Abstract Background: To evaluate the antimicrobial activity of the fast-setting bioceramic iRoot Fast Set Root Repair Material (iRoot FS) and two other calcium silicate cements. Methods: The antimicrobial activity of iRoot FS, ProRoot MTA and Biodentine against E. faecalis and P. gingivalis were evaluated in this study. The materials were freshly mixed or set for 1 and 7 days on 5mm diameter sterile filter papers. The agar diffusion test, direct contact test and carry-over effect test were conducted, and the pH values (using a digital pH meter) were also evaluated. The data were analyzed by an analysis of variance and two-way ANOVA (α=0.05).Results: In the agar diffusion experiment, no obvious inhibition zone was observed for iRoot FS, ProRootMTA or Biodentine at any time interval. In the direct contact test, all three materials showed good antibacterial activity after setting for 20 minutes. The antibacterial properties of the three materials decreased with the increase of setting time. None of the three materials showed carry-over antibacterial effect. The pH measurement showed that the suspension of all the three materials showed high pH values (11-12). With the extension of setting time, the pH of iRoot FS and Biodentine slightly decreased.Conclusions: Fresh iRoot FS, Biodentine, and MTA killed E. faecalis and P. gingivalis effectively, and the antimicrobial effect of all the three materials decreased over 1 and 7 days after mixing. All three materials showed a tendency of alkalinity which last for at least 7 days after setting.

AbstractChitosan films containing distilled pyroligneous extracts of Eucalyptus grandis (DPEC), characterized and developed by Brazilian Agricultural Research Corporation—Embrapa Temperate Agriculture (EMBRAPA-CPACT), were evaluated for antimicrobial activity against Candida albicans, Streptococcus mutans, and Lactobacillus acidophilus by direct contact test. Further, their capacity for the prevention of teeth enamel demineralization and cytotoxicity in vitro were also determined. The natural polymers were tested at different concentrations (1500–7500 µg mL−1) and the formulation of an experimental fluoride varnish with antimicrobial activity was evaluated by direct contact test, whereas cytotoxicity was analyzed through the colorimetric MTT assay. Preliminary data showed no statistically significant differences in cytotoxicity to NIH/3T3 cell line when DPEC is compared to the control group. On the other hand, the antimicrobial capacity and demineralization effects were found between the test groups at the different concentrations tested. Chitosan films containing distilled pyroligneous extracts of E. grandis may be an effective control strategy to prevent biofilm formation related to dental caries when applied as a protective varnish. They may inhibit the colonization of oral microorganisms and possibly control dental caries through a decrease in pH and impairment of enamel demineralization.

Aims and Objectives: The aim of the present study was to evaluate and to compare the antibacterial activity of AH Plus, Tubliseal and MTA Fillapex against E.faecalis with and without incorporation of Chitosan nanoparticles. Methodology: The antibacterial effect against Enterococcus faecalis was evaluated by i) direct contact test (DCT) and ii) agar diffusion test (ADT). The nanoparticles were incorporated into the sealers at concentration of 2 % weight/ weight. Bacterial growth rates were analysed using ANOVA followed by Fischer Exact Test Results: The antibacterial tests demonstrated significant E.faecalis growth inhibition in the DCT with the sealer samples incorporated with chitosan nanoparticles. Direct contact test showed total growth inhibition of up to 8 logs in count in AH Plus, up to 7 logs in count in Tubliseal, and up to 6 logs in count in MTA Fillapex which were incorporated with chitosan nanoparticles. Significant differences were found between the unmodified sealers and the modified sealers. No antibacterial effect was observed in the ADT, indicating Chitosan nanoparticles were not diffusing into the agar. Conclusion: The endodontic sealers incorporating Chitosan nanoparticles exhibited significant and stable antimicrobial properties.

Objective: The objective of this study was to evaluate the antimicrobial activity of mineral trioxide aggregate (MTA) and calcium hydroxide (Ca(OH)2)sealers on Enterococcus faecalis at various time points after the preparation of the sealer.Methods: Antimicrobial activity of MTA and Ca(OH)2 sealer on E. faecalis (ATCC 29212) was assessed by the direct contact test immediately, 1 day,and 7 days after the preparation of the sealer.Results: The antimicrobial effect of MTA sealer was greatest 1 and 7 days after the preparation of the sealer. The effect of Ca(OH)2 was greatest 1 dayafter the preparation of the sealer.Conclusion: MTA sealer showed constant antimicrobial activity on E. faecalis overtime. By contrast, Ca(OH)2 sealer showed decreasing antimicrobialactivity overtime, with the greatest antimicrobial activity being 1 day after the preparation of the sealer.

ABSTRACT Aim To evaluate pH and antibacterial activity of pastes with calcium hydroxide [Ca(OH)2] and zinc oxide (ZnO) microparticles (micro) or nanoparticles (nano) and association with 0.4% chlorhexidine against Enterococcus faecalis. Materials and methods The following pastes were analyzed: Ca(OH)2/ZnO micro, (2) Ca(OH)2/ZnO nano, (3) Ca(OH)2/ ZnO micro + 0.4% chlorhexidine, (4) Ca(OH)2/ZnO nano + 0.4% chlorhexidine. Antibacterial activity against E. faecalis was evaluated by agar diffusion test. The direct contact test on planktonic cells of E. faecalis was performed for 30 and 60 seconds. Root canals from bovine teeth were filled with the pastes and pH was evaluated after 1, 7, 14, 21, 30 and 60 days. The data obtained were submitted to the statistical tests analysis of variance (ANOVA) and Tukey or Kruskal-Wallis and Dunn test, with a 5% significance level. Results Calcium hydroxide and zinc oxide nano, and the pastes with 0.4% chlorhexidine were more effective in agar diffusion test. In the direct contact test, the pastes with chlorhexidine showed the highest effect after 30 seconds. All pastes eliminated E. faecalis after 60 seconds. All pastes promoted an increase in pH. The highest increase in pH was observed with nanoparticle medications after 1 and 7 days (p < 0.05). After this period, the pastes presented similar pH increase. Conclusion It was concluded that calcium hydroxide and zinc oxide nanoparticles promoted greater initial alkalinization. The antimicrobial activity of the pastes against E. faecalis is favored by the association with chlorhexidine. Clinical significance Although nanoparticles of calcium hydroxide and zinc oxide promoted antibacterial effect, the activity against E. faecalis is favored by association with chlorhexidine. Root canal sealers that possess good antimicrobial property can prevent residual and recurrent infection and contribute to successful endodontic therapy. This study evaluated the antimicrobial activity of four endodontic sealers, AH Plus, Tubliseal EWT, EndoRez, and iRoot SP, against three different microorganisms,E. faecalis, C. albicans, andS. aureus, by direct contact test. 10 μL microbial suspensions were allowed to directly contact the four endodontic sealers for 1 hr at 37°C. Subsequently microbial growth was measured spectrophotometrically every 30 min for 18 hours. The microbial suspensions were simultaneously tested to determine the antimicrobial effect of components which are capable of diffusing into the medium. The results revealed that AH Plus and iRootSP had significantly higher antimicrobial activity againstE. faecalis. AH Plus and Tubliseal EWT showed significantly higher antimicrobial activity againstC. albicansandS. aureuscompared to iRoot SP and EndoRez. EndoRez showed the least antimicrobial activity against all the three microorganisms. Inhibition of microbial growth is related to the direct contact of microorganisms with the sealers. In conclusion AH Plus had significantly higher antimicrobial activity againstE. faecalis, C. albicans, andS. aureus.

The volatile oil composition of four Baccharis species ( Baccharis papilosa, B. polycephala, B. buxifolia, and B. convawyi) growing wild in Bolivia were studied by GC and GC/MS. The antimicrobial activity of the oils was determined by the agar diffusion and bioautography methods. Solvent extracts, polar and non-polar, of the same species were also tested for antimicrobial and antioxidant activities. Extracts from B. polycephala, B. papilosa and B. convawyi presented significant antioxidant activity, determined using the DPPH radical scavenging method, showing EC50 values of 4.74, 7.17 and 7.97 μg/mL, respectively. Screening for antimicrobial activity was conducted using the agar diffusion test. The most active extracts (% inhibition ≥ 50) were further examined by the dilution method. All the plant extracts studied showed higher antimicrobial activity against S. aureus (Gram-positive) than against E. coli and P. fluorescens (Gram-negative).

Elimination of microorganisms from the root canal system and the prevention of subsequent reinfection are of importance for long-term endodontic treatment. The application of a sealer with antibacterial properties may reduce the reinfection and improve the success rate of the root filling treatment. The aim of this paper is to evaluate the antibacterial properties of a novel root canal sealer based on injectable self-curing polyurethane with silver phosphate (PU/Ag3PO4). The antibacterial abilities were assessed by direct contact test, anti-bacterial adhesion assay and bacteriostatic rate test. The results show that the fabricated PU/Ag3PO4sealer can completely inhibit the bacterial growth and prevent bacterial adhension effectively. The bacteriostatic rate is 93.6% and 98.1% when the bacteria incubated with PU/Ag3PO4 sealer for 7 and 24h respectively. The strong antibacterial abilities suggest that PU/Ag3PO4 sealer has great application potential in the field of root canal filling.Key words: Root canal sealer, polyurethane, silver phosphate, antibacterial properties, direct contact test
