Abstract Fungi of the genus Fusarium display inter- and intraspecific variation in their pathogenic capabilities. Although no resistance genes against dry rot have been identified, some indications of the possible defense responses are noticeable, since a single isolate might cause lesions of various sizes when infecting tubers of different potato cultivars. Pathogenicity tests are routinely used to assess potato tubers resistance against dry rot; a setup of pathogenicity test performed at Młochów Research Center is described in this work.

Potato tubers (cultivar ‘Austrian Crescent’) displaying a dry rot decay symptom and internal sclerotia were received for diagnosis. Isolations were attempted from symptomatic material, and Sclerotinia sclerotiorum was frequently recovered. Pathogenicity tests for S. sclerotiorum in potato tubers were completed using agar plugs with S. sclerotiorum placed over tuber wounds and through injecting stolon end with a S. sclerotiorum hyphal suspension. Both inoculation methods induced decay symptoms, although these were observed less frequently with the agar plug inoculation (37.5%) compared with hyphal injection (100%). S. sclerotiorum was consistently reisolated from inoculated symptomatic tubers. This symptom is rare, but potato growers should be aware that tuber infection is possible when white mold is present.

Abstract Fungi of the genus Fusarium cause dry rot, a potato disease which develops during long-term storage of tubers. The disease-inducing capabilities differ among Fusarium spp., but may also vary within species universally considered main dry rot agents. Identification of Fusarium spp. present on diseased tubers in a surveyed area can help minimize crop losses and mycotoxin contamination by, for example, applying proper fungicides or planning crop rotation. Here, we present a procedure of obtaining single spore colonies of Fusarium spp. from potato tubers infected by dry rot, their identification using molecular methods and ways of preservation.

Potato tubers, under storage conditions, suffer from Fusarium dry rot disease caused by Fusarium spp. leading to devastating losses of stored tubers.  Newly harvested tubers are resistance toward Fusarium infection, but disease becomes more vigorous by increasing storage period.  Inoculation of potato tuber slices of Desiree cv. (highly susceptible) or Spunta cv. (highly resistant) either by a weak pathogen (F. moniliforme) or severe pathogen (F. solani) and tracking accumulation of phytoalexin rishitin in inoculated tissues at different intervals from inoculation revealed that Desiree cv. accumulated high amounts of rishitin in a very short time in comparison to another three tested situations. Dipping of potato tuber slices of both cvs. in different concentrations of the known antibiotic chloramphenicol prior to inoculation with a weak pathogen (F. moniliforme) led to increased disease severity associated with accumulation huge amounts of rishitin in inoculated tissues. The correlation coefficient between disease severity and rishitin production was +0.83.  Moreover, the reaction of spunta cv. toward inoculation by a weak pathogen was changed from the state of resistance to very severe susceptibility.  Since chloramphenicol affects protein synthesis on 70S ribosome’s, it was postulated that it prevents synthesis of factor(s) take a part in cell death during infection. Mitochondria extracted from potato tuber tissues secret compound(s) decreased rishitin synthesis in potato tuber slices treated with mycelia extract of F. solani. Treatment of isolated mitochondria with this antibiotic led to the elimination of this factor(s) from their secretions. Results obtained revealed that the elicitation of phytoalexins may consider one of pathogenicity factor of such system.

AbstractFusarium is one of the most important genera of phytopathogenic fungi, causing potato wilt in the field and potato tuber dry rot during storage. The objectives of this study were to identify Fusarium species associated with both potato diseases in different growing regions in Algeria, and to assess their pathogenicity. Among the 152 isolates collected from symptomatic potato plants and tubers in different provinces in Algeria, 13 species of Fusarium and Neocosmospora were identified. Among these three species were isolated only from plants showing symptoms of Fusarium potato wilt (F. oxysporum, F. venenatum, Neocosmospora solani). Two species (F. culmorum, N. tonkinensis) and an isolate of Neocosmospora sp. were found exclusively in tubers with potato dry rot and the remaining ones (F. redolens, F. cf. tricinctum, F. sambucinum, F. cf. incarnatum-equiseti, F. nygamai, F. brachygibbosum and N. falciformis) were associated with both sample types. Fusarium sambucinum was the most frequent species (52.6% of isolates). Fusarium oxysporum and F. nygamai isolates were the most aggressive in the potato wilt pathogenicity test, and F. sambucinum isolates were the most aggressive in the potato tuber pathogenicity test. This is the first study identifying and characterizing potato dry rot and potato wilt pathogens in Algeria.

In recent years, different postharvest alterations have been detected in garlic. In many cases, the symptoms are not well defined, or the etiology is unknown, which further complicates the selection of bulbs during postharvest handling. To characterize the different symptoms of bulb rot caused by fungi, garlic bulb samples were collected from six Spanish provinces in two consecutive years. Eight different fungal species were identified. The most prevalent postharvest disease was Fusarium dry rot (56.1%), which was associated with six Fusarium species. Fusarium proliferatum was detected in more than 85% of symptomatic cloves, followed by F. oxysporum and F. solani. Pathogenicity tests did not show a significant correlation between virulence and mycotoxin production (fumonisins, beauvericin, and moniliformin) or the mycelial growth rate. Penicillium allii was detected in 12.2% of the samples; it was greatly influenced by the harvest season and garlic cultivar, and three different morphotypes were identified. Stemphylium vesicarium and Embellisia allii were pathogenic to wounded cloves. Some of the isolated fungal species produce highly toxic mycotoxins, which may have a negative impact on human health. This work is the first to determine the quantitative importance, pathogenicity, and virulence of the causative agents of postharvest garlic rot in Spain.

Lamb's lettuce or corn salad (Valerianella olitoria) is increasingly grown in Italy and used primarily in the preparation of mixed processed salad. In the fall of 2005, plants of lamb's lettuce, cv Trophy, exhibiting a basal rot were observed in some commercial greenhouses near Bergamo in northern Italy. The crown of diseased plants showed extensive necrosis, progressing to the basal leaves, with plants eventually dying. The first symptoms, consisting of water-soaked zonate lesions on basal leaves, were observed on 30-day-old plants during the month of October when temperatures ranged between 15 and 22°C. Disease was uniformly distributed in the greenhouses, progressed rapidly in circles, and 50% of the plants were affected. Diseased tissue was disinfested for 1 min in 1% NaOCl and plated on potato dextrose agar amended with 100 μg/liter of streptomycin sulfate. A fungus with the morphological characteristics of Rhizoctonia solani was consistently and readily isolated and maintained in pure culture after single-hyphal tipping (3). The five isolates of R. solani, obtained from affected plants successfully anastomosed with tester isolate AG 4, no. RT 31, received from R. Nicoletti of the Istituto Sperimentale per il Tabacco, Scafati, Italy (2). The hyphal diameter at the point of anastomosis was reduced, and cell death of adjacent cells occurred (1). Pairings were also made with AG 1, 2, 3, 5, 7, and 11 with no anastomoses observed between the five isolates and testers. For pathogenicity tests, the inoculum of R. solani (no. Rh. Vale 1) was grown on autoclaved wheat kernels at 25°C for 10 days. Plants of cv. Trophy were grown in 10-liter containers (20 × 50 cm, 15 plants per container) on a steam disinfested substrate (equal volume of peat and sand). Inoculations were made on 20-day-old plants by placing 2 g of infected wheat kernels at each corner of the container with 3 cm as the distance to the nearest plant. Plants inoculated with clean wheat kernels served as controls. Three replicates (containers) were used. Plants were maintained at 25°C in a growth chamber programmed for 12 h of irradiation at a relative humidity of 80%. The first symptoms, consisting of water-soaked lesions on the basal leaves, developed 5 days after inoculation with crown rot and plant kill in 2 weeks. Control plants remained healthy. R. solani was consistently reisolated from infected plants. The pathogenicity test was carried out twice with similar results. This is, to our knowledge, the first report of R. solani on lamb's lettuce in Italy as well as worldwide. The isolates were deposited at the AGROINNOVA fungal collection. AbstractSoft rot is a bacterial disease that causes heavy losses in potato production worldwide. The goal of this study was to identify quantitative trait loci (QTLs) for potato tuber resistance to bacteriumDickeya solaniand for tuber starch content to study the relationship between these traits. A highly resistant diploid hybrid of potato was crossed with a susceptible hybrid to generate the F1 mapping population. Tubers that were wound-inoculated with bacteria were evaluated for disease severity expressed as the mean weight of rotted tubers, and disease incidence measured as the proportion of rotten tubers. Diversity array technology (DArTseq™) was used for genetic map construction and QTLs analysis. The most prominent QTLs for disease severity and incidence were identified in overlapping regions on potato chromosome IV and explained 22.4% and 22.9% of the phenotypic variance, respectively. The second QTL for disease severity was mapped to chromosome II and explained 16.5% of the variance. QTLs for starch content were detected on chromosomes III, V, VI, VII, VIII, IX, XI, and XII in regions different than the QTLs for soft rot resistance. Two strong and reproducible QTLs for resistance toDickeya solanion potato chromosomes IV and II might be useful for further study of candidate genes and marker development in potato breeding programs. The relationship between tuber resistance to bacteria and the starch content in potato tubers was not confirmed by QTL mapping, which makes the selection of genotypes highly resistant to soft rot with a desirable starch content feasible.

In the winter of 2007 in Piedmont (northern Italy), symptoms of a previously unknown disease were observed on beet (Beta vulgaris L. subsp. vulgaris) (garden beet group) grown under a tunnel on several commercial farms near Cuneo. First symptoms appeared on 1-month-old plants, occurring as brown, round-to-oval spots as much as 2 cm in diameter with dark concentric rings near the perimeter. Small, dark pycnidia were present throughout the spots in concentric rings. Generally, older, lower leaves were affected more than the younger ones. Ten to fifteen percent of the plants were affected. Symptoms on the roots began near the crown as small, dark, sunken spots that became soft and water soaked. Eventually, spots on the roots turned dark brown to black and black lines separated diseased and healthy tissues. Older infected tissues were black, dry, shrunken, and spongy. Pycnidia were not observed on affected roots. From infected leaves and roots, a fungus was consistently isolated on potato dextrose agar (PDA) amended with 25 mg/l of streptomycin. The fungus was grown on PDA and maintained at 22°C (12 h of light, 12 h of dark). After 10 days, black pycnidia (130 to 328 [204] μm in diameter) developed, releasing abundant hyaline, elliptical, nonseptate conidia measuring 3.9 to 6.7 (5.1) × 2.4 to 5.9 (3.6) μm. On the basis of its morphological characteristics, the fungus was identified as a Phoma sp. (1). The internal transcribed spacer (ITS) region was amplified using primers ITS4/ITS6 (2) and sequenced. BLASTn analysis of the 557 bp obtained showed an E-value of 0.0 with Phoma betae. The nucleotide sequence has been assigned GenBank Accession No. EU003450. Pathogenicity tests were performed by spraying leaves of healthy 20-day-old potted B. vulgaris plants with a spore and mycelial suspension (1 × 106 spores or mycelial fragments per ml). Noninoculated plants sprayed only with water served as controls. Fifteen plants (three per pot) were used for each treatment. Plants were covered with plastic bags for 5 days after inoculation and kept in a growth chamber at 20°C. Symptoms previously described developed on leaves of all inoculated plants 5 days after inoculation, while control plants remained healthy. Later, pycnidia and conidia, with the same dimensions and characteristics previously described, were observed on the infected leaves. The fungus was consistently reisolated from the lesions of the inoculated plants. The pathogenicity test was carried out twice. P. betae on B. vulgaris var. cycla has been reported in Canada (3) as well as in other countries. The same pathogen was reported in Italy on sugar beet (2). References: (1) G. H. Boerema and G. J. The complex biological phenomenon “resistance” can be reduced to single Mendelian traits acting on both the plant and the pathogen side in a number of pathosystems. According to the “gene-for-gene hypothesis”, the outcome of a plant/pathogen interaction in these cases is incompatibility if a plant carrying a particular resistance gene and a pathogen with the complementary avirulence gene meet. This suggests a causal role of resistance genes in a recognition process initiating active plant defense responses. Fundamentally different strategies are followed to identify these genes molecularly depending on the plant and pathogen species involved. Fungal diseases of crop plants, especially those of cereals, cause dramatic yield losses worldwide. It is assumed that a molecular characterization of plant genes conferring resistance to fungal pathogens will lead to a better understanding of the plant defense system in general permitting the development of new methods of crop plant protection.
