Vernonia cinerea, belonging to the family Asteraceae, is of wide medicinal application. This study investigated the antimicrobial, antioxidant activities and the total phenolic contents of wild plant of Vernonia cinerea with its respective shoot cultures.Nodal explants of V. cinerea were cultured on Murashige and Skoog (MS) medium supplemented with various combinations of BAP (0.5-2.5 mg/L) and NAA (0.1-0.5 mg/L) including various concentrations of Gibberllic acid for subculturing. The antimicrobial activity of plant extracts was tested using agar well diffusion and macrodilution methods for zone of inhibition (ZI), minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC).The free radical scavenging test was conducted using 2,2, diphenyl-1-picryl hydrazyl (DPPH) reagent while total phenolic contents (TPC) were measured by Folin–Ciocalteau reagent. Maximum shoot proliferation of 11.42 ± 2.26 cm was obtained from MS medium supplemented with 0.5 mg/L BAP + 0.1 mg/L NAA. All extracts displayed moderate antimicrobial potential against the tested pathogens in the range of 9.0-13.1 mm ZI, with highest MIC of 0.78 mg/mL and 1.56 mg/mL fromMPE and WPE, respectively.The IC50 values of 4.49 and 5.10 μg/mL was obtained in WPE and MPE. The MPE had TPC of 6.66 ± 0.83 mg GAE/g compared to the WPE with TPC of 5.43 ± 1.31 mg GAE/g. The crude methanol extracts of wild and micropropagated plants of V. Cinerea showed high amounts of phenolic compounds, which could present them as candidates for future search for antimicrobial and antioxidant agents for different ailments.Keywords: Micropropagation, Antioxidant, DPPH, Phenolic Compound, Vernonia cinerea

The composition and properties of natural honeys differ with plant species on which the bees forage and the climatic conditions of the production areas. In Ethiopia, Amhara and Tigray are neighboring regions consisting of different agricultural activities and blossoms from different types of vegetations which may influence the natural composition and hence the properties of honey. So, the aim of the current study was to assess the quality of honey from selected districts of the two regions. In the study, 18 composited honey samples were collected from six selected districts and analyzed for selected physicochemical parameters and antioxidant and antimicrobial activities. The analyses of physicochemical parameters were carried out following standard procedures of IHC and QSAE. The antioxidant activity was determined by analyzing the RSA using DPPH while the antibacterial activities were determined by the agar well diffusion method. The moisture, ash content, electrical conductivity, pH, free acidity, reducing sugar, and sucrose content of the honey samples were found to be in the range 16.34 ± 0.26 to 19.83 ± 0.43 %, 0.08 ± 0.00 to 0.45 ± 0.03 %, 0.19 ± 0.00 to 0.89 ± 0.03 mS/cm, 3.79 ± 0.04 to 4.20 ± 0.01, 19.56 ± 1.13 to 38.11 ± 1.54 meq/kg, 62.10 ± 0.48 to 66.37 ± 0.20 %, and 1.35 ± 0.08 to 5.96 ± 0.10 %, respectively. The total phenolic content ranged from 1165.60 ± 23.45 to 1854.83 ± 10.47 mg/kg with antioxidant activity of 21.64 ± 0.26 to 36.12 ± 0.52 AEAC/100 g. The total phenolic contents showed strong correlation with RSA. Furthermore, all honey samples showed an antibacterial activity varying from 23.23 ± 0.12 to 28.84 ± 0.24 mm.

Background: Coffee is the most preferred morning beverage throughout the world due to its pleasant flavor and stimulating properties. It contains a multipart combination of chemicals constituents, which associated with health benefits. In the Kingdom of Saudi Arabia there are many types of coffee in the local markets with different characteristics. Aims: The present study was designed to identify the total phenolic contents, antioxidant properties and antimicrobial activities for three of the most commonly consumed coffee brews; Intenso (Arabica), Reebass (Turkish) and Lavazza (Brazilian) purchased from some local markets at the Northern region in Kingdom of Saudi Arabia. Study Design: It was an in-vitro study. Methodology: The purchased coffee types were extracted using the boiling water method. Determination of total phenolic content of the obtained extracts was carried out using the Folin-Ciocalteu method. Antioxidant activity was measured using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging assay. Measurement of antimicrobial activity was determined by disk diffusion method against number of microbial test strains. The obtained data were statistically analyzed and results with p < 0.05 were considered statistically significant. Results: The obtained results showed that, the estimated phenolic contents were arranged as 745.50 ± 10.5, 668.86 ± 11.2 & 651.25 ± 13.0 mg/g of gallic acid/1 g of coffee for Lavazza (Brazilian), Reebass (Turkish) & Intenso (Arabica) respectively. DPPH radical scavenging activity was higher 78.83% for Turkish coffee than Arabica 71.59% and Brazilian 65.90% types. Regarding antimicrobial activity; Arabica coffee extract was the highest antimicrobial activity compared to the other two extracts of both Turkish and Brazilian types where the mean diameter of inhibition zone for three coffee types were ranged from 11.0 ± 0.15 to 16.0 ± 0.40; 9.0 ± 0.25 to 11.0 ± 0.10 and 9.5 ± 0.10 to 13.0 ± 0.50 mm for Arabica, Turkish and Brazilian coffee extracts respectively. Conclusion: The present study concludes that coffee types under investigation showed high phenolic content and strong antioxidant activity as well as promising antimicrobial activity.

Abstract This study was carried out to determine the phenolic contents as well as to evaluate the antioxidant and antimicrobial activities of Genista microcephala. Total phenolic and flavonoid compounds were quantified. Antioxidant activity was evaluated by various in vitro tests, including Ferric Ion Reducing Activity (FRP), Cuprac Ion Reducing Antioxidant Capacity (CUPRAC), and total antioxidant capacities (TAC). The antimicrobial activity of ethyl acetate and butanolic extracts of G. microcephala in addition of fatty acids content of different parts of the studied plant was evaluated against human pathogenic bacteria, fish bacteria and two yeast. The used methods were agar diffusion method and the determination of minimal bactericidal and fungicidal concentrations (MBC and MFC). No significant difference was found between the two used solvents on total phenolic content (TPC). While, difference in total flavonoid content (TFC) were found to be significant. Ethylene acetate extract was the most actif regarding antioxidant assay. The tested activity seems to be due to the high content of this extract on isoflavonoids. The tested extracts demonstrated the lowest inhibition concentration for B.cereus RSKK 863.
