This study was aimed to find out the morphology and carbohydrates content of lingual and sub-lingual glands of duck (Anas plathyrinchos). Sample used in this study was lingual and sublingual glands of a one year old female duck. Anatomy morphology and histology of lingual and sublingual glands were observed after processed into histological preparation and stained with hematoxylin-eosin (HE) then stained further with alcian blue (AB) pH 2.5 and periodic acid Schiff (PAS) to detect carbohydrate contents. The results showed that the lingual gland of duck cannot be observed due to its location below the epithelial layer of the tongue. While the sublingual gland of duck has an elongated shape and not granulated like typically glands, located at ventral tongue (dorsoventral lingual) between os mandibulae and covered with musculus digastricus venter oral. Staining with AB pH 2.5 and PAS showed that cytoplasm of secretory cell and secrete of lingual and sublingual glands contain acidic and neutral carbohydrates which indicated a positive reaction with intensity ranging from weak (+) to strong (+++). This study concluded that the lingual and sublingual gland found in female duck is a complex tubular gland with mucous secretory cells type. ____________________________________________________________________________________________________________________

The vomeronasal organ (VNO) detects specific chemicals such as pheromones and kairomones. Hedgehogs (Eulipotyphla: Erinaceidae) have a well-developed accessory olfactory bulb that receives projections from the VNO, but little is known about the hedgehog VNO. Here, we studied the histological features of the VNO in five individual African pygmy hedgehogs by hematoxylin-eosin, periodic acid-Schiff, and Alcian blue stains. The hedgehog VNO comprises a hyaline cartilage capsule, soft tissue and epithelial lumen, and it branches from the site just before the incisive duct opening into the nasal cavity. The soft tissues contain several small mucous (or mucoserous) glands and a large serous gland, and many venous sinuses all around the lumen. The VNO lumen is round to oval throughout the hedgehog VNO, and the sensory epithelium lines almost the entire rostral part and medial wall of the middle part. These findings indicate that the VNO is functional and plays an important role in the hedgehog. Notably, the VNO apparently has a characteristic flushing mechanism with serous secretions like those of gustatory glands, which the hedgehog might frequently use to recognize the external environment.

Abstract Periapical actinomycosis caused by a gram-positive anaerobic pathogen characterizes a typical extra-radicular infection. This study determined the frequency and correlated the content of bacteria colonies with the of periapical actinomycosis size. The study comprised a total of 218 periapical lesions (PL) (cysts, granulomas or abscess). The specimens embedded in paraffin were sliced into 4-µm sections and stained with hematoxylin-eosin, Gram, Periodic Acid-Schiff (PAS) and Grocott’s stain. The presence of bacterial colonies composed of filamentous structures labeled with the histochemical stains were described as Actinomyces, and for each case, the bacterial colonies were counted and measured. The correlation between the number and size of bacterial colonies and the size of PL was tested using Pearson’s adjusted correlation coefficient. From 218 PL, bacterial colonies were identified in 64 biopsies. Seven cases (0.3%) fulfill the criteria for diagnosis of periapical actinomycosis. All of cases were therapy-resistant and did not showed periapical repair after 12 months of follow-up. Periapical surgery or dental extraction was performed. The correlation test indicated no correlation between the number of bacterial colonies and the lesion size (p=0.752, r=-0.148). However, a larger bacterial colony size generally resulted in a larger periapical lesion (P=0.000, r=0.657). The frequency of periapical actinomycosis was low, and this lesion should be included in the differential diagnosis of PL. The size of the Actinomyces colonies seemed to contribute to increase the size of the periapical lesion.

Holocrine secretory cells have been identified in the epithelium of the epididymal canal of the mouse. These cells develop from basal cells. During their differentiation they grow toward the lumen of the epididymal canal and come to form club-shaped structures with an expanded apical portion, a central nucleus and a thin stalk-like connection to the basement membrane. Mature holocrine cells are characterized by their high acid phosphatase and aliesterase activity. They also are highly active for succinic dehydrogenase, nicotinamide adenine dinucleotide diaphorase, and nicotinamide adenine dinucleotide phosphate diaphorase. Nucleoside diphosphatase, thiamine pyrophosphatase, adenosine triphosphatase, and alkaline nucleoside phosphatase are also found in these cells. These cells are also characterized by their reactivity with the Aoyama and periodic acid-Schiff reactions. They react moderately with the molybdate and Luxol Fast Blue MBS reactions for choline containing compounds. Mature holocrine cells may disintegrate in situ or may be discharged in toto into the lumen of the epididymal canal. Glycerylphosphorylcholine was identified in extracts prepared from sperm-free epididymides of mice. Glycerylphosphorylcholine reacts with Aoyama and periodic acid-Schiff reactions as do mature holocrine cells. This fact coupled with the identification of choline containing material in holocrine cells suggests that they may be one site for the formation of glycerylphosphorylcholine.

PURPOSE: The purpose of this study was to identify the possible alteration in the thickness of the epithelium basal membrane of the vocal cords and correlate it with the cause of death. METHOD: Larynxes collected from adult autopsies during the period of 1993 to 2001 were utilized. We used the hematoxylin-eosin and periodic acid-Schiff staining methods for the morphological and morphometric analysis. RESULTS: Sixty-six vocal cords were analysed; increased thickness was identified in 14 cases (21.2%), with equal proportions between the genders. Increased vocal-cord thickness was more frequent in patients of the white ethnicity (12 cases, 85.7%). Respiratory alterations were found in 10 (71.4%) of the cases with increased vocal-cord thickness. Of the patients that were maintained with mechanical ventilation before death, 7 (18.4%) had thickening of the basal membrane. Among the smokers, 9 (19.63%) had basal membrane thickening. CONCLUSION: No statistically significant differences were found between the cases in which the cause of death was related to respiratory diseases as compared to non-respiratory diseases and the thickening of the basal membrane of the vocal cords. However, new studies are needed in order to verify the etiopathogenesis of this thickening.

AIMS: Rapidly progressive glomerulonephritis (RPGN) presents with rapidly deteriorating renal function (> 50% loss of glomerular ltration rate /GFR within 3 months) associated with nephritic urinary sediments and crescents in biopsy. Crescentric IgA Nephropathy, Anti-GBM (Glomerular basement membrane) disease and combined IgA Nephropathy with Anti-GBM disease are three uncommon reasons of RPGN. We have compared clinicopathological. Immunouorescence (DIF) and renal outcome of three groups. Setting and designs: Prospective, cross-sectional, single centre study. METHODS AND MATERIALS :Ultrasonography guided core biopsies obtained, one stained with hematoxylin-eosin, periodic acid-Schiff, Masson's trichrome, and silver methenamine stain another one with immunouorescence conjugated IgG, IgM, IgA, C3, C1q, kappa and lambda stain. Demographic, clinicopathological and therapeutic parameters with survival data were collected. STATISTICAL ANALYSIS USED: Done using software (GraphPad PRISM 6). RESULTS: We have included 9 cases of crescentric IgA Nephropathies, 6 cases of AntiGBM diseases and 2 cases of combined IgA Nephropathy and AntiGBM diseases. Signicant difference seen in the incidences of hypertension, hemoptysis, serum creatinine, anti GBM antibody, total number of crescents and mesangial hypercellularity, fragmentation of GBM etc. IgG, IgA and kappa positivity in DIF show signicant difference. Survival analysis and mortality versus dialysis dependence and complete and partial remission versus no remission showed no difference between these three groups. CONCLUSIONS: Proper and early clinicopathological diagnosis is important since all are of poor renal outcome. Further renal outcome of the combined disease is same as that of individual ones.

We hypothesized that foreign bodies in airways cause inflammation leading to goblet cell metaplasia. Instilled agarose plugs lodged in the bronchi of pathogen-free rats caused a time-dependent increase in Alcian blue-periodic acid-Schiff staining that was detected within 24 h and markedly increased at 72 h. Control bronchi contained no pregoblet or goblet cells, but plugged bronchi contained many pregoblet and goblet cells and a decrease in nongranulated secretory cells. In situ hybridization showed no expression of MUC5AC in control airways, but plugged airways showed a marked expression. Control bronchi showed sparse staining for epidermal growth factor receptor (EGFR) protein, but plugged bronchi showed intense EGFR staining in the epithelium. Pretreatment with an EGFR tyrosine kinase inhibitor (BIBX1522) prevented Alcian blue-periodic acid-Schiff staining and MUC5AC gene expression in plugged bronchi. Pretreatment with tumor necrosis factor-α neutralizing antibody or pretreatment with cyclophosphamide abolished plug-induced EGFR protein expression and goblet cell metaplasia. Thus instillation of agarose plugs induces profound goblet cell metaplasia by causing EGFR expression and activation.

The female prostate is a reproductive gland that typically presents a morphology similar to that of the male gland and is highly developed in female Mongolian gerbils. Two main cell populations compose the epithelium gland: basal and secretory luminal cells. However, during postnatal development, diverse secretory cell phenotypes are distributed among the typical ones. Prostate homeostasis is under the control of sexual hormones, such as oestrogen and progesterone. After hormonal deprivation the female gland undergoes several morphophysiological changes. The objective of this study was to identify and characterise, structurally and ultrastructurally, the cellular heterogeneity of the female prostate epithelium in normal conditions and after ovariectomy. Histological routine stains, such as haematoxylin–eosin, periodic acid–Schiff and silver impregnation, as well as immunocytochemical techniques were used to enable identification of the different cell types. Some secretory cells types were identified and characterised as mucinous, basophil, clear, ciliated, droplet, spumous and neuroendocrine cells. Population tally data showed that the hormonal suppression caused by ovariectomy resulted in a decrease in the proportions of basophil and clear cells and an increase in spumous cells. Thus, the secretory epithelial cells of the female gerbil prostate are not morphologically and functionally uniform, presenting a phenotypical plasticity according to the hormonal environment in which they operate.

A convenient method is described for the removal of Epon 812 from thin sections, utilizing a saturated solutions of sodium hydroxide. The tissue architecture and ultrastructural details are preserved. Hematoxylin-eosin, periodic acid-Schiff and phosphotungstic-hematoxylin staining modifications are suggested which result in differentiation similar to that seen in paraffin sections. The technique is applicable to ultrathin sections suitable for examination with the electron microscope, allowing comparison of staining characteristics and fine structure of adjacent thin sections.

This study investigated the protective effect of vitamin D against carbon tetrachloride (CCl4)-induced nephrotoxicity in rats. Adult male Wistar albino rats were divided into four groups ((A) control; (B) 10-week exposure to CCl4; (C) 10-week exposure to CCl4 + vitamin D treatment; and (D) 10-week exposure to CCl4 + 12 weeks of vitamin D treatment). The CCl4 dose (1.5 ml kg−1) was injected subcutaneously twice a week, while the 0.5 mg kg−1 dose of vitamin D was administered intraperitoneally every day, as appropriate for each group. Whole animal and kidney weights as well as serum urea, creatinine, and glucose levels were measured. Kidney tissue sections were stained with hematoxylin–eosin, Masson’s trichrome, and periodic acid–Schiff. Tubular and glomerular degeneration were detected in the kidney tissues of CCl4-treated rats, together with dilatation and vacuolization within the tubules and hemorrhage in the intertubular region. In the kidney glomeruli; congestion, atrophy, and adhesion to parietal layer were observed. Tissue disorganization and aggregation of Bowman’s capsules were noted. Mononuclear cell infiltration was observed between the glomeruli and the tubules. In contrast, the kidney sections and functional parameters of vitamin D-treated rats were similar to the controls, suggesting that vitamin D treatment is able to reduce renal damage.
