Abstract Background Silybum marianum L. Gaertn is a medicinal plant of unique pharmaceutical properties in the treatment of liver disorders and diabetic nephropathy. Biochemical (SDS-PAGE) and molecular markers such as randomly amplified polymorphic DNA (RAPD) and inter-simple sequence repeats (ISSR) technologies were used in this work to detect genetic diversity of 14 collections of Silybum marianum population in Egypt. Results The electrophoretic pattern of seed protein gave different molecular weight bands, ranging from 24 to 111 KDa with the presence of unique bands. RAPD results revealed a high level of polymorphism (73.2%) using 12 RAPD primers, but only eight of them gave reproducible polymorphic DNA pattern. Sixteen primers were used in the ISSR method; only ten of them yielded clearly identifiable bands. The percentage of polymorphism is about 80% of the studied samples. Conclusion The obtained data confirmed that SDS-protein, RAPD, and ISSR markers are important tools for genetic analysis for Silybum marianum and recommended to give accurate results.

In this study, the phylogenetic relationship within the selected Eleven Indian (Dolichos biflorus (horse gram) varieties was analyzed for total soluble seed protein. Twenty-five bands were documented through SDS PAGE based on 100 seed weight of each variety and were studied for genetic diversity. Jaccard’s similarity matrix was acquired and used in UPGMA cluster analysis based on the polymorphism generated by the presence (1) or absence (0) of protein bands. Thus, the dendrogram showed four major groups that corre-spond to an earlier study on polymorphisms of 11 accessions of Indian Dolichos. Signifi-cant correspondence between the clustering pattern and the pedigree was observed; thus, a high genetic diversity could be kept within the Dolichos varieties. A similarity matrix among the targeted genotypes and phylogenetic analysis is considered a unique feature in the present work. Therefore, the current investigation was carried out to analyse Protein diversity of unexplored Dolichos genotypes at the molecular level, construct a dendro-gram based on similarity band matrix and generate efficiency in genetic divergence analy-sis among Dolichos. This study underlines the importance of using genetic diversity based in the Dolichos breeding program.

A total of 60 genotypes of Brassica campestris (L) germplasm were acquired from National Research Centre (NARC) Islamabad, Pakistan for the evaluation of genetic diversity using the morphological characters and SDS-PAGE analysis. These genotypes were grown-up in Plant Garden Department of plant sciences, University of Malakand, Khyber Pukhtunkhwa, Pakistan. For the assessment of genetic diversity total of 18 morphological traits were scored and their phylogenetic relationship were plot through cluster diagram. For the estimation of genetic diversity, we used the most influential technique SDS PAGE. We observed six loci (bands) from the collected genotypes; locus-1 (band-1) contained 100% bands of protein and obvious as monomorphic locus while loci i.e. 2,3,4,5 and 6 showed 28%, 42%, 36%, 56% and 25%, respectively variation and were polymorphic. The inter species locus influence toward genetic assortment (LCTGA) was 83.33% in the attained genotypes. Profiling through SDS PAGE based on two-way cluster plottingresolved effectively the collected genotype into 4 cluster. We consider that this work provides a baseline for the understanding genetic diversity in the common crops used in Pakistan and warrants further investigation in this newly evolving field of study.

Alnus nitida (Spach) Endl. is an ethnobotanically important threatened plant species. The genetic diversity among the 50 different genotypes of Alnus nitida was carried out using sodium dodecyl sulfate poly acrylamide gel electrophoresis (SDS-PAGE) characterization. A considerable amount of genetic diversity (90%) was observed among the genotypes of A. nitida. The protein characterization was carried out on 12% gel electrophoresis. A total of 10 protein bands were detected in A. nitida genotypes. SDS-PAGE procedure is a useful method for the investigation of both genetic diversity and phylogenetic relationship. Especially, B-5 was monomorphic in A. nitida genotypes and was considered as species specific. All other bands/loci were polymorphic. These polymorphic bands displayed 12, 16, 72, 88, 2, 44, 84, 54 and 12 percent variation respectively. In the present examination, the high intra-specific diversity was observed representing SDS-PAGE is a powerful tool for determining the genetically diverse germplasms in A. nitida. The results obtained by this study could be helpful in the identification and selection of desired genotypes of Alnus nitida for conservation programmes in future. Today, there is still a need to assess genetic variation and protect genetic resources, especially of wild species for prospective benefits in plant conservation programmes.
