Muyocopronales, ord. nov., (Dothideomycetes, Ascomycota) and a reappraisal of Muyocopron species from northern Thailand

Muyocopron species are associated with a wide variety of plant substrates worldwide and presently 57 species epithets are listed in Index Fungorum. Species in this genus form distinctive black, dull, rounded regions on the surface of plants and the genus is probably polyphyletic. The present study clarifies the phylogenetic placement of Muyocopron and related species, using fresh tropical collections from northern Thailand. Three Muyocopron species are characterized based on analyses of combined LSU and SSU sequence datasets. Phylogenetic analyses indicate that Muyocopron species form a distinct lineage with the Dyfrolomycetales and Acrospermales lineages. The new order Muyocopronales with three new Muyocopron species is introduced based on its distinct phylogeny and unique morphological characteristics. The taxonomy and phylogenetic relationships of tropical Muyocopron species are reappraised with suggestions for future work.

Amanita submelleialba sp. nov. in section Amanita, is described from northern Thailand based on both multi-gene phylogenetic analysis and morphological evidences. It is characterized by having small to medium-sized basidiomata; a yellow to yellowish pale pileus covering pyramidal to subconical, white to yellow white volval remnants; globose stipe base covered conical, white to yellow white volval remnants; fugacious subapical annulus; and absent clamps. Multi-gene phylogenetic analyses based on partial nuclear rDNA internal transcribed spacer region (ITS), partial nuclear rDNA larger subunit region (nrLSU), RNA polymerase II second largest subunit (RPB2), partial translation elongation factor 1-alpha (TEF1-α) and beta-tubulin gene (TUB) indicated that A. submelleialba clustered together with A. elata and A. mira, but represented as a distinct lineage from other extant species in section Amanita. The detailed morphological characteristics, line-drawing illustration and comparisons with morphologically similar taxa are provided.

Lasiodiplodia species are commonly as endophytes, saprobes and pathogens in tropics and subtropics. During an investigation of Botryosphaeriaceae in Thailand, two Lasiodiplodia taxa were isolated. Morphological characteristics and phylogenetic analyses based on combined ITS, tef and tub2 sequence data support the establishment of a novel species, Lasiodiplodia chiangraiensis, isolated from woody hosts. Lasiodiplodia chiangraiensis is phylogenetically close to L. iraniensis and L. thailandica, but represents a distinct lineage. The new species could be distinguished from extant Lasiodiplodia species by its mature conidial dimensions. A detailed description and illustration are provided, as well as an updated phylogenetic tree (ITS, tef and tub2) including all species (with available molecular data) of Lasiodiplodia. In addition, the accepted genera in Botryosphaeriaceae based on recent studies are given.

A new species of rust fungus, Puccinia marrubii sp. nov. is described from Turkey. The new species is the first Puccinia species to be identified on genus Marrubium (Lamiaceae). In order to determine the phylogenetic relationships of the new species, ITS region was amplified by PCR, sequenced, and analyzed. In addition to the ITS region of seven Puccinia species, P. annularis, P. jasmini, P. malvacearum, P. nigrescens, P. phlomidis, P. salviae, and P. serpylli were sequenced for the first time. Phylogenetic analyses performed by using distance, Bayesian methods confirmed that the new rust was closely related to P. serpylli, P. malvacearum, P. salviae, and P. annularis. The diagnostic morphological characteristics of the new species were discussed with the phylogenetically related species.

Abstract “Dok Hin” is the Thai local name for Selaginella species that form rosettes. They commonly distributes in Siberia, Manchuria, southern China, Japan, the Philippines and Thailand. Morphology of Dok Hin is very resemble leading to misidentification. So, exactly number of species of Dok Hin in Thailand and their differences in morphological characteristics is not well understood. Thus, revision of morphological characters and phylogenetic confirmation of the taxonomic identification are needed. This study aims to examine morphological charateristics and phylogenetic patterns in eight populations of the Dok Hin in Northern Thailand. Morphology of Dok Hin from each populations was quantitatively examined using 15 vegetative and 6 reproductive characters meanwhile phylogenetic analyses was explored by DNA barcode ITS2. The results of the phylogenetic analysis revealed the existence of two species of Dok Hin, S. tamariscina and S. pulvinata. Selaginella tamariscina can be distinguished from S. pulvinata by its presence of a pseudotrunk above ground and ridges of dorsal leaves. On the other hand, the results of phylogenetic analysis indicated the differences among populations of S. pulvinata as well. Chiang Mai populations of S. pulvinata was characterized by peculiar set of characters long leaves and leaf apices look like caudate, while the rest of their populations have shorter leaves and leaf apices look like aristate. It indicates that S. pulvinata has genetic and phenotypic divergence among populations. However, additional studies of Dok Hin populations in other parts of Thailand and studies on different genetic markers are necessary to confirm the taxonomic status of S. pulvinata. This study presents one telamonioid species new to science based on morphological characteristics and molecular phylogenetic analyses. Cortinarius khinganensis was collected from the Greater Khingan Mountains, Northeast China and it is characterized by hygrophanous, vivid brownish red and striate pileus, white universal veil, and subglobose spores. According to phylogenetic analyses results, C. khinganensis belongs to the section Illumini, which is a lineage distantly related from subgenus Telamonia sensu stricto. Detailed descriptions of the new species and the comparisons with morphologically similar species are provided. The phylogenetic relationships within the section Illumini are also discussed.

During a faunal survey in southern Xizang, we collected 27 specimens of voles that could not be identified as any knownspecies in the Arvicolinae. These specimens shared the following morphological characteristics, not corresponding withany other arvicoline species: the first lower molar possessed five closed triangles, the third upper molar exhibited eitherfour or three inner angles, and the tails of all specimens measured 30% of the body length. Their proximal baculum of theglans was very sturdy and trumpet-shaped, the distal baculum was tongue-like and sturdy, and the lateral bacula were veryshort. Molecular phylogenetic analyses based on nucleotide sequences of the mitochondrial cytochrome b (cyt b) geneclustered these specimens as a distinct lineage within the genus Neodon. According to the morphological and moleculardata, we described them as a new species, Neodon linzhiensis. Our phylogenetic analysis strongly supported that Lasio-podomys fuscus, Phaiomys leucurus, Neodon sikimensis, N. irene and the new species formed a monophyletic group, notincluding N. juldaschi. We suggested that L. fuscus and P. leucurus should be transferred to Neodon and that N. juldaschishould be removed from this genus. Following our new delineation of Neodon, we proposed a redefinition of the morphological diagnostic characters of the genus.

The soft coral family Xeniidae, commonly found in tropical and subtropical regions, consists of 20 genera and 162 species. To date, few studies on this family have been conducted in Japan, especially at higher latitudes. Although molecular phylogenetic analyses have recently been used to distinguish soft coral species, it is difficult to identify species and genera in this family due to the limited taxonomic indices and high morphological variation. In this study, we found a large Xeniidae community off the coast of Oshima Island (31°31.35'N, 131°24.27'E) at Miyazaki, Kyushu Island, located in the temperate region of Japan. The species composition and molecular phylogenetic relationships were investigated to uncover the species diversity of Xeniidae in this community. A total of 182 xeniid specimens were collected and identified to the species level, after which the samples were molecularly analyzed using a mitochondrial marker (ND2) and a nuclear marker (ITS) to infer the phylogenetic relationships. A total of 14 xeniid species were identified, including five undescribed species from five genera (Anthelia, Heteroxenia, Sympodium, Xenia, and Yamazatum). Miyazaki was identified as having the highest xeniid species diversity in Japan. The molecular phylogenetic trees inferred from each marker recovered very similar topologies: four genera (Anthelia, Heteroxenia, Sympodium, and Yamazatum) were monophyletic, whereas one (Xenia) was polyphyletic. Thus, except for Xenia, the morphological characteristics used for traditional taxonomy well reflected the phylogeny of the Xeniidae at the genus level. On the other hand, our results show that further taxonomic revisions of Xenia are needed.

The fungal genus Conidiobolus sensu lato was delimited into four genera based on morphology and phylogeny. However, the taxonomic placement of C. parvus has not been determined until now. Here, we show that C. parvus belongs to a distinct lineage based on mitochondrial (mtSSU) and nuclear (TEF1 and nrLSU) phylogenetic analyses. Phylogenetic analyses further revealed a new species as sister to C. parvus. We identified a synapomorphy uniting these lineages (azygospore production) that was not observed in other allied genera of the family Ancylistaceae, and erected a new genus Azygosporusgen. nov. for this monophyletic group, with a new combination, A. parvuscomb. nov. as the type species. Within Azygosporus, the novel species A. macropapillatussp. nov. was introduced from China based on morphological characteristics and molecular evidence, which is characterized by its prominent basal papilla, in comparison to other closely related species, measuring 7.5–10.0×5.0–10.0 µm. Our study resolved the phylogenetic placement of C. parvus and improved the taxonomic system of the Ancylistaceae family.

Abstract Background The genus Ligusticum consists of approximately 60 species distributed in the Northern Hemisphere. It is one of the most taxonomically difficult taxa within Apiaceae, largely due to the varied morphological characteristics. To investigate the plastome evolution and phylogenetic relationships of Ligusticum, we determined the complete plastome sequences of eight Ligusticum species using a de novo assembly approach. Results Through a comprehensive comparative analysis, we found that the eight plastomes were similar in terms of repeat sequence, SSR, codon usage, and RNA editing site. However, compared with the other seven species, L. delavayi exhibited striking differences in genome size, gene number, IR/SC borders, and sequence identity. Most of the genes remained under the purifying selection, whereas four genes showed relaxed selection, namely ccsA, rpoA, ycf1, and ycf2. Non-monophyly of Ligusticum species was inferred from the plastomes and internal transcribed spacer (ITS) sequences phylogenetic analyses. Conclusion The plastome tree and ITS tree produced incongruent tree topologies, which may be attributed to the hybridization and incomplete lineage sorting. Our study highlighted the advantage of plastome with mass informative sites in resolving phylogenetic relationships. Moreover, combined with the previous studies, we considered that the current taxonomy system of Ligusticum needs to be improved and revised. In summary, our study provides new insights into the plastome evolution, phylogeny, and taxonomy of Ligusticum species.

The family Salifidae is a predaceous leech taxon in the suborder Erpobdelliformes. Although Salifidae is widely distributed in the African, Oriental, Indo-Malayan, Sino-Japanese and Australasian regions, the phylogenetic relationships of the family Salifidae have never been tested using molecular data obtained from leeches collected from the family distributional range. A salifid species was collected for the first time in Vietnam, and relevant morphological and molecular data are presented here. Because the Vietnamese salifid species possesses unique morphological characteristics among the known salifid species, this species is herein described as a new species, Salifa motokawai, sp. nov. Phylogenetic analyses based on nuclear 18S rRNA and histone H3, as well as mitochondrial cytochrome c oxidase subunit I, tRNACys, tRNAMet, 12S rRNA, tRNAVal, 16S rRNA, tRNALeu and NADH dehydrogenase subunit 1 markers demonstrate that the Vietnamese salifid species is a close congener with the African Salifa perspicax and the Malagasy Linta be. Furthermore, molecular data revealed non-monophyly of the Asian salifid leeches. According to the observed phylogenetic relationships and morphological characteristics of the Vietnamese Salifa motokawai, sp. nov., the current classification of salifid taxa should be revised.
