Pitangueira (E. uniflora) is considered a diploid species with n = 11 and 2n = 22 chromosomes, in genotypes with the presence of seeds in the fruits. With the pitangueira production of apyrenic fruits existence, such behavior may be related to the ploidy level. The objective of this study was to determine the ploidy level of the pitangueira accessions producing fruit with and without seed, as well as to observe the meiotic behavior and possible chromosomal abnormalities. To check the ploidy level of the pitangueira, freshly expanded mature leaves were collected from the pyrenic pitangueira and from two pyrenic accessions, with suspensions of intact nuclei being prepared. Samples were analyzed in a flow cytometer equipped with multiple parameters data acquisition and UV laser. All analyses were performed using peak-height detection (>6000 fluorescent events, for example, nuclei, were analyzed per sample) and logarithmic amplification. The data were presented as histograms of the number of nuclei along the y-axis and the relative fluorescence intensity on the x-axis. The pyrenean pitangueira and other two pyrenic accessions were characterized as diploid.

Meiotic behavior of ten pteridophyte species from Bangladesh was analyzed in the present investigation. In SMCs of Adiantum capillus-veneris, thirty distinct bivalents were enumerated. In A. caudatum 16 SMCs resulting 64 spores were observed in sporangium which appeared to be diploid sexual. A. lunulatum was found to be triploid apogamous and as diploid sexual forms, thus this species of Adiantum exhibited wide range of morphological variations. Pteris biaurita, P. graffithi and P. vittata were recorded to be n = 29 as diploid. In thelypterids, all the four species were observed to be diploid sexual except C. dentata when it was tetraploid (n = 36, 4x = 144). In C. arida, meiosis was almost regular with 2n = 72 chromosomes. C. cylindrothrix and Ampelopteris prolifera were also found to be diploid sexual, with 2n = 72 chromosomes. J. Bio-Sci. 29(1): 135-142, 2021 (June)

Original meiotic or both meiotic and mitotic chromosome numbers are reported for ten endemic and one non endemic species in nine vascular plant families from Iran. The chromosome numbers of Acantholimon schahrudicum, A. truncatum, Anthochlamys multinervis, Campanula perpusilla, Cousinia calcitrapa var. interrupta, Dorema ammoniacum, Euphorbia gedrosiaca, and Hyocyamus orthocarpus were determined for the first time. The chromosome counts for Astrodaucus persicus and Hedysarum criniferum agree with previous ones. The gametic chromosome numbers for Hedysarum criniferum and Allium stipitatum are reported here for the first time. The occurrence of accessory chromosomes are also reported for Acantholimon schahrudicum and Dorema ammoniacum, being the first records of B chromosomes in the genera Acantholimon and Dorema.

Abstract The genus Schinus (Anacardiaceae) comprises 30 species found in Peru, Bolivia, Chile, Paraguay, Uruguay, Argentina and Brazil. Schinus terebinthifolia (Brazilian pepper tree) is socioeconomically important in the Brazilian state of Espírito Santo. Dried fruit is used as a pepper-like spice and natural populations are exploited. Studies on the meiotic behavior and pollen stainability from an ecological and evolutionary point of view are essential, since they can promote future sustainable management strategies for this species. The objective was to evaluate meiotic behavior and characteristics of pollen grains of individuals from a natural population of S. terebinthifolia localized in the municipality of Vitória, Espírito Santo state, Brazil. Young anthers were squashed in 1% acetocarmine dye to obtain the pollen mother cells. The pollen stainability with 1% acetocarmine, Lugol’s iodine solution and Sudan IV and pollen shape were used to assess the pollen potential viability, cytochemistry and morphology. All individuals had a gametic number of n = 7, a regular meiotic behavior and so the somatic number is 2n = 2x = 14 chromosomes. Median pollen stainability was 94%. Pollen grains are dispersed as monads, 3 - colporate and subspheroidal. The exine has striate-reticulate ornamentation. Pollen grains are starch positive and lipid negative. This is the first report of the occurrence of diploidy for the species. The high pollen stainability is the result of the occurrence of regular meiosis. The presence of starch as pollen reserve can increase the resistance of pollen grains to hostile environments in this species.

(1)Allopolyploids have globally higher fitness than their diploid progenitors however, by comparison, most resynthesized allopolyploids have poor fertility and highly unstable genome. Elucidating the evolutionary processes promoting genome stabilization and fertility is thus essential to comprehend allopolyploid success. (2)Using the Brassica model, we mimicked the speciation process of a nascent allopolyploid species by resynthesizing allotetraploid B. napus and systematically selecting for euploid individuals over eight generations in four independent allopolyploidization events with contrasted genetic backgrounds, cytoplasmic donors and polyploid formation type. We evaluated the evolution of meiotic behavior, fertility and identified rearrangements in S1 to S9 lineages, to explore the positive consequences of euploid selection on B. napus genome stability. (3)Recurrent selection of euploid plants for eight generations drastically reduced the percentage of aneuploid progenies as early as the fourth generation, concomitantly with a quasi disappearance of newly fixed homoeologous rearrangements. The consequences of homoeologous rearrangements on meiotic behavior and seed number strongly depended on the genetic background and cytoplasm donor. (4)The combined use of both self-fertilisation and outcrossing as well as recurrent euploid selection, allowed identification of genomic regions associated with fertility and meiotic behavior, providing complementary evidence to explain B. napus speciation success.

ABSTRACT This study was performed to evaluate the meiotic behavior and fertility of four hybrids obtained from the crossing of Capsicum baccatum and its botanical forms with Capsicum chinense and Capsicum frutescens. We aimed to identify the causes of the low fertility in interspecific hybrids involving species from different gene pools of Capsicum. Hybrid flower buds were fixed in fixative solution and the slides were prepared using 1%-acetic carmine solution. Meiosis was observed in all phases and the main meiotic abnormality observed was the complete or partial lack of chromosome pairing, indicating that the species are either genetically distant or that some asynaptic gene was present in the hybrids. Meiotic index (MI), the frequency of unreduced gametes (type 2n), and pollen viability were estimated. MI and pollen viability of the hybrids were low. We concluded that due to the complete or partial lack of chromosome pairing, the species used in the hybrid combinations are genetically distant. We also noticed that the low-fertility hybrids are a consequence of the lack of chromosome homology between the two genomes involved. The hybrids were considered partially sterile since their pollen viability percentage was lower than 50%.

In this paper, we present new cytogenetic data for three species of the family Pentatomidae: Dichelops melacanthus (Dallas, 1851), Loxa viridis (Palisot de Beauvois, 1805), and Edessa collaris (Dallas, 1851). All studied species presented holocentric chromosomes and inverted meiosis for the sex chromosomes. D. melacanthus has 2n = 12 (10A + XY); L. viridis showed 2n = 14 (12A + XY); and E. collaris showed 2n = 14 (12A + XY). C-banding was performed for the first time in these species and revealed terminal and interstitial heterochromatic regions on the autosomes; DAPI/CMA3 staining showed different fluorescent patterns. In all species, fluorescence in situ hybridization (FISH) with 18S rDNA probe identified signals on one autosomal bivalent, this being the first report of FISH application in the species D. melacanthus and L. viridis. The results obtained add to those already existing in the literature, enabling a better understanding of the meiotic behavior of these insects.
