Microbiological quality of groundwater sources used by rural communities in Limpopo Province, South Africa

A survey of the microbiological quality of water from 194 boreholes (97 privately owned and 97 communal boreholes) in the rural Thitale-Hlanganani area of the Limpopo Province, South Africa was carried out between August 2002 and August 2003. Very little information on the microbiological quality of privately-owned boreholes in rural communities is available raising concerns about the safety of these groundwater supplies. In this study, levels of total coliforms, thermotolerant (faecal) coliforms, faecal enterococci, Clostridium perfringens (vegetative cells and spores) and somatic coliphages were determined for community and privately-owned borehole water. The average counts for total coliforms, faecal coliforms, faecal enterococci and Clostridium perfringens exceeded the South African recommended guideline limits of 0–10 counts.100 ml−1 for total coliforms and 0 counts.100 ml−1 for faecal coliforms, faecal enterococci and Clostridium perfringens respectively. Comparisons between the levels of indicator bacteria present in private and communal boreholes during dry seasons indicated a statistical difference for faecal enterococci bacteria (p=0.005) and Clostridium perfringens (p=0.08). Comparisons between the levels of indicator bacteria present in private and communal boreholes during rainy seasons indicated statistical differences between total coliforms (p=0.05), faecal coliforms (p=0.03) and Clostridium perfringens (p=0.009) bacteria. No significant differences in the presence of somatic coliphages in both private and communal borehole water were see. The results indicated the need for environmental impact assessment studies to monitor the microbiological quality of groundwater sources in rural communities.

Monitoring the microbiological quality of water used for recreational activities is very important to human public health. Although the sanitary quality of recreational marine waters could be evaluated by standard methods, they are time-consuming and need confirmation. For these reasons, faster and more sensitive methods, such as the defined substrate-based technology, have been developed. In the present work, we have compared the standard method of membrane filtration using Tergitol-TTC agar for total coliforms and Escherichia coli, and Slanetz and Bartley agar for enterococci, and the IDEXX defined substrate technology for these faecal pollution indicators to determine the microbiological quality of natural recreational waters. ISO 17994:2004 standard was used to compare these methods. The IDEXX for total coliforms and E. coli, Colilert®, showed higher values than those obtained by the standard method. Enterolert® test, for the enumeration of enterococci, showed lower values when compared with the standard method. It may be concluded that more studies to evaluate the precision and accuracy of the rapid tests are required in order to apply them for routine monitoring of marine and freshwater recreational bathing areas. The main advantages of these methods are that they are more specific, feasible and simpler than the standard methodology.

There are problems associated with sludge management in small treatment plants (<10 L/s) located in rural communities, due to costly conventional technology for sludge stabilization. Many of these plants have only sludge drying beds. Mexican Institute of Water Technology has proposed developing suitable low-cost technologies, one of which is vermicomposting a biodegradation system using earthworms of the species Eisenia foetida (earthworm) which stabilize sludge and reduce its pathogenicity. The objective of this work is to present two case studies where vermicomposting technology has been applied in Mexico. The first study corresponds to a plant where 4.8 m3/month of sludge are produced; for these wastes, a vermicomposting system was built and installed. The second study is a treatment plant where 9 m3/month of sludge are produced; experimental tests were conducted with sludge and water hyacinth and a vermicomposting system was designed. The vermicomposts were analyzed using parameters defined by Mexican standards. In regards to stabilization, TVS was reduced by 38% and the microbiological quality of the vermicompost was Class A and B, with a reduction in fecal coliforms and Helminth eggs according to NOM-004-SEMARNAT-2002. A CRETI (Corrosivity, Reactivity, Explosivity, Toxicity and Ignitability) analysis (NOM-052-SEMARNAT-2005) was used to show that the process reduced the concentration of releasable sulfides. The agronomic quality of the vermicompost exhibited a high content of organic matter comparable to many organic manures and high content of nutrients such as nitrogen and phosphorus. It is concluded that it is possible to improve the conditions of sewage sludge management in small plants of rural communities with a minimum investment (less than $10,000.00 USD) and with a requirement of a minimum area of 60 to 70 m2 for a production of less than 9 m3/month of dehydrated sludge (80% humidity).

This article looked at the quantity and storage time of attiéké produced and sold in the town of Daloa. It also examines the microbiological characteristics of attiéké-femme and attiéké-garba from this locality during storage. The study was carried out on the one hand through a field survey carried out on the producers and sellers. On the other hand, a sample was taken from 10 sellers of attiéké-garba and 10 sellers of attiéké-woman. The study found that the women producers sell 87% of their production in the city of Daloa and export 13%. In addition, attiéké can be kept for 2 days at the producers and beyond 2 days at the sellers before their stock runs out. Therefore, a weekly production of more than 200 kg for the majority of the producers is observed. Microbiological analyses showed high levels of germs (MAG: 6.106 CFU/g; Yeasts and moulds: 2.7.106 CFU/g) for attiéké-women and (2.106 CFU/g of GAM and 1.6.103 CFU/g of Yeasts and moulds) for attiéké-garba. Total coliforms and faecal coliforms were only found in attiéké-women. No salmonella was observed. In view of the results, it should be noted that female attiéké is the most contaminated type of attiéké.

A survey of the general microbiological quality of ready-to-eat food served in schools was undertaken across Wales, United Kingdom. Of the 2,351 samples taken, four were identified as containing unsatisfactory counts of Escherichia coli, four contained unsatisfactory counts of Staphylococcus aureus, and one contained an unacceptable count of Bacillus cereus when compared with guidelines for the microbiological quality of ready-to-eat food published by the United Kingdom Public Health Laboratory Service in 2000. No samples contained detectable levels of Salmonella, Listeria species, or Clostridium perfringens. When compared with data on the general microbiological quality of food available in Wales, the food sampled from schools was of relatively better microbiological quality.

The objective of the present study was to determine the microbiological quality of bottled mineral water marketed in commercial establishments and by street vendors and to evaluate the influence of the storage and maintenance conditions on the microbiological quality of the product. Ten samples from the same batches of five different brands of water were analyzed, for a total of 50 samples. Of the five brands analyzed, only one (brand A), when collected in a commercial establishment, complied with the legal Brazilian standards for mineral water with respect to the presence of total coliforms, fecal coliforms, and Pseudomonas aeruginosa. The remaining samples failed to comply with these microbiological standards for at least one of the parameters evaluated. The water samples obtained from street vendors were inferior in microbiological quality to samples from the same batch that were obtained from commercial establishments.

The objective of this study was to evaluate the microbiological quality of oysters and mussels grown in Cananéia, Brazil, by analysing mesophiles, psychrothophic bacteria, moulds and yeasts, Staphylococcus aureus, and Salmonella spp., and to compare the efficiency of Compact Dry EC method and the conventional method for counting of total coliforms and Escherichia coli. The microbial analysis showed that the mean values of mesophilic counts were 3.14±0.81 log CFU g−1 for oysters and 3.92±0.90 for mussels; the mean values of psychrophilic counts were 2.78±0.75 log CFU g−1 for oysters and 3.22±0.75 log CFU g−1 for mussels; the mean values of mould and yeast counts were 3.70±0.58 log CFU g−1 for oysters and 3.33±0.81 log CFU g−1 for mussels. Salmonella spp. did not present positive results, and the maximal count of Staphylococcus aureus was 1.7 log CFU g−1, therefore, within the limits established in the legislation. The correlation coefficients between the Compact Dry EC method and conventional method were >0.87 for total coliform and E. coli counts for both types of shellfish. The data in this study show that the Compact Dry EC method is an acceptable alternative to conventional methods for enumeration of total coliforms and E. coli in shellfish.

Objective of the study: to evaluate the microbiological quality of lettuce (Lactuca sativa L.) in three different cultivation: organic, hydroponic and traditional. Sixty lettuce samples commercialized in Videira – SC were analyzed. Microorganisms investigated: Salmonella spp., Listeria spp., total coliforms e Escherichia coli. The results showed the absence of Salmonella spp. for all samples. Present of Listeria spp. was found in 16,7% of the samples. Total coliforms e Escherichia coli count varied from <1x101 - 2,8x105 and <1x101 - 1x103, respectively. Traditional and hydroponic farming lettuce samples presented 100% of total coliforms. Escherichia coli analysis occurred in 22,2% of the samples traditional farming, 11,1% hydroponic farming and none in organic farming vegetables. Statistically, we found no significant relation between microbiological contamination and king of growing used in the vegetables production. The strains of the Listeria, total coliforms and Escherichia coli found in the samples indicated hygienic and sanitary quality and so it is important that good hygiene practices are taken from the manufacturing process until the moment of consumption.

The present work aims to assess the physico-chemical and microbiological parameters of the waters of the three sources (Ain Bergougaya, Ain Sigleb and Siporex) in the region of Oum el Teboul wilaya of El Tarf. Parameters such as temperature, pH, conductivity and salinity were measured in situ with a field multimeter. Hardness (62 mg/L to 266 mg/L), salinity (0.01 to 1 mg/L). From a microbiological point of view, there has been no evidence of the complete absence of germs of contamination such as total coliforms and fecal coliforms. This study compared to the drinking water standards shows that the waters in the area studied are of good quality.

Foram avaliadas as condições higiênico-sanitárias de lingüiças mistas do tipo frescal, provenientes de 4 frigoríficos submetidos à inspeção sanitária estadual, e comercializadas em Pelotas (RS). Os resultados obtidos foram interpretados pela legislação vigente (RDC no 12/01) e pela anterior (Portaria 451), ambas do Ministério da Saúde (Brasil). Verificou-se que 17,86% das 32 amostras estavam contaminadas com Salmonella sp e 3,57% apresentaram Coliformes fecais acima dos parâmetros permitidos pela legislação em vigor. Com a alteração da legislação, a probabilidade de se encontrar amostras de lingüiça mista frescal contaminada por Coliformes fecais passou de 23,27% para 5,6%. MICROBIOLOGICAL QUALITY OF MIXING SAUSAGES OF THE FRESCAL TYPE PRODUCED IN PELOTAS - RS Abstract The hygienic-sanitary conditions of mixing sausages of the frescal type were evaluated, proceeding from 4 slaughterhouses with state sanitary inspection and commercialized in Pelotas (RS). The results were interpreted by the current legislation or the previous one, both of the Brazilian Health Ministry, in the interpretation of the results. It was verified that, 17,86% of the 32 samples were contaminated with Salmonella sp and 3.57% presented faecal coliforms above of the parameter allowed by the current legislation. With the change of the legislation, the probability to find samples of frescal mixing sausage with countings of faecal coliforms above of the maximum limits allowed by the legislation, passed of 23,27% to 5,6%.

An evaluation of the sanitary quality of the sand beaches of Azores archipelago was undertaken May-October 1994 and 1995. The samples were analysed for total and faecal coliforms, faecal enterococci, Clostridium, Pseudomonas aeruginosa and fungi (keratinolytic, yeasts, potential pathogenic and allergic and/or environmental saprophytes). The results were evaluated according to a “Proposal Guideline for the Microbiological Quality of Sand”. The data showed good or satisfactory sanitary quality of the sand beaches. The results indicated that the faecal enterococci can be a better indicator organism than the total and faecal coliforms. The fungi could be used as indicator organisms. The application of the “Proposal” should complete the criteria used in the “Blue Flag” distinction for high environmental quality beaches.
