This ex vivo study evaluated the antibacterial effect of intracanal medications in root canals contaminated with Enterococcus faecalis. Fifty single-rooted human teeth were contaminated with E. faecalis (ATCC 29212) and incubated at 37°C for 21 days. The specimens were randomly divided into 5 groups according to the intracanal medication used: OZ-PG: ozonized propylene glycol; CH/CPMC: calcium hydroxide/camphorated paramonochlorophenol; OZ-PG/CH ozonized PG/CH; PC: positive control group (no medication); and NC: negative control group (no contamination). The samples were collected after 7 days (post-medication) and 14 days (final). Bacterial growth was checked by counting the colony-forming units (CFU). OZ-PG and CH/CPMC reduced significantly the CFU counts compared with PC in the post-medication and final samples, with no statistically significant differences among them. On the other hand, OZ-PG/CH did not reduce significantly the number of bacteria compared with PC. In conclusion, among the evaluated medications OZ-PG and CH/CPMC were the most effective against E. faecalis.

The aim of this study was to evaluate the sealing ability of three different canal filling techniques. Sixty-four roots of extracted human maxillary anterior teeth were prepared using ProTaper® rotary instruments. The specimens were then randomly divided into 3 experimental groups (n=16) and 2 control groups (n=8). The root canals were filled using cold lateral compaction (CLC group), continuous wave condensation technique using the Elements Obturation Unit® (EOU group), and ProTaper obturators (PT group). For the negative control group, 8 roots were filled using lateral compaction as in the CLC group, and the teeth were covered twice with a layer of nail varnish (NCG group). Another 8 roots were filled using lateral compaction, but without sealer, and these were used as the positive control (PCG group). A glucose leakage model was used for quantitative evaluation of microleakage for 24 hours and 1, 2, 3, 4, 5, 6, 7, 8, 9, 10, 11, and 12 weeks. No significant difference in the cumulative amount of leakage was found between the three experimental groups at all observation times. The lateral condensation of cold gutta-percha can guarantee a similar seal of canal fillings as can be achieved by using thermal methods, in the round canals.

Aim. The study aimed at evaluating the incidence of dentinal defects after root canal retreatment with ProTaper Universal retreatment (PTUR) and XP-endo Shaper and Finisher R (XP). Materials and Methods. Sixty extracted single-rooted human premolars were selected and divided into 4 groups of 15 teeth each. In the negative control group, the teeth were left unprepared. In the positive control group, the teeth were prepared with ProTaper Next and obturated with no further retreatment. In the PTUR and XP groups, the teeth were prepared and obturated followed by removal of the filling material at body temperature using PTUR and XP instruments, respectively. The roots were then sectioned at 3, 6, and 9 mm from the apex and observed under a digital microscope to detect defects. Results. PTUR group showed significantly higher ( p value <0.05) incidence of defects than the other groups. Comparison of no defects versus defects between groups in different areas of root canals demonstrated significant difference among the groups in the apical and cervical regions. Conclusion. Within the limitations of the present study, PTUR files created significantly more dentinal defects than XP files, with most of those defects at the cervical and apical areas of the root canals.

The purpose of this study was to compare the effect of a self-etching primer and a non-rinse conditioner with the effect of a conventional adhesive system on the penetration depth in dentin of human teeth, using scanning electronic microscopy (SEM). Fifteen human third molar teeth were sectioned into 2 pieces. All pieces were flattened with grade 600 to 1200 silicon carbide paper and divided into 6 groups: group 1 - Prime & Bond NT (NT), negative control; group 2 - 37% phosphoric acid + Prime & Bond NT (PANT), positive control; group 3 - Non-rinse Conditioner (NRC) + Prime & Bond NT (NRCNT); group 4 - NRC + Prime & Bond 2.1 (NRCPB); group 5 - NRC + Scotchbond MP (NRCSB); group 6 - Prompt L-Pop (PLP). All teeth were covered with Dyract AP. The tooth fragments were decalcified, and its resin replicas were evaluated by SEM by three previously standardized examiners. The penetration was scored from 0 (no penetration) to 3 (maximum penetration). The Kruskal-Wallis and Mann-Whitney U tests (p<0.05) showed three statistically homogeneous groups: {NT, NRCPB, NRCSB and PLP}, {NRCNT} and {PANT}. The authors concluded that the self-etching primer and the non-rinse conditioner provide a lower penetration depth in human tooth dentin than the conventional adhesive system.

Abstract Obtaining the cleanest canal possible before obturation is one of the goals of endodontic treatment. The purpose of this study was to compare the smear layer removal capability of ethylenediamine tetra-acetic acid (EDTA) application with passive ultrasonic and cotton wrapped on reamer activation. Twelve extracted human teeth with single root canals were used for the study. They were conventionally hand instrumented using reamers and K files. The apical aspect of the canals was enlarged to a #40 file. The teeth were divided into 4 four groups, instrumented and irrigated as follows: Group-A EDTA agitated at the end of preparation with cotton wrapped on a reamer for 1 min; Group-B EDTA applied with ultrasonic agitation for 1 min; Group-C irrigated with EDTA+sodium hypochlorite (NaOCl) (negative control group); and Group-D irrigated with distilled water (positive control group). After scanning electron microscopic study at three different levels, smear layer and dentinal tubules were scored. Means were tested for significance using the Z test. When the techniques were compared, the cotton wrapped on reamer agitation method was as successful as the ultrasonic activation of the files. The study aimed to evaluate the antimicrobial effect of photodynamic therapy (PDT) with the use of Toluidine Blue (TB) on extracted teeth infected with biofilms of Enterococcus faecalis. Fifty-four extracted teeth with single-roots and single canals were mechanically shaped, autoclaved, and contaminated with E. faecalis. They were randomly divided into six groups: two groups were negative and positive control groups, two groups were subjected to mechanical instrumentation and PDT with different pre-irradiation times and irradiation times, and two groups were subjected to chemo-mechanical endodontic treatment and PDT with different pre-irradiation times and irradiation times. In PDT groups, after the application of TB, the canals were irradiated with a diode laser of wavelength 635 nm, with a fiber diameter of 200 μm and 100 mW of power in continuous mode. The bacterial load was evaluated using a BioTimer Assay protocol. The greatest reduction of bacterial load was observed in groups of combined PDT with chemo-mechanical treatment. The reductions of bacterial load in groups of combined PDT with chemo-mechanical treatment, and in the positive control group, were significant (p < 0.01) when compared to that of the negative control group. Photodynamic therapy as an adjunctive modality may improve the disinfection capacity of conventional endodontic treatment against E. faecalis.

Despite the excellent properties of mineral trioxide aggregate (MTA), the condensation technique may have some influence in its sealing ability. The purpose of this study was to compare the sealing ability of sonic and ultrasonic setting of MTA. Thirty-four extracted human teeth had their canals prepared and filled with Sealapex sealer and gutta-percha using the active lateral condensation technique. The teeth were rendered waterproof and apicoectomy performed at 3 mm from the apex. Root-end cavities (3.0 mm deep and 1.4 mm diameter) were prepared with diamond ultrasonic tips. The root-end cavities were filled with Pro-Root MTA® with ultrasonic vibration, sonic vibration or no vibration. The positive control group did not receive any material while the negative control group was totally rendered waterproof. After material set, the specimens were immersed in Rodhamine B for 24 h, under vacuum in the first 15 min, then washed, dried and split longitudinally for evaluating the infiltration at the dentin/material interface. Data were analyzed using ANOVA and Tukey's tests at 5% significance level. Sonic vibration promoted the lowest infiltration values (p<0.05). It was concluded that sonic vibration could be considered an efficient aid to improve the sealing ability of MTA when used as root-end filling material.

Aim: To evaluate the effectiveness of different endodontic irrigants against Enterococcus faecalis (ATCC 29212).Methods: Seventy bovine mandibular incisors were prepared, inoculated with a bacterial strain for 60 days and divided into the following groups: positive control; negative control; 2.5% NaOCl; 17% EDTA; 0.2% chitosan; 2.5% NaOCl + 0.2% chitosan; and 2.5% NaOCl + 17% EDTA. The irrigation protocol was performed using an experimental peristaltic pump device, with the irrigating solutions circulating within the apparatus at a constant flow for 10 min. Paper-point samples were then collected from the root canals and immersed in 7 mL of brain heart infusion broth, followed by incubation at 37°C for 48 h. Bacterial growth was assessed by turbidity of the culture medium.Results: E. faecalis was present in all samples after the use of different irrigants.Conclusion: The different irrigants tested were not effective in completely eliminating dentin bacterial contamination with E. faecalis.

BACKGROUND: Enterococcus faecalis (E. faecalis) is the most commonly detected micro-organism in asymptomatic and persistent endodontic infections. Thorough disinfection of the root canal is more important than proper shaping for a successful endodontic treatment. This study is an attempt to implement nanoparticles which have proven anti-bacterial efficacy as irrigating solutions against E. faecalis. AIM: To compare and evaluate the anti-bacterial efficacy of 5% sodium hypochlorite (NaOCl), 0.2% chitosan nanoparticles solution (ChNP) and 0.01% silver nanoparticles solution (AgNP) against E.faecalis with and without diode laser activation. MATERIALS AND METHODS: 70 single rooted mandibular premolars were included in this study. Access opening, working length determination and biomechanical preparation were standardized. Samples were embedded in putty material inside an eppendorf tube to simulate periodontal ligament and autoclaved followed by inoculation of  E.faecalis (MTCC 439) and incubated at 37℃ for 7 days. The samples were divided into 7 groups (10 samples in each group). Group 1- No treatment (Negative control), group 2 - 5% NaOCl without activation (Positive control), group 3 - 5% NaOCl with diode laser activation, group 4-  0.01% AgNP without activation, group 5- 0.01% AgNP with diode laser activation, group 6- 0.2% ChNP without activation and group 7- 0.2% ChNP with diode laser activation. A final rinse of the respective irrigants and activation were performed, followed by counting the colony forming units.  Statistical analysis used were one-way analysis of variance (ANOVA) followed by Tukey’s post-hoc tests. Results showed that diode laser in combination with 5% NaOCl and 0.2% ChNP had significant effects in the reduction of microbial colony counts of E. faecalis in comparison to other experimental groups.  Keywords: Nanoparticles, Irrigation, LASER, Enterococcus faecalis

<p><strong><em>Background:</em></strong><em> Orthodontic tooth movement is a </em><em>interaction prosess</em><em> of resorption and deposition of bone remodeling. Orthodontic tooth movement by mechanical strength causes changes in alveolar bone. Osteocyte is an essential cell to respond bone remodelling. Hyperbaric Oxygen Therapy affects production of osteocyte because it can release Reactive Oxygen Species (ROS) and Nitrid Oxide (NO).  <strong>Purpose: </strong>To determine the difference number  of osteocyte in pressure and tension area during tooth movement by adjuvant of Hyperbaric Oxygen 2,4 ATA during 7 days starting on day 8 to day 14. <strong>Materials and Methods</strong>: This research used Completery Randomized Control Group Post Test Only Design. 36 cavia cobaya (male)  were divided into 3 groups randomly : the negative control groups, positive control group, and treatment group. Preparat staining used Hematoxylin Eosin (HE) and calculated on microscop 1000x with 20 field of view. Data analyses used one way ANOVA and LSD test then compared each area by using paired T test. <strong>Result:</strong> The data showed that the treatment group (P=10,67) tension area has the highest number of osteocyte than  negative control group (K-=3,67), positive control (K+=7,42). <p>Penelitian ini bertujuan untuk melihat pengaruh pemberian ekstrak etanol kulit petai (Parkia speciosa Hassk) terhadap penurunan kadar glukosa darah mencit jantan yang diinduksi aloksan. Hewan percobaan dibagi atas 5 kelompok diantaranya kelompok kontrol negatif, kelompok kontrol positif,dosis I (280 mg/kgBB mencit), dosis II (560 mg/kg BB mencit), dosis III (840 mg/kg BB mencit). Penelitian dilakukan selama 21 hari. Persentase penurunan kadar glukosa darah mencit jantan setelah diberikan ekstrak etanol kulit petai pada hari ke-21 adalah dosis I (77,52 %) lebih besar dibandingkan dengan dosis II (69,5 %) dan dosis III (73,37 %). Data yang diperoleh dianalisis dengan uji Two Way Anova dengan program SPSS 17. Hasil penelitian ini menunjukkan bahwa pemberian ekstrak etanol kulit petai untuk tiga variasi dosis menyatakan perbedaan yang bermakna secara statistik terhadap penurunan kadar glukosa darah mencit jantan.</p><p><em>Petai (Parkia speciosa Hassk) has a compound β-sitosterol and stigmasterol that have efficacy to decreased blood glucose levels. This study aimed to determine the effect of ethanol extract of petai peel for decrease blood glucose levels of male mice induced by alloxan. Experimental animals were divided into 5 groups including negative control group, positive control group, the first dose (280 mg/kg in mice), the second dose (560 mg/kg in mice), the third dose (840 mg/kg in mice). The study was conducted for 21 days. After 21 days, the result found that the percentage of blood glucose levels after the male mice given the ethanol extract of petai peel was, the first dose (77.52%) biger than the second dose (69.5%) and the third dose (73.37%). The data obtained were analyzed by Two Way ANOVA using SPSS 17. The results showed that have signicantly difference between three dose variation of ethanol extract of petai peel in blood glucose levels.</em></p>
