The objective of this study was to assess the sperm membrane integrity in extended boar semen during storage time using specific spectrum laboratory methods. Boar semen was diluted with the long-term liquid commercial extenders Androhep (A), Androstar (AS), Androstar plus (AS<SUP>+</SUP>), LD and M III and was stored up to 96 h. The sperm membrane integrity was evaluated by motility, viable spermatozoa, short hypoosmotic swelling test (sHOST) and by the activity of the enzyme aspartate aminotransferase (AST). Negative changes in the quality of sperm membrane in relation to storage time were observed after 48 h for sHOST, after 72 h for viable spermatozoa and after 72 h for motility. The percentage of viable spermatozoa was decreased by 0.27% each hour. A statistically significant difference between extenders A and LD was observed in sHOST after 72 h and 96 h storage (<I>P</I> &lt; 0.05). The AST activity did not show any statistically significant differences in extenders and in storage time. In overall assessment Androhep was the best of the tested extenders, followed by AS, AS<SUP>+</SUP>, M III and LD in terms of motility, viable spermatozoa and sHOST. The correlations among laboratory methods were highly significant (<I>P</I> &lt; 0.001). In conclusion, the results documented that the sperm membrane integrity was statistically significantly affected by extenders and storage time (<I>P</I> &lt; 0.001). Boar semen quality was the best in extender A. sHOST is a very sensitive and relatively simple method for the assessment of sperm membrane integrity in diluted semen.

Boar semen is voluminous and ejaculated as jets or fractions of pre-sperm, sperm rich (SRF) and post-sperm rich fractions. Recent studies have reported more resilient characteristics of sperm in initial portions of SRF towards cold shock and cryopreservation. The present study was conducted to assess the quality of specific fractions of SRF, namely, first 10mL of SRF (F1) and rest of SRF (F2) in Large white Yorkshire (LWY) boar semen. Ejaculates were collected using gloved-hand technique and were subjected to quality assessments of volume, pH, sperm progressive motility, concentration, plasma membrane integrity, abnormality, acrosome integrity and sperm membrane cholesterol. Upon statistical analysis, significant differences were noticed in volume, pH, sperm concentration and sperm membrane cholesterol between fractions of the ejaculate.

The experiment was conducted to find out the effect of salt and storage temperature on the physico-chemical properties of beef nugget. For this purpose nugget samples were divided into two parts; one is called fresh nugget and another is preserved nugget at different temperatures. Then the fresh samples as well as the preserved samples were divided into four subdivisions. Then these are treated with different salt levels (0, 1.5, 3 and 5% salt level). The preserved samples were stored at 4oC and -20oC. Samples preserved at 4oC were stored in the refrigerator for 21 days and were analyzed on 7th day, 14th day and 21th day and samples preserved at -20oC were stored in the refrigerator for 60 days and were analyzed on 15th day, 30th day, 45th day and 60th day of preservation. Dry matter and Ash content of all the samples increased significantly (P<0.01) with the advancement of storage time and salt level. CP% of fresh samples was 22.31, 20.55, 20.13 and 20.55 at 0, 1.5, 3 and 5% salt concentration. CP, DM, Ash and Fat also varied among the samples significantly (P<0.01). Highly significant difference is observed in preserved samples than in fresh samples at different salt levels. Fresh nugget treated with 1.5% salt found to be more acceptable in terms of sensory evaluation. Abstract This study was aimed to analyze the metabolic activity and membrane integrity of boar spermatozoa following storage in long-term semen extenders. Boar semen was diluted with AndrohepR EnduraGuardTM (AeG), DILU-Cell (DC), SafeCell PlusTM (SCP) and Vitasem LD (VLD) extenders and stored for 10 days at 17oC. Parameters of the analyzed sperm metabolic activity included total motility (TMOT), progressive motility (PMOT), high mitochondrial membrane potential (MMP) and ATP content, whereas those of the membrane integrity included plasma membrane integrity (PMI) and normal apical ridge (NAR) acrosome. Extender type was a significant (P < 0.05) source of variation in all the analyzed sperm parameters, except for ATP content. Furthermore, the storage time had a significant effect (P < 0.05) on the sperm metabolic activity and membrane integrity during semen storage. In all extenders the metabolic activity and membrane integrity of the stored spermatozoa decreased continuously over time. Among the four analyzed extenders, AeG and SCP showed the best performance in terms of TMOT and PMI on Days 5, 7 and 10 of storage. Marked differences in the proportions of spermatozoa with high MMP were observed between the extenders, particularly on Day 10 of storage. There were not any marked differences in sperm ATP content between the extenders, regardless of the storage time. Furthermore, the percentage of spermatozoa with NAR acrosomes decreased during prolonged storage, being markedly lower in DC-diluted semen compared with semen diluted with either AeG or SCP extender. The results of this study indicated that components of the long-term extenders have different effects on the sperm functionality and prolonged semen longevity by delaying the processes associated with sperm ageing during liquid storage.

The experiment was conducted to find out the effect of salt and storage temperature on the quality of sausage. For this purpose sausage samples were divided into two portions. One is called fresh sausage and another is preserved sausage at different temperature. Then the fresh samples as well as the preserved samples were divided into four subdivisions, treated with different salt levels and e.g. control group-0% and the others are 1.5%, 3% and 5% of salt concentration. The preserved samples were stored at 4°C and -20°C. Samples preserved at 4°C were stored in the refrigerator for 21 days and were analyzed on 7th, 14th and 21th day and on the other hand samples preserved at -20°C were stored in the freezer for 60 days and were analyzed on 15th, 30th, 45th and 60th day. Dry matter and Ash content of all the samples increased with the advancement of storage time and salt concentration level. Dry matter in fresh sample was less compared to preserved samples. Crude protein (CP) percent of fresh samples were 23.13, 22.63, 22.48 and 22.44 at different salt concentration level. The values of CP, DM, Ash & Fat also varied among the samples significantly (P<0.01). Sausage can be preserved for 60 days in different techniques with different changes in the quality. Highly significant difference was observed in preserved samples than in fresh samples at different salt levels. AbstractThe objective of the present study was to determine the seasonal changes in boar semen quality by the assessment of sperm membrane integrity, analysis of chromatin structure, assessment of oxidative stress and of apoptotic changes in spermatozoa. Semen from 16 boars (172 ejaculates) was investigated. The males were aged between 7 months and 7 years. Semen was extended with BTS diluent and stored at +17°C. During seven days of storage, the semen was subjected to standard evaluation and novel methods for semen assessment. In the autumn-winter period, the semen had higher evaluations than in the spring-summer period, but only sperm membrane integrity examination showed significantly lower (P≤0.01) percentage of moribund spermatozoa and the semen had a significantly (P≤0.05) lower (by 0.5%) percentage of sperm with damaged chromatin. Examination performed after 7-day storage showed significantly (P≤0.01) higher percentage of live spermatozoa and with high mitochondrial membrane potential for the autumn-winter period. The level of apoptotic cells was significantly (P≤0.01) lower for the autumn-winter period. Examination of sperm membrane integrity after 7 days of storage showed a lower (P≤0.05) percentage of moribund spermatozoa for the autumn-winter period. In our opinion, novel methods for sperm assessment may be used to monitor new parameters of sperm function.

Seminal cryopreservation allows the long-term conservation of gametes of various species, including endangered species, such as Prochilodus brevis. However, the application of this biotechnology can cause damage to sperm cells, reducing seminal quality. Thus, we have sought substances that minimize the damage caused by this process, such as antioxidants. Thus, this study aimed to evaluate the association between two cryoprotectants and two vitamins, in different concentrations, on the quality of cryopreserved semen of P. brevis. For cryopreservation, the experiment was performed in two stages. In the first stage, the semen of 10 animals was submitted to six different freezing means, coming from the combination of 5% glucose, two cryoprotectants (Dimethyl sulfoxide [DMSO] or Methyl glycol) and two vitamins (C or E to 0.0001 mg) for cryopreservation. In the second stage, semen samples of eight animals were diluted in 5% glucose and the best cryoprotectant found in the first stage, associated with three different concentrations of vitamins C or E (0.01, 0.001, and 0.0001 mg). In both steps, the in natura and post-thawed samples were submitted to kinetic analysis, morphology, and sperm membrane integrity. The cryopreserved semen with DMSO presented significantly higher results (p < 0.05) than that frozen with Methyl glycol, regardless of the vitamin used. The morphologically normal spermatozoa rate was higher (p < 0.05) in the vitamin-containing samples, however, vitamin E reduced sperm motility rates, independent of the cryoprotectant used. As for vitamin concentrations, higher motility rates were obtained when cryopreserved semen with 0.01 and 0.0001 mg of any of the vitamins. However, the higher concentration had a deleterious effect on the spermatic morphology of P. brevis. Therefore, the glucose associated with DMSO and the lower concentration of vitamin C provides good quality for the post-thawed semen of P. brevis.

Introduction After extensive mediastinal dissection fails to achieve adequate intra-abdominal esophageal length, a Collis gastroplasty(CG) is recommended to decrease axial tension and reduce hiatal hernia recurrence. However, concerns exist about staple line leak, and long-term symptoms of heartburn and dysphagia due to the acid-producing neoesophagus which lacks peristaltic activity. This study aimed to assess long-term satisfaction and GERD-related quality of life after robotic fundoplication with CG (wedge fundectomy technique) and to compare outcomes to patients who underwent fundoplication without CG. Outcomes studied included patient satisfaction, resumption of proton pump inhibitors (PPI), length of surgery (LOS), hospital stay, and reintervention. Methods This was a single-center retrospective analysis of patients from January 2017 through December 2018 undergoing elective robotic hiatal hernia repair and fundoplication. 61 patients were contacted for follow-up, of which 20 responded. Of those 20 patients, 7 had a CG performed during surgery while 13 did not. There was no significant difference in size and type of hiatal hernias in the 2 groups. These patients agreed to give their feedback via a GERD health-related quality of life (GERD HRQL) questionnaire. Their medical records were reviewed for LOS, length of hospital stay (LOH), and reintervention needed. Statistical analysis was performed using SPSS v 25. Satisfaction and need for PPIs were compared between the treatment and control groups using the chi-square test of independence. Results Statistical analysis showed that satisfaction with outcome and PPI resumption was not significantly different between both groups ( P > .05). There was a significant difference in the average ranks between the 2 groups for the question on postoperative dysphagia on the follow-up GERD HRQL questionnaire, with the group with CG reporting no dysphagia. There were no significant differences in the average ranks between the 2 groups for the remaining 15 questions ( P > .05). The median LOS was longer in patients who had a CG compared to patients who did not (250 vs. 148 min) ( P = .01). The LOH stay was not significantly different ( P > .05) with a median length of stay of 2 days observed in both groups. There were no leaks in the Collis group and no reoperations, conversions, or blood transfusions needed in either group. Conclusion Collis gastroplasty is a safe option to utilize for short esophagus noted despite extensive mediastinal mobilization and does not adversely affect the LOH stay, need for reoperation, or patient long-term satisfaction.

Abstract Communication and interactions are an integral part of care in long-term care settings. Resident variables, such as race and gender, shape communication and interaction between staff and residents. The Quality of Interactions Schedule (QuIS) was developed to measure the quality of verbal and nonverbal interactions among nursing staff and older adults initially for those in acute care and later used as well in a variety of long term care settings. A quantified measurement of the quality of interactions between residents and staff was created to quantify the QuIS. The purpose of this study was to describe the gender and racial differences in scored quality of interactions. Data for the present study was based on baseline data from the Evidence Integration Triangle for Behavioral and Psychological Symptoms of Dementia (EIT-4-BPSD) implementation study. A total of 535 residents from 55 settings were included in the analyses. An analysis of covariance was conducted to determine a difference in QuIS scores between males and females while controlling for age. The second model tested for differences in QuIS scores between blacks and whites while controlling for age and gender. There was not a statistically significant difference in QuIS scores between male and female residents. There was a significant difference in QuIS scores between those who were black versus white, such that those who were black received more positive interactions from staff than those who were white. Future work should focus on a deeper examination of resident factors and staff factors that may influence these interactions.
