<p>Kualitas biji kakao fermentasi rendah  karena kualitas starter mikroba untuk fermentasi biji kakao rendah. Seleksi starter mikroba diperlukan untuk mendapatkan starter yang unggul. Ada dua jenis starter, yaitu starter cair dan starter kering. Starter cair banyak digunakan untuk fermentasi biji kakao. Starter yang diuji adalah starter cair, yaitu starter Inoka, starter cair BB-Pasca, dan starter <em>yoghurt</em>. Seleksi starter mikroba dilakukan dengan memfermentasi pulp biji kakao selama 24 jam pada berbagai suhu fermentasi (20<sup>o</sup>C, 30<sup>o</sup>C, dan 40<sup>o</sup>C). Parameter yang diamati adalah jumlah total mikroba, laju pertumbuhan mikroba starter, tingkat konsumsi gula pereduksi oleh mikroba starter, total asam yang diproduksi, tingkat penurunan pH, dan peningkatan suhu fermentasi serta korelasi antara parameter pengamatan penelitian. Starter cair unggul yang terpilih adalah starter cair Inoka. Karakteristik starter Inoka adalah memiliki tingkat laju pertumbuhan μ = 0.470, konsumsi gula pereduksi oleh mikroba starter 12%, peningkatan asam total 7%, penurunan pH 5,2, dan peningkatan suhu fermentasi 1,56<sup>o</sup>C  serta korelasi antara parameter penelitian di atas 0,61.</p><p> </p><p><strong>Selection of Superior Liquid Starters for Cocoa Beans Fermentation</strong></p><p>The quality of fermented cocoa beans varies because the microbial starter for fermented cocoa beans varies. The selection of starter is needed to get a superior starter. The starter tested is a liquid starter, that is the Inoka starter, the BB-Pasca liquid starter, the yoghurt. The selection of starter is done by fermenting cocoa bean pulp for 24 hours at various fermentation temperatures (20<sup>o</sup>C, 30<sup>o</sup>C and 40<sup>o</sup>C). The parameters observed were total microbial count, microbial growth rate of starter, consumption of reducing sugars by starter microbes, total acid produced, decrease of pH, and increase of fermentation temperature and the correlation between the parameters of the study. The selected superior liquid starter is the Inoka liquid starter. The characteristics of Inoka starter are to have a growth rate of μ = 0.470, consumption of reducing sugars by starter microbes 12%, total acid increase of 7%, decrease in pH 5.2, and increase in fermentation temperature of 1.56<sup>o</sup>C and correlation between research parameters above 0.61.</p>

The effects of sex and age of castration of males on growth rate, feed efficiency and carcass characteristics in bacon type swine were measured using 16 pigs each as gilts, entire boars, and boars castrated at birth, 6, 12, 16, and 20 weeks of age. Castration at 20 weeks was too late since some pigs reached 200 pounds live weight before that age. Boars consumed less feed daily than barrows and gilts, were thinner in loin fat, and yielded a higher percentage of shoulder than gilts. The carcasses of boars and gilts were longer, had less fat covering over the shoulder and back, a larger area of loin, and a higher iodine number than those castrated late. Acceptability tests showed that sex or age of castration did not affect the flavour, odour, or tenderness of the meat.Purebred breeders can delay castration for better selection of boars without any serious effect on growth, physical carcass characteristics, and feed requirements. Although the market discriminates against boars, it appears questionable whether castration is necessary when boars are slaughtered at 200 pounds before they reach the age of 150 days.

Suppression ofP. bergheiinfections but not those ofN. rodhainiorT. bruceiin mice on milk diet were noted. The suppression ofP. bergheiwas variable. This was not due to differences of the quality of the milk or to the mice eating faeces, sawdust, etc. which may be rich in PABA. The variability was at least partially due to selection of a strain of parasite probably requiring a lower level of PABA than normal. One such strain was isolated and its pattern of infection compared with the normal strain. The ‘milk diet’ strain caused a higher fatality rate in mice on milk diet but always showed a lower parasitaemia growth rate in mice on normal diet. When repassaged through mice on normal diet, the ‘milk diet’ strain did not revert immediately to the infection pattern of the normal strain.

Pinrang Regency is one of the regencies that contributes the largest production in South Sulawesi. The fermentation process is the stage of processing cocoa beans that must be carried out to guarantee the taste and aroma of good chocolate. The purpose of research is to study the effect of fermentation time with different cocoa clones on the quality characteristics of cocoa beans during fermentation. Characterization was held by observing the shape and color of the cocoa beans visually and measuring the temperature, pH, total acid, water content and fat content of the beans during fermentation. The results was showed that cocoa beans from local clones were better than Sulawesi 2 clones based on the quality characteristics of the beans produced. The best beans quality is on the 4 and 5 days of fermentation which produces an optimum temperature of around 45-47°C, pH around 5-5.4, total acid around 0.90%. Cocoa beans with a physical appearance and color that is bright enough, fat content around 50.45 %, and the water content is quite low around 4.97%. The quality of the cocoa beans produced is still in accordance with SNI 2323-2008. The use of styrofoam can be used as an alternative besides fermentation with wooden boxes that are usually done by farmers. The results of the study are expected to be used to obtain data and information on the characteristics of fermented cocoa beans, especially from Pinrang Regency, South Sulawesi.

Microbial activity involved in the cocoa beans fermentation process is essential to maintain and improve the organoleptic and nutritional qualities of chocolate; therefore, the aim of this investigation was to search and select microbial isolates with the potential to improve the quality of cocoa beans. Fermentation experimentswere conducted on farms located in Maceo (Antioquia), San Vicente de Chucurí (Santander), and Rivera and Algeciras (Huila), Colombia. Yeast, lactic acid bacteria (LAB), acetic acid bacteria (AAB), and mesophilic aerobic microorganisms were obtained from different fermentation batches. The growth of these microorganismswas tested in six treatments as follows: 50% cocoa pulp agar (CPA), high concentrations of glucose (10%), ethanol (5%), and acetic acid (7%), an acidic pH of 3.0, and a high temperature of 50oC for 24 h. The isolates with the highest growth were identified by 18S and 16S rRNA gene analysis, revealing a high diversity ofspecies associated with cocoa fermentation, including eight species of yeasts (Debaryomyces hansenii, Meyerozyma guillermondii, Wickerhanomyces anomalus, Pichia guillermondii, Pichia kudriavzevii, Trichosporon asahii, Candida parapsilosis, and Pichia manshurica), six species of LAB (Pediococcus acidilactici, Lactobacillus brevis, Lactobacillus plantarum, Lactobacillus farraginis, Lactobacillus rhamnosus, and Leuconostoc mesenteroides), four species of AAB (Gluconobacter japonicus, Acetobacter tropicalis, Acetobacter pasteurianus, and Acetobacter malorum/tropicalis), and three species of Bacillus spp. (Bacillusaryabhattai /megaterium, Bacillus subtilis, and Bacillus coagulans). In general, microbial populations increased in cocoa batches after 12 h of fermentation and decreased after 84-96 h. All the yeast isolates grew in 10% glucose and CPA, 85.7% in 5% ethanol, and 95% at a pH of 3.0. All the yeast isolates were affectedby 7% acetic acid and incubation at 50oC for 24 h. Eighty-five percent of the LAB grew in 10% glucose, 100% in 5% ethanol, 42.8% in CPA, 64% at a pH of 3.0, and 35.7% grew after being exposed to 50oC for 24 h; all were affected by 7% acetic acid. As for the AAB, 100% grew in 10% glucose, 71% in 7% ethanol, 100% grew in CPA, in 7% acetic acid, and at a pH of 3.0, while 100% were affected by incubation at 50oC. Three yeast isolates, W. anomalus, D. hansenii and M. guillermondii, three LAB isolates, P. acidilactici, L. brevis, and L. plantarum, and three AAB isolates, A. tropicalis, A. pasteurianus and G. japonicus, were selected as promising strains to be used in a microbial starter culture for cocoa bean fermentation to improve the organoleptic quality of cocoa.

The purpose of this research is to investigate the quality-related physical, chemical, and microbiological changes in Thai cocoa beans during fermentation in 2 types of wooden containers. The results will compose a book of guidelines for good Thai cocoa fermentation in order to educate Thai farmers. Fresh Thai cocoa beans have a low pH value (5.0 - 5.5) compared to those from other countries in general (6.0 - 7.0). However, fermented temperature is able to reach 40 - 45 °C in 6 days, which is a main criteria for finishing cocoa fermentation. The color of fresh cocoa beans changes from white to brown within 2 days; after that, cocoa beans are mixed from the top to the bottom of the containers. Three groups of microorganism are evaluated with 3 different sampling points in wooden containers. The results reveal that yeast is grown quickly in 2 days on the top of containers, and then acetic acid bacteria and lactic acid bacteria are grown by the utilized yeast’s metabolites. These behaviors were found in both of the 2 wooden containers; however, a heap of cocoa beans (200 - 250 kgs) in a wooden box showed better quality of cocoa fermentation than a small volume (40 - 50 kgs) in a wooden tray. Cocoa bean is a rich source of polyphenols, which are the largest group secondary metabolite with natural antioxidant property. Polyphenols from cocoabeans was reported to possess health benefits. Fermentation, one step in cocoa processing is needed to improve the quality of cocoa in which the concentration of cocoa bean polyphenols might decrease significantly through oxidation and exudation. Cocoa polyphenols content among different cocoa clones mightalso vary. The aims of this study were to determine total polyphenols, total flavanoid, epicatechin, and catechin content in several cocoa clones, those wereSulawesi 1, Sulawesi 2, ICCRI 03, and KW 617. Until now, characterization of polyphenols from those clones has not been reported. The effect of five daysfermentation to those parameters was also studied. The results of the study showed that fermentation and type of clones significantly affected total of polyphenols, total of flavanoids, epicatechin, and catechin content of the cocoa, there is also an interaction between fermentation and type of clones. Unfermented of Sulawesi 1 had the highest total polyphenols of 96.94±5.83 mg/g, total flavanoids of 90.92±1.89 mg/g, epicatechin of 52.50±0.46 mg/g, and catechin of 1.99±0.02 mg/g content compared to other clones. Among five days fermented cocoa beans, Sulawesi 2 showed the highest total polyphenols and total flavanoids content, while ICCRI 03 had the highest epicatechin and catechin content than other clones. Thus, in can be concluded that although fermentation is required to improve the flavor quality of cocoa, it significantly reduced the content of bioactive compounds. This effect varied amongdifferent cocoa clones.

Most cocoa beans in Indonesia are traditionally produced by farmers using non fermented and sun drying method. The quality of cocoa beans produced by farmer in yogyakarta may be improved by the fermentation method. However, it needs optimization for best fermentation process. The aims of the study was to improve quality of cocoa bans by fermentation of sun dried cocoa beans. The characteric fermented cocoa beans was determined by maesuring changes pH, acidity and fermentation indexs of cocoa beans during fermentation. This study used preconditioned cocoa beans to resamble obtain from the farmer. Preconditioning was done in order to get 15% mouisture content of pulp at same level as moisture content of pulp from traditional process. Before fermentation, therefor sun dried cocoa beans was rehydrated to obtain a moisture content of pulp similiar to fresh beans pulp, and then fermentation was conducted for 120 hours. Changes in acidity and fermentation indexs of cocoa beans during fermentation were measured. The fermentation process used 3 level treatment i.e. control (without inoculum), mixed culture of microbies added at the begining fermentation. The result show all cocoa beans acidity increase during fermentation from 4,48% to 6,45% for control, 4,64% to 6,39% for addition of inoculum at begining of fermentation and from 4,45% to 6,59% for addition of inoculum at the begining and midle of fermentation and fermentation indexs of cocoa beans increase for all level of inoculum addition i.e. 0,31 to 0,88 for control, 0,32 to 0,99 for addition of inoculum at the beginning fermentation and 0,33 to 1,03 for addition of inoculum at the beginning and midle of fermentation. The study indicated that addition of mix culture microbies in fermetation improved the quality of cocoa beans that characterized by pH, acidity and fermentation indexs of cocoa beans.

Abstract MAPPING THE EFFECTS OF COCOA POST HARVEST PROCESSING ON COCOA BEAN QUALITY BY METABOLIC PROFILING METHOD USING GC/MS. The quality of cocoa is strongly effected by the implemented post harvest processes. The fermented cocoa beans is presumed to have higher quality than the unfermented ones, whereas roasting strongly affects the organoleptic quality of the cocoa products. However, what exactly differentiates those cocoa beans are not known yet. This research was foccused on the mapping of post harvest processing method on the quality of cocoa beans, using metabolic profiling method. Different types of cocoa bean samples, that were fermented, roasted, fermented and roasted, as well as unfermented and unroasted (as control) were analysed by GC-MS. The obtained metabolic profiles were further mapped using Principal Component Analysis (PCA). The results showed that there were significant different on samples processed through different processing methods. The fermentation and roasting of cocoa beans direct the process such that more homogenous metabolic composition were obtained. Kata kunci: cocoa beans; fermentation; metabolomic; PCA; processing; roasting  Abstract Proses pengolahan biji kakao sangat menentukan kualitas kakao yang dihasilkan. Biji kakao terfermentasi diyakini memiliki kualitas yang lebih tinggi dibandingkan biji kakao tanpa fermentasi. Proses penyangraian juga diyakini sangat menentukan kualitas organoleptik produk olahan kakao. Akan tetapi, para pelaku bisnis kakao masih belum mengetahui secara pasti apa yang membedakan hal tersebut. Penelitian ini berfokus pada pemetaan pengaruh proses pengolahan pada kualitas biji kakao, menggunakan metode metabolik profiling. Sampel biji kakao dengan berbagai proses pengolahan: difermentasi, disangrai, difermentasi dan disangrai, serta kontrol (tidak difermentasi dan tidak disangrai) dianalisa dengan GC/MS untuk memetakan komposisi metabolitnya. Spektrum metabolit yang diperoleh kemudian dipetakan menggunakan metode Principal Component Analysis (PCA). Hasil yang diperoleh menunjukkan terdapat perbedaan yang signifikan antara sampel dengan berbagai proses pengolahan tersebut, pengolahan biji kakao yang terdiri dari fermentasi dan penyangraian mengakibatkan komposisi biji kakao yang lebih seragam. Keywords: biji kakao; fermentasi; metabolomik; PCA; pemrosesan; penyangraian

The objective of this study was to present empirical evidence about the sustainability of cocoa farming in Indonesia and how decisions are made in producing cocoa beans. This study used a survey method involving a questionnaire for collecting data. The results showed that the sustainability of cocoa farming was determined by weak sustainable ecological factors (46.07%), moderately sustainable socioeconomic factors (54.43%), moderately sustainable technological factors (55.95%), moderately sustainable factors that help farmers in cocoa farming (59.60%), and weak sustainable factors in cocoa farming families' futures (47.52%). To increase the sustainability of cocoa farming, the current study found that farmer education, cocoa crop rejuvenation, cocoa pest and disease control, cleanliness and quality of cocoa beans, cocoa productivity, institutions, extension, technology, credit availability, and stability of cocoa bean prices are the most important factors to be improved.

Cocoa is a plantation commodity that requires fermentation to improve the quality of flavor and aroma. Research on changes in physical and chemical properties during the spontaneous fermentation process of fresh cocoa beans for weight scales of 20, 40 and 60 kg have been carried out. This study aims to obtain information as an indicator of the success rate of the fermentation process of fresh cocoa beans, both on a small and large scale. The fermentation process lasts for 6 days with stirring starting on the second day until the sixth day, with an interval of 24 hours. The study design uses a randomized block design method (RBD) with two factors, namely the weight scale of seeds per box and fermentation time with three replications. The data obtained were analyzed qualitatively and quantitatively. Based on the observed parameter indicators, namely temperature, pH and total acid, there was no significant difference in the profile of the physico-chemical properties during the fermentation process between the three weight scale levels. The physico-chemical condition shows a straight relationship with the quality of fermented cocoa beans, in accordance with the results of the cut test.
