Phytochemical, Heavy Metals and Antimicrobial Study of the Leaves of Calopogonium Mucunoides

Abstract Background: In this study, the phytochemical, heavy metals, and antimicrobial characteristics of the leaves of Calopogonium mucunoides were investigated in other to determine its therapeutic potentials. The phytochemicals present in the leaves were extracted with n-hexane, methanol and ethyl acetate, and the extracts were used to investigate for the phytochemical constituents and antimicrobial activity. The methanol extract was used to test for the concentration of five heavy metals.Results: The result of the phytochemical screening confirmed the presence of alkaloids, anthocyanins, flavonoids, glycosides, phenols, reducing sugars, saponins, steroids, tannins, and terpenoids in various quantities. The heavy metals analysis result revealed the presence of lead (0.08 mg/kg) and iron (0.08 mg/kg) well below the acceptable limits set by the World Health Organization for heavy metals in plants, while cadmium, nickel and zinc were found to be below detectable limits. The extracts were tested against thirteen human pathogens (ten bacteria and three fungi) using the disk diffusion method. The extracts possessed a broad range of microbial activities, with the methanolic extract reportedly showing the highest zone of inhibition (31 mm) against Bacillus sp. while the n-hexane extract did not show any antimicrobial activity in the whole test organisms.Conclusion: The results obtained revealed that the leaves of C. mucunoides has some therapeutic values and could be exploited in the preparation of herbal drugs for the treatment of various ailments.

Objective: To estimate the antimicrobial activity of methanolic extract of Adenium obesum (family Apocyanaceae) in association with phytochemical analysis. Methods: The methanolic extract of the flowers of Adenium obesum was prepared and investigated for phytochemical constituents using standard methods. Antimicrobial activity of methanolic extract Adenium obesum has been carried out against one gram-positive bacteria Staphylococcus aureus and one gram-negative bacteria Escherichia coli and the antifungal activity of the plant extract was evaluated on Candida albicans species. The testing was done by the disc diffusion method to visualize the zone of inhibition of methanolic extract was compared with that of standard drug i.e., Ciprofloxacin for antibacterial activity and Fluconazole for antifungal activity. The minimum inhibition concentration was determined by micro dilution method (cup-plate method). Results: The present investigation shows the phytochemical analysis confirmed the presence of alkaloids, saponin glycosides, flavonoids, phenols, proteins and amino acids, cardiac glycosides, steroids, tannin, and phenolic compounds. The anti-microbial activity of the methanolic extract of the plant showed significant results against all the three of the test organisms in compare to the standard drugs. Conclusion: The present study concluded that methanolic extract of the flowers of adenium obesum proved the existence of phytochemical constituents. The methanolic extract of the was found to possess promising antimicrobial activity when compared with the standards and it can be useful for the development of antibiotics.

Background: Environmental contamination with elevated levels of copper (Cu), cobalt (Co), iron (Fe), zinc (Zn), lead (Pb), chromium (Cr6+), cadmium (Cd), and nickel (Ni)—all states of which are found in Uganda—raises health risk to the public. Pb, Cr6+, Cd, and Ni for instance are generally considered nonessential to cellular functions, notwithstanding the importance of the oxidative state of the metals in bioavailability. As such, we aimed in this study (i) to evaluate heavy metal concentrations in four vegetables from a typical open-air market in Uganda, (ii) to assess the safety of consuming these vegetables against the World Health Organization (WHO) recommended limits of heavy metals consumption, and (iii) to formulate a model of estimated daily intake (EDI) among consumers in the country. Methods: This was a cross-sectional study conducted in five georeferenced markets of Bushenyi district in January 2020. Amaranthus, cabbages, scarlet eggplants, and tomatoes were collected from open markets, processed, and analyzed by atomic absorption spectrometry. Modeled EDI, principal component (PCA) and cluster analysis (CA) were conducted to identify relationships in the samples. Results: The levels of essential elements in the four vegetables were found to fall from Co > Cu > Fe > Zn. Those of non-essential metals were significantly higher and followed the pattern Cd > Cr > Pb > Ni. The highest EDI values were those of Cu in scarlet eggplants, Zn in amaranthus, Fe in amaranthus, Co in amaranthus, Pb in cabbages, total Cr in scarlet eggplant, Cd in cabbages and tomatoes, and Ni in cabbages. In comparison to international limits, EDIs for Zn, Cu, Co and Fe were low while Ni in cabbages were high. PCA showed high variations in scarlet eggplant and amaranthus. The study vegetables were found to be related with each other, not according to the location of the markets from where they were obtained, but according to their species by CA. Conclusion: The presence of non-essential elements above WHO limits raises policy challenges for the consumption and marketing of vegetables in the study area. Furthermore, low EDIs of essential elements in the vegetables create demand for nutritious foods to promote healthy communities.

ABSTRACT Aim The present study was conducted to evaluate the efficacy of commercially available herbal toothpastes against the different periodontopathogens. Materials and methods Six herbal toothpastes that were commonly commercially available were included in the study. Colgate herbal, Babool, Meswak, Neem active, Dabur red toothpastes were tested for the study whereas sterile normal saline was used as control. Antimicrobial efficacies of dentifrices were evaluated against Streptococcus mutans and Actinobacillus actinomycetemcomitans. The antimicrobial properties of dentifrices were tested by measuring the maximum zone of inhibition at 24 hours on the Mueller Hinton Agar media inoculated with microbial strain using disk diffusion method. Each dentifrice was tested at 100% concentration (full strength). Results The study showed that all dentifrices selected for the study were effective against the entire test organism but to varying degree. Neem active tooth paste gave a reading of 25.4 mm as the zone of inhibition which was highest amongst all of the test dentifrices. Colgate Herbal and Meswak dentifrices recorded a larger maximum zone of inhibition, measuring 23 and 22.6 mm respectively, compared to other toothpastes. All other dentifrices showed the zone of inhibition to be between 17 and 19 mm respectively. Conclusion The antibacterial properties of six dentifrices were studied in vitro and concluded that almost all of the dentifrices available commercially had antibacterial properties to some extent to benefit dental health or antiplaque action. The level of heavy metals (Fe, Cu, As, Pb, Cd, Mg, Ca, Hg, Ni, Cr, Zn, Ag, Co, Mo, Se and Al) in soils and food crops (okra, cassava and rice) cultivated within selected mining sites in Ebonyi State, Nigeria were determined using FS240AA Atomic Absorption Spectrophotometer (AAS) according to the method of American Public Health Association (APHA). Soil samples were collected from Enyigba mining site, Ikwo mining site, AmeriAmekamining site, Izza mining site, MkpumeAkwatakwa mining site and MpumeAkwaokuku mining site while the food crop samples (okra, cassava and rice) were collected from the farmlands within the mining sites. Control samples were collected 500m away from the mining destinations were there was no evidence of mining activities on the soils. A total of sixty sub-samples and six control soil samples were collected for this study. Generally, the values of all the heavy metals analyzed for soil and food crop samples were higher than the values recommended by the World Health Organization (WHO), and those from the control site suggesting possible mobility of the metals from mining sites to farmlands through leaching and runoffs. The findings in this study also revealed that the food crops contain heavy metals exceeding the maximum permissible concentration, and could be detrimental to human health when they are consumed.

Determination of  heavy  metal  content  (Hg, Pb, Cd, Cu, Cr, Zn) in some seafoods collected from  Muara  Angke  Fish  Auction  Place  were carried out during February to July 1979. The results showed  that Zn  content  in seafoods a nalyzed  was always much higher than the other heavy metals. The concentration levels of  the  six  heavy  metals observed were low. It is almost of the same level as the metal concentration of  other  seafoods  observed  in various parts of the world oceans, but still lower  than the maximum standard set  for human  consumption  as  established  by   Tasmania, Australia and  World Health Organization. The  mean  per capita  consumption of  fish  in Jakarta is only  about 165 g  per week, while  the  Provisional Tolerated Weekly Intake for Hg, set by WHO/FAO at 300 ug level, would be attained by consuming 1500 g Rastrelliger sp., or 698 gPenaeus setiferus, or 545 g Sepia sp., and or 1250 g Pecten sp.  Therefore,  at   present the  level of metal content in fishes sold at TPI  Muara  Angke  can be  considered  as  not  dangerous  to human health as long as these quantities are not surpassed.

Solenostemon monostachyus P. Beauv (Lamiaceae family) is an important herb that is widespread in West and Central Africa.The leaves are used in various decoctions traditionally to treat diseases most especially microbial infections associated with the foot. In this study, the phytochemicals (qualitative and quantitative) of the plant were determined. The antimicrobial and antioxidant activities of the plant were also evaluated by standard methods. Results showed alkaloids, tannins, flavonoids, saponins phenolics in the methanolic extract of Solenostemon monostachyus. Cardiac glycosides and steroids absent in the methanolic extract were found present in the dichloromethane extract. The quantitative phytochemicals recorded were alkaloids (18.05 ± 0.87%), saponins (11.3 ± 0.56 %), tannins (3.3 ± 0.19 GAE). The antioxidant activity testing showed that the methanolic extract has higher DPPH scavenging ability over dichloromethane extract and standard ascorbic acid. In the antimicrobial activity testing, the cup-plate diffusion method was used and the result showed that the dichloromethane extract inhibited the growth of Bacillus species at concentrations 100 mg/ml, 50 mg/ml, 20 mg/ml and 10 mg/ml while methanolic extract inhibited the growth of bacillus species only at 100 mg/ml with 5 mm zone of inhibition. However, both methanol and dichloromethane extract showed no antimicrobial activity on the other test organisms such as Psudomonas aeruginosa, Escherichia coli and Staphylococcus aureus. In conclusion both methanolic and dichloromethane extracts of Solenostemon monostachyus are potential sources of antimicrobial and antioxidant.

In this and the next four chapters, we examine five change agents which have facilitated the emergence and re-emergence of infectious human diseases. Each agent—microbial and genetic adaptation, technology and industry, changes in host populations, environmental and ecological change, and war as a disease amplifier—has underpinned over the centuries both the appearance of new diseases and the waxing and waning of familiar infections. As shown in Figure II.1, the agents are not independent and commonly interact in complex ways to facilitate microbe emergence and re-emergence at different times and in different geographical locations. Accordingly, we also explore these interactions in our account. We begin here with microbial and vector adaptation. Disease microbes are in a continuous state of evolution, responding and adapting to the challenges and opportunities afforded by their hosts and their environments (Morse 1995). New pathogens are evolving, old pathogens are developing enhanced virulence and new clinical expressions, and susceptible pathogens are acquiring resistance to antimicrobial agents. In parallel, the environmental tolerance bands of both old and new pathogens are also changing (Cohen 1998). Not only are disease microbes in a continuous state of evolution. So, too, are the arthropod vectors that transmit many human pathogens. In the second half of the twentieth century, many of these vectors have developed tolerance to an expanding range of insecticides, larvicides, pupicides, and other chemical agents used in their control (World Health Organization 1992c). Against this background, our examination of microbial change and vector adaptation is structured around the three interlinked themes shown in Figure 4.1. We begin in Section 4.2 by examining the issue of natural variation in pathogens and illustrate this with special reference to the emergence and spread of novel subtypes of influenza A virus. We then examine the topic of selective pressure and genetic change in the context of the man-made problems of pathogen resistance to antimicrobials (Section 4.3) and vector resistance to insecticides (Section 4.4). The processes of microbial change and vector adaptation are not intrinsically geographical but they take place within, and are inextricably linked to, specific geographical environments. This gives a strong geographical emphasis to our discussion.

The drinking water quality is associated with the conditions of the water supply networks, the pollution and the contamination of groundwater with pollutants of both anthropogenic and natural origin. In this study, water samples were taken from four different waterworks in Tunceli, Turkey and heavy metals concentrations (As, Cu, Cd, Cr, Pb, Ni and Hg) were measured. Four sampling sites were pre-defined in different locations of the city. The obtained results showed that, the heavy metals concentrations in water samples did not exceed the values of WHO (World Health Organization), EC (Europe Community), EPA (Environment Protection Agency) and TSE-266 (Turkish Standard) guidelines.

Genus Mentha presents group of plants which are the most studied in family Lamiaceae. Aboveground parts are used for different purposes in pharmacy, food industry or confectionery. Most important is natural product extracted from leaves - essential oil (EO). The aim of presented experiment was to demonstrate different chemotype and compare antibacterial activity of two Mentha species EO. Plant samples were obtained from various environments – from Slovakia and from Italy. Dominant compounds were determined by GC/MS. The results showed high amount of menthol and menthone in tested Slovak peppermint EO. On the other hand, carvone and 1,8-cineole were determinate as dominant compounds in Italian spearmint EO. The antimicrobial activity of the EO was investigated by disc diffusion and broth micro dilution methods. EO was evaluated for their antibacterial activity against 7 microorganisms: Enterobacter cloacae, Salmonella spp., Klebsiella pneumoniae, Escherichia coli, Staphylococcus aureus, Streptococcus pyogenes. The determination results of antibacterial activity by agar disk diffusion method ranged from 7 to 14 mm of the growth inhibition zone. MIC of tested mint EO varied from 0.625 to 2.5 μg/mL. In addition, both EO showed relatively the same antibacterial activity against the selected Gram-negative bacteria. However, there is a variation in the antibacterial activity against Gram-positive bacteria.
