In present study, the total phenolic, flavonoid, tannin contents and the antioxidant activity of various solvent extracts of Sphaeranthus indicus collected from three different regions of central India were assessed. Plants extracts were prepared using Soxhlation method, while the total phenols, flavonoids and tannins were measured by the spectrophotometric method. DPPH, metal chelating, nitric oxide, superoxide oxide scavenging activity and FRAP, reducing power, total antioxidant assays were also evaluated. The highest phenolic contents 268.22 GAE equivalent, flavonoids 441.33 QE equivalents and tannin content was 120.32 tannic acid equivalents (mg/g) obtained from ethanolic extracts of sample SIEE-1 compared to other two plants extract. Similarly, the highest flavonoid contents was observed in SIAE-1 aqueous extract and lowest in SIAE-2 and SIAE-3. Extract of SIEE-1 possessed maximum antioxidant potentiality and SIAE-2 shown the least antioxidant activity in all assays. It could be concluded that different agroclimatic conditions have effects on the total phenolics, flavonoids, tannin contents and antioxidant potentiality of S. indicus plant.

Exudates (DE) secreted from two shiitake mushroom mycelia (strains 1358 and L5458) were evaluated for their antioxidative properties and phenolic content. 1358DE and L5458DE showed distinct antioxidant activity in different in vitro assays, including scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, hydroxyl radical, superoxide anions and hydrogen peroxide; the ability to chelate ferrous ions; reducing power; hemolysis inhibition activity in rat erythrocyte; and lipid peroxidation inhibition (IC50 values of 1358DE and L5458DE were 3.3 and 132.6; 44.5 and > 1000; 26.9 and 53.7; 153.6 and >175.0; 176.0 and 521.0; 26.7 and 746.4; 47.8 and 736.9; and 3.1 and > 1000 μg/mL, respectively). Their total phenolic content was 237.33 and 24.08 mg gallic acid equivalent (GAE)/g of dry DE, respectively. Overall, these results show that 1358DE generally possesses better antioxidant properties than L5458DE, possibly due to its larger total phenolic content. Shiitake mushroom mycelial exudates, particularly of 1358DE, could be a good source of natural antioxidants.

The aim of this study was to determine the impending antioxidant properties of different extracts of crude methanolic extract (CME) of leaves ofLannea coromandelica(L. coromandelica) and its two ethyl acetate (EAF) and aqueous (AqF) subfractions by employing various establishedin vitrosystems and estimation of total phenolic and flavonoid content. The results showed that extract and fractions possessed strong antioxidant activityin vitroand among them, EAF had the strongest antioxidant activity. EAF was confirmed for its highest phenolic content, total flavonoid contents, and total antioxidant capacity. The EAF was found to show remarkable scavenging activity on 2,2-diphenylpicrylhydrazyl (DPPH) (EC5063.9 ± 0.64 µg/mL), superoxide radical (EC508.2 ± 0.12 mg/mL), and Fe2+chelating activity (EC506.2 ± 0.09 mg/mL). Based on ourin vitroresults, EAF was investigated forin vivoantioxidant assay. Intragastric administration of the EAF can significantly increase levels of superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), and glutathione peroxidase (GSH-Px) levels, and decrease malondialdehyde (MDA) content in the liver and kidney of CCl4-intoxicated rats. These new evidences show thatL. coromandelicabared antioxidant activity.

The present study was designed to confirm the traditional use of the fruits of Ficus hispida Linn. (Moraceae) as an antioxidant agent. Fruits of the plant extracted with methanol and crude methanol extract (CME) were further fractionated with n-hexane, chloroform, and ethyl acetate. All the fractions, n-hexane (NHF), chloroform (CHF), ethyl acetate (EAF), aqueous (AQF) and CME were preliminary screened for in vitro antioxidant activity and total phenolic and total flavonoid content. In DPPH radical scavenging assay, CME exhibited highest scavenging activity (IC50 = 11.20 µg/mL) as compared to other fractions. In this assay, IC50 of reference standard BHT was 5.10 µg/mL. The reducing power of the samples was in the order as AQF > CME > CHF > EAF > NHF. The results for hydrogen peroxide scavenging activity indicated that CME, EAF and AQF had almost the same scavenging activity except NHF. Total antioxidant capacity of CME and other fractions were ranked as CHF > AQF > CME > EAF > NHF.  In the assay of antioxidant constituents (total phenol and total flavonoids content), the CME had highest phenolic and flavonoids content. The results indicate that Ficus hispida fruits could be considered as a potential source of natural antioxidant.

Abstract Background Wedelia chinensis has been reported as a folk medicine for the treatment of different diseases including neurodegenerative disease. Although the plant has been studied well for diverse biological activities, the effect of this plant in neurological disorder is largely unknown. The present study was undertaken to evaluate the cholinesterase inhibitory and antioxidant potential of W. chinensis. Methods The extract and fractions of the plant were evaluated for acetylcholinesterase and butyrylcholinesterase inhibitory activity by modified Ellman method. The antioxidant activity was assessed in several in vitro models/assays such as reducing power, total antioxidant capacity, total phenolic and flavonoid content, scavenging of 2,2’-diphenyl-1-picrylhydrazyl (DPPH) free radical and hydroxyl radical, and inhibition of brain lipid peroxidation. Chromatographic and spectroscopic methods were used to isolate and identify the active compound from the extract. Results Among the extract and functions, aqueous fraction (AQF) and ethylacetate fraction (EAF) exhibited high inhibition against acetylcholinesterase (IC50: 40.02 ± 0.164 µg/ml and 57.76 ± 0.370 µg/ml) and butyrylcholinesterase (IC50: 31.79 ± 0.182 µg/ml and 48.41 ± 0.053 µg/ml). Similarly, the EAF and AQF had high content of phenolics and flavonoids and possess strong antioxidant activity in several antioxidant assays including DPPH and hydroxyl radical scavenging, reducing power and total antioxidant activity. They effectively inhibited the peroxidation of brain lipid in vitro with IC50 values of 45.20 ± 0.995 µg/ml and 25.53 ± 0.042 µg/ml, respectively. A significant correlation was observed between total flavonoids and antioxidant and cholinesterase inhibitory activity. Activity guided chromatographic separation led to the isolation of a major active compound from the EAF and its structure was elucidated as apigenin by spectral analysis. Conclusions The potential ability of W. chinensis to inhibit the cholinesterase activity and peroxidation of lipids suggest that the plant might be useful for the management of AD.

Abstract: Helicteres hirsuta, which belongs to the Malvaceae family, is used in traditional medicine to treat malaria, diabetes and cervical cancer. While these uses have not been validated in clinical studies thus far, extracts from H. hirsuta exhibit confirmed antioxidant and potential anti-cancer activity that warrant critical assessment. The aim of this study was to determine the antioxidant properties of different parts of the plant H. hirsuta. The antioxidant capacities of methanol extracts of different plant parts, and specific solvent fractions thereof were determined on the basis of 2,2-diphenyl1picrylhydrazyl (DPPH) radical scavenging and total antioxidant activity assays. The ethyl acetate fraction from leaf material of H. hirsuta has the most potent antioxidant activity with the lowest half maximal inhibitory con-centration (IC50) value of 9.50 μg/mL, that is 4 times lower than the IC50 of curcumin. The amounts of methyl gal-late and rutin dominated the extracts at 8.62 ± 0.01 and 6.76 ± 0.02 mg/g, respectively, out of a total of 21.00 ± 0.01 mg/g active phenolic fraction. The amount of methyl gallate strongly correlated with total phenolic content and total antioxidant capacity, thus this compound may be useful as a marker of antioxidant activity in common medicinal plants

<p>The present study was conducted to evaluate the antioxidant activity and chemical composition of aqueous methanolic extract of <em>Trichodesma indicum </em>(L.) R.Br. The extract was tested for antioxidant activity using various <em>in vitro</em> models viz., DPPH, ABTS, NO, FRAP, total antioxidant activity and reducing power. The total phenolic and flavonoid contents were found to be equivalents to 97.83 ± 0.08 µg of gallic acid and 45.45 ± 0.19 µg of rutin /mg of dried aqueous methanolic extract, respectively. Among various antioxidant assays performed, maximum inhibition was observed in the ABTS (IC<sub>50</sub> 17.13 ±  0.23 μg) followed by DPPH (IC<sub>50 </sub>80.64  ±  0.29 μg) and NO (IC<sub>50 </sub>4.12 ± 0.02  μg) assay. GC-MS analysis revealed the presence of over 38 compounds, the prevailing compounds were hexadecanoic acid, β-sitosterol, ethyl iso-allocholate and 9,12,15,-Octadecatrienoicacid.  The HPLC analysis further confirmed the presence of rutin, salicylic acid and benzoic acid. This confirms that <em>T. indicum</em> could be the good source of natural antioxidant for industrial and pharmaceutical preparations.</p>

Cordia macleodii, an important medicinal plant traditionally used for the treatment of various disorders, including wound healing, jaundice, gastric ulcer, hepatoprotective activity and antivenom potential, thus the plant showed the high antioxidant properties. Successive extraction of leaf and barks powder with methanol and water was tested for quantitative determination of total phenolics, total flavonoids and various in vitro antioxidant activities. Phytochemical screening of crude plant extracts revealed the presence of sterols, alkaloid, flavonoid, phenolic, Reducing sugar, glycosides, tannins and saponins. All extracts showed the significant total phenolic and flavonoid contents and as well as radical scavenging and iron-chelating activities. Among all, methanol leaf extract was observed to be higher antioxidant activity than that of other extract. The methanol extracts showed greater antioxidant activity by DPPH scavenging the free radical with IC50 values of 7.63 ± 0.38 μg gm-1 along with scavenged free radical in a concentration depended manner, which is compare to the standard ascorbic acid.  Our results revealed that the presence of grater antioxidant activity of C. macleodii extract may be due to the high level of flavonoid and phenolic content in the plant. These results clearly indicated that C. macleodii is an effective natural antioxidant. Key words: Cordia macleodii, antioxidant, radical scavenging, phenolics, flavonoid, phytochemical.

Abstract Background The objective of the study is to evaluate the hepatoprotective activity of methanolic extract fractions of Lindernia ciliata (LC) and development of qualitative analytical profile of the bioactive fraction using HPLC fingerprinting analysis. All the fractions of methanolic extract of Lindernia ciliata (LCME) are assessed for their total phenolic, flavonoid contents and in vitro antioxidant properties by using DPPH, superoxide, nitric oxide, hydroxyl radical scavenging activities and reducing power assay. Acute toxicity study was conducted for all the fractions and the two test doses 50 and 100 mg/kg were selected for the hepatoprotective study. Liver damage was induced in different groups of rats by administering 3 g/kg.b.w.p.o. paracetamol and the effect of fractions were tested for hepatoprotective potential by evaluating serum biochemical parameters and histology of liver of rats. The effective fraction was evaluated for its antihepatotoxic activity against D-Galactosamine (400 mg/kg b.w. i.p.) and in vivo antioxidant parameters viz., Glutathione (GSH), Melondialdehyde (MDA) and Catalase (CAT) levels are estimated using liver homogenate. Results Among all the fractions, butanone fraction of LCME, (BNF-LCME) has shown better hepatoprotective activity and hence it is selected to evaluate the antihepatotoxicity against D-GaIN. The activity of BNF-LCME is well supported in in vitro and in vivo antioxidant studies and may be attributed to flavonoidal, phenolic compounds present in the fraction. Hence, BNF-LCME was subjected to the development of qualitative analytical profile using HPLC finger printing analysis. Conclusions All the fractions of LCME exhibited significant hepatoprotective activity and BNF-LCME (50 mg/kg) was identified as the most effective fraction.

The antioxidant activities of crude extract ofPhaulopsis fascisepalaleaf were evaluated and compared with α-tocopherol and BHT as synthetic antioxidants and ascorbic acid as natural-based antioxidant.In vitro, we studied its antioxidative activities, radical-scavenging effects, Fe2+-chelating ability and reducing power. The total phenolic content was determined and expressed in gallic acid equivalent. The extract showed variable activities in all of thesein vitrotests. The antioxidant effect ofP. fascisepalawas strongly dose dependent, increased with increasing leaf extract dose and then leveled off with further increase in extract dose. Compared to other antioxidants used in the study, α-Tocopherol, ascorbic acid and BHT,P. fascisepalaleaf extract showed less scavenging effect on α,α,-diphenyl-β-picrylhydrazyl (DPPH) radical and less reducing power on Fe3+/ferricyanide complex but better Fe2+-chelating ability. These results revealed thein vitroantioxidant activity ofP.fascisepala.Further investigations are necessary to verify these activitiesin vivo.
