Yellow-tailed fish (Caesio cuning) have morphologically similarities with Lutjanidae families, it causes ambiguity on species authentication process. The process of species identification using morphological characteristic does not provide a precise information related to the species.This study was aimed to identify the morphometric and molecular of Yellow-tailed fish (Caesio cuning) which landed on Muara Baru Fish Market, Jakarta using COI gene. A total 30 fishes were observed their nineteen morphometric characters, and 1 fish sample was taken from the fins for DNA extraction, amplification using PCR method, electrphoresis visualization, and sequensing. Sample was analyzed by MEGA 6 software. Based on morphological analysis showed that sampel are Yellow-tailed fish which part of Caesio genus and Caesionidae family. While, genetic analysis using COI gene showed has similarities with database of Genbank NCBI. It can be concluded that identification using morphological character and DNA barcoding methode showed the species belong to Yellow-tailed fish (Caesio cunning).  Ikan Ekor Kuning (Caesio cuning) memiliki kemiripan morfologi dengan anggota famili Lutjanidae lainnya, hal tersebut menyebabkan kesulitan dalam proses autentikasinya.  Proses identifikasi pada suatu spesies menggunakan karakteristik morfologinya belum mampu memberikan informasi yang akurat terkait spesies tersebut. Penelitian ini bertujuan untuk mengidentifikasi ikan ekor kuning (Caesio cuning) yang didaratkan di Pasar Muara Baru, Jakarta melalui kajian karakteristikmorfometrik dan DNA barcoding menggunakan marka gen COI. Total 30 ikan diamati karakter morfometriknya, dan 1 sampel ikan diambil bagian siripnya untuk dilakukan ekstraksi DNA, amplfikasi PCR, elektroforesis, sekuensing dan dianalisis menggunakan aplikasi MEGA 6. Hasil analisis morfologi menunjukkan sampel ikan berasal dari genus Caesio dan termasuk famili Caesionidae. Sedangkan berdasarkan analisis secara molekuler menggunakan marka gen COI, didapatkan hasil bahwa spesies yang diamati (Caesio cuning) memiliki kemiripan dengan database GenBank NCBI. Dapat simpulkan bahwa identifikasi secara morfologi dan DNA menunjukkan bahwa spesies yang di peroleh yaitu Ikan ekor kuning (Caesio cuning).

A morphological identification of fish species is still considered inaccurate, so the molecular anal- ysis is necessary. This study aims to identify commercially important fish species deriving from Muara Baru Modern Fish Market, North Jakarta, employing mor- phological, and molecular analysis. A total of 30 specimens were collected from the field. The morphological analysis utilized a visual identification method, and morphometric measurement, while molecular analysis with DNA barcoding em- ployed the mitochondrial cytochrome oxidase subunit I (COI) gene. The results of the morphological analysis indicated that the fish species were yellowstripe scad (S. leptolepis). It also followed the results of molecular analysis of DNA barcoding that the fish was a S. leptolepis species. Therefore, the combination of morphological and genetic analysis has succeeded in identifying the fish species of S. leptolepis.

DNA barcoding is a modern method for the identification of different species, including insects. Among animals, the major emphasis of DNA barcoding is on insects. Due to this global trend we addressed this approach for surveying a group of insects. The parasitic wasps (including primary and hyperparasitoids) of pome fruit orchard aphids were collected from Iran-Mashhad during 2009-2010. Preliminary identification of this group was performed by using morphological and morphometric characters and SEM. The COI gene in the specimens was amplified and sequenced. In this survey, Aphidius matricariae, Binodoxys angelicae, Diaeretiella rapae, Ephedrus persicae, Lysiphlebus fabarum and Praon volucre parasitoids and Alloxysta sp., Asaphes suspensus, Dendrocerus carpenteri, Pachyneuron aphidis, Syrphophagus aphidivorus hyperparasitoids were studied. Based on intra-interspecies distances and phylogenetic analysis using NJ, all species possess diagnostic barcode sequences. The results of this study show that the COI sequence could be useful in identification study of this group of insects. Here we have provided the first GenBank data for the COI gene of the above-mentioned hyperparasitoids as well as an initial attempt toward preparing DNA barcodes for Iranian parasitoid and hyperparasitoid aphids.

Grunts (family Haemulidae) are among the most commercially important fish in Bangladesh. This paper provides brief diagnostic characters of five previously reported grunt species: Plectorhinchus pictus (Thunberg, 1792); Pomadasys andamanensis McKay et Satapoomin, 1994; Pomadasys argenteus (Forsskål, 1775); Pomadasys argyreus (Valenciennes, 1833); Pomadasys maculatus (Bloch, 1793) and two new distributional records: Pomadasys guoraca (Cuvier, 1829) and Plectorhinchus macrospilus Satapoomin et Randall, 2000. The fishes were collected from the Saint Martin’s Island coral reef-like ecosystem and the adjacent sea of the Sundarbans mangrove forest of Bangladesh. The examined specimens were identified and diagnosed based on their morphometric characters and DNA barcoding COI gene. The new records of Pomadasys guoraca and Plectorhinchus macrospilus from Bangladesh greatly extend their distributional range in the Bay of Bengal. An updated checklist of grunts of Bangladesh is provided.

Species list of economical fish landed in Muara Baru Modern Fish Market, Jakarta needs to be examined. Fish species information needs to know the species of fish being traded. However, many species are difficult to identify morphologically. The aim of this research was to identify fish based on their morphological characters and DNA barcoding. The methods of this research were morphologic, morphometric, and molecular identification with DNA barcoding using PCR. Morphological analysis results showed that fish samples have unique characteristics in the presence of spots on their bodies and fading in the abdomen. Morphometric observations were made with 19 different characters and weight measurements. The 19 fish characters observed have a standard deviation of <1, mean that the fish samples taken have a size that was not much different. A comparison ratio of 18 morphometric characters to total length (PT) showed variable results. Genetic analysis of the fish studied had the Max Score and Total Score was same, 1201, Query Coverage was 95%, and Ident was 100%. Based on morphological analysis and DNA barcoding used by fish species identified as Scatophagus argus species. Both methods were successfully carried out and the two methods complement each other to identify fish species correctly and accurately. Jumlah ikan ekonomis yang banyak diperjualbelikan di Pasar Muara Baru, Jakarta perlu dilakukan penelitian. Informasi spesies ikan perlu diketahui untuk mengetahui jenis ikan yang diperjualbelikan. Penelitian ini bertujuan untuk mengidentifikasi morfologi ikan dengan pendekatan studi morfometrik dan DNA barcoding. Metode yang digunakan dalam penelitian ini yaitu pengamatan morfologi dan pengukuran morfometri serta identifikasi molekuler dengan DNA barcoding menggunakan PCR. Hasil analisis morfologi menunjukkan bahwa sampel ikan memiliki karakteristik yang unik dengan adanya corak totol di seluruh tubuhnya dan memudar di bagian perut. Pengamatan morfometrik dilakukan dengan 19 karakter yang berbeda serta pengukuran berat. 19 karakter ikan yang diamati memiliki standar deviasi yang <1, artinya sampel ikan yang terambil memiliki ukuran yang tidak jauh berbeda. Rasio perbandingan 18 karakter morfometri terhadap panjang total (PT) menunjukkan hasil yang bervariasi. Analisis genetik dari ikan  yang diteliti memiliki Max Score dan Total Score sama yaitu 1201 dengan Query Coverage 95%, dan Ident 100%. Berdasarkan analisis morfologi dan DNA barcoding yang digunakan spesies ikan teridenfikasi spesies Scatophagus argus.  Kedua metode tersebut berhasil dilakukan dan kedua metode tersebut saling melengkapi untuk melakukan identifikasi spesies ikan secara tepat dan akurat.

Fisheries products are some of the most traded commodities world-wide and the potential for fraud is a serious concern. Fish fraud represents a threat to human health and poses serious concerns due to the consumption of toxins, highly allergenic species, contaminates or zoonotic parasites, which may be present in substituted fish. The substitution of more expensive fish by cheaper species, with similar morphological characteristics but different origins, reflects the need for greater transparency and traceability upon which which the security of the entire seafood value-chain depends. Even though EU regulations have made significant progress in consumer information by stringent labelling requirements, fraud is still widespread. Many molecular techniques such as DNA barcoding provide valuable support to enhance the Common Fisheries Policy (CFP) in the protection of consumer interests by unequivocally detecting any kind of fraud. This paper aims to highlight both the engagement of EU fishery policy and the opportunity offered by new biotechnology instruments to mitigate the growing fraud in the globalized fish market and to enforce the food security system to protect consumers’ health. In this paper, after a presentation of EU rules on fish labeling and a general overview on the current state of the global fish market, we discuss the public health implications and the opportunities offered by several techniques based on genetics, reporting a case study to show the efficacy of the DNA barcoding methodology in assessing fish traceability and identification, comparing different species of the Epinephelus genus, Mottled Grouper (Mycteroperca rubra) and Wreckfish (Polyprion americanus), often improperly sold with the commercial name of “grouper”.

Abstract Bats are mammals of great ecological and medical importance, which have associations with different pathogenic microorganisms. DNA barcoding is a tool that can expedite species identification using short DNA sequences. In this study, we assess the DNA barcoding methodology in bats from the Colombian Northern region, specifically in the Córdoba department. Cytochrome oxidase subunit I (COI) gene sequences of nine bat species were typified, and their comparison with other Neotropic samples revealed that this marker is suitable for individual species identification, with ranges of intra-species variation from 0.1 to 0.9%. Bat species clusters are well supported and differentiated, showing average genetic distances ranging from 3% between Artibeus lituratus and Artibeus planirostris, up to 27% between Carollia castanea and Molossus molossus. C. castanea and Glossophaga soricina show geographical structuring in the Neotropic. The findings reported in this study confirm DNA barcoding usefulness for fast species identification of bats in the region.

We adopted DNA barcoding technique using a 658-bp fragment of the mitochondrial cytochrome c oxidase I (COI) gene to identify shrimp species collected from the different areas of Bangladesh. A total of 24 sequences were generated belonging to 14 species including four new records- Macrobrachium nipponense, Macrobrachium kistnense, Exopalaemon carinicauda and Alpheus malleator. Genetic distance measured with Kimura 2 parameter showed that genetic divergence increased with higher taxonomic rank. The mean genetic divergence was evaluated and found to be 0.935%, 22.67% and 30.92% within species, genus and family, respectively. In addition to the barcode-based species identification system, phylogenetic relationships were established where individuals belonging to the same species were grouped under the same clade. Maximum likelihood (ML) was preferred as the statistical method and as expected, the phylogenetic tree complemented and ensured the conventional taxonomy. The present study evidently showed that DNA barcoding can be served as an effective tool to discriminate the shrimp species and this will enhance the understanding on evolution and conservation biology. Red devil has been extensively researched due to extremely high morphometric and chromatophore variations, including the phenomena of polychromatic and trophic polymorphism. The purpose of this study was to identify and observe the phenomenon of sex dimorphism that occurs in polychromatic red devil species which caught in Sermo Reservoir. Fish sample as much as 240 individual was collected from fishermen catch during November 2016-September 2017 by using gill nets, cest nets, and fishing rods. Subsequently, the fish sample was grouped based on color and sex, the namely group of red devil red, red devil black, and red devil reddish black. The morphometric character data of 21 units were measured in each fish sample, whereas the DNA samples were taken from the gill tissue on each species of two individuals. The morphometric character data was analyzed for discriminant distance, while DNA was analyzed by BLAST (Basic Local Allignment Search Tools) from NCBI. The result of the principal component analysis and discriminant function showed the three group of fish can be distinguished by the characters around the head. The T-test of male and female morphometric characters in each fish group showed the phenomenon of sex dimorphism. Canonical and dendrogram diagrams consistently classify the three group of fish as the same species but are segregated by sex so that significantly dimorphism occurs. The mitochondrial sequencing results of the DNA control region show that among the red devil fish group found in the Sermo Reservoir is a species of A. amarillo.AbstrakRed devil telah banyak diteliti karena variasi morfometrik dan kromatofora yang sangat tinggi, termasuk di dalamnya fenomena polikromatisme dan trofik polimorfisme. Penelitian ini bertujuan untuk mengamati polikromatik dan dimor-fisme seksual serta mengidentifikasi spesies red devil di perairan Waduk Sermo Yogyakarta. Contoh ikan sebanyak 240 ekor diambil dari hasil tangkapan nelayan selama bulan November 2016-September 2017 dengan menggunakan jaring insang, jala dan pancing. Selanjutnya contoh ikan dikelompokkan berdasarkan warna dan jenis kelamin, yaitu kelompok red devil merah, red devil hitam, dan red devil merah kehitaman. Data karakter morfometrik sebanyak 21 unit diukur pada setiap contoh ikan, sedangkan contoh DNA diambil dari jaringan insang pada masing-masing kelompok ikan seba-nyak dua ekor. Data karakter morfometrik dianalisis pembedanya, sedangkan DNA dianalisis dengan BLAST (Basic Local Allignment Search Tools) dari NCBI. Hasil analisis komponen utama dan fungsi diskriminan menunjukkan ketiga kelompok ikan dapat dibedakan berdasarkan karakter yang berada di sekitar kepala. Uji t terhadap karakter morfome-trik jantan dan betina pada masing-masing kelompok ikan menunjukkan fenomena dimorfisme seksual. Diagram kanonikal dan dendrogram konsisten mengelompokkan ketiga kelompok ikan tersebut sebagai spesies yang sama, namun terpisah berdasarkan jenis kelamin sehingga sangat nyata terjadi dimorfisme. Hasil sekuensing mitokondria DNA control region menunjukkan seluruh kelompok ikan red devil yang ditemukan di Waduk Sermo merupakan spesies Amphilophus amarillo.

We submitted first results of the DNA studies of the Central Asiatic owlet moths of the genus Euchalcia. Standard cytochrome C oxidase subunit I (COI) gene fragments were sequenced for DNA barcoding of six specimens belonging to Euchalcia herrichi and Euchalcia gyulai. We compared the received sequences between discussed species and with two European Euchalcia species (E. variabilis and E. consona). We found no variability within the COI sequences of the samples collected in the same locality (Alai Mts., Kyrgyzstan), whereas the difference in COI sequences between two populations (Ketmen Mts., Kazakhstan and Alai Mts., Kyrgyzstan) was 0.005.
