Background and Objectives: Bacteria that attach to the surfaces and produce polymeric matrix resulting in the biofilms formation are involved in a wider range of human infections. Biofilms forming Staphylococcus aureus and Escherichia coli are considered to be highly antibiotics resistant. This study aimed to analysis the antibiogram profile of biofilm forming S. aureus and E. coli in Mukalla city, Hadhramaut,Yemen. Methods: Sixty clinical isolates of S. aureus and E. coli were isolated from different clinical samples, and identified by standard bacteriological methods, then subjected to biofilm formation detection by tissue culture plate (TCP) method. The antibiotics susceptibility test was performed by disc diffusion (Kirby-Bauer) method. Chi-square test was used to analyze the data and p value< 0.05 was taken as significant. Results: Among the total isolates S. aureus and E. coli, TCP method detected 55% of isolates as strong, 25% as moderate and 25% as weak/non-biofilm producers. Biofilm forming of S. aureus developed significantly higher degrees of antibiotic resistance of amoxicillin/clavulanic acid100%, ceftazidime 95.8%, cefotaxime62.5%, cefadroxil 45.8%, ciprofloxacin 41.7% and ceftriaxone 25% with significant statistics correlation the resistance of amoxicillin/clavulanic acid and ceftazidime and bacterial biofilm production (p-value< 0.05). The rates of antibiotics resistance biofilm E. coli were 100%, 91.7%, 75%, 70.8%, 66.7%, 62.5% and 33.3%for amoxicillin/clavulanic acid, cefadroxil, cefotaxime, ceftazidime, ceftriaxone, ciprofloxacin and co-trimoxazole respectively with statistically significant correlation of cefadroxil resistance (p-value < 0.05). Conclusion: TCP method showed that S. aureus and E. coli isolates have high degree of biofilm forming ability. Abstract Objective Antibiotics misuse and a high level of antibiotics resistance is observed worldwide, but particularly in developing countries. Kosovo in the last decade is facing challenges regarding antimicrobial resistance. The purpose of the present study was to investigate patterns of antibiotics prescriptions of dentists in Kosovo's major dental clinics. Materials and Methods For Kosovo's prescribing pattern, data collection was obtained from 10 Regional Dental Clinics and a Tertiary Health Center regarding patients who were prescribed antibiotics in the years 2015 to 2019. Data analysis was performed by using descriptive statistics and was processed by using MS Excel. Results Most prescribed antibiotic during the observed period from 2015 to 2019 in Kosovo was amoxicillin, although a drastic increase of amoxicillin with clavulanic acid—as a broad-spectrum antibiotic—is observed. The trend of antibiotics use in tertiary health institutions is in an overall decrease in Kosovo with an exception in the year 2017. Despite this overall decrease, inconsistency in prescribing is observed when the pattern is analyzed for each region separately. The highest number of patients in health care dental clinics received antibiotics for maxilla-related health conditions and the lowest number of them for oncologic ones. Conclusion The patterns of antibiotics prescriptions by dental practitioners in Kosovo during the years 2015 to 2019 are fluctuating. Compared with the global health care standards, the irrational use of antibiotics in dental health care clinics in Kosovo still exist and this issue should be further addressed by respective actors.

Abstract Baiyangdian Lake is a typical and largest multi-habitat lake in the North plain of China. To understand the generation and transmission of antibiotics resistance genes (ARGs) in multi-habitat lakes, the contents of nutrients (TC, TOC, TN, TP and TS), heavy metals (Zn, Cr, Ni, Cu, Pb, As, Cd and Hg), 22 antibiotics, 16S-rRNA(16S), Class I integron (intI1) and 20 ARGs were determined. Samples were taken from the Fuhe river, river estuaries, reed marshes, living area, fish poods and open water of Baiyangdian Lake. The results showed that quinolones (QNs) were the main pollutants, and the content range was ND-104.94 ng/g. Thereinto, aac (6') -IB, blaTEM-1, ermF, qnrA, qnrD, tetG, sul1, sul2 and tetM were detected in 100%. The absolute abundance of sul1 was the highest (5.25×105copies/g-6.21×107 copies/g), which was the dominant ARGs. In these different habitats, the abundance of antibiotics and ARGs in river estuary was the highest, and that in reed marshes was the lowest. There was a significant positive correlation between the abundance of heavy metals (Cu, Pb, Zn, Ni, Cd, Hg) and the absolute abundance of 11 ARGs (P<0.01). Redundancy analysis showed that Cu, Zn, intI1, TP and macrolides (MLs) were the important factors affecting the distribution of ARGs. Our findings provides a more likely driving and influencing factor for the transmission of ARGs in lakes with complex and diverse habitats.

Milk is an ideal food for all age groups. The current study was carried out to identify the microorganisms to assess the raw milk quality and the antibiotic resistance of those identified micro-organisms. Five raw milk samples along with two high treatment (UHT) milk samples from different locations of Noakhali district of Bangladesh were analysed. Bacterial isolation was performed by Nutrient Agar (NA) and MacConkey (MCA), Eiosin Methylene Blue (EMB) and Genital menital salt agar (GMSA). The isolates were then identified by Kliger’s Iron Agar (KIA) test, Motility Indole Urease (MIU) test, Catalase and Oxidase tests. Antibiotics resistance tests were done for 13 different antibiotics. Among all these samples, Maijdee Bazar (S4) contained the highest load as 1.87×106 and the UHT samples contained no bacterial contamination. E. coli covered 47.05% whereas Listeria, Bacillus and Yersinia were in the same percentage as 5.88% among all isolates. Salmonella and Staphylococcus were 23.53% and 11.76%, respectively. Listeria and Salmonella were resistant to five different antibiotics by 46.15% and 38.46% of multiple antibiotic resistance index (MRI), correspondingly. However, E. coli and Yersinia were resistant to three antibiotics namely, Rifampcin (RIF), Cefotaxime (CTX), Amoxycillin (AMX) by about 23% as MRI percentage. Bacillus and Staphylococcus both were resistant to Cefepime (CPM) by 7.69% of MRI. Hence, it can be concluded that Rifampcin and Cefepime were most common antibiotics which were resisted by most of the isolates. Therefore, hygiene aspect of these milk sources needs to be taken into consideration with high priority. Also, the antibiotics which are resisted by different organisms will be detrimental for public health aspects.

The problem of antibiotic resistance remains a major concern among the patients and health care provider. This study was carried out therefore to determine the antibiotics resistance susceptibility pattern bacteria with urinary tract infections among patients attending Federal Teaching Hospital Ido-Ekiti. A total of 200 patients suspected of having a UTI were recruited for the study. The urine samples were cultured on Cysteine Lactose Electrolyte Deficient Agar (CLED) and the colonies were identified using colonial morphology and biochemical test. Antibiotics susceptibility testing was carried out using Kirby-Bauer disk diffusion techniques. Out of 200 urine samples analysed 82 (41.0%) yielded significant bacteria growth belonging to 4 different genera with Escherichia coli having the highest isolation rate 42 (81.2%) followed by Pseudomonas aeruginosa 16 (19.5%), Staphylococcus aureus 14 (17.0%) and Klebsiella aerogenes 10 (12.1%). Female patients had the highest isolation rate of 48 (58.5%) compare to their male counterpart with 34 (41.5%) isolation rate. Age group 31-40yrs had the highest isolation, rate of 20 (24.4%). The antibiotic resistance pattern exhibited by all the bacteria ranged from 14.3% to 90.5%. Gentamicin exhibited the least resistance rate. However, antibiotics resistance is frightening; therefore, there is a need for antibiogram before prescription of antibiotics for the treatment of urinary tract infection.

Pharmaceuticals, among the emerging contaminants, are pseudopersistent and recently of serious concern due to universal use, toxicity, and resistance development at low concentrations. This study was aimed at assessing the prevalence and risk of eight pharmaceuticals in surface water used for vegetable irrigation in Ghana’s Kumasi and Sunyani metropolises, which are influenced by hospitals, sewage treatment facilities, and market effluents. Samples were concentrated via solid-phase extraction (SPE) while liquid chromatography was used to identify and quantify the analytes. Ibuprofen, acetaminophen, and diclofenac were the detected analgesics in this study, with concentrations stretching from below detection limit (not detected) to 319.0 ng/L, while amoxicillin, trimethoprim, and cefuroxime were the detected antibiotics with a concentration range of no detection to 840.0 ng/L. Based on the available long-term data, an environmental risk assessment was conducted. Because of the presence of ibuprofen, the lowest trophic level and fish were shown to be at risk. The estimated risk quotient values for antibiotics resistance were above 1 for all the antibiotics investigated in surface water impacted by the wastewater of hospitals and pharmaceutical companies’ except surface water impacted by sewage treatment plants (STPs) and market wastewater. The existence of these pharmaceuticals in surface water does not only point to a general concern for the environment but also a potential health risk on humans and other lives as a result of their impact on drinking water and vegetable production in Ghana.

Livestock, particularly pigs, have increasingly been recognized as important reservoirs for zoonotic transmission of pathogenic bacteria, including staphylococci. Livestock production systems in developing countries of sub-Saharan Africa, including Nigeria, are characterized by high misuse/abuse of antimicrobials and a close association between humans and these animals, which promotes the emergence and transmission of resistant and potentially virulent bacteria. In the present study, we investigated the occurrence and characteristics (species distribution, virulence and resistance profile) of staphylococci from smallholder backyard pig farms, slaughter slabs and pig handlers in Makurdi, Nigeria. A total of 330 nasal swabs originating from 300 pigs and 30 in-contact humans were collected and processed. One hundred and thirteen samples [34.2 %; 95 % confidence interval (CI): 29.1–39.6] comprising 103 (34.3 %; 95 % CI: 29.0–40.0) and 10 (33.3 %; 95 % CI: 17.3–52.8 %) samples from pigs and humans, respectively, were positive for staphylococci, yielding 120 isolates (pigs n=110, humans n=10). The 120 isolates were distributed into 15 species with Staphylococcus aureus (n=25) followed by Staphylococcus cohnii (n=19) and Staphylococcus sciuri (n=14) occurring more frequently. All isolates were resistant to β-lactam (100 %) antibiotics. Resistance to some critical antimicrobials, including linezolid (22 %), vancomycin (19.2 %), gentamicin (7.5%) and the fluoroquinolones ciprofloxacin (75.8 %) and enrofloxacin (66.7 %), was also observed. Majority (99.2 %) of the isolates displayed a multidrug resistance phenotype with the AMP-C-CIP-E-ENR-FOX-OX-P-S-SXT-TE phenotype being predominant. Overall, 70 % of the isolates expressed the methicillin resistance phenotype, out of which 20 % (n=17) were MRSA. Resistance to serum bactericidal activity and biofilm production were respectively observed in 45 (100 %) and 5 (11.3 %) of the coagulase-positive staphylococci. Our findings demonstrated the occurrence of a high diversity of staphylococci expressing multidrug resistance and potentially virulent phenotypes among healthy swine and pig handlers in small-scale backyard farms in North-Central Nigeria. These findings underscore the potential role of pig production settings in the emergence and dissemination of potentially virulent staphylococci and the importance of the development of antimicrobial resistance monitoring systems/implementation of control measures in developing countries. Proper hygienic practices and control of indiscriminate use and misuse of antibiotics are recommended.

Fusobacterium nucleatum is a typical periodontal opportunistic pathogen that contributes to inflammatory diseases at oral or extra-oral sites. The emergence of antibiotics resistance calls for a substitute therapeutic strategy to control bacterial infections effectively. In addition, macrophage is an essential part of host defense against microorganism invasion in vivo. Herein, we synthesized gold nanoclusters (AuNCs) as potent antibacterial material to kill F. nucleatum through bactericidal behavior and pro-phagocytic activity. Firstly, we evaluated the bactericidal effect of AuNCs on F. nucleatum by the means of plate counting, growth curve measurement, bacterial viability analysis, and live/dead fluorescent imaging. Morphological changes and the intracellular reactive oxygen species levels of bacteria were further analyzed to explore the possible mechanisms. Additionally, the impact of AuNCs on the ability of macrophages to uptake bacteria under normal and inflammatory circumstance was also evaluated by fluorescent imaging. We found that AuNCs exhibited superior antibacterial performance on F. nucleatum in vitro by a combination of membrane disruption and oxidative stress. Furthermore, in the presence of AuNCs, macrophages displayed enhanced phagocytosis against F. nucleatum without inducing any significant cytotoxicity. Therefore, AuNCs are providing a new platform for the prevention and treatment of F. nucleatum-associated diseases.

Abstract Background: Recent studies on the urine microbiome have highlighted the importance of the gut-vagina-bladder axis in recurrent cystitis (RC). In particular, the role of Gardnerella as a covert pathogen that activates E. coli in animal experiments has been reported. Herein, we conducted a human bladder microbiome study to investigate the effect of Gardnerella on RC. Methods: Urine 16S ribosomal RNA gene sequencing via transurethral catheterization was conducted in the normal control group (NC) (n=18) and RC group (n=78). Results: The positive detection rate of Gardnerella species did not differ between the NC and RC groups (22.2% vs. 18.0%, p = 0.677). In addition, the Gardnerella-positive NC and Gardnerella-positive RC groups showed similar levels of microbiome diversity. The Gardnerella-positive group was categorized into three subgroups: Escherichia-dominant group, Gardnerella-dominant group, and Lactobacillus-dominant group, respectively. All of the Escherichia-dominant groups were associated with RC. Gardnerella-dominant or Lactobacillus-dominant groups expressed RC with symptoms when risk factors such as degree of Gardnerella proliferation or causative agents of bacterial vaginosis were present.Conclusion: Gardnerella can act as a covert pathogen of RC depending on other risk factors, and Gardnerella infection should be considered in patients with RC. New guideline recommendations regarding antibiotics selection based on a novel method to detect the cause of RC may be required to reduce antibiotics resistance.

AbstractImproper use of the antimicrobials as E. coli giving the microorganism multi-resistance against many antimicrobials by gene mutation on integrons, transposons and plasmids. Therefore, our aim in this study is to 1)examine antibiotics resistance phenotype and genotype in Escherichia coli, 2) identifying the structure of bacterial resistance genes on whole-genome sequencing against multi-drug resistant of Escherichia coli in marketed poultry meat. Samples collected, prepared and Bacteriological examination, Antimicrobial sensitivity test performed, Serological identification of Escherichia coli isolates. Results declared that; the prevalence of E. coli from tested chicken meat samples of 100 chicken meat samples surveyed against E. coli the result declared that about 40%. Antimicrobial susceptibility was; antibiotics of choice against E. coli Sulfonamides, Cephalosporins,Tetracyclines, Quinolones. Serologically, STEC (O157:H7) 30%, ETEC (O142) 10%, EHEC (O26:H11). The subunit B of shiga-like toxin (SLT) gene appeared as a homogenous band. Heat-labile toxin (LT) gene was screened in both genomic DNA and plasmid preps in tested strains. Control STEC as it represents a danger to the poultry consumers. We recommended to increase the hygienic measures during slaughtering, processing and/or handling of chicken carcasses and avoidance unnecessary usage of any antimicrobials to avoid appearance of new antimicrobials resistant.
