Genus Claviceps is a plant pathogen able to produce a group of toxins, ergot alkaloids (EAs), whose effects have been known since the Middle Ages (ergotism). Claviceps purpurea is the most important representative specie, known to infect more than 400 monocotyledonous plants including economically important cereal grains (e.g., rye, wheat, triticale). EAs are not regulated as such. Maximum limits are in the pipeline of the EU Commission while at present ergot sclerotia content is set by the Regulation (EC) No. 1881/2006 in unprocessed cereals (0.05% as a maximum). This study aimed to investigate the presence of the six principal EAs (ergometrine, ergosine, ergocornine, α-ergocryptine, ergotamine and ergocristine) and their relative epimers (-inine forms) in rye- and wheat-based products. Of the samples, 85% resulted positive for at least one of the EAs. Wheat bread was the product with the highest number of positivity (56%), followed by wheat flour (26%). Rye and wheat bread samples showed the highest values when the sum of the EAs was considered, and durum wheat bread was the more contaminated sample (1142.6 μg/kg). These results suggest that ongoing monitoring of EAs in food products is critical until maximum limits are set.

Cows were fed ration for 9 wk containing 5, 48, 201, and 822 μg kg−1 ergot alkaloids. The objective was to evaluate the impact of ergot consumption in beef cow–calf operations. Ergot alkaloids up to 822 μg kg−1 did not alter the weight of peripartum and postpartum beef cows (P = 0.93) or nursing calves (P = 0.08), rectal temperature (P = 0.16), or plasma prolactin concentrations (P = 0.30) at moderate ambient temperatures. Ergot did not influence the time (>1 ng mL−1; P = 0.79) or the progesterone concentration (P = 0.38) at the time of first postpartum rise or the size of the first (14 ± 0.6 mm; P = 0.40) and second (13 ± 0.5 mm; P = 0.41) follicles to ovulate. The maximum size of the first postpartum corpus luteum (CL) was 4 mm larger in the 822 μg kg−1 ergot group compared with the control (P = 0.03) for the first ovulation post partum, but not for the second (P = 0.11). There was no effect of ergot exposure on the number of days until the appearance of the first (43 ± 4 d; P = 0.95) or second (52 ± 4 d; P = 0.98) CL post partum. Ergot alkaloid concentrations up to 822 μg kg−1 did not affect pregnancy rates (X2 = 0.36). In conclusion, ergot alkaloid exposure for 9 wk to concentrations as high as 822 μg kg−1 did not alter performance in pregnant and postpartum beef cattle at moderate ambient temperatures.

Isolates of Claviceps purpurea (Fr.) Tul. originally isolated from many different grass hosts in the northern Great Plains and several other areas in the United States and England were tested for their pathogenicity to selected cultivars or lines of male-sterile wheat (Triticum aestivum L.) and barley (Hordeum vulgare L.). While there was a great range in the level of virulence, no clear-cut evidence of specific races was obtained. A few isolates were weakly virulent on two cultivars of male-sterile spring wheat but were highly virulent on the other two cultivars tested. Wheat and barley breeders are advised to use a mixture of isolates in screening germ plasm for resistance to ergot.

O trabalho teve como objetivos, identificar e quantificar os fungos associados a sementes de azevém, comparar a incidência em diferentes meios de cultura, e determinar o número de escleródios de Claviceps purpurea presentes em amostras de sementes. Foram analisadas 37 amostras de sementes de azevém provenientes de municípios do Rio Grande do Sul, Santa Catarina e Paraná. As sementes foram plaqueadas em três meios de cultura: BDA, semi-seletivo de Reis e semi-seletivo de Segalin & Reis, analisando-se a incidência dos fungos. Para detecção de C. purpurea, foram pesados 100g de sementes por amostra e, através de exame visual, foi determinado o número de escleródios. Os fungos detectados foram Alternaria alternata, Bipolaris sorokiniana, Drechslera spp., D. siccans, Fusarium graminearum, Fusarium spp., Aspergillus spp. e Penicillium sp. A incidência de A. alternata variou de 0,0% a 33,7% e freqüência de 89,2% nas amostras analisadas. Para B. sorokiniana a incidência foi de 0,0% a 2,2% e frequência de 62,2%, Drechslera spp., apresentou incidência de 0,0% a 40,3% e frequência de 78,4%. D. siccans a incidência foi de 0,1% a 20,0% e frequência de 100%.Para Fusarium spp., e F. graminearum a incidência foi de 0,0% a 31,0% e 0,0% a 11,3% e frequência de 81,1% e 64,9%, de 0,0% a 43,7% de incidência e 94,6% de frequência para Aspergillus spp. e Penicillium sp. com incidência entre 0,0% a 51,7% e frequência de 91,9%, respectivamente. O fungo C. purpurea foi encontrado em 81,1% das amostras em estudo.

Evidence suggests that sucrose is the main carbon source for growth of Claviceps spp. in the parasitic condition. The sucrose acts as substrate for an active β-fructofuranosidase, produced by the fungus, which in the first instance converts the disaccharide into glucose and an oligofructoside. In this way, 50% of the glucose, supplied as sucrose, is made available to the parasite for assimilation. Subsequent action of the enzyme on both sucrose and the oligofructoside leads to the release of more glucose and the formation of additional oligosaccharides. The structures of the main oligosaccharides formed have been elucidated and the interactions of each compound studied. In experiments with purified enzyme in vitro the interaction of the oligosaccharides is rapid but in culture they are assimilated only slowly; in each case some free fructose is liberated. Free fructose is not assimilated in the presence of glucose and, further, inhibits growth at concentrations which might be expected to occur in the parasitic condition. A dual role has been suggested for the enzyme, with sucrose as substrate, in which glucose is made available to the growing parasite, while at the same time transfer of the fructose to form oligosaccharides prevents it from accumulating at inhibitory concentrations. Ultimately, when glucose becomes limiting, the fungus will adapt to fructose assimilation.
