<p>This study was aimed at determining the phytochemical composition, antioxidant effect and acute toxicity of Irvingia gabonensis (O’Rorke) baill (IG) ethanolic leaf extract. Qualitative phytochemcal analysis was carried out on the ethanolic leaf extract using standard procedures. Different concentrations of the plant extract (20 µg/ml- 100µg/ml) were used to assess its effect on 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical activity, It's reducing power and it total phenolic content. Lorke’s method of acute toxicity was adopted for the acute toxicity study. Results obtained showed the presence of saponins, tannins, flavonoids, cardiac glycosides, steroids, and carbohydrates in the leaf extract. The ethanolic leaf extract of IG significantly (P&lt; 0.05) inhibited the activity of DPPH when compared with the vitamin C standard. IG leaf extract also showed a higher reducing power as compared with the Vitamin C standard. The total phenolic content of IG ethanolic leaf extract was however, significantly (P&lt;0.05) lower than that of gallic acid, the standard used. Futhermore, the LD50 of the ethanolic leaf extract was found to be above 5000mg/kg body weight. Irvingia gabonensis (O’Rorke) baill ethanolic leaf extract is a rich source of important phytochemicals and possesses a high antioxidant activity. Also, the administration of the ethanolic leaf extract in wistar rats is safe up to a dose of 5000mg/kg body weight.</p>

AbstractIn this study, the extraction conditions extracted maximize amounts of phenolic and bioactive compounds from the fruit extract of Ficus auriculata by using optimized response surface methodology. The antioxidant capacity was evaluated through the assay of radical scavenging ability on DPPH and ABTS as well as reducing power assays on total phenolic content (TPC). For the extraction purpose, the ultrasonic assisted extraction technique was employed. A second-order polynomial model satisfactorily fitted to the experimental findings concerning antioxidant activity (R2 = 0.968, P < 0.0001) and total phenolic content (R2 = 0.961, P < 0.0001), indicating a significant correlation between the experimental and expected value. The highest DPPH radical scavenging activity was achieved 85.20 ± 0.96% at the optimum extraction parameters of 52.5% ethanol (v/v), 40.0 °C temperature, and 22 min extraction time. Alternatively, the highest yield of total phenolic content was found 31.65 ± 0.94 mg GAE/g DF at the optimum extraction conditions. From the LC–ESI–MS profiling of the optimized extract, 18 bioactive compounds were tentatively identified, which may regulate the antioxidant activity of fruits of F. auriculata.

Antioxidant and free radical scavenging activity of methanol, ethanol, ethyl acetate and chloroform extracts of aerial parts of Lamium purpureum L. was determined by DPPH, ABTS, FRAP and TRP assays. Contents of flavonoids and phenols were also investigated. The total phenolic content in the extracts, determined using Folin?Ciocalteu assay, ranged between 8.57 to 128.00 mg GAE/g d.e. while concentrations of flavonoids in the extracts varied from 24.20 to 39.80 mg QuE/g d.e. The highest phenolic content was found in methanol extract (128.00 mg GAE/g d.e.). The highest content of total flavonoids was identified in the methanol extract (39.80 mg QuE/g d.e.) and the lowest was in the chloroform (24.30 mg QuE/g d.e.). DPPH scavenging of the extracts was determined and obtained IC50 values ranged from 0.12 to 3.12 mg/mL of solution. The values of ABTS radical scavenging activity ranged from 0.35 to 1.80 mg AA/g. The highest ABTS antiradical activity was registered for methanol extract. The FRAP value was found within the range 0.08 to 1.04 ?mol Fe/mg. The best radical scavenger was methanol (1.04 ?mol Fe/mg). In reducing power assay different extracts of L. purpureum showed increasing of activity with increased concentration, and all extracts possessed substantial dose dependent antioxidant activity. The best reducing capacity was obtained with methanol extract of L. purpureum (0.0132 mg AA/mL). The results in this study confirmed that L. purpureum possesses moderate antioxidant properties.

Seaweeds are widely used in the life science as source of compounds with diverse structural forms and biological activities, therefore, potential source of new antioxidant activity. The algae of the Black Sea have not been adequately explored for their potential as a source of bioactive substances. In this context, Brown algae Cystoseira barbata isolated from Black Sea coastal waters of Romania, were evaluated for their bioactivity potential. The results from Cystoseira barbata were obtained: for total phenolic content obtained in ethanolic extracts (TPC): 385.6 � 1.85 mg GAE/100 g f.w. Phenolic Acids quantified by HPLC-DAD from hydroethanolic extract of Cystoseira barbata from Black Sea was 80.14% from total phenolic content identified. We showed that Cystoseira barbata from the Romanian shore of the Black Sea contains vanillic acid in highest quantity (99.5�0.08 mg/100 g f.w.), followed by benzoic acid (65.7�0.06 mg/100 g f.w.) and feluric acid (54.5�0.01 mg/100 g f.w.). The smallest quantities were for gallic acid (3.5�0.03 mg/100 g f.w.), pyrogallol acid (4.2�0.05 mg/100 g f.w.) and 4-amino-benzoic acid (5.2�0.09 mg/100 g f.w.). Cystoseira barbata has a total content of large phenols and also has strong antioxidant and antimicrobial activity. Antioxidant activity (ACL) was studied using DPPH radical scavenging activity and reducing power. The extracts containing high levels of total phenolic content were also potent DPPH radical scavengers. The IC50 (�g/mL) for Cystoseira barbata in methanolic extract was 942.92� 2.56 �g/mL. High absorbance obtained from reducing power assay indicates high reducing power, measured values of absorbance varied from 0.0901 at 0.9223.

Background: Herbal medicine mostly contains wide range of chemical compounds responsible for medicinal therapeutic use. Costus woodsonii is commonly called as Red Button Ginger and synonyms of the botanical name are Costus spiralis, Alpinia spiralis and Costus pisonis. In Malay, it is known as Setawar Halia Merah. In Chinese, it is known as Hong Bi Qiao Jiang. Objective: This research was conducted to study the pharmacognostical, phytochemical, antioxidant and antimicrobial activity of C. woodsonii leaf extracts. Method: Macroscopy, microscopy, phytochemical analysis, thin layer chromatography, antioxidant activity and antimicrobial activity of C. woodsonii leaf were carried out. Total flavonoids were estimated in the leaf extract. The total phenolic content of C. woodsonii leaf was determined using Folin-Ciocalteu reagent. The antioxidant activity of leaf extract of C. woodsonii was determined by performing DPPH radical scavenging. The microbial activity was determined by Well diffusion test, MIC (Minimum Inhibitory Concentration) test and MBC (Minimum Bactericidal Concentration) test. Result and Discussion: C. woodsonii belongs to costaceae with elliptical green leaves. Till now are no extensive studies on C. woodsonii. Preliminary phytochemical analysis revealed the presence of flavonoid, steroid, fat, phenol, tannin and mucilage in leaf extract. Physicochemical studies further revealed the ash value of leaf as 8.7%. Among the three extractions, alcohol extractive values showed the highest as 13%. Loss on drying at 105 degree Celsius in leaf was found to be 12.67%. The plant extract showed total phenolic content of 7.941 mg GAE/g at concentration of 5µg/ml. As for flavonoids content, plant extract showed 21.7 mg RE/g at concentration of 200µg/ml and 43.4 mg RE/g at concentration of 400µg/ml. For antioxidant activity, the plant extract showed weak antioxidant activity in DPPH scavenging activity assay. For antimicrobial test, the leaf extract of C. woodsonii showed weak antimicrobial activity. Conclusion: From this study, it can conclude that C. woodsonii leaf extract possess weak antioxidant activity and weak antimicrobial activity which need to be further validated by using more antioxidant assays and antimicrobial tests.

Exudates (DE) secreted from two shiitake mushroom mycelia (strains 1358 and L5458) were evaluated for their antioxidative properties and phenolic content. 1358DE and L5458DE showed distinct antioxidant activity in different in vitro assays, including scavenging activity on 1,1-diphenyl-2-picrylhydrazyl (DPPH) radicals, hydroxyl radical, superoxide anions and hydrogen peroxide; the ability to chelate ferrous ions; reducing power; hemolysis inhibition activity in rat erythrocyte; and lipid peroxidation inhibition (IC50 values of 1358DE and L5458DE were 3.3 and 132.6; 44.5 and > 1000; 26.9 and 53.7; 153.6 and >175.0; 176.0 and 521.0; 26.7 and 746.4; 47.8 and 736.9; and 3.1 and > 1000 μg/mL, respectively). Their total phenolic content was 237.33 and 24.08 mg gallic acid equivalent (GAE)/g of dry DE, respectively. Overall, these results show that 1358DE generally possesses better antioxidant properties than L5458DE, possibly due to its larger total phenolic content. Shiitake mushroom mycelial exudates, particularly of 1358DE, could be a good source of natural antioxidants.

A raw extract and four extractive fractions were obtained from Cedrus brevifolia (Cyprus cedar) bark. They were all studied regarding the phenolic content and profile using spectrophotometry and HPLC-DAD-ESI-MS. The antioxidant activity was investigated using in vitro assays: DPPH and ABTS radicals scavenging and reducing power assays. The ethyl acetate fraction had the highest total phenolic and proanthocyanidin contents; a taxifolin-O-hexoside, catechin, epicatechin and procyanidin oligomers (three dimers, two trimers) were identified in this fraction. The ethyl acetate fraction was found to possess the highest DPPH and ABTS radicals scavenging effects (EC50=13.9 ± 0.3 and 2.3 ± 0.0 μg/mL, respectively) and reducing capacity (EC50=9.1 ±0.1 μg/mL). Antioxidant effects were highly correlated with total phenolic and proanthocyanidin contents (r=0.89-0.99). These results suggest that Cedrus brevifolia bark is a new source of antioxidants.

This study aims to determine the total phenolic content and antioxidant activity in gayam leaf extract (Incarpus fagiferus Fobs). The research method used is a quasi-experiment that aims to predict the situation to be achieved through actual experiments but no treatment. The sample used is old gayam leaves, with the characteristic of dark green leaf and rough leaf surface. The process of preparing simplicia, ie preparing fresh gayam leaves, dried in an oven temperature 45-50oC, and then dried to produce gayam leaf powder. Samples were extracted with methanol solvent and ethanol for 5 days. The total phenol assay method uses Folin-Ciocalteau method and antioxidant activity test using DPPH free radical retardation method (1,1-diphenyl-2-picrylhydrazyl). The results showed that the total phenolic content of gayam leaf extract with ethanol and methanol solvent was 313,704 GAE (Gallic Acid Equivalent) and 273,913 GAE, respectively. Antioxidant activity as a free antidote to free radical DPPH is known to be valued with IC50 (inhibitory concentration).

Glutinous rice-based foods have a long history are consumed worldwide. They are also in great demand for the pursuit of novel sensory and natural health benefits. In this study, we developed a novel fermented glutinous rice product with the supplementation of Fu brick tea. Using in vitro antioxidant evaluation and phenolic compounds analysis, fermentation with Fu brick tea increased the total phenolic content and enhanced the antioxidant activity of glutinous rice, including scavenging of 1,1-Diphenyl-2-picryl-hydrazyl (DPPH) radical, 2,2′-azino-bis-3-ethylbenzthiazoline-6-sulphonic acid (ABTS) radical, and hydroxyl radical, ferric-reducing antioxidant power, and ferric ion reducing power and iron chelating capability. Besides, compared with traditional fermented glutinous rice, this novel functional food exhibited a stronger activity for protecting DNA against hydroxyl radical-induced oxidation damage. Quantitative analysis by HPLC identified 14 compounds covering catechins and phenolic acids, which were considered to be positively related to the enhanced antioxidant capability. Furthermore, we found that 80% ethanol was a suitable extract solvent compared with water, because of its higher extraction efficiency and stronger functional activities. Our results suggested that this novel fermented glutinous rice could serve as a nutraceutical food/ingredient with special sensory and functional activities.
