Abstract Polyethylene terephthalate (PET) is an important synthetic polymer accumulating in nature 2 and recent studies have identified microorganisms capable of degrading PET. While the majority of 3 known PET hydrolases originate from the Actinobacteria and Proteobacteria, here we describe the 4 first functional PET-active enzymes from the Bacteroidetes phylum. Using a PETase-specific 5 Hidden-Markov-Model (HMM)-based search algorithm we identified two promiscuous and cold6 active esterases derived from Aequorivita sp. (PET27) and Chryseobacterium jeonii (PET30) acting 7 on PET foil and powder. Notably, one of the enzymes (PET30) was able to hydrolyze PET at 8 temperatures between 4° - 30°C with a similar turnover rate compared to the well-known Ideonella 9 sakaiensis enzyme (IsPETase). 10 PET27 and PET30 homologues were detected in metagenomes encompassing a wide range 11 of different global climate zones. Additional transcript abundance mapping of marine samples imply 12 that these promiscuous enzymes and source organisms may play a role in the long-term 13 degradation of microplastic particles and fibers.

Certain members of the Actinobacteria and Proteobacteria are known to degrade polyethylene terephthalate (PET). Here, we describe the first functional PET-active enzymes from the Bacteroidetes phylum. Using a PETase-specific Hidden-Markov-Model- (HMM-) based search algorithm, we identified several PETase candidates from Flavobacteriaceae and Porphyromonadaceae. Among them, two promiscuous and cold-active esterases derived from Aequorivita sp. (PET27) and Kaistella jeonii (PET30) showed depolymerizing activity on polycaprolactone (PCL), amorphous PET foil and on the polyester polyurethane Impranil® DLN. PET27 is a 37.8 kDa enzyme that released an average of 174.4 nmol terephthalic acid (TPA) after 120 h at 30°C from a 7 mg PET foil platelet in a 200 μl reaction volume, 38-times more than PET30 (37.4 kDa) released under the same conditions. The crystal structure of PET30 without its C-terminal Por-domain (PET30ΔPorC) was solved at 2.1 Å and displays high structural similarity to the IsPETase. PET30 shows a Phe-Met-Tyr substrate binding motif, which seems to be a unique feature, as IsPETase, LCC and PET2 all contain Tyr-Met-Trp binding residues, while PET27 possesses a Phe-Met-Trp motif that is identical to Cut190. Microscopic analyses showed that K. jeonii cells are indeed able to bind on and colonize PET surfaces after a few days of incubation. Homologs of PET27 and PET30 were detected in metagenomes, predominantly aquatic habitats, encompassing a wide range of different global climate zones and suggesting a hitherto unknown influence of this bacterial phylum on man-made polymer degradation.

Quantitative estimates of future Antarctic climate change are derived from numerical global climate models. Evaluation of the reliability of climate model projections involves many lines of evidence on past performance combined with knowledge of the processes that need to be represented. Routine model evaluation is mainly based on the modern observational period, which started with the establishment of a network of Antarctic weather stations in 1957/58. This period is too short to evaluate many fundamental aspects of the Antarctic and Southern Ocean climate system, such as decadal-to-century time-scale climate variability and trends. To help address this gap, we present a new evaluation of potential ways in which long-term observational and paleo-proxy reconstructions may be used, with a particular focus on improving projections. A wide range of data sources and time periods is included, ranging from ship observations of the early 20th century to ice core records spanning hundreds to hundreds of thousands of years to sediment records dating back 34 million years. We conclude that paleo-proxy records and long-term observational datasets are an underused resource in terms of strategies for improving Antarctic climate projections for the 21st century and beyond. We identify priorities and suggest next steps to addressing this.

<p>One of the most used metrics to gauge the effects of climate change is the equilibrium climate sensitivity, defined as the long-term (equilibrium) temperature increase resulting from instantaneous doubling of atmospheric CO2. Since global climate models cannot be fully equilibrated in practice, extrapolation techniques are used to estimate the equilibrium state from transient warming simulations. Because of the abundance of climate feedbacks – spanning a wide range of temporal scales – it is hard to extract long-term behaviour from short-time series; predominantly used techniques are only capable of detecting the single most dominant eigenmode, thus hampering their ability to give accurate long-term estimates. Here, we present an extension to those methods by incorporating data from multiple observables in a multi-component linear regression model. This way, not only the dominant but also the next-dominant eigenmodes of the climate system are captured, leading to better long-term estimates from short, non-equilibrated time series.</p>


This volume in the Long-Term Ecological Research Network Series would present the work that has been done and the understanding and database that have been developed by work on climate change done at all the LTER sites. Global climate change is a central issue facing the world, which is being worked on by a very large number of scientists across a wide range of fields. The LTER sites hold some of the best available data measuring long term impacts and changes in the environment, and the research done at these sites has not previously been made widely available to the broader climate change research community. This book should appeal reasonably widely outside the ecological community, and because it pulls together information from all 20 research sites, it should capture the interest of virtually the entire LTER research community.

Outputs of studies of current and scenario changes of the river runoff in the Volga and the Don basins probable in the current century first third part under influence of the global climate change and social/economic transformations are presented. Long-term phases of low and high annual and seasonal runoff of the Volga and the Don over the period from late XIX and early XXI centuries and the human factors’ influence on them have been found. Their duration varies in the wide range reaching eighty years and more. Noticeable runoff changes are characteristic for contrast phases. Integral scenario assessment of the river runoff changes on the basis of various climatic scenarios and water consumption has been given. Rather wide range of the probable runoff changes in comparison with the current situation has been obtained on the basis of scenarios of social/economic development and specific water use changes. Expected increase of the Volga water content under the influence of climatic factors will facilitate improvement of the water/economic situation at this river, while there is no chance to expect water content in the Don. At the same time there is a probability that the current phase of climate worming is going to come to an end and to be replaced by the long-term cold phase and this might cause the corresponding hydrological changes.

In the last decade, extensive fires have occurred on most continents, affecting a wide range of ecosystems. We convened a Symposium at the 3rd International Fire Ecology and Management Congress in 2006 to address the issue of large fires and their ecological consequences in landscapes. The 10 papers presented here variously discuss the place of large fires in the context of historical fire regimes, the heterogeneity of fire regime components that are associated with large fires, and the ecological consequences of large fires. The discussions cover a range of biomes, from tropical to temperate, across the world. Three consistent themes emerged: firstly, large fires are usually a part of the Historical Range of Variability; secondly, large fires are inherently heterogeneous, leaving footprints of spatial and temporal diversity that may influence landscapes for decades; and thirdly, large fires have been perceived as socially and ecologically ‘disastrous’, due to obvious and significant deleterious effects on life and property, and the scale of immediate environmental impact. However, the papers presented here indicate that the long-term ecological impacts of individual large fires are not necessarily disastrous. Crucial impacts of large fires on ecosystems may depend largely on their rate of recurrence as well as landscape-scale variation in severity. The incidence and characteristics of large fires may change in the future, as a consequence of global climate change, and other social drivers of landscape change.

Conceptual approaches to the formation of an integrated program for early detection and prevention of civilization diseases based on these technologies are given. A vision of the prospects for the development of this area in domestic and foreign medicine has been formed.

Based on the latest data, paper investigates the dynamics of global climate change and its impact on economic growth in the long-term. The notion of climate risk is considered. The main directions of climate risk management policies are analyzed aimed, first, at reducing anthropogenic greenhouse gas emissions through technological innovation and structural economic shifts; secondly, at adaptation of population, territories and economic complexes to the irreparable effects of climate change. The problem of taking into account the phenomenon of climate change in the state economic policy is put in the context of the most urgent tasks of intensification of long-term socio-economic development and parrying strategic challenges to the development of Russia.
