Plenodomus dezfulensis sp. nov. causing leaf spot of Rapeseed in Iran

Rapeseed (Brassica napus subsp. napus) is one of the autumn crops in Iran. A new fungal species isolated from internal tissues associating a typical leaf spot of Rapeseed in Dezful (southwestern Iran) was described and illustrated. The sampling and isolation were performed in the late fall of 2019 (during December), which led to obtain the colonies with the similar morphology. Molecular phylogenetic analyses based on a combined matrix of the internal transcribed spacer regions 1 and 2 including the intervening 5.8S nuclear ribosomal DNA (ITS), and partial regions of the β-tubulin (tub2) and RNA polymerase II second largest subunit (rpb2) indicated that two isolated strains represent a new Plenodomus species in family Leptosphaeriaceae (Pleosporales). These novel strains formed a sister lineage with P. biglobosus in our phylogenetic analyses, while, observed morphology is distinguished the taxa from P. biglobosus by its pycnidia in having hyphal outgrowths and lacking cylindrical neck. Accordingly, P. dezfulensis sp. nov. was introduced and its pathogenicity was investigated. Pathogenicity test with both P. dezfulensis strains on three-week seedlings of Rapeseed reproduced leaf spot symptoms. These results approve P. dezfulensis as the causal agent of leaf spot observed on Rapeseed in Dezful.

In our ongoing research on lepiotaceous taxa (Agaricaceae s.l.) in Laos and northern Thailand, we focus here on Chlorophyllum, Clarkeinda, Macrolepiota, Pseudolepiota, and Xanthagaricus. Collections were obtained from various habitats, including agricultural habitats, grasslands, and rainforests. A total of 12 taxa were examined and investigated. Of these 12, two are new for science; viz. Xanthagaricus purpureosquamulosus with brownish-grey to violet-brown squamules on a pale-violet to violet background; it shares the pileus color with X. caeruleus and X. ianthinus, but differs in other characters; and Macrolepiota excelsa, rather similar to M. procera but related toM. detersa. Two species, Pseudolepiota zangmui and Xanthagaricus necopinatus are recorded for the first time in Thailand. Four species of Chlorophyllum and a total of four species of Macrolepiota were found, viz., C. demangei and C. hortense with white basidiospores, C. molybdites and C. globosum with green basidiospores, M. detersa, M. dolichaula, the new M. excelsa, and M. velosa. Another rather common striking species is Clarkeinda trachodes, with yellow-green basidiospores. Each species is described in detail, with color photographs and line drawings. Phylogenetic analyses based on internal transcribed spacer (nrITS) region, the large subunit nuclear ribosomal (nrLSU) DNA and RNA polymerase II second largest subunit (rpb2) genes provide evidence for the placement of the species covered.

A new species, Calocybe decurrens, is illustrated and described in detail based on morphological characteristics and phylogenetic analyses. Calocybe decurrens is mainly characterized by its decurrent gills and by its stipe that discolors upon maturation. Molecular phylogenetic analyses were based on the internal transcribed spacer (ITS1-5.8S-ITS2) and the large subunit of the nuclear ribosomal DNA (nrLSU) sequences. The results indicated that its affiliation is in genus Calocybe, where it occupies an isolated position. A full description, color images, illustrations and a phylogenetic tree to show the placement of the new species are provided.

Russula darjeelingensis is characterized by its small sized white pileus with a tall and narrow stipe, white spore print, basidiospores with amyloid suprahilar spot and a pileipellis containing encrusted pileocystidia and absence of primordial hyphae. The combination of all these characters and molecular phylogenetic analyses of internal transcribed spacer sequences of nuclear ribosomal DNA confirmed it as a new species in genus Russula Pers., subg. Russula Romagn. emend. sect. Polychromae (Maire) Sarnari subsect. Paraintegrinae Sarnari. A comprehensive morphological description, illustrations, and comparisons with morphologically similar and phylogenetically related species are provided in the present study.

A new species of agaricomycetes, Clitopilus lampangensis, is described based on collections from northern Thailand. This species was distinguished from previously described Clitopilus species by its pale yellow to grayish yellow pileus with the presence of wider caulocystidia. Molecular phylogenetic analyses, based on the data of the internal transcribed spacers (ITS) and the large subunit (LSU) of the nuclear ribosomal DNA, and the second largest subunit of RNA polymerase II (rbp2) genes, also support the finding that C. lampangensis is distinct from other species within the genus Clitopilus. A full description, color photographs, illustrations and a phylogenetic tree showing the position of C. lampangensis are provided.

Eggplant (Solanum melongena L.) is an economically important solanaceous crop in Iran with fruits used for food and traditional medicine. Despite the importance of Alternaria leaf spot and blight disease of solanaceous crops which is commonly seen in the fields, our knowledge about the causal agents on eggplant is limited. In this study, a set of large-spored Alternaria isolates was recovered from eggplant with leaf spot and blight symptoms in Somehsara region, Guilan province, Iran. All recovered isolates shared conspicuous morphological characteristics e.g. production of large, solitary conidia with several transverse disto- and eusepta and long tapering filamentous beak resemble those seen in the members of Alternaria section Porri. Multi-locus phylogenetic analyses based on the internal transcribed spacer region of nrDNA (ITS-rDNA) and parts of glyceraldehyde-3-phosphate dehydrogenase (GAPDH), second largest subunit of RNA Polymerase II (RPB2), translation elongation factor 1-alpha (TEF1-α) and Alternaria major allergen (Alt a 1) gene sequences provided further evidence supporting not only their exact placement in Alternaria sect. Porri, but also in a distinct lineage representing a new species. The new species was named, described and illustrated herein as Alternaria guilanica sp. nov.. The phylogenetic and morphological comaprisions of the new species with other closely related species were also provided. Pathogenicity test conducted for the new strains revealed that they were capable to induce disease symptoms on eggplant leaves under greenhouse conditions, and re-isolation of the inoculated isolates confirmed Koch’s postulates.

Taxonomic and phylogenetic studies of Chlorophyllum were carried out on the basis of morphological differences and molecular phylogenetic analyses. Based on the phylogeny inferred from the internal transcribed spacer (ITS), the partial large subunit nuclear ribosomal DNA (nrLSU), the second largest subunit of RNA polymerase II (rpb2) and translation elongation factor 1-α (tef1) sequences, six well-supported clades and 17 phylogenetic species are recognised. Within this phylogenetic framework and considering the diagnostic morphological characters, two new species, C.africanum and C.palaeotropicum, are described. In addition, a new infrageneric classification of Chlorophyllum is proposed, in which the genus is divided into six sections. One new combination is also made. This study provides a robust basis for a more detailed investigation of diversity and biogeography of Chlorophyllum.

Amanita rufobrunnescens is described as a new species in the subgenus Lepidella section Amidella from Guangdong Province, China. It is characterized by whitish basidiomes that bruise reddish brown, brownish sub-membranous to fibrillose volval remnants on the pileus, a striate pileus margin, white lamellae with truncate lamellulae, grayish orange to light brown volva, amyloid basidiospores that are ellipsoid to elongate (9.5–)10–12(–13) × (5–)5.5–6.5(–7) µm, and 5–10 μm wide pileipellis hyphae with yellowish vacuolar pigments. There are no clamp connections. Molecular phylogenetic analyses based on the nuclear ribosomal large subunit of nuclear ribosomal DNA (LSU) and internal transcribed spacer (ITS) sequences reveal that A. rufobrunnescens is a distinct species.

The taxonomic status of Euryops jaberiana Abedin & Chaudhary (tribe Senecioneae, family Asteraceae), endemic to northern Saudi Arabia was evaluated based on molecular phylogenetic analyses of internal transcribed spacer sequence (ITS) of nuclear ribosomal DNA (nrDNA) in order to ascertain its position within the genus. The phylogenetic tree constructed by the Neighbour Joining, Maximum Parsimony and Maximum Likelihood analyses showed a clear resolution of taxon included in the analyses at the level of sections, and E. jaberiana nested within the clade of the section Angustifoliae. E. jaberiana showed proximity with the allied species E. arabicus; however, a total number of eight nucleotide differences were evident between E. jaberiana and E. arabicus, indicating E. jaberiana as distinct from its allied species.Bangladesh J. Relationships within the family Phytolaccaceae sensu lato were examined based on internal transcribed spacer (ITS) sequences of nuclear ribosomal DNA (nrDNA). The study revealed Phytolacca L. as taxonomically the most difficult genus in the family with completely unknown phylogeny. Molecular evidence was used from nrDNA ITS sequences of about 90% of the species for maximum parsimony analyses, and the molecular phylogenetic analyses defined a monophyletic Phytolacca. This first molecular phylogenetic study of Phytolacca concludes that the relationships among the species within the genus do not show harmony with the generic classification based on morphology. Camellia sinensis is one of the major crops grown in Taiwan and has been widely cultivated around the island. Tea leaves are prone to various fungal infections, and leaf spot is considered one of the major diseases in Taiwan tea fields. As part of a survey on fungal species causing leaf spots on tea leaves in Taiwan, 19 fungal strains morphologically similar to the genus Diaporthe were collected. ITS (internal transcribed spacer), tef1-α (translation elongation factor 1-α), tub2 (beta-tubulin), and cal (calmodulin) gene regions were used to construct phylogenetic trees and determine the evolutionary relationships among the collected strains. In total, six Diaporthe species, including one new species, Diaporthe hsinchuensis, were identified as linked with leaf spot of C. sinensis in Taiwan based on both phenotypic characters and phylogeny. These species were further characterized in terms of their pathogenicity, temperature, and pH requirements under laboratory conditions. Diaporthe tulliensis, D. passiflorae, and D. perseae were isolated from C. sinensis for the first time. Furthermore, pathogenicity tests revealed that, with wound inoculation, only D. hongkongensis was pathogenic on tea leaves. This investigation delivers the first assessment of Diaporthe taxa related to leaf spots on tea in Taiwan.
