The aim of the study was to evaluate the informative value of various diagnostic methods for spontaneous drainage of the pancreatic pseudocyst into the stomach.Materials and methods: based on the clinical observation of a patient who received complaints of pain in the epigastrium, an episode of melena, after suffering acute pancreatitis several years ago with the development of pancreatic necrosis with an outcome in the cyst of the tail of the pancreas, drainage of the omentum bag. The patient underwent a comprehensive laboratory and instrumental examination.Results: ultrasound, endoscopic examination, MSCT suspected a malignant process in the stomach with ulceration and germination into the pancreas. After performing endoscopic ultrasonography, it was possible to differentiate the layers of the formation and make the correct diagnosis, which was confirmed by subsequent MSCT with oral contrast.Conclusions: a spontaneously opened pancreatic pseudocyst in the lumen of the stomach is a rare and difficult to diagnose disease. An integrated approach, including endoscopic, ultrasound, X-ray, and laboratory methods of investigation when compared with the clinical picture, allows us to verify the formation of the pancreas.

Összefoglaló. A cardialis troponinok biomarkerként való alkalmazása az akut myocardialis infarctus diagnosztikájában nagy múltra tekint vissza, és ez idő alatt jelentősen megváltoztak a cardialis troponinok diagnosztikai jelentőségével kapcsolatos elképzelések. Ez pedig a szívtroponinok és mindenekelőtt az érzékenység mérésére szolgáló laboratóriumi módszerek fejlődésének köszönhető. A legelső laboratóriumi módszerek, amelyeket több mint 30 évvel ezelőtt fejlesztettek ki a cardialis troponinok vérszérumban való kimutatására, még rendkívül alacsony érzékenységűek voltak, ezért csak 12–24 órával az akut myocardialis infarctus jelentkezése után tudták kimutatni a diagnózis felállításában jelentőséggel bíró troponinszintet. Ezenkívül a szívtroponin molekuláinak diagnosztikai reagensei nem specifikus módon kölcsönhatásba léphetnek a vázizombeli troponin molekuláival, ami a vázizmok betegségei és sérülései esetén a cardialis troponin koncentrációjának gyakori hamis pozitív növekedéséhez vezetett. Ahogy a cardialis troponinok mérési módszerei javultak, érzékenységük növekedésével (nagy érzékenységű cardialis troponinok) új lehetőségek nyíltak meg az akut myocardialis infarctus korai diagnózisának felállítására, és többek között modern diagnosztikai algoritmusokat is kifejlesztettek: 0–1 órás, 0–2 órás, 0–3 órás. Ez a korai diagnózis felállítását engedte meg azoknál a betegeknél, akiknek mellkasi fájdalmaik voltak, s ezzel korábban dönthettek a páciens gyógyításának optimális taktikájáról. Miután bevezetésre kerültek a cardialis troponin szintjének nagy érzékenységű mérési módszerei, a cardialis troponin molekuláinak biológiájával kapcsolatos elképzeléseink némileg megváltoztak, különösképpen, hogy innentől kezdve már nem tekinthetők szigorúan intracelluláris molekuláknak, mivel minden egészséges emberben kimutathatók, valamint nemi, életkori és cirkadián tulajdonságokat is felfedeztek a cardialis troponin koncentrációiban, amelyek ugyancsak szerepet játszhatnak az akut myocardialis infarctus diagnózisának felállításában. Arról nem is beszélve, hogy a nagy érzékenységű cardialis troponinok alkalmazási lehetőségei messze túlmutatnak az akut myocardialis infarctus diagnózisán. A jelen cikk egy történeti és egy modern perspektívát kíván bemutatni a cardialis troponinok legfontosabb analitikai jellemzőiről, érintve az azok biológiájára vonatkozó új adatokat és felhasználásuk új diagnosztikai lehetőségeit is. Throughout a very long history of using cardiac troponins in clinical practice as biomarkers of acute myocardial infarction, there have been significant changes in ideas about their diagnostic value. Such a circumstance is caused by the improvement of laboratory methods of cardiac troponin determination and, first of all, sensitivity. Thus, the very first laboratory methods designed to detect cardiac troponins in serum, developed over 30 years ago, had extremely low sensitivity, due to which they could detect diagnostically significant levels of troponins only 12–24 hours after the development of acute myocardial infarction. In addition, diagnostic antibodies directed against cardiac troponin molecules could interact nonspecifically with skeletal troponin molecules, leading to frequent false-positive increases in cardiac troponin concentrations in skeletal muscle disease and injury. As the methods of determining cardiac troponins improved, increasing their sensitivity (high-sensitivity cardiac troponins), the prospects for early diagnosis of acute myocardial infarction opened up, in particular, modern diagnostic algorithms: 0–1 hour, 0–2 hours, 0–3 hours. This allowed earlier diagnosis of patients who came with chest pain, and earlier decision-making on the choice of optimal tactics for the treatment of patients. With the introduction of high-sensitivity cardiac troponin assays, our understanding of the biology of cardiac troponin molecules has changed somewhat, in particular, they are no longer considered strictly intracellular molecules, as they are now detectable in all healthy individuals, and gender, age and circadian patterns in cardiac troponin concentrations have been discovered, which may have an impact on the diagnosis of acute myocardial infarction. Moreover, the possibilities of using highly sensitive cardiac troponins go far beyond the diagnosis of acute myocardial infarction. This article presents a view of the main analytical characteristics from a historical and contemporary perspective, covering some new data on the biology of cardiac troponins and new diagnostic possibilities for their use. The coronavirus disease 2019 (COVID-19) which has become the pandemic par excellence of our time places pressure on various aspects of human endeavor and as such requires detailed study to better combat it. However, diagnostic tests were used to provide data on the incidence of COVID-19 and to assess the immune status of infected individuals. The objective of this chapter is to describe the diagnostic methods currently used to identify SARS-CoV-2 infection. Obtaining the first SARS-CoV-2 genome sequence was decisive for the development of molecular diagnostic assays that currently make it possible to diagnose and screen for the Sars-CoV-2 infection. Their uses depend on the target to be detected. Antigenic tests detect the presence of a virus antigen, which usually makes a proteinaceous part of the virus surface. The serology tests detect the presence of antibodies generated against SARS-CoV-2 and are also a relevant tool for epidemiological studies.

Purpose: The inconsistencies and variations of creatine kinase level due to modifiable and non-modifiable factors were the basis of this study. The aim was to find out the relationships between creatinine and creatine kinase in the blood of somatotypes.    Methods: The 122 males, aged 10 to 20 years, were classified according to their somatotypes. Somatotypes were measured by the ISAK method. By standard laboratory methods, creatinine and creatine kinase estimate. The IBM SPSS version 24 is used for calculation. One way ANOVA followed by post hoc tests was performed to compare the variables among the three groups (p<0.05).   Results: Creatinine level in the blood insignificantly deferred among the three somatotypes. The significant differences (p<0.05) were found in creatine kinase level in the blood and creatinine/creatine kinase ratio among the three dominant Somatotypes. Creatine Kinase was significantly higher in Ectomorphs (212 U/L) than Endomorphs. Ectomorphs and mesomorphs have crossed normal creatine kinase levels (35 -175 U/L). The creatinine/creatine kinase ratio was found highest in endomorphs and lowest in the ectomorphs and significantly differed in three Somatotypes.   Conclusion: Creatinine production remains the same, indicating production of Creatinine is independent of specific body types. A significant higher Creatine Kinase level in Ectomorphs over Endomorphs showed fat content was not associated with it. Significant differences in Creatinine / Creatine Kinase ratio among Somatotypes suggested its relevance between cellular and morphological relationships and might uses as biomarkers.

The aim of the paper is to assess dynamics of water quality in four main recreational areas in Kharkiv and to develop recommendations for water quality improvement. Methods. Field and laboratory methods and analytical data processing. Results. The paper presents analysis of dynamics in water quality for four main recreational areas located in Kharkiv: Zhuravlevskiy Hydropark, Oleksiivskiy Lugopark, Udyanskiy Hydropark and Osnovyanske Lake. Period of study: 2006-2020, time: May-June and August-September. Average data are presented. Samples were analysed at certified Laboratory of Analytical Chemical Research of the Karazin Institute of Environmental Sciences. Laboratory data were compared with national UA standards and European ones. Conclusions were made for the following key parameters: dissolved oxygen and BOD5, total mineralization, biogenic substances content, oil products and surfactants, toxic heavy metals content. There is no specific dynamics on the pollution level. Conclusions. The following recommendations for improvement of water quality were developed: (i) to preform regular monitoring of water quality; (ii) to introduce regular and systematic control by one local authority; (iii) to improve infrastructure of recreational areas; (iv) to introduce regular cleaning of the recreational areas; (v) to install bio-WC; (vi) to install additional waste containers (preferably with separate waste collection); (vii) plant trees for better isolation from highways; (viii) to improve level of water treatment on WWTP; (ix) to create nature protected areas along rivers

Introduction: An atopic status assessment (skin prick test or specific immunoglobulin (sIgE)) in asthmatic children is considered a milestone in identifying potential risk factors and triggers provoking loss of asthma control and asthma exacerbation. Objective: The study aims to perform a comparative analysis of different laboratory methods for a serological assessment of an atopic status in asthma and respiratory allergies in children. Material and methods: A total of 86 children were included, all of whom were diagnosed with bronchial asthma, aged from 5 to 17 years and screened for total IgE level using enzyme-linked immunosorbent assay (ELISA). In 48 randomly selected children, we performed a semi-quantitative serological in vitro assessment of the specific IgE antibodies against food and aeroallergen, using two different laboratory methods—Euroimmun Immunoblot and ImmunoCAP (Phadiatop/fx5). Results: In 70% of the children with a history of allergies, and 65.3% without clinically manifested allergies, multiscreen test ImmunoCAP Phadiatop/fx5 showed positivity and confirmed atopy. Our results showed a significant moderate to strong correlation between multiscreen ImmunoCAP Phadiatop/fx5, and Euroimmun specific IgE titers against aero-allergens—cats, mites, tree mix and food allergens—soy, wheat (р = 0.006), rice, р = 0.090), apple р = 0.007) and peanut. A sensitivity of 63% and specificity of 73.5% was observed for EUROIMMUN Pediatric (food allergens, IgE titer > 1) compared with the gold standard ImmunoCap/fx5. The mean value of total IgE is significantly higher in children with asthma and concomitant with allergic rhinitis compared to those without allergic rhinitis (mean 202.52 U/mL, IQR 102.50 (24.20–363.95) vs. 316.68, IQR 261.00 (109.20–552.50), p = 0.005). Conclusion: Establishing the spectrum of the most common respiratory and food allergens is an essential factor for maintaining asthma control, both through a strategy to avoid allergen exposure and by developing a recommendation plan. The immunoblotting technique is easily applicable in daily clinical and laboratory practice. It is also a cost-effective and reliable alternative to the “gold standard” ImmunoCAP Phadiatop/fx5 in diagnosing atopy in children.

Introduction. Circadian rhythms help the body to anticipate changes in environmental conditions and adapt to the daily fluctuations of external factors. The presence of such mechanisms, which provide early preparation of the organism, gives an advantage in the struggle for survival. The dynamics of light is one of the main factors, which determines the level of growth, development, nutrition, metabolism, affects hormonal activity. Aim. The aim of the study was to study the circadian rhythms of motor activity of Pinchuk's goby Ponticola cephalargoides (Pinchuk, 1976) in the laboratory. Methods. To study the activity of the fish individuals of the Pinchuk's goby caught in the Gulf of Odesa on a fishing rod were selected. The experiments were performed in an aquarium with two lighting options – under natural light and in the complete round-the-clock absence of lighting. Measurements of the fish activity were recorded at 1 hour intervals for 5 days. Results. The study revealed that the activity in constant darkness is generally lower than in the natural light. In both cases, the maximum value of the motor activity occurs in the first half of the day. Under the natural light, the activity of the Pinchuk's goby reaches the maximum value from 06:00 to 07:00, in constant darkness the activity reaches the maximum from 07:00 to 08:00. The difference in the activity of the fish between the two lighting modes is most noticeable from 16:00 to 22:00. Conclusion. It is established that the activity of Pinchuk's goby is cyclical both under the natural light conditions and in the absence of light. From 00:00 to 16:00 Pinchuk's goby has an endogenous circadian rhythm, from 16:00 to 22:30 the rhythm is exogenous.

The analysis of laboratory equipment offered on the Belarusian market for high‑temperature testing of molding materials and mixtures is carried out, the methodology for conducting such tests using devices from leading world manufacturers is described. The results of high‑temperature test tests of mixtures based on furan binders are presented.

Prerequisite to any biological laboratory assay employing living animals is consideration about its necessity, feasibility, ethics and the potential harm caused during an experiment. The imperative of these thoughts has led to the formulation of the 3R-principle, which today is a pivotal scientific standard of animal experimentation worldwide. The rising amount of laboratory investigations utilizing living animals throughout the last decades, either for regulatory concerns or for basic science, demands the development of alternative methods in accordance with 3R to help reduce experiments in mammals. This demand has resulted in investigation of additional vertebrate species displaying favourable biological properties. One prominent species among these is the zebrafish (Danio rerio), as these small laboratory ray-finned fish are well established in science today and feature outstanding biological characteristics. In this review, we highlight the advantages and general prerequisites of zebrafish embryos and larvae before free-feeding stages for toxicological testing, with a particular focus on cardio-, neuro, hepato- and nephrotoxicity. Furthermore, we discuss toxicokinetics, current advances in utilizing zebrafish for organ toxicity testing and highlight how advanced laboratory methods (such as automation, advanced imaging and genetic techniques) can refine future toxicological studies in this species.
