In this research, with the aim of maximizing amino acid content by different hydrolization procedures of fish meat were carried out and analysed with high performance liquid chromatography (HPLC) equipped with diode array detector (DAD). Response surface methodology (RSM) was used to determine factors that used in the experiments. The RSM suggested 16 different hydrolization trials between the ranges of normality as 3 N - 8 N, temperature as 90°C-110°C and duration as 12-24 hours were run. In the following, hydrolysed samples were analysed with HPLC-DAD and obtained data were evaluated with Box-Behnken method in RSM. Our results show that, the most effective experiment was found as hydrolysing by 3.79 N at 110°C in 24 hours for total amino acid content as well as maximized essential amino acids with 0.981 desirability. For sensitive ones, hydrolysing in 3.42 N at 106.8°C in 12.02 hours was found as most effective with maximized Asn, Gln and Trp with 0.849 desirability.

Native bean populations (Phaseolus vulgaris L.) provide bioactive and nutrient compounds; however, their amino acid profiles are unknown. Therefore, the aim of this study is to evaluate the protein content and amino acid profile of 46 native bean populations cultivated by small farmers in Oaxaca, Mexico, and compare them with that of commercial beans. Through high-performance liquid chromatography (HPLC), 16 amino acids were identified and quantified in all samples. The region of origin influenced the concentrations of amino acids. The Santa Lucia Miahuatlan populations stood out for their high content of isoleucine, threonine, methionine, arginine, serine, alanine, tyrosine, and cysteine. Amino acid content showed high variability among the populations; accessions labeled as FSLM22, FSLM27, FSLM28, and FSLM32 were enriched in aliphatic, hydroxylated, aromatic, acidic, and basic amino acids, while the FSLM14, FSLM17, and FSLM18 populations had the highest concentrations of sulfur amino acids. The FSLM01, FSLM22, FSLM27, FSLM28, FSLM30, and FSLM32 populations frequently displayed the highest concentrations of essential amino acids. The findings show that samples of native populations are highly variable in amino acid content due to the genetic characteristics of cultivated beans, environmental and agroecological influences, and crop management by farmers. The beans populations stood out can be used for direct use or a basis for the initiation of a breeding program.

Kawal is made by a natural or uncontrolled fermentation of Senna obtusifolia leaves. It is a food condiment, and appreciated because of its particular flavor. It is also used as a meat or fish substitute. Kawal is an important source of protein in the diet of low-income families. The present work was done to determine the total amino acid content in raw and fermented Senna obtusifolia leaves. The high performance liquid chromatography (HPLC) with The Pico-Tag method was used for the analysis and determination of amino acid profiles. The results obtained showed that the kawal protein is rich in essential amino acids and semi-essential for humans, with different concentrations. Leucine gave a maximum value of 208.56 mg/100 g (DM) and 538,29 mg/100 g (DM); valine showed the value of 173,46 mg/100 g (DM) and 476.25 mg/100 g (DM), for lysine this value is 205.58 mg/100 g (DM) and 456.13 mg/100 g (DM). In the background, we have histidine, threonine, tyrosine, isoleucine and phenylalanine, whose best concentrations are between 157.36 mg/100 g and 397.93 mg/100 g (DM) in fermented leaves. For semi-essential amino acids, the fermented leaves have concentrations of between 121.96 mg/100 g and 1260.92 mg/100 g (DM).

Both inorganic and organic selenium (Se) can prevent and treat various diseases caused by Se deficiency. However, organic Se has less toxicity and a higher absorption rate than inorganic Se. In this study, inorganic Se (Na2SeO3) was bio-transformed into Se-enriched discarded beer yeast (Se-enriched DB-yeast) through fermentation accumulation by re-using discarded Saccharomyces cerevisiae from the beer industry for Se-enriched fodder application. Through a single-factor experiment and L9(34)-orthogonal test for optimization of fermentation conditions, the Se content and biomass of Se-enriched DB-yeast were calculated as 14.95 mg/L and 7.3 g/L, respectively, under the optimized condition. The total amino-acid content of Se-enriched DB-yeast was increased by 9.9% compared with that from DB yeast. Additionally, alkaline amino-acid content was increased, whereas acidic amino-acid and sulfur-containing amino-acid contents were decreased. Reducing capacity, hydroxyl radical removal capacity, and sulfhydryl content after treatment with H2O2 of the Se-enriched DB-yeast extracted protein were obviously increased compared with those of the DB-yeast extracted protein. Mouse and genetically improved farmed tilapia (Oreochromis niloticus) (GIFT) bioassays showed that the Se sedimentation of organs and serum indexes after feeding Se-enriched DB-yeast-containing fodder were higher than those of DB-yeast-containing fodder. The half lethal dose (LD50) of Se-enriched DB-yeast (9260.0 mg/kg body weight (BW), 18.97 mg/kg of Se content, non-toxic level) was considerably higher than that of Na2SeO3 (20.0 mg/kg BW, 5.08 mg/kg of Se content, highly toxic level) against mouse. Therefore, Se-enriched yeast prepared by re-using discarded S. cerevisiae from beer industry fermentation accumulation has the potential to be a safe and effective Se-enriched fodder additive.

Objective: To determine the protein quality, especially the amino acid content of 8 tropical fruits both raw and boiled samples. Eight different tropical fruits were used in the study (Apricot, Jamun, Dragonfruit, Pomegranate, Mangustan, Litchi, Jackfruit, and Kiwi.Methods: Ninhydrin method was used for the estimation of the concentration of amino acids present in the above fruits. Raw and boiled fruits were used for the study.Results: Both raw and boiled forms which showed thats Jamun and Mangustan contained highest concentration amino acids whereas apricot shows the lowest concentration of amino acids except in Jamun which showed higher values in the raw fruit whereas in others the boiled samples showed higher values.Conclusion: It was evident that tropical fruits have a good balance of the essential amino acids (both raw and boiled fomr) which provide significant sources of protein in our diet.
