A previously undescribed polyketide (1) and 3 known analogs (2-4) were obtained from cultures of the potato endophytic fungus Aspergillus carneus. The structures were elucidated on the basis of extensive nuclear magnetic resonance spectroscopic data. The absolute configuration of 1 was further determined by electronic circular dichroism and optical rotation calculations. Compounds 1-4 showed moderate antifungal activity against plant pathogens. Compounds 1, 2, and 4 inhibited nitric oxide production in lipopolysaccharide-stimulated RAW264.7 cells, with half-maximal inhibitory concentration values of 13.36, 30.16, and 51.47 µM, respectively. Compound 4 showed effective antioxidant activity.

Four undescribed compounds, guhypoxylonols A (1), B (2), C (3), and D (4), were isolated from the mangrove endophytic fungus Aspergillus sp. GXNU-Y45, together with seven previously reported metabolites. The structures of 1–4 were elucidated based on analysis of HRESIMS and NMR spectroscopic data. The absolute configurations of the stereogenic carbons in 1–3 were established through a combination of spectroscopic data and electronic circular dichroism (ECD). Compounds 1–11 were evaluated for their anti-inflammatory activity. Compounds 1, 3, 4, and 6 showed an inhibitory activity against the production of nitric oxide (NO), with the IC50 values of 14.42 ± 0.11, 18.03 ± 0.14, 16.66 ± 0.21, and 21.05 ± 0.13 μM, respectively.

Abstract Three previously undescribed cytochalasins, named xylariasins A‒C (1‒3), together with six known ones (4‒9) were isolated from Xylaria sp. CFL5, an endophytic fungus of Cephalotaxus fortunei. The chemical structures of all new compounds were elucidated on the basis of extensive spectroscopic data analyses and electronic circular dichroism calculation, as well as optical rotation calculation. Biological activities of compounds 1, 4‒9 were evaluated, including cytotoxic, LAG3/MHC II binding inhibition and LAG3/FGL1 binding inhibition activities. Compounds 6 and 9 possessed cytotoxicity against AGS cells at 5 μM, with inhibition rates of 94% and 64%, respectively. In addition, all tested isolates, except compound 6, exhibited obvious inhibitory activity against the interaction of both LAG3/MHC II and LAG3/FGL1. Compounds 1, 5, 7, and 8 inhibited LAG3/MHC II with IC50 values ranging from 2.37 to 4.74 μM. Meanwhile, the IC50 values of compounds 1, 7, and 8 against LAG3/FGL1 were 11.78, 4.39, and 7.45 μM, respectively. Graphic Abstract

Piper solmsianum C. DC. compounds exhibit several properties, including antimicrobial activity. The aim of the present study was to investigate whether conocarpan alters Candida albicans growth or killing of the yeast by macrophages. Conocarpan showed strong activity against the yeast with minimal inhibitory concentration (MIC) of 20 µg/mL and minimal fungicidal concentration (MFC) of 30 µg/mL. Mice peritoneal cells (macrophages) were cultured for 24 and 48 hours in supplemented RPMI 1640 medium. Cellular activation was assessed by determining MTT reduction and nitric oxide production. Standardized tests were conducted to select the optimal parameters for the subsequent killing test. Results showed that conocarpan exhibited antifungal activity and that C. albicans cultivated in the presence of the compound had greater susceptibility to death by macrophages. These findings suggest that conocarpan may have potential as an antimicrobial agent for C. albicans infections, promoting macrophagic immune support by altering growth of the yeast.

Three new lavandulylated flavonoids, (2S,2′’S)-6-lavandulyl-7,4′-dimethoxy-5,2′-dihydroxylflavanone (1), (2S,2′’S)-6-lavandulyl-5,7,2′,4′-tetrahydroxylflavanone (2), and (2′’S)-5′-lavandulyl-2′-methoxy-2,4,4′,6′-tetrahydroxylchalcone (3), along with seven known compounds 4–10 were isolated from culture broth of Streptomyces sp. G248. Their structures were established by spectroscopic data analysis, including 1D and 2D nuclear magnetic resonance (NMR), and high-resolution electrospray ionization mass spectrometry (HR-ESI-MS). The absolute configurations of 1–3 were resolved by comparison of their experimental and calculated electronic circular dichroism spectra. Compounds 1–3 exhibited remarkable antimicrobial activity. Whereas, two known compounds 4 and 5 exhibited inhibitory activity against Mycobacterium tuberculosis H37Rv with minimum inhibitory concentration (MIC) values of 6.0 µg/mL and 11.1 µg/mL, respectively.

One new diterpenoid, diaporpenoid A (1), two new sesquiterpenoids, diaporpenoids B–C (2,3) and three new α-pyrone derivatives, diaporpyrones A–C (4–6) were isolated from an MeOH extract obtained from cultures of the mangrove endophytic fungus Diaporthe sp. QYM12. Their structures were elucidated by extensive analysis of spectroscopic data. The absolute configurations were determined by electronic circular dichroism (ECD) calculations and a comparison of the specific rotation. Compound 1 had an unusual 5/10/5-fused tricyclic ring system. Compounds 1 and 4 showed potent anti-inflammatory activities by inhibiting the production of nitric oxide (NO) in lipopolysaccharide (LPS)-induced RAW264.7 cells with IC50 values of 21.5 and 12.5 μM, respectively.

AbstractGrowing crops in sabulous soils is often challenging due to their limited oligotrophy and weak water retention. However, some plants adapt to these adverse growth conditions, and in some cases, favorable properties are imparted to the fruit. This study investigated the influence of the cultivation environment on Capsicum by assessing the levels and functions of both pungent components and antioxidants when cultivated in sandy soils in Southern Japan; these parameters were then compared to those in traditional tropical-origin Capsicum. In seven varieties of Capsicum, the distribution of capsaicin and dihydrocapsaicin differed between the pericarp and seeds within the placenta. The leaves and fruits of Habanero orange and Tabasco, which are the most suitable for cultivation in sandy soil, were collected during the cultivation period and analyzed in terms of their size, color, and pungent component composition. Pungent components were detected in fruits only, and not in leaf or flower samples. In particular, we found that pungent components were generally present within the seeds and placenta. Antioxidant activity and nitric oxide production within human vascular endothelial cells were also evaluated to compare the differences in their functionality. Satsuma-Habanero orange cultivated under adverse conditions possessed the highest antioxidant activity. Furthermore, Satsuma-Capsicum cultivated under adverse conditions exhibited higher levels of antioxidants than traditional tropical-origin peppers, and induced similar levels of nitric oxide production in the vascular endothelial cells. We concluded that Capsicum cultivated in harsh environments produced beneficial effects such as higher levels of antioxidants and capsaicinoids in seeds and placenta. Moreover, the fruits from these plants could be harvested for a significantly longer period and took longer to spoil than traditional Capsicum; thus, they show merit as a viable commercial crop in Japan.
