B-Cell Leukemia in a Sheep

Necropsy examination was performed on an 8.5-year-old Finnish ewe euthanatized because of progressive respiratory distress, cachexia, and weakness. Significant postmortem findings included a diffusely enlarged, dark-red friable liver, mild splenomegaly, and mild mesenteric lymphadenopathy. Examination of multiple tissue sections revealed intravascular atypical mononuclear cells in all tissues examined, with a leukemic pattern of infiltration of mesenteric lymph nodes, liver, lung, and spleen. Neoplastic cells were positive for CD79a and negative for CD-3, BLA.36, and CD68 leukocytic markers. The final diagnosis was B-cell leukemia.

Pathological evaluations were done on 205 rats with mononuclear cell leukemia. Leukemia was diagnosed in 22.2% of males and 20.4% of females with significant risk beginning at 20 months of age. Mononuclear cell leukemia was responsible for 50% of early deaths in two-year studies. Clinically, rats became depressed, pale, icteric and had palpably enlarged spleens. Gross lesions included splenomegaly, enlarged mesenteric lymph nodes, and mottled livers. Hemorrhages occurred in the lungs, brain, and lymph nodes. Histological examination demonstrated that spleen and liver were most consistently and seriously involved, although numerous other organs contained leukemic infiltrates of variable severity. Spleens exhibited diffuse leukemic infiltration of the red pulp, follicular lymphoid depletion, and decrease in both extramedullary hematopoiesis and hemosiderin. Liver lesions consisted of diffuse centrilobular degeneration and necrosis. Erythrophagocytosis by tumor cells was common in the spleen and observed in liver, lymph nodes, and adrenals. The disease appeared to originate in the spleen. Bone marrow infiltration occurred late relative to spleen involvement and was present in less than half of the rats.

To understand the pathology of natural cases of rotavirus (RVA) in bovine calves, a total of 40 cases below 6 months died due to diarrhoea were studied, out of which 7 cases (17.5%) turned positive for RVA by RT-PCR. Histopathology of small intestine showed loss of villous enterocytes, blunting and fusion of villi, elongation of crypts and mononuclear cells infiltration in the lamina-propria. The mesenteric lymph nodes were severely depleted of lymphocytes. These changes were corroborated with presence of RVA antigen in sections by dFAT and nucleic acid by RT-PCR. The fluorescent signals were more in mesenteric lymph nodes than in intestine. Besides, 115 rectal fecal samples were also collected from calves for RVA detection by RT-PCR using VP6 gene specific sets of primers. Dead carcasses of calves (n= 40) belonged to organized dairy farm of Bareilly, while rectal fecal samples belonged to both organized (n= 38) and unorganized farms (n= 77) of Bareilly and Gwalior. The overall occurrence of RVA was 19.3% (30/155), comprising 5/37 cases (13.5%) from Gwalior (MP) and 25/118 cases (21.1%) from Bareilly (UP). These findings suggest the infection of RVA widely prevalent in calves and have potential to escape from the intestinal site to mesenteric lymph nodes.

In this study, the pathological lesions and occurrence of subclinical Johne's disease in Awassi sheep is investigated using histopathological (HP) and immunohistochemical (IHC) examinations, Acid Fast Staining (AFS) of tissue sections, direct smears from tissues and culture. Histopathological examination of 202 ilea and the corresponding mesenteric lymph nodes (179) was conducted. In addition, IHC examination, using rabbit polyclonal antiserum, of 134 ilea and 123 mesenteric lymph nodes was also conducted. The occurrence of the disease was observed in 50% and 93% of the ilea examined using histopathology and IHC techniques, respectively. Fifty nine percent of lymph nodes were positive by IHC. The histopathological lesions were graded from І–IV, I being the least severe, based on the type of cellular infiltrate (lymphocytes, macrophages and epithelioid cells) and the severity of the lesions. Grades III and IV (SP) were considered positive while I and II were considered suspected. Analysis of the results also revealed that the majority of suspected cases (grades I and II) reacted positive with the IHC. Furthermore, the IHC reactions were classified into three categories depending on the number of stained cells and the intensity of the staining (I-mild, II-moderate and III-strong). Direct smears, and tissue sections obtained from the ilea and stained with AFS revealed that out of 202 tissue samples, 53 (26%) and 22 (11%) were positive, respectively. Results of the culture revealed that 22 (11%) out of 202 were positive. These results showed that the occurrence of paratuberculosis (Johne's disease) in Awassi sheep is very high in Jordan and needs further study in order to develop rational methods of control effective for the Jordanian sheep population.

The large mononuclear cells showed the features of mononuclear phagocytes and were mainly characterised by a large number of vacuolar structures. The results suggest that there is storage of lipids and mucosubstances, probably in enlarged lysosomes.

Pulmonary bactericidal activity, macrophage phagocytic activity, alveolar macrophage (AM) enzyme activity, and T- and B-cell mitogenesis of lymphocytes from lung associated lymph nodes (LALN) or mesenteric lymph nodes (MESLN) were assessed in Sprague-Dawley rats exposed 4 hr/d, 4 days/wk for 1, 4, or 16 days to hexachlorobenzene (HCB) aerosols. Pulmonary bactericidal activity was depressed after 1 or 4 but not 16 exposures to 35 mg/m3 of HCB. AM phagocytosis of 51Cr-RBC in vitro was increased after 4 but not 1 or 16 exposures to HCB, and no effect was observed in peritoneal macrophages. HCB significantly enhanced mitogenesis in MESLN to the B-cell mitogen Salmonella typhimurium lipopolysaccharide (STM) after 4 exposures; LALN STM mitogenesis and LALN and MESLN mitogenesis to phytohemagglutinin (PHA) were not affected. After 16 exposures, however, the PHA responses in LALN and MESLN were significantly increased and decreased, respectively.

In this study was aimed to investigate spread and pathological findings of Linguatula serrata nymphs in mesenteric lymph nodes (MLNs) in sheep. A total of 1000 MLNs from 140 sheep (125 male and 15 female) were collected from a commercial (Konet) abattoir. Fourteen of 140 sheep (10%) were infested with nymph stages of L. serrata.Infested lymph nodes were swollen, firm, edematous, many yellowish-white pinpoint and with congestion. In histopathological examinations, these nodules were consist of one or more nymphal stages of L. serrata and these were surrounded by infiltration mononuclear cells and neutrophyl leucocyte. Lymphoid follicules were hyperplastic and sinus catarrh was also observed in medulla.
