1.0–9.0, and faster sedimentation velocity and larger flocs size. Meanwhile, MeMsp-24 exhibited 92.12% clarification efficiency in treating railway tunnel construction effluent. The flocculation kinetic and mechanism analysis revealed that the most effective particle collision occurred at the optimal dosage, with charge neutralization and adhesion played irreplaceable roles in different environment, respectively. Therefore, through the extraction and methylation modification, the MeMsp could be a promising eco-friendly flocculant for high turbidity wastewater treatment.

Background: The paper explored the mechanism of Poriaic acid-containing serum interfering with EMT and apoptosis of A549/DDP cells. The aim is to find experimental evidence of Poriaic acid intervening cisplatin resistance in lung cancer, searching for effective targets, and to explore the mechanism of cisplatin resistance in lung cancer. Material and methods: Immunochemistry and western blotting were employed to detect the effects of Poriaic acid-containing serum on the expressions of p-GSK-3β (ser9), Snail protein and mRNA in GSK-3β/Snail signaling pathway, and the effects of Poriaic acid-containing serum on the expressions of EMT markers and related apop-totic factors. Results: The results of immunoblotting and immunocytochemistry rendered that the expressions of p-GSK-3β (ser9), Snail protein and mRNA decreased in the administration group as contrast to the blank group. As to the effect of Poriaic acid-containing serum on EMT markers, the immunoblotting results showed that the E-cadherin protein and mRNA expressions increased while the expressions of N-cadherin protein and mRNA decreased. Poriaic acid-containing serum can up-regulate the expressions of P53, Bax protein and mRNA, and down-regulate the expressions of Bcl-2 protein and mRNA. Conclusion: Poriaic acid-containing serum can affect EMT and apoptosis of A549/DDP cells by interfering with GSK-3β/Snail signaling pathway.

ABSTRAK Penelitian ini bertujuan untuk mengetahui pembuatan sate dan semur keong mas sebagai makanan sumber protein bagi anak sekolah dasar. Penelitian ini bersifat Quasi Eksperimen, menggunakan 2 produk olahan keong mas yaitu sate dan semur. Uji kesukaan pada panelis agak terlatih sebanyak 25 orang dan uji daya terima dari produk terpilih pada 25 orang siswa SD Inpres Negeri Siboi-Boi Kabupaten Jayapura. Hasil uji kesukaan menunjukkan bahwa yang paling disukai adalah olahan sate keong mas yaitu warna (98%), aroma (98%), rasa (98%) dan tekstur (98%). Hasil uji daya terima menunjukkan bahwa siswa yang memiliki daya terima >80% untuk hari ke-1 hingga hari ke-2 sebanyak 100% siswa. Kata kunci : Keong Mas, Sumber Protein, Uji Daya Terima, Uji Kesukaan ABSTRACT This research aims to study the making of satay and stew as a protein source for elementary school children. The research was quasi-experiment. We used two processed golden apple snail products i.e. satay and stew. . Preference test was conducted to 25 semi-trained panelists; while acceptance test was conducted to 25 students of SD Inpres Negeri Siboi-boi Jayapura. According to the result of preference test, the most preferable processed golden apple snail product was satay (color 98%, aroma 98%, taste 98%, and texture 98%). The result of acceptability test indicated that all students showed acceptability of >80% on the first and second days (100%).
