AbstractAn efficient system for Agrobacterium tumefaciensmediated transformation of Jatropha curcas was developed in this study. Several factors affecting the transformation efficiency were optimized, including the explant type, preculture and coculture periods, usage of acetosyringone and density of A. tumefaciens. Compared with other explants, 2-day precultured and cocultured hypocotyl explants showed a significant GUS transient expression efficiency (67.7%). Moreover, adding AS showed a remarkable increase in transformation efficiency. After infecting with A. tumefaciens, hypocotyl explants were subjected to expansion and proliferation on MS medium with 1.0 mg· l-1 IBA and 0.5 mg· l-1 BA. Transformants were demonstrated by the GUS assay and PCR analysis. Rate of hypocotyl explants forming resistant calli reached 67.08%. Furthermore, in the transformed calli development, morphologic changes (calli superficial development and internal structure) were observed by SEM and LM. The present study has provided a fundamental information and research approach for the future study of inducing new traits to J. curcas and genetic modification.

This study illustrates the contribution of plant and faecal microfossil records to interdisciplinary approaches on the identification, composition, taphonomy and seasonality of livestock dung materials. The focus is on the taphonomy of opal phytoliths and calcitic dung spherulites embedded within modern faecal pellets collected from pasture grounds and pens from a range of animals, including cattle, sheep and pigs from three different farms and seasons of the year in Menorca (Balearic Islands, Spain) declared a Biosphere Reserve by UNESCO. Modern reference materials provide comparative plant and dung microfossil indicators on factors affecting the formation, composition, preservation and decay of animal faeces, as well as on the diverse environmental and anthropogenic aspects influencing these. The reported results show relevant changes in phytolith and spherulite composition according to animal species and age, livestock management, seasonality, and grazing and foddering regimes. Both microfossil records provide fundamental information on taphonomic issues that are understudied, such as the variation in the digestibility among different species, including under investigated animals such as pigs, as well on the seasonality of plant and faecal microfossils that are excreted with dung as an important material for reconstructing human-environment interactions which is commonly overlooked in archaeology.

Astragalus membranaceus is an important medicinal plant widely cultivated in East Asia. MicroRNAs (miRNAs) are endogenous regulatory molecules that play essential roles in plant growth, development, and the response to environmental stresses. Cold is one of the key environmental factors affecting the yield and quality of A. membranaceus, and miRNAs may mediate the gene regulation network under cold stress in A. membranaceus. To identify miRNAs and reveal their functions in cold stress response in A. membranaceus, small RNA sequencing was conducted followed by bioinformatics analysis, and quantitative real time PCR (qRT-PCR) analysis was performed to profile the expression of miRNAs under cold stress. A total of 168 conserved miRNAs belonging to 34 families and 14 putative non-conserved miRNAs were identified. Many miRNA targets were predicted and these targets were involved in diversified regulatory and metabolic pathways. By using qRT-PCR, 27 miRNAs were found to be responsive to cold stress, including 4 cold stress-induced and 17 cold-repressed conserved miRNAs, and 6 cold-induced non-conserved miRNAs. These cold-responsive miRNAs probably mediate the response to cold stress by regulating development, hormone signaling, defense, redox homeostasis, and secondary metabolism in A. membranaceus. These cold-corresponsive miRNAs may be used as the candidate genes in further molecular breeding for improving cold tolerance of A. membranaceus.

This study was conducted with the objective of identifying the influence of service quality, university image on agricultural students’ satisfaction and loyalty. The quantitative research approach was applied. Data were collected using questionnaires with a sample size of 313 students who majored in Veterinary Medicine, Animal Husbandry, Agronomy and Plant Protection at Nong Lam University Ho Chi Minh City. The sample has been selected by convenient method. The research results show that agricultural students’ satisfactions and loyalty are affected by service quality and school image. In which, the school image has the stronger impact on student satisfaction and loyalty. From the findings, the researcher has made conclusions and some suggestions to contribute to improving student loyalty.

The genetic transformation via pollen-tube pathway is an alternative method to overcome the constraints imposed by genotype specificity in transformation and regeneration in jatropha (Jatropha curcas L.) tissue culture. Therefore, it is necessary to establish important parameters for efficient genetic transformation of jatropha via pollen-tube pathway. The objective of the research was to study the efficiency of direct transformation of jatropha via pollen-tube pathway based on histochemical and molecular analysis. Solution of purified pCAMBIA1301 DNA plasmid carrying a hptII marker gene and a gus reporter gene with concentration level of 0.05, 0.25, 0.50 µg µl-1 were applied to stigma of flowers at 1, 2, 4, 7, 10 h after pollination. Seedling of IP3A, IP3P and JcUMM18 jatropha’s genotypes derived from 15 combination treatments of plasmid DNA concentration and application time, also wild type was subjected to histochemical and molecular analyses. Based on those analyses, the efficiency of transformation via pollen-tube pathway of three jatropha genotypes ranged from 1.5-16.7%. PCR analysis showed that a number of positive plants were identified by using specific primers hptII and gus, i.e. 1-3 and 3-7 plants of the 15 combined treatments, respectively. AbstractThe purpose of the work is to analyze the risk for surface water intake, taking into account factors affecting the water quality. A three-parameter risk definition was proposed. It was found that the analyzed water intake has an efficient system that protects consumers against drinking water of inadequate quality (multibarier system). The task of the water supply company is to maintain its security measures (including a caution and warning station, biomonitoring) in a state of efficiency.

Purpose This paper explores a type of organizing that can be found in tourist destinations that are administratively bound to a specific geographic area in the intersection of public and private context. The purpose of this paper is to contribute to the understanding of the organizing of activities within destinations and also to contribute theoretically and conceptually to how place dependency and public/private can be understood from an industrial marketing and purchasing (IMP) network perspective. Design/methodology/approach The research approach has its origin in an ongoing multi-disciplinary and longitudinal case study. Findings By applying a network approach to the organizing of destinations, where interaction of relationships, resources, actors and activities play an essential role, a number of propositions have been put forth so as to provide for a better understanding of place-specific organizing, in the intersection between public and private interests. Research limitations/implications The paper is conceptual and more empirical studies are needed to test the findings. One implication to consider in future empirical studies is the tensions between created and organic networks that exist in public and private place partnerships. Practical implications The paper provides insights into factors affecting destination management. Social implications With an emphasis on a socio-political context, the opportunities and limitations that exist between public and private sectors are discussed. Originality/value The paper sheds light on a neglected aspect of a contemporary phenomenon where the IMP network approach could contribute to the understanding of destination marketing or management organization that are bound to a specific place in the intersection between the public and private context. The area of public-private organizing is a topic that may also add new aspects to the IMP community.

Due to the advancement of technology, many businesses moved from physical locations to online businesses where they do business online. The objective of this paper is to investigate the factors affecting online shopping behavior among residents of Lasanod Somalia. This paper used a quantitative research approach. Data were collected from residents of Lasanod Somalia through questionnaires. 160 questioners were collected from respondents who were selected using a simple random sampling technique. The results showed that factors such as trust, firm reputation, functionality, privacy, reliability, and firm value have an influence on consumers’ online buying behavior. The study found out there is a strong positive correlation between trust and firm reputation with an R-value of .612 and a significant level of .000 which is way below 0.01. This research focused on Lasanod city and cannot be generalized to the whole of Somalia. Furthermore, the sample size was not large.

In the present study, an efficient system for the in vitro regeneration of adventitious shoots from the peach rootstock Hansen 536 leaves has been established. Twenty regeneration media containing McCown Woody Plant Medium (WPM) as a basal salt supplemented with different concentrations and combinations of plant growth regulators (PGRs) were tested. Expanded leaves along with their petiole from 3-week-old elongated in vitro shoot cultures were used as starting explants. The highest regeneration rate (up to 53%) was obtained on WPM basal medium enriched with 15.5 μM N6-benzylaminopurine (BAP). The influences on leaf regeneration of the ethylene inhibitor silver thiosulphate (STS) and of different combinations of antibiotics added to the optimized regeneration medium were also investigated. The use of 10 μM STS or carbenicillin (238 μM) combined with cefotaxime (210 μM) significantly increased the average number of regenerating shoots per leaf compared to the control. In vitro shoots were finally elongated, rooted and successfully acclimatized in the greenhouse. The results achieved in this study advances the knowledge on factors affecting leaf organogenesis in Prunus spp., and the regeneration protocol described looks promising for the optimization of new genetic transformation procedures in Hansen 536 and other peach rootstocks and cultivars.
