Lipid utilisation during storage of the entomopathogenic nematodes Steinernema feltiae and Heterorhabditis megidis from Greece and the UK

Abstract The total lipid (TL) content and the fatty acid composition of the different lipid classes in infective juveniles of Steinernema feltiae and Heterorhabditis megidis isolates from Greece (GR) and from the United Kingdom (UK) were compared, and the changes during storage were determined. Steinernema feltiae GR had a greater amount of TL than S. feltiae UK, which was reflected in a greater neutral lipid (NL) content. The phospholipid and free fatty acid fractions of the two Steinernema populations were similar. The two Heterorhabditis populations did not differ in their lipid content. In both S. feltiae populations, NL decreased by ca 60% during the first 4 weeks but remained more or less stable during the next 16 weeks. In the two Heterorhabditis populations, NL decreased by ca 70% during the first 4 weeks and continued to decrease, with 3-5% remaining after 12 weeks. The fatty acid profiles and the percentages of saturation of NL and phospholipid fractions in each population were similar before and after storage.

AbstractThe infectivity of populations of the entomopathogenic nematodes Steinernema feltiae and Heterorhabditis megidis from Greece (GR) and the UK was compared using Galleria mellonella larvae as hosts. Dose-response tests showed that the two Steinernema populations did not differ in their establishment rates but they were more infective than H. megidis UK 211. The temperature range for infectivity was greater than that for development. However, the optimal temperature for infection and development for all populations was 23°C. Infectivity of Steinernema populations was not affected by storage for 12 weeks. However, 12 week-old H. megidis UK 211 infective juveniles (IJ) were less infective than fresh IJ. H. megidis GR showed very low establishment rates at all the doses and temperatures tested, before and after storage. The results are discussed in relation to the nematodes' climatic origin and lipid content. Pouvoir infestant de populations des nématodes entomopathogènes Steinernema feltiae et Heterorhabditis megidis suivant la température, l'âge et le contenu lipidique - Le pouvoir infestant de populations des nématodes entomopathogènes Steinernema feltiae et Heterorhabditis megidis provenant de Grèce et du Royaume Uni a été comparée, utilisant comme hôte Galleria mellonella. Les tests de dose/réaction ont montré que les taux d'établissement des deux populations ne diffèrent pas mais que leur pouvoir infestant était plus élevée que celle de H. megidis UK211. La plage des températures permettant l'infestation était plus étendue que celle relative au développement. Cependant, les températures optimales pour l'infestation et pour le développement étaient l'une et l'autre de 23°C pour toutes les populations. L'infestivité des populations de Steinernema n'a pas été affectée par un stockage de 12 semaines. Les juvéniles infestants de H. megidis UK211 âgés de 12 semaines montraient toutefois une infestivité plus faible que celle d'individus frais. Les specimens de H. megidis provenant de Grèce présentaient - que ce soit avant ou après le stockage - des taux d'établissement très faibles pour toutes les doses et les températures testées. Ces résultats sont discutés en relation avec l'origine climatique et le contenu lipidique des nématodes.

The aquaculture of macroalgae for human consumption and other high-end applications is experiencing unprecedented development in European countries, with the brown algae Saccharina latissima being the flag species. However, environmental conditions in open sea culture sites are often unique, which may impact the biochemical composition of cultured macroalgae. The present study compared the elemental compositions (CHNS), fatty acid profiles, and lipidomes of S. latissima originating from three distinct locations (France, Norway, and the United Kingdom). Significant differences were found in the elemental composition, with Norwegian samples displaying twice the lipid content of the others, and significantly less protein (2.6%, while French and UK samples contained 6.3% and 9.1%, respectively). The fatty acid profiles also differed considerably, with UK samples displaying a lower content of n-3 fatty acids (21.6%), resulting in a higher n-6/n-3 ratio. Regarding the lipidomic profile, samples from France were enriched in lyso lipids, while those from Norway displayed a particular signature of phosphatidylglycerol, phosphatidylinositol, and phosphatidylcholine. Samples from the UK featured higher levels of phosphatidylethanolamine and, in general, a lower content of galactolipids. These differences highlight the influence of site-specific environmental conditions in the shaping of macroalgae biochemical phenotypes and nutritional value. It is also important to highlight that differences recorded in the lipidome of S. latissima make it possible to pinpoint specific lipid species that are likely to represent origin biomarkers. This finding is relevant for future applications in the field of geographic origin traceability and food control.

The fatty acid composition of the plasma free fatty acid and adipose tissue triacylglycerol fractions was determined in lean and fat selection lines of Texel-Oxford and Scottish Blackface sheep at the 6th year of divergent selection. The mean proportion of 18[ratio ]2n-6 in the triacylglycerol of subcutaneous backfat was 1·3-fold higher in the phenotypically fatter sheep in the fat lines than in the phenotypically leaner sheep in the lean lines. Regression analysis indicated a positive relationship between this fatty acid and backfat depth whereas the proportion of 18[ratio ]1n-9 in the tissue triacylglycerol was negatively related to fatness. The proportions of 18[ratio ]2n-6 and of other polyunsaturated fatty acids in the plasma free fatty acid fraction were much higher than in adipose triacylglycerol. For the Scottish Blackface sheep in the fed state, the mean proportion of 18[ratio ]2n-6 in plasma free fatty acid (measured on all sheep) was 1·4-fold greater in the fat line than in the lean line. Regression analysis indicated a positive relationship between the plasma content of this fatty acid and backfat thickness whereas the proportion of 18[ratio ]1n-9 in plasma free fatty acid showed a negative relationship with fatness. The relationship between the plasma proportion of 18[ratio ]2n-6 and fatness was not observed after 48 h of fasting; instead, the plasma proportion of 18[ratio ]0 was found to be positively related to fatness in the fasted state. In summary, this paper shows how plasma and adipose tissue fatty acid profiles differ, and it quantifies the effects of selection on the plasma profiles. Possible reasons for the difference in fatty acid profiles between adipose tissue and plasma are discussed in the paper. It is suggested that plasma 18[ratio ]2n-6 levels during the early post-weaning growth period should be investigated as indicators of future fatness.

AbstractThe desiccation survival and ability to control water loss during drying of individual infective juveniles (IJs) of populations of Steinernema feltiae and Heterorhabditis megidis from Greece and the UK were examined at different relative humidities. Survival at all relative humidities was for minutes only and there was no evidence of enhanced survival of the populations of either species from Greece compared with those from the UK. The survival of individuals of both populations of H. megidis was superior to that of S. feltiae and the rate of drying of H. megidis was significantly slower. The sheath surrounding the IJs of H. megidis appears to be involved in slowing the rate of water loss of the enclosed nematodes.

Although emerging data suggests a greater influence of gluconeogenic precursors, endurance sled dogs have long appeared to rely heavily on fatty acid oxidation for sustained energy production. However, much of the research investigating lipid utilization during exercise in sled dogs has been carried out with dogs subjected to extended bouts of endurance exercise. Less is known about changes in fatty acid composition in endurance training sled dogs subjected to short bouts of exercise, and fewer data define how fatty acid composition may change in distinct lipid fractions. As such, the study objective was to assess whether short bouts of submaximal exercise would affect fatty acid profiles of serum lipid fractions in endurance training sled dogs. Fifteen privately-owned Siberian huskies were used (8 females: 4 intact, 4 spayed; 7 males: 2 intact, 5 neutered), with an average age of 4.6 ± 2.5 years and body weight of 24.8 ± 4.2 kg. Throughout the diet acclimation and remainder of the study, all dogs were fed a dry extruded diet that met or exceeded all AAFCO nutrient recommendations. Dogs were weighed weekly and fed to maintain baseline body weight. A 12-week exercise regimen was designed to incorporate weekly increases in running distance, but weather played a role in setting the daily distance. On weeks 2, 5, and 11, an exercise challenge was implemented whereby dogs would run 4 km at 15 km/h in teams of 4. Pre- and post-exercise blood samples were taken, and gas chromatography was used to evaluate fatty acid profiles of all identified serum lipid fractions (cholesterol ester, diacylglycerol, free fatty acid, phospholipids, triglyceride). Data were analyzed using PROC MIXED of SAS, with dog as a random effect and week and sampling time point as fixed effects. Composition of oleic (18:1n9), linoleic (18:2n6), and alpha-linolenic (18:3n3) acids in the free fatty acid fraction decreased by ~9, 10, and 60%, respectively, following exercise (P ≤ 0.05). The results presented herein suggest that aside from a degree of depletion of these 18-carbon unsaturated fatty acids, short bouts of submaximal exercise do not induce considerable changes to sled dog fatty acid profiles.

Abstract Background Coeliac disease (CD) is characterised by diverse clinical symptoms, which may cause diagnostic problems and reduce the patients’ quality of life. A study conducted in the United Kingdom (UK) revealed that the mean time between the onset of coeliac symptoms and being diagnosed was above 13 years. This study aimed to analyse the diagnostic process of CD in Poland and evaluate the quality of life of patients before and after CD diagnosis. In addition, results were compared to the results of the original study conducted in the UK. Methods The study included 2500 members of the Polish Coeliac Society. The patients were asked to complete a questionnaire containing questions on socio-demographic factors, clinical aspects and quality of life, using the EQ-5D questionnaire. Questionnaires received from 796 respondents were included in the final analysis. Results The most common symptoms reported by respondents were bloating (75%), abdominal pain (72%), chronic fatigue (63%) and anaemia (58%). Anaemia was the most persistent symptom, with mean duration prior to CD diagnosis of 9.2 years, whereas diarrhoea was observed for the shortest period (4.7 years). The mean duration of any symptom before CD diagnosis was 7.3 years, compared to 13.2 years in the UK. CD diagnosis and the introduction of a gluten-free diet substantially improved the quality of life in each of the five EQ-5D-5L health dimensions: pain and discomfort, anxiety and depression, usual activities, self-care and mobility (p < 0.001), the EQ-Index by 0.149 (SD 0.23) and the EQ-VAS by 30.4 (SD 28.3) points. Conclusions Duration of symptoms prior to the diagnosis of CD in Poland, although shorter than in the UK, was long with an average of 7.3 years from first CD symptoms. Faster CD diagnosis after the onset of symptoms in Polish respondents may be related to a higher percentage of children in the Polish sample. Introduction of a gluten-free diet improves coeliac patients’ quality of life. These results suggest that doctors should be made more aware of CD and its symptoms across all age groups.

The aim of this study was to evaluate the polymorphic effects of two single nucleotide polymorphisms (SNPs) of fatty acid binding protein (FABP4) and stearoyl-CoA desaturase (SCD) genes on intramuscular fatty acid profiles in the longissimus muscle in two cattle breeds. Two previously reported SNPs of bovine FABP4 (7516G>C) and SCD (878C>T) were in turn assessed for their associations with intramuscular fatty acid profiles from the upper sirloin cuts of Aberdeen Angus and Blonde d’Aquitaine cattle. In total, 33 animals were genotyped using PCR-RFLP. Intramuscular fatty acid composition was evaluated using two complementary statistical approaches: a classical univariate regression model and a multivariate approach using a combination of Principal Component Analysis and Random Forests. Significant effect of FABP4 SNP genotypes was found for several fatty acids including C15:0, C17:0, C18:0, C14:1, C17:1, C18:2n6, C20:4n6, C20:5n3, C22:5n3, total n-3, n-6 and total SFA (P < 0.05). These results suggest that FABP4 is a potential candidate gene affecting fatty acid composition in beef cattle.
