ABSTRACT Objective: To evaluate whether vitamin C can help to prevent obesity and hyperglycemia in Wistar rats treated with excess invert sugar to induce prediabetes. Methods: One hundred-day-old Male Wistar rats with a mean weight of 336.58±23.43g were randomly assigned to the following groups: (1) control, receiving water (C); (2) invert sugar control, receiving a 32% watery solution of invert sugar; (3) vitamin C control, receiving a watery solution of vitamin C (60mg/L), and (4) vitamin C plus invert sugar, receiving a watery solution of vitamin C and invert sugar. All animals had access to chow and water ad libitum and were treated for 17 weeks. Prediabetes was assessed according to two criteria: obesity (based on body mass indexand peritoneal fat content) and impaired glucose tolerance (assessed by the intraperitoneal glucose tolerance test and expressed as area under the curve) . Results: Group invert sugar control gained significantly more weight (p=0.035) and visceral fat (p<0.001) than groups vitamin C control and vitamin C plus invert sugar. Consequently, groups vitamin C control and vitamin C plus invert sugar had gained as little body mass index as group C by the end of the experiment. Vitamin C decreased the fasting glycemia of both groups supplemented with vitamin C and normalized the glucose tolerance of group vitamin C plus invert sugar, whose area under the curve matched that of group C. Conclusion: Vitamin C has anti-obesogenic and glycemia-lowering effects in Wistar rats, which might be promising to prediabetics. Future studies are needed to understand the anti-obesogenic and anti-hyperglycemic mechanisms of vitamin C in prediabetes.

Hepatosteatosis is a complex disorder, in which insulin resistance and associated dyslipidemic and inflammatory conditions are fundamental. Dietary habit, especially regular consumption of fat and sugar-rich diet, is an important risk factor. Coconut and mustard oils (CO and MO) are medium-chain saturated and monounsaturated fats that are common dietary ingredients among the Indian populations. Present study analyzed the effect of prolonged consumption of the fresh and thermally oxidized forms of these oils on glucose tolerance and hepatosteatosis in male Wistar rats. Thermally oxidized CO (TCO) and MO (TMO) possessed higher amount of lipid peroxidation products and elevated p-anisidine values than their fresh forms. Dietary administration of TCO and TMO along with fructose altered glucose tolerance and increased hyperglycemia in rats. Dyslipidemia was evident by elevated levels of triglycerides and reduced high density lipoprotein cholesterol (HDLc) levels in fructose and edible oil-fed group ( p < 0.05). Additionally, hepatic antioxidant status was diminished and oxidative stress markers were elevated in TCO- and TMO-fed rats. Substantiating these, hike in liver function marker enzyme activities were also observed in these animals. Supporting this, histological analysis revealed higher incidence of microvesicles and hepatocellular ballooning. Results thus suggest that consumption of thermally oxidized fats may cause hepatic damage.

The present study was carried to develop and analyze the consequences of hypercaloric pellet-diet cycle that promotes obesity in rats. Male Wistar rats were randomly distributed into two groups that received either normal diet (ND; n =32; 3,5 Kcal/g) or a hypercaloric diet (HD; n =32; 4,6 Kcal/g). The ND group received commercial Labina rat feeding while the HD animals received a cycle of five hypercaloric diets over a 14-week period. The effects of the diets were analyzed in terms of body weight, body composition, hormone-metabolite levels, systolic arterial pressure and glucose tolerance at the 5% significance level. The hypercaloric pellet diet cycle promoted an increase in body weight and fat, systolic arterial pressure and a high serum level of glucose, triacylglycerol, insulin and leptin. The HD group also presented an impaired glucose tolerance. In conclusion, the results of this study show that the hypercaloric pellet-diet cycle promoted obesity in Wistar rats and displayed several characteristics that are commonly associated with human obesity, such as high arterial pressure, insulin resistance, hyperglycaemia, hyperinsulinaemia, hyperleptinaemia and dyslipidaemia.

Objective To evaluate the preventive effects of Moringa oleifera on metabolic syndrome (MS) in male Wistar rats. Methods MS was induced by feeding rats a high-fat diet and drinking water containing 10% fructose for 6 weeks. In the preventive group, M. oleifera was orally administered for 3 weeks prior to the induction of MS, while in the treatment group, M. oleifera was administered for 3 weeks after the onset of MS. The treatment groups were compared with a control group of untreated rats with induced MS. Fasting glucose, oral glucose tolerance, insulin tolerance, total cholesterol, triglycerides, abdominal circumference, and systolic and diastolic blood pressure were measured before and after MS induction and/or M. oleifera treatment. Results After the induction of MS, the control group had higher fasting glucose levels than the preventive group. No significant differences were observed in insulin tolerance, oral glucose tolerance, cholesterol, triglycerides, abdominal circumference, or systolic or diastolic blood pressure. Compared with untreated controls, rats in the treatment group had significantly improved glucose tolerance, triglycerides, and abdominal circumference. Conclusions M. oleifera treatment attenuates MS in Wistar rats.

Under experimental conditions severe reduction of renal mass results in the hyperfiltration of the remaining nephrons leading to a progressive renal insufficiency. Similar changes are observed in patients with various renal disorders associated with a loss of the functioning nephrons. The progression of renal damage is accelerated by high protein and phosphate intake, and may be modified by the dietary restrictions.We studied 50 five-sixth nephrectarrized male Wistar rats on a standard diet (Rodent Laboratory Chow 5001 Ralston Purina Co., Richmond, Indiana; containing 23.4% protein) over a 20 week period.
