Base-to-tip profiles of sucrose, glucose, fructose, and respiration rate were measured for asparagus (Asparagus officinalis L.) spears stored at 0C. Fructose content was ≈3-fold and 4-fold higher than glucose and sucrose, respectively. The highest level of fructose was found in the base and was ≈15-fold higher than the tip. The changes in asparagus metabolism were characterized by loss of sucrose and a high rate of respiration within the first hours after harvest. Sucrose was more rapidly lost than the other sugars during this period. The respiration rate was measured along the length of intact spears at 0.5, 1, 2, and 3 h after harvest. Subsequent measurements were taken after larger time intervals for 23 days. The respiration rate declined rapidly to ≈60% of the initial rate within 12 h, decreasing more slowly thereafter. Initially, the respiration rate of the tip was about four times that of the base, but, after 23 days, the respiration rate of the tip was only twice that of the base. Sucrose content and respiration rates were closely correlated.

The influence of the superoxide-generating system, xanthine oxidase, on the release of iron from various vertebrate ferritins was determined both in the presence and absence of superoxide dismutase. The initial rate of iron release in the presence of this system was higher for ferritins from human, trout and rat liver than for those from lamprey liver and horse spleen. The proportion of this iron release that was superoxide-dependent in the case of rat, human and trout ferritins was 92, 86 and 84% respectively, whereas no such superoxide-dependent iron release occurred from the ferritins of lamprey liver and horse spleen. On the other hand, the rate of superoxide-independent iron release was of comparable magnitude for all of the species examined. The rate of superoxide-dependent iron release was related neither to the iron: protein ratios nor to the subunit size of the ferritins. However, it is significant that the ferritins with a high rate of superoxide-dependent iron release came from tissues known to be susceptible to iron damage. It is thus proposed that the resistance of lamprey liver ferritin to the mobilization of iron by superoxide ions accounts in part for the tolerance of the lamprey liver to high iron loads.

Asparagus spears (Asparagus officinalis L.) were placed in solutions of six different concentrations of sucrose (0%, 1%, 2%, 4%, 8%, and 16%) plus citric acid at 0C for 24 h following harvest. The profiles of sucrose, fructose, glucose, and respiration rate along the length of the spear were evaluated throughout storage. The effect of carbohydrate loading on the rate of respiration, sucrose loss, and the shelf life of asparagus was determined. For all treatments, sugars decreased and respiration rate increased from the butt to the tip. The 4% sucrose treatment enhanced the sugar level in the tip ≈5-fold relative to the control. For the 8% and 16% treatments, sucrose tended to accumulate in the base. Spears loaded with higher sucrose concentrations had higher respiration rates than controls up to 3 h after loading. After this time, no significant differences were observed between treatments. For all treatments, respiration rates declined rapidly following harvest, stabilizing in ≈24 h. Weight gain and growth increased as the treatment sucrose concentration decreased. Solution uptake was enhanced by loading at lower humidity levels.

We measured CO2 efflux from root systems of mycorrhizal and nonmycorrhizal conifer seedlings planted in silica sand and maintained in darkness for 11 to 15 days. We first compared methods of isolating the root system for root respiration measurement using Douglas-fir seedlings (Pseudotsugamenziesii (Mirb.) Franco). Shoot excision effectively isolated the root system and affected total root respiration only slightly. We used this technique to compare respiration rates of mycorrhizal and nonmycorrhizal root systems of ponderosa pine (Pinusponderosa Laws.) seedlings. Some seedlings were uninoculated; others were infected with an ectomycorrhizal species (Laccahalaccata or Hebelomacrustuliniforme) or an unidentified ectendomycorrhizal species. After shoot excision, respiration rates declined within 48 h to a "maintenance" respiration rate. Over the next several days, the respiration rate continued to decline to a much lower "basal" rate typical of nonphotosynthetic seedlings depleting their reserves. The maintenance respiration rate of the Laccaria-infected seedlings was significantly lower than those of the other treatments. The basal respiration rates of the two ectomycorrhizal treatments (Laccaria and Hebeloma) were lower than those of the other groups. Such reductions in respiration rate might strongly affect root longevity and whole-plant carbon budgets.

ABSTRACT Relatively large doses of vasopressin administered intraperitoneally to the trout significantly enhanced the kidney respiration rate. In contrast to vasopressin a single dose of oxytocin depressed the kidney Qo2 value. This depression continued throughout the observed 24 hour period after injection. Cortisol enhanced the kidney Qo2 values significantly and to a greater extent than vasopressin. These results are discussed in relation to possible adaptive mechanism in euryhaline species of teleosts.

An on-line respiration meter is presented to monitor three types of respiration rates of activated sludge and to calculate effluent and influent short term biochemical oxygen demand (BODst) in the continuous activated sludge process. This work is to verify if the calculated BODst is reliable and the assumptions made in the course of developing the proposed procedure were acceptable. A mathematical model and a dynamic simulation program are written for an activated sludge model plant along with the respiration meter based on mass balances of BODst and DO. The simulation results show that the three types of respiration rate reach steady state within 15 minutes under reasonable operating conditions. As long as the respiration rate reaches steady state the proposed procedure calculates the respiration rate that is equal to the simulated. Under constant and dynamic BODst loading, the proposed procedure is capable of calculating the effluent and influent BODst with reasonable accuracy.

The New Delhi Metallo-β-lactamase (NDM) is among the most threatening forms of carbapenemases produced by K. pneumoniae, well-known to cause severe worldwide infections. The molecular epidemiology of blaNDM-1-harboring K. pneumoniae is not well elucidated in Pakistan. Herein, we aim to determine the antibiotics-resistance profile, genes type, molecular type, and plasmid analysis of 125 clinically isolated K. pneumoniae strains from urine samples during July 2018 to January 2019 in Pakistan. A total of 34 (27.2%) K. pneumoniae isolates were carbapenemases producers, and 23 (18.4%) harbored the blaNDM-1 gene. The other carbapenemases encoding genes, i.e., blaIMP-1 (7.2%), blaVIM-1 (3.2%), and blaOXA-48 (2.4%) were also detected. The Multi Locus Sequence Typing (MLST) results revealed that all blaNDM-1-harboring isolates were ST11. The other sequence types detected were ST1, ST37, and ST105. The cluster analysis of Xbal Pulsed Field Gel Electrophoresis (PFGE) revealed variation amongst the clusters of the identical sequence type isolates. The blaNDM-1 gene in all of the isolates was located on a 45-kb IncX3 plasmid, successfully transconjugated. For the first time, blaNDM-1-bearing IncX3 plasmids were identified from Pakistan, and this might be a new primary vehicle for disseminating blaNDM-1 in Enterobacteriaceae as it has a high rate of transferability.

1. A prolyl-s-RNA synthetase (prolyl-transfer RNA synthetase) has been purified about 250-fold from seed of Phaseolus aureus (mung bean), a species not producing azetidine-2-carboxylic acid, and more than 10-fold from rhizome apices of Polygonatum multiflorum, a liliaceous species containing azetidine-2-carboxylic acid. The latter enzyme was unstable during ammonium sulphate fractionation. 2. The enzymes exhibited different substrate specificities towards the analogue. That from Phaseolus, when assayed by the ATP-PP(i) exchange, showed azetidine-2-carboxylic acid activation at about one-third the rate with proline. Both labelled imino acids gave rise to a labelled aminoacyl-s-RNA. The enzyme from Polygonatum, however, activated only proline. 3. The enzyme from Polygonatum also formed a labelled prolyl-s-RNA with Phaseolus s-RNA but at a lower rate than when the Phaseolus enzyme was used. No reaction occurred when the Phaseolus enzyme was coupled with Polygonatum s-RNA, and only a very slight one was observed when both enzyme and s-RNA came from Polygonatum. 4. Protein preparations from seeds of Pisum sativum, another species not producing azetidine-2-carboxylic acid, also activated the analogue in addition to proline, whereas those from rhizome and seeds of Convallaria, the species from which the analogue was originally isolated, failed to activate it. However, a liliaceous species not producing the analogue, Asparagus officinalis, activated it. 5. Of the other proline analogues investigated, only 3,4-dehydro-dl-proline and l-thiazolidine-4-carboxylic acid were active with the enzyme preparation from Phaseolus. 6. pH optima of 7.9 and 8.4 were established for the enzymes from Phaseolus and Polygonatum respectively. 7. The Phaseolus enzyme was specific for ATP and PP(i). Mn(2+) partially replaced the requirement for Mg(2+) as cofactor. Preincubation with p-chloromercuribenzoate at a concentration of 0.5mm or higher produced over 99% inhibition of the Phaseolus enzyme. One-half the enzymic activity was destroyed by preheating for 5min. at 62 degrees in tris-hydrochloric acid buffer, pH7.9. 8. All experimental evidence supports the hypothesis that azetidine-2-carboxylic acid and proline are activated by the same enzyme in Phaseolus preparations, whereas the analogue was inactive in all Polygonatum preparations. The possible nature of this different substrate behaviour is discussed.

Microbial biomass and soil respiration rate decreased after application of 150 kg NH4NO3–N∙ha−1 to different coniferous forest podzols. The decrease was already found 3 months after fertilization and was still evident after 3–5 years. Changes in pH, organic matter, or water content in the soils could not explain the decreases. In laboratory experiments, several unfertilized forest soils were treated with 2 mg of NH4NO3–N or of urea–nitrogen∙g wet soil−1. The ammonium nitrate addition resulted in severe depressions of the respiration rates during and up to 175 days of incubation and the decrease was evident after about 1 week. The urea treatment initially increased the respiration rate of the soils, but this appeared to be a transitory effect.

The author develops a multiregional growth model with endogenous amenity and capital accumulation for any number of regions. The simulation results demonstrate that the national dynamics have a unique equilibrium. Comparative statics analysis shows that, if environmental improvement occurs in the technologically advanced (less advanced) region, the national output rises (falls). As a region improves its technology, the other two regions' aggregated output levels fall—not only in relative, but also in absolute, terms. This implies that if any region has a high rate of technological change and the other regions remain technologically stationary, then economic activities will tend to be concentrated in the technologically advancing region. It is also shown that technological differences appear to play only a small role in accounting for spatial wage disparities and endowments.

During satellite tagging of harbour porpoises (Phocoena phocoena), heart rate, respiration rate and cortisol value were measured to evaluate stress effects during handling and tagging. Respiration rates were obtained using video recordings, heart rates were recorded and serum cortisol levels were analysed from blood samples. Differences in heart rates, respiration rates and cortisol levels before and during the tagging events were investigated. An overall significant decrease of 31.5% in respiration rate was found during the tagging event period, while mature porpoises respired significantly more often than immature individuals. Though significant differences in heart rates were found for some individuals, no general significant change for all animals was detected. We found no correlation between cortisol concentration and either heart rate or respiration rate, nor did we find any relationships between cortisol and month of year, sex and body length. As high individual variations occurred in response to tagging of harbour porpoises, it is not possible to give general advice based on the factors investigated, on how to reduce stress during handling. However, pouring water over the animal and lowering it into the water seem to stabilize a stressed animal. Therefore, general precaution and individual judgement based on experience is essential when handling wild harbour porpoises.
