The Alchornea triplinervia specie belongs to the Euphorbiaceae family, one of the main families of the Brazilian flora. In order to contribute to a better understanding of the specie, a counting of chromosome number and the microsporogenesis analysis of A. triplinervia were done. The inflorescences were collected in the municipalities of Paranavaí and Diamante do Norte, State of Paraná, Brazil, and the slides were prepared by squashing technique and staining with 1% acetic carmine. The analysis were performed using an optical microscope and showed a chromosome number for the specie equal to 2n=8x=72. Irregularities in the chromosome segregation process were the main meiotic abnormalities, presenting typical polyploid behavior. Other irregularities were observed; however, at low frequency without compromising the pollen grain formation of the analyzed plants.

Despite the importance of soybeans little cytogenetic work has traditionally been done, due to the small size and apparent similarity of the chromosomes. Fifteen soybean [Glycine max (L.) Merrill] varieties adapted for cultivation in two distinct regions of Brazil were analyzed cytogenetically. A low frequency of meiotic abnormalities was noted in all varieties, although they were not equally affected. Irregular chromosome segregation, chromosome stickiness, cytoplasmic connections between cells, cytomixis and irregular spindles were the main abnormalities observed, none of which had been described previously in soybeans. All of these abnormalities can affect pollen fertility. Pollen fertility was high in most varieties and was correlated with meiotic abnormalities. Although soybean is not a model system for cytological studies, we found that it is possible to conduct cytogenetic studies on this species, though some modifications in the standard methods for meiotic studies were necessary to obtain satisfactory results.

Some African species of Brachiaria have been introduced into the Americas and became the most important forage for pastures in the tropics. New cultivars can be obtained either from direct selections from the natural existing variability in the germplasm collections or from interspecific hybridizations. Polyploidy is predominant in the genus Brachiaria and correlated with apomixis which complicates hybridization. The objective of cytological studies underway on the Brachiaria germplasm collection at Embrapa Beef Cattle is to determine the chromosome number and meiotic behavior of accessions. For the breeding of Brachiaria species, compatible sexual and apomictic accessions need to be identified. Microsporogenesis was evaluated in two accessions of Brachiaria bovonei (BO01 and BO05) and one accession of B. subulifolia (SU01). BO01 is pentaploid (2n = 5x = 45), BO05 is tetraploid (2n = 4x = 36), and SU01 is hexaploid (2n = 6x = 54), derived from x = 9. Meiotic abnormalities typical of polyploids, characterized by precocious chromosome migration to the poles in metaphases, laggard chromosomes in anaphases, and micronuclei in telophases and tetrads, were recorded in high frequency in all the accessions generating unbalanced gametes. Both accessions of B. bovonei presented chromosome stickiness. The results are discussed in the view of the Brachiaria breeding program objectives.

The two accessions of B. dura analyzed (DU01 and DU02) are hexaploid (2n = 6x = 54), derived from x = 9. Meiotic abnormalities, such as precocious chromosome migration to the poles, laggards and micronuclei, were recorded in low frequency in both accessions. The few multivalent chromosome association at diakinesis and meiotic stability suggested that hexaploidy probably resulted from chromosome doubling. In DU02, chromosome transfer (cytomixis) among meiocytes, involving part or the entire genome was observed. The implication of these findings for the Brachiaria breeding is discussed.

To clarify events occurring during pollen grain formation in yellow passion-fruit (Passiflora edulis f. flavicarpa), floral buds were collected at different stages of development. After bracket, petal and sepal removal the anthers were fixed, dehydrated, embedded in paraffin wax, sectioned at 10 mum and after differential dying with safranin O and fast green, mounted in Canada balsam and observed under optical microscope. Formation of the male gamete followed the normal pattern for angiosperms. Observation covered final sporogenic mass phase up to pollen grain formation; microsporangium tissue modifications were also observed. Microsporogenesis was characterized by sporogenic tissue differentiation in microsporic mother cells, followed by meiosis and resulting in tetrads. Microgametogenesis began with callose microspore release, subsequent mitosis, in addition to radial and tangential tapetum wall degradation, parietal layer compression nearer to the tapetum and endothecium widening, terminating in mature pollen grain formation.

Despite of economic importance of the genus Paspalum, little or no cytologic information is available for many species. This is the first report about chromosome number and meiotic behavior for P. maritimum. The three accessions collected in Amapá State (North Region of Brazil) were tetraploid (2n=4x=40) with the chromosomes associating predominantly as bivalents. The low frequency of multivalents suggested that they were segmental allotetraploids. All accessions showed a low rate of meiotic irregularities, and as a consequence the pollen fertility was high. The results suggested that these accessions presented potential for use in a hybridization program.

Tapetum, orbicule, and pollen grain ontogeny inColletia paradoxaandDiscaria americanawere studied with transmission electron microscopy (TEM). The ultrastructural changes observed during the different stages of development in the tapetal cells and related to orbicule and pollen grain formation are described. The proorbicules have the appearance of lipid globule, and their formation is related to the endoplasmic reticulum of rough type (ERr). This is the first report on the presence of orbicules in the family Rhamnaceae. Pollen grains are shed at the bicellular stage.

Cytological and cytogenetic studies were carried out on populations of leafy spurge (Euphorbia esula L. s.l.) and its allies primarily from North America but also from Europe. Chromosome numbers were determined for 126 samples of E. esula, 11 of E. cyparissias L., 1 of the hybrid (E. ×pseudoesula Schur) between these two species, and 1 of E. agraria Bieb. All plants of E. esula were hexaploid. Of the total, 125 leafy spurge accessions had a chromosome number of n = 30; 1 of n = 25++. Very few meiotic abnormalities were observed. Euphorbia cyparissias was primarily tetraploid (n = 20), although occasional diploids (n = 10) were encountered. Tetraploids were fertile; diploids were sterile. The hybrid between the two foregoing species had a chromosome number of n = 25, indicating that the E. cyparissias parent was a tetraploid; meiosis in the hybrid was abnormal. Euphorbia agraria was found to have a gametic number of n = 20, which is the first chromosome number determination for this species; meiosis was normal. Artificial crosses were made successfully between 31 accessions of leafy spurge. Seed germination of the F1 progeny slightly exceeded that reported for natural populations, and meiosis was normal. Pollen stability studies were carried out on herbarium material. Stainability was 100% for most of the samples studied. Extensive pollen size polymorphism was found. It is suggested that this phenomenon supports the hypothesis that E. esula is of allopolyploid origin. No cytological or cytogenetic basis was found for considering the leafy spurge accessions examined in this study as other than as a single, albeit somewhat polymorphic, species.

A basic chromosome number of x = 9 has been confirmed for Cenchrus ciliaris L. Polyploidy is common and levels vary from tetraploid to hexaploid. Aneuploidv is reported for a single specimen, where two chromosomes of a single genome were lost. Various meiotic irregularities were observed. The highest incidence of meiotic abnormalities was observed in the pentaploid specimens. This was attributed to their uneven polyploid level All specimens varied from segmental alloploid to alloploid.

The cytology, development of aposporous embryo sac, pro-embryo and pseudogamy in Paspalum thunbergii Kunth ex Steud. was studied. P. thunbergii was found to be a tetraploid cytotype, with a chromosome number of 40. Meiosis of the pollen mother cells was irregular, pollen viability was low and multiporate pollens were often observed. Megasporogenesis began normally; however, the megaspore deteriorated at the developmental stage of tetrad, while one to five specific nucellar cells became active and began enlarging, and then developed into aposporous embryo sacs. The mature aposporous sacs usually had three nuclei characterised by one egg cell and two polar nuclei. The egg cell developed spontaneously to form pro-embryos prior to anthesis. When several aposporous sacs occurred in the same ovule, usually one sac near the micropyle was involved in pseudogamy, while the other sacs were not involved. Low frequency of twin-embryo seedlings was observed after seeds matured. Examination of three successive generations by paraffin-section and clearing methods revealed that no sexual sac was present. Therefore, the species P. thunbergii is considered to be an obligate apomict that reproduces by apospory.
