In the present study, morphological and genetic diversity is revealed among the four species of Ocimum. Among the morphological characters plant height, the number of leaves, leaf area, leaf width, leaf length, stem circumference and mitotic index studied and contributed a greater proportion of variations. RAPD markers revealed a high degree of polymorphism (88.89%) among the species of Ocimum. Dendrogram study revealed that the species O. gratissimum and O. basilicum are very closely related whereas O. kilimandscharicum and O. sanctum are distantly related in the phylogenetic history.

The genus Brassica is one of the most important oil seed crops in India with high degree of genetic diversity. In present study, genetic diversity was studied in forty germplasm lines and eight cultivars of Indian mustard using morphological traits and SSR markers. Morphological characters were taken for days to 50% flowering, days to maturity, plant height (cm), length of main raceme (cm), number of primary branches/plant, number of secondary branches/plant, number of silique per plant, number of seeds per silique, 1000 seed weight (g) and seed yield per plant (g). Total 50 SSR markers were used for characterization of these lines, out of which 7 SSR markers were highly polymorphic between all the germplasms of mustard. An UPGMA phonogram was constructed for all 48 Germplasms and the similarity coefficient ranged from 0.00 to 0.91. Number of alleles ranged from 3 to 4, genetic diversity ranged from 71% to 65% with average value of 67%, heterozygosity raged from 20 to 10% with average of 12% and PIC value for markers ranged from 0.65 to 0.59 with mean PIC value 0.61. All seven SSR primers showed PIC value above 0.5 (50%) indicating high genetic diversity in the studied plant material.

Essential oils extracted from basil (Ocimum spp.) by steam distillation are used to flavor foods, oral products, in fragrances, and in traditional medicines. The genus Ocimum contains around 30 species native to the tropics and subtropics, with some species naturalized and/or cultivated in temperate areas. Interand intraspecific hybridization have created significant confusion in the botanical systematics of this genus. Taxonomy of basil (O. basilicum) is also complicated by the existence of numerous varieties, cultivars, and chemotypes within the species that do not differ significantly in morphology. In this study we are using RAPD markers and volatile oil composition to characterize the genetic diversity among the most economically important Ocimum species. We hypothesize that the genetic similarity revealed by molecular markers will more accurately reflect the morphological and chemical differences in Ocimum than essential oil composition per se. Preliminary research using five Ocimum species, four undetermined species, and eight varieties of O. basilicum (a total of 19 accessions) generated 107 polymorphic fragments amplified with 19 primers. RAPDs are able to discriminate between Ocimum species, but show a high degree of similarity between O. basilicum varieties. The genetic distance between nine species and among 55 accessions within the species O. americanum, O. basilicum, O. campechianum, O. × citriodorum, O. gratissimum, O. kilimandscharium, O. minimum, O. selloi, and O. tenuiflorum will be analyzed by matrix of similarity and compared to the volatile oil profile. This research will for the first time apply molecular markers to characterize the genetic diversity of Ocimum associate with volatile oil constituent.

ABSTRACT This study aimed to determine the genetic variability and heritability of morphological characters of some genotypes of potato at the Seed Station in Linelean Village, Modoinding of South Minahasa Regency. The results showed that, the relatively narrow range of genotypes variance for all the characters observed. The coefficient of variance genotypes for plant height, at 14, 42 and 56 DAP (Days After Planting) was relatively narrow, whereas at 7, 21, 28, 35 and 42 DAP relatively wide. For the character of the number of leaves at 7 and 14 DAP are wide and at 21, 28, 35, 42 and 49 DAP relatively narrow. For the character of leaf area at 7 and 14 DAP were wide and at 21, 28, 35, 42 and 49 DAP relatively narrow. For the character of the number of harvest tubers per plant and weight per tuber harvest were wide. Wide sense heritability were high for all the characters observed, except for plant height at 7 and 56 DAP were classified as moderate. Hasil penelitian menunjukkan bahwa ragam genotipe tergolong sempit untuk semua karakter yang diamati. Koefisien ragam genotipe untuk karakter tinggi tanaman, pada 14, 42 dan 56 HST (Hari Setelah Tanam)  tergolong sempit, sedangkan pada 7, 21, 28, 35 dan 42 HST tergolong luas. Untuk karakter luas daun pada 7 dan 14 HST tergolong luas dan pada 21, 28, 35, 42 dan 49 HST tergolong sempit. Untuk karakter jumlah umbi panen per tanaman dan berat per umbi panen tergolong luas. A total of 60 genotypes of Brassica campestris (L) germplasm were acquired from National Research Centre (NARC) Islamabad, Pakistan for the evaluation of genetic diversity using the morphological characters and SDS-PAGE analysis. These genotypes were grown-up in Plant Garden Department of plant sciences, University of Malakand, Khyber Pukhtunkhwa, Pakistan. For the assessment of genetic diversity total of 18 morphological traits were scored and their phylogenetic relationship were plot through cluster diagram. For the estimation of genetic diversity, we used the most influential technique SDS PAGE. We observed six loci (bands) from the collected genotypes; locus-1 (band-1) contained 100% bands of protein and obvious as monomorphic locus while loci i.e. 2,3,4,5 and 6 showed 28%, 42%, 36%, 56% and 25%, respectively variation and were polymorphic. The inter species locus influence toward genetic assortment (LCTGA) was 83.33% in the attained genotypes. Profiling through SDS PAGE based on two-way cluster plottingresolved effectively the collected genotype into 4 cluster. We consider that this work provides a baseline for the understanding genetic diversity in the common crops used in Pakistan and warrants further investigation in this newly evolving field of study.

Molecular markers are the most ideal approach to study genetic diversity. Consequently, we utilized both ISSR and RAPD markers to assess genetic diversity and relationships among three different populations of Farafra, Ossimi and Rahmani Egyptian sheep breeds. Both ISSR and RAPD gave moderate polymorphism 41.3% and 48.51%, respectively. Besides, this value was consistent with the moderate value of the mean of polymorphism information content (0.16 and 0.20, respectively). Farafra-F and Farafra-D populations had the highest similarity which was 0.92 for ISSR and 0.90 for the RAPD marker. Furthermore, ISSR and RAPD constructed dendrogram separated all the studied sheep into two main clusters. All the three populations of Farafra breed combined into one main cluster, while the second cluster contained both Rahmani and Ossimi breeds. The used molecular markers were able to discriminate among evaluated sheep and displayed that Farafra breed more closely related to Ossimi than Rahmani breed.

The continuous destruction of our rainforest has threatened the existence of many useful and multipurpose plant species such as Tetrapleura tetraptera. To revert this situation, efforts must be made towards its sustainable use by encouraging reforestation which entails raising of seedlings for mass propagation of this crop. This study was aimed at comparing the effects of organic (chicken litter) and inorganic (NPK 15:15:15) fertilizers on seedling growth in four accessions of Tetrapleura tetraptera and hence determining fertilizer preference for this forest species. The Complete Randomized Design (CRD) was used in a factorial arrangement with four accessions and three fertilizer types in eight replicates. One month after transplanting, the fertilizers (organic and inorganic) were uniformly applied at the rate of 45 kg/hectare according to the experimental design. Data were collected monthly for a period of three months on plant height, stem diameter, leaf area, leaf length, member of branches, number of leaves, primary and secondary internode length per plant. Data on seedling emergence were also taken. Data collected were subjected to a two-way analysis of variance. Least significant difference (LSD) was used to separate significant means at p = 0.05. Result obtained revealed that the soil was rich in calcium, phosphorus and organic matter but low in nitrogen and potassium which were adequately complemented in mean days to seedling emergence in four accessions studied. Our findings also revealed that the organic (chicken litter) fertilizer did better (P<0.05) in all accessions than inorganic (NPK 15:15:15) fertilizer for all morphological traits studied. The performance of Cross River accession was outstanding as illustrated by the significant interaction obtained in most parameters. With chicken, litter fertilizer, Cross River accession produced the tallest plant (27.38 cm), largest leaf area (1.99 cm2), highest number of branches (20.13), highest number of leaves (66.75), longest stem diameter (0.93 cm), longest leaf length (1.94 cm), lowest primary (0.80) and lowest secondary (0.83) internode length. The use of organic fertilizers is therefore recommended for improved seedling growth of Tetrapleura tetraptera.

Khaya senegalensis (Desr) A. Juss is a perennial deciduous tree which has many uses in developing nations. The population of the tree is rapidly declining due to no conservation efforts to prevent the species from endanger and extinction. At the moment there is dearth information on manure types required to raise healthy seedlings of the tree species in the study area. To conserve this genetic resource, germination and early growth rate experiment was carried out using seeds collected from three locations (eco-zones) (Alkaleri, Bauchi and Toro). Matured seeds of K. senegalensis were collected from mother tree stand in the three locations. One hundred and fifty seeds (150) from each location were sown in perforated polythene pots at an average depth of 2-3 cm. After germination i.e. 30 days after planting, different manures were applied to the seedlings. The experiment was laid out in a Completely Randomized Design (CRD) with three treatments, namely: poultry dung, cow dung and NPK manures; the experiment was replicated four times. Data on seeds germination variables of interest (seedling heights, number of leaves, collar diameter, leaf area, leaf width, leaf length and fresh weights) were measured. Seedlings height were measured using meter rule, collar diameters using a venier caliper, numbers of leaves were determined by manual counting the number of leaves on the seedlings, leaf area was measured by tracing leaves of seedling on a graduated graph sheet. Data collected on seed germination and early growth rate were subjected to descriptive statistics and inferential statistics (ANOVA). Where significant differences occurred, Tukey test was used to separate the means. Based on this finding, germination of seeds commenced after 10 to 14 days. Locations and manure types had significant effects on the mean heights, number of leaves, collar diameter, leaf area, leaf width, length and fresh weights of K. senegalensis seedlings in the study area. Thus, for species conservation and genetic heredity, seeds sourced from Bauchi eco-zone was the best with poultry manure as the best performed manure for the raising of K. senegalensis seedlings in the study area.
