Photodynamic therapy (PDT) has been commonly used in treating many diseases, such as cancer and infectious diseases. We investigated the different effects of PDT on three main pathogenic bacteria of periodontitis — Prevotella melaninogenica (P.m.), Porphyromonas gingivalis (P.g.) and Aggregatibacter actinomycetemcomitans (A.a.). The portable red light-emitting diode (LED) phototherapy device was used to assess the exogenous PDT effects with different light doses and photosensitizer concentrations (Toluidine blue O, TBO). The portable blue LED phototherapy device was used to assess the endogenous PDT effects with the use of endogenous photosensitizers (porphyrin) under different light doses. We found out that both exogenous and endogenous PDT were able to restrict the growth of all the three bacteria significantly. Moreover, the optimal PDT conditions for these bacteria were obtained through this in vitro screening and could guide the clinical PDT on periodontitis.

Comprehensive research has led to significant preclinical outcomes in modified human adipose-derived mesenchymal stem cells (hADSCs). Photobiomodulation (PBM), a technique to enhance the cellular capacity of stem cells, has attracted considerable attention owing to its effectiveness and safety. Here, we suggest a red organic light-emitting diode (OLED)-based PBM strategy to augment the therapeutic efficacy of hADSCs. In vitro assessments revealed that hADSCs basked in red OLED light exhibited enhanced angiogenesis, cell adhesion, and migration compared to naïve hADSCs. We demonstrated that the enhancement of cellular capacity was due to an increased level of intracellular reactive oxygen species. Furthermore, accelerated healing and regulated inflammatory response was observed in mice transplanted with red light-basked hADSCs. Overall, our findings suggest that OLED-based PBM may be an easily accessible and attractive approach for tissue regeneration that can be applied to various clinical stem cell therapies.

Effects of concurrent vs. alternating blue and red light using light-emitting diodes (LEDs) on the photomixotrophic growth of potato plantlets in vitro were investigated. All seven treatments had the same 5.53 mol·m-2 daily light integral (DLI), photoperiod (16-hour day/8-hour night) and similar proportion of red light (45%) and blue light (55%). Results showed that the fresh/dry weight accumulation of potato plantlets in vitro under the concurrent blue and red light was superior than that under the alternating blue and red light, indicating that the simultaneous coexistence of blue and red light are necessary for optimum plantlet growth. Low PPF with long duration was better than high PPF with short duration under same DLI. Within the concurrent blue and red light treatments, when the duration of blue light was shorter than that of red light, timing of the blue light affected the growth of potato plantlets in vitro. Providing blue and red light together at the beginning of the photoperiod resulted in optimal growth, however plantlets illuminated with alternately blue and red light had significantly less fresh/dry weight accumulation.

Abstract Balanced levels of macronutrients and sucrose may ensure the success of micropropagation of the endangered ones. This study aimed to evaluate the effect of levels of salts in the culture medium on in vitro culture of Lomatozona artemisiifolia, as well as to determine the influence of light emitting diode (LED) on the shoot proliferation and rooting of the species. Nodal segments were used to evaluate the different macronutrient concentrations of MS medium (25, 50 and 100%), as well as sucrose concentrations (0.0 mM; 0.34 mM; 0.68 mM and 1.03 mM). Five light conditions were evaluated at shoots proliferation and rooting [100% blue (455 nm); 100% red (630 nm); 30% blue + 70% red; 30% red + 70% blue or fluorescent white]. Low levels of macronutrients in MS (50% and 25%) and sucrose (0.00 mM and 0.34 mM) resulted in plants with higher height, number of shoots and higher production of photosynthetic pigments. The 100% red light promoted rooting of 100% of the plants, and in 100% red or 70% red + 30% blue, higher plants were observed. These results demonstrate that under in vitro conditions, L. artemisiifolia has low nutritional needs, typical of plants that live in rupestrian fields.

The aim of this study is to develop a light-emitting diode- (LED-) based illumination system that can be used as an alternative light source forin vitrophotodynamic therapy (PDT). This illumination system includes a red LED array composed of 70 LEDs centered at 643 nm, an air-cooling unit, and a specific-designed case. The irradiance as a function of the irradiation distance between the LED array and the sample, the homogeneity and stability of irradiation, and the effect of long-time irradiation on culture medium temperature were characterized. Furthermore, the survival rate of the CNE1 cells that sensitized with 5-aminolevulinic acid after PDT treatment was evaluated to demonstrate the efficiency of the new LED-based illumination system. The obtained results show that the LED-based illumination system is a promising light source forin vitroPDT that performed in standard multiwell plate.
