Comparison of extraction efficiency of various methods to extract L-DOPA from Mucuna pruriens (L.) DC.

Mucuna pruriens seeds are noted to be a natural source of L-DOPA and are also used as a substitute for the synthetic L-DOPA. In the present study; attempts are made to develop suitable method(s) for extraction of L-DOPA from the powdered seeds of Mucuna pruriens using different solvents and conditions. The Seed powder was subjected to 7 different extraction methods and Method 1 was subjected to various solvent concentrations. Some methods used de-fatting procedure, either the method was cold maceration or in high temperature. Soxhlet extraction was also used in one of the extraction methods. All the extracts were analyzed using RP-HPLC. Mobile Phase used was Water: Methanol: AcetoNitrile (100:60:40) (v/v) containing 0.2% Triethylamine, pH = 3.3 and monitored at 280 nm with variable wavelength UV detector. The extraction was best with Methanol Water mixture in a cold maceration technique and overall gives good extraction efficiency of 13.36 % L-DOPA and id the best method giving highest extraction efficiency. The De-fatting method was the 2nd best methods giving approximately 8.8% L-DOPA and Method 5 viz, heat reflux method gives 8.7% L-DOPA making it the 3rd best method. There are not many studies done for optimization of extraction technique for L-DOPA despite an extensive work is reported for isolation, identification and pharmacological activities of L-DOPA from various plant sources. Keeping this in view, present investigation was done to study the extraction efficiency of various extraction methods of L-DOPA content in seed extracts of Mucuna pruriens and compare it.

In the present study, attempts are made to develop suitable method(s) for extraction of L-DOPA from the powdered seeds of 3 species of Mucuna using different solvents and conditions. The seed powder of both plants was subjected to 6 different extraction methods, with different solvent ratios. All the extracts were analyzed using RP-HPLC and was validated according to The International Conference on Harmonisation of Technical Requirements for Registration of Pharmaceuticals for Human Use guidelines. The L-DOPA extraction was best with Methanol Water mixture in a cold maceration technique and overall gives good extraction efficiency in all the three plants giving concentrations of 5.03%, 13.36 % and 16.78% of L-DOPA in Mucuna gigantea, Mucuna pruriens and Mucuna monosperma, respectively. The present investigation was done to study the extraction efficiency of various extraction methods of L-DOPA content in seed extracts of Mucuna and compare it.

To raise the extraction efficiency of the organic components of coal, samples were selected from tectonic coal and original bituminous coal in symbiotic seam of coal in Pingdingshan 8th coal mine and 5th coal mine. For comparing the extraction time required under the same extraction rate, two different extraction methods, ultrasonic-microwave coordination extraction and traditional soxhlet extraction, was adopted. The experimental results showed that the extraction proportion of ultrasonic-microwave coordination extraction was 10.7911% when adopted tectonic coal of 8th coal mine as sample, selected 80mL pyridine as solvent, set 95W as microwave power and 3min as extraction time. Under the condition of same extraction proportion, soxhlet extraction time is 39.43h.So the extraction efficiency of the ultrasonic-microwave coordination extraction was as 788.6 times as soxhlet extraction. The ultrasonic-microwave coordination extraction was a simplify method with fast extraction speed, large volume sample, energy saving and environmental friendly.

AbstractThe purpose of this review is to describe and compare selected traditional and modern extraction methods employed in the isolation of isoflavonoids from plants. Conventional methods such as maceration, percolation, or Soxhlet extraction are still frequently used in phytochemical analysis. Despite their flexibility, traditional extraction techniques have significant drawbacks, including the need for a significant investment of time, energy, and starting material, and a requirement for large amounts of potentially toxic solvents. Moreover, these techniques are difficult to automate, produce considerable amount of waste and pose a risk of degradation of thermolabile compounds. Modern extraction methods, such as: ultrasound-assisted extraction, microwave-assisted extraction, accelerated solvent extraction, supercritical fluid extraction, and negative pressure cavitation extraction, can be regarded as remedies for the aforementioned problems. This manuscript discusses the use of the most relevant extraction techniques in the process of isolation of isoflavonoids, secondary metabolites that have been found to have a plethora of biological and pharmacological activities.

Five methods were used for the extraction of hexachlorobutadiene and chlorobenzenes from a contaminated estuarine sediment. The following extraction methods were used: Soxhlet extraction, sonication and solvent extraction, sequential solvent extraction, saponification and solvent extraction, and supercritical fluid extraction. Soxhlet extraction resulted in better contaminant extraction efficiency and reproducibility. Most of the problems associated with the extraction methods were related to high water content and the inherent heterogeneity of the sediment samples. Drying and grinding of the sample greatly improved both extraction efficiency and reproducibility. Analyses of wet sieved and fractionated sediment samples indicated that the contaminants are not evenly distributed throughout the sediment organic fraction but rather predominate in the larger particle sediment fraction. The implications of quantification difficulties of sediment contaminants on the environmental fate and hazard assessment are discussed.

Background: The official compendium of the quantification of ginkgo flavonoids from Ginkgo biloba extract has been proposed using HPLC. The drawbacks of this technique appear to be due to the restricted efficiency in terms of the recovery results and suitability of the system for the quantification of these compounds. This study investigated the potential advantages and limitations of the development of efficient extraction methods for the recovery of flavonol glycosides (quercetin, kaempferol and isorhamnetin) and terpene trilactones (bilobalide, ginkgolide A, ginkgolide B and ginkgolide C) using extraction, quantification and detection techniques, namely, GC-FID and UHPLC-DAD, which are alternatives to those techniques available in the literature. Methods: Two different extraction methodologies have been developed for the determination of flavonoids (quercetin, kaempferol and isorhamnetin) and terpene trilactones (bilobalide, ginkgolide A, ginkgolide B and ginkgolide C) using ultra-high-pressure liquid chromatography coupled to a diode array detector and gas chromatography coupled to a flame ionization detector. Results: In this study, the Ginkgo biloba extract mass, hydrolysis preparation method (with or without reflux), and volume of the extraction solution seemed to affect the ginkgo flavonoid recovery. The UHPLC-based method exhibited higher extraction efficiency for ginkgo flavonoid quantification compared to the pharmacopoeial method. The developed method exhibited higher extraction efficiency for terpene quantification compared to the previous method that used extractive solution without pH adjustment, with less time of extraction and less amount of the sample and organic solvent aliquots. Conclusion: The UHPLC and GC analysis methods established in this study are both effective and efficient. These methods may improve the quality control procedures for ginkgo extract and commercial products available in today´s natural health product market. The results indicate that redeveloped extraction methods can be a viable alternative to traditional extraction methods.

Background: Eugenol is the main constituent of clove essential oil. Past studies have found that clove oil has diverse uses in the pharmaceutical field due to its antioxidant, antibacterial and anesthetic properties. Objective: This work compares the performance of different extraction methods and factors and identifies the effect of the treatments on oil yields and eugenol content. Materials and Methods: Maceration, Hydro distillation, microwave-assisted extraction (MAE), and Soxhlet were performed. The best technique was identified according to yield and content. Further studies were conducted to examine the effects of different factors, such as solvent types (ethanol and methanol) and sample-to-solvent ratio (1:10 and 1:15). HPLC UV-Vis was utilized in the analysis of eugenol concentration. Results and Discussion: Soxhlet extraction provided the highest yield (39.98%) and eugenol content (15.83%), compared to other methods. The results observed from several Soxhlet extraction factors showed that there is no significant difference between the different factors. In the meantime, methanol 1:15 provided the greatest amount of yields (57.83%) and eugenol content (22.21%). In this regard, the higher ratio resulted in higher eugenol content. Conclusion: The results obtained are less comparable because the processing time, the working solvent, and the separation technique were carried out differently for each method. In the meantime, as there is no past study that compared the selected methods and factors, this study’s findings will contribute substantially to fill the gap in this field.

Objective: The aim was to study the antiparkinson activity in the seed extracts of four species of Mucuna.Methods: The hydroalcoholic extracts of seeds of four species of Mucuna were evaluated for antiparkinson activity of after a preliminary phytochemical study. The activity was measured in rats by indirectly measuring the decrease in malondialdehyde level, decrease in tongue protrusion frequency, and reduction in vacuous chewing movement after administering reserpine at the dose of 1 mg/kg. The dose levels of four species of Mucuna seed extract were kept at 100, 200, and 300 mg/Kg.Results: Extracts exhibited potent antiparkinson activity and achieved statistically significant p values compared with control group. The study corroborates and compares all four species of Mucuna. Conclusion: Among the extracts, the highest percentage of antiparkinson activity was recorded for Mucuna pruriens.

The aim of this research is to know the extraction method of caterpillar pupa without detrimental to chemical composition to know the amount of pupa production of German caterpillar with two extraction methods per 100 grams of pupa To know the content of dry matter, organic matter, crude protein, crude fat, crude fiber and amino acid lysin, methionine, and vitamin E pupa oil of German caterpillar. The study was conducted from February to April 2017, at the Lab. Central University Tribhuwana Tunggadewi. Oil extraction analysis was conducted in the chemistry laboratory of Tribhuwana Tunggadewi University and chemistry laboratory of Poltek Malang. German caterpillar research material, German caterpillar pupa, pupa caterpillar germany extracted by maceration and sokletasi. German caterpillar pupa is obtained from German caterpillar breeders in Amadanom, Dampit, Malang Regency. This research uses two different extraction methods, namely cold extraction method (maseration method) and using heat method (soxhlet method). The conclusions of both research methods are known that the quantity of German caterpillar yield using soxhlet extraction method is more than the maceration extraction method. However, for the quality of the two methods it is still necessary to do further analysis to know the exact comparison of the two methods.
