This study aims to determine the type and potency of egg parasitoid in controlling brown planthopper (Nilaparvata lugens Stal.) pests in Banyumas regency after the pest’s explosion. This research has been conducted in five districts of the endemic area of brown planthopper in Banyumas Regency, i.e. in  Jatilawang, Cilongok, Kebasen, Sumpiuh, and Kembaran. Each of the  districts was taken 5 sample villages. The testing and calculation of the level of parasitization  were done in the laboratory of Plant Protection, Faculty of Agriculture, University of Jenderal Soedirman, Purwokerto. The research  used nest plot design, where  the first factor was the district and the second factor was the village. Village nested in  district. The data were  analyzed using F 5% test,  followed by 5% DMRT, if there were  any differences found . The results of the study were as follows: (1) The parasitoids found in Banyumas Regency were Gonatocerus sp. and Oligosita sp. with the ability to parasite 26.8−64.73%, and 1.82−31.40%; (2) the presence parasitoid has the potency  to suppress the intensity of brown planthopper attack on the vegetative phase, the intensity of attacks ranged between 6.96−23.58%, with brown planthopper population ranged from 0.84 to 27.36 individuals per hill. IntisariPenelitian ini bertujuan untuk mengetahui jenis dan potensi parasitoid telur dalam mengendalikan hama wereng batang coklat (Nilaparvata lugens Stal.) di Kabupaten Banyumas pasca terjadinya ledakan. Penelitian ini dilaksanakan di lima kecamatan daerah endemik wereng batang cokelat di wilayah Kabupaten Banyumas yaitu Kecamatan Jatilawang, Cilongok, Kebasen, Sumpiuh, Kembaran. Masing-masing kecamatan diambil 5 desa sampel. Pengujian dan penghitungan tingkat pemarasitan dilakukan di laboratorium Perlindungan Tanaman Fakultas Pertanian, Universitas Jenderal Soedirman, Purwokerto. Penelitian menggunakan rancangan petak tersarang,  dengan kecamatan sebagai faktor pertama  dan desa sebagai faktor kedua. Desa tersarang pada kecamatan. Data dianalisis menggunakan uji F 5%, apabila ada perbedaan dilanjutkan dengan uji banding ganda DMRT 5 %. Adapun hasil penelitian adalah sebagai berikut: (1) Parasitoid yang ditemukan di Kabupaten Banyumas adalah Gonatocerus sp. dan Oligosita sp. dengan kemampuan memarasit 26,8−64,73%, dan sebesar 1,82−31,40 %, (2) keberadaan parasitoid berpotensi menekan intensitas serangan hama wereng batang cokelat pada fase vegetatif, intensitas serangan berkisar antara 6,96−23,58%, dengan populasi wereng batang cokelat berkisar 0,84−27,36 individu per rumpun.

ABSTRACTPopulation of Brown Plant Hopper (Nilaparvata lugens Stal.) and the Diversity of Its Natural Enemyin Highland Paddy Rice Field in the Village of Panyocokan, Ciwidey District, Bandung RegencyBrown lanthopper/BPH (Nilaparvata lugens Stal.) is a main pest of rice in the field. This researchwas aimed to study the population of BPH and the diversity of its natural enemy in highland paddyrice field. The survey was conducted in three paddy plots (15 m x 20 m) located in the village ofPanyocokan, Ciwidey District, Bandung Regency, West Java. The experiment was conducted in thegreenhouse of the Department of Plant Pests and Diseases, Faculty of Agriculture, UniversitasPadjadjaran. The samples of insects were collected systematically using yellow trap and net trap,and visually observed in each rice cluster. The number of BPH were recorded and accumulatedeach week. Fecundity, life cycle, and sex ratio of the new generation of BPH were observed in 3replications by using 1 pair of WBC on each replication. The result showed that the population ofBPH in the highland was below 10, which means that BPH population was still below theeconomic threshold and control threshold. Temperature, humidity, and rainfall did notsignificantly affect the BPH population. This was indicated by the regression analysis of eachtemperature (Y = 0.557-8.167x; R2 = 0.039; P = 0.465), humidity (Y = -0.077+ 9.112x; R2 = 0.045; P =0.428), and rainfall (Y = -0.118 + 3.412x; R2 = 0.136; P = 0.159). Natural enemy diversity indextended to fluctuate widely from low to moderate. In the temperature range from 21.1°C to 34.8°C,BPH could produce 127-207 new generation during its lifetime. BPH needed an average of 37.66days to produce a new generation. Penelitian dilakukan dengan metode survei dan eksperimen.Survei dilakukan pada 3 petak lahan percobaan berukuran 15 mx 20 m bertempat di DesaPanyocokan, Kecamatan Ciwidey, Kabupaten Bandung dan eksperimen dilakukan di rumah kacaDepartemen Hama dan Penyakit Tumbuhan, Fakultas Pertanian, Universitas Padjadjaran. Metodepengambilan sampel dilakukan secara diagonal sistematis. Populasi WBC ditentukan denganmenggunakan perangkap kuning dan perangkap jaring dan dengan pengamatan langsung padarumpun padi. Eksperimen dilakukan dengan meletakkan sepasang WBC dalam wadah, lalu diamatikeperidian, siklus hidup, dan sex ratio keturunannya. WBC yang tertangkap dihitung jumlahnyadan diakumulasikan setiap minggunya. Hasil penelitian menunjukkan bahwa kepadatan populasi dilahan survey di bawah 10 ekor/rumpun yang artinya populasi WBC masih di bawah ambangekonomi atau ambang kendali. Suhu, kelembaban, dan curah hujan tidak memberikan pengaruhyang signifikan terhadap populasi WBC. Hal tersebut ditunjukkan dengan analisis regresi masingmasingpada suhu (Y= 0,557 – 8,167x; R2 = 0,039; P= 0,465), kelembaban (Y = -0,077 + 9,112x; R2 =0,045; P = 0,428), dan curah hujan (Y = -0,118 + 3,412x; R2 = 0,136; P = 0,159). Indeks keragamanmusuh alami cenderung mengalami fluktuasi dari rendah sampai sedang. Pada kisaran suhu 21,1oC–34,8oC, hasil pengamatan keperidian menunjukkan WBC dapat menghasilkan 127-207 individubaru selama masa hidupnya. Pada pengamatan siklus hidup, WBC memerlukan rata-rata 37,66 harisampai menghasilkan generasi baru. Anagrus nilaparvatae is a potential egg parasitoid to control the rice brown planthopper (Nilaparvata lugens Stal.) The parasitoid needs to consume suitable food to maximize its biotic potential and parasitizing ability. This study was aimed to determine the benefits provided by the presence of Turnera subulata and Cosmos sulphureus flowers on the life of A. nilaparvatae. This study consisted of two experiments. The first experiment was designed to determine the effects of the tested flowers on the parasitism and hatching rate of A. nilaparvatae on N. lugens eggs. The flowers were set inside the rearing cage of parasitoid in the presence of N. lugens eggs in Ciherang rice seedlings. In addition, honey and the control (no feed added) were included into the treatments, totalling of four treatments. The parasitism rate of A. nilaparvatae fed with the flowers or honey was similar to those unfed. However, the hatching rate of A. nilaparvatae was much higher on those fed with flower or honey than those unfed. The number of A. nilaparvatae unable to eclose from eggs of N. lugens for the unfed parasitoid was 37.4% in comparison with 8.19 to 15.67% for those fed with flower or honey. The second experiment was a follow-up to address the question on the fitness of progeny of A. nilaparvatae fed with the tested flowers. The flowers and honey did not increase the longevity of A. nilaparvatae progeny. However, A. nilaparvatae fed with flowers or honey produced progeny that resulted in higher number of offspring compared to those unfed. C. sulphureus flower significantly increased the number of offspring produced by A. nilaparvatae. This suggest that the diet of the parental parasitoid determines the fitness of the progeny. Improving the hatching rate and the fecundity of progeny produced by the adults of A. nilaparvatae fed with the flower of T. subulata and C. sulphureus would contribute to the increasing population of this parasitoid which could lead to a better control of N. lugens in the rice field. IntisariAnagrus nilaparvatae merupakan parasitoid telur yang potensial untuk mengendalikan serangan wereng batang cokelat (Nilaparvata lugens Stal.). Parasitoid perlu mendapatkan sumber pakan yang sesuai untuk memaksimalkan potensi biologis dan kemampuan memarasit inang. Penelitian ini bertujuan untuk mengkaji manfaat pemberian bunga Turnera subulata dan Cosmos sulphureus pada kehidupan A. nilaparvatae. Penelitian ini terdiri dari dua percobaan. Percobaan pertama dirancang untuk mengetahui pengaruh bunga yang diuji terhadap tingkat parasitasi dan penetasan telur A. nilaparvatae pada telur N. lugens. Bunga diletakkan di dalam tabung rearing parasitoid yang di dalamnya terdapat bibit padi varietas Ciherang yang mengandung telur N. lugens. Selain itu, terdapat pula perlakuan madu dan kontrol (tanpa bunga dan tanpa madu), dengan total empat perlakuan. A. nilaparvate yang diberi pakan bunga atau madu memiliki tingkat parasitasi yang sama dengan yang tidak diberi pakan. Namun, tingkat penetasan telur A. nilaparvatae yang diberi pakan bunga atau madu jauh lebih tinggi daripada yang tidak diberi pakan. Jumlah A. nilaparvatae yang tidak menetas dari telur N. lugens pada parasitoid yang tidak diberi pakan adalah 37,4% dibandingkan dengan parasitoid yang diberi pakan bunga atau madu yang berkisar 8,19 - 15,67%. Percobaan kedua adalah tindak lanjut untuk menjawab pertanyaan tentang kebugaran keturunan A. nilaparvatae yang diberi pakan dengan bunga yang diuji. Bunga dan madu tidak meningkatkan lama hidup keturunan A. nilaparvatae. Namun, A. nilaparvatae yang diberi pakan dengan bunga atau madu menghasilkan keturunan yang memiliki fekunditas lebih tinggi dibandingkan dengan keturunan dari A. nilaparvatae yang tidak diberi pakan. Bunga C. sulphureus secara signifikan mampu meningkatkan jumlah keturunan yang dihasilkan oleh A. nilaparvatae. Ini menunjukkan bahwa jenis pakan induk parasitoid menentukan kebugaran keturunannya. Peningkatan penetasan dan fekunditas dari keturunan yang dihasilkan oleh induk A. nilaparvatae yang diberi pakan bunga T. subulata dan C. sulphureus akan berkontribusi pada peningkatan populasi parasitoid sehingga dapat mengendalikan serangan N. lugens di pertanaman padi dengan lebih baik.

An extensive study about egg parasitoid biodiversity in different plant growth stages is an important need. This study was aimed to determine the diversity and abundance of egg parasitoids Nilaparvata lugens on different rice growth stages in Saketi, Pandeglang, Banten. The parasitoids sample was collected three times with two days interval sampling in three villages, i.e. Sodong (location I), Sindanghayu (location II), and Sukalangu (location III) from February to September 2018. Ten plants (as bait: rice that infected by N. lugens eggs) were put during two days along the transect line (100 meters) with 10 m space in between. The findings revealed that the abundance richness (S), the diversity index (H'), and similarity (J') of egg parasitoids of N. lugens were higher in the location I than those in the locations II and III. The study indicated that the diversity and abundance of egg parasitoids of N. lugens decreased in the generative phase of rice growth.

Brown planthopper (BPH) is one of the important pest insects that attack rice plants, which indirectly becomes a vector for the spread of grassy dwarf and empty dwarf diseases caused by viruses, and can consume a lot of food in a short time so that it can cause damage explosions and great losses. The aim of this study was to compare the eating response of brown planthopper to resistant and vulnerable rice varieties through the measurement of secreted honeydews. The study was conducted at the Toxicology Laboratory, Department of Plant Protection, Faculty of Agriculture, Bogor Agricultural University. Rice seedlings for honeydew testing were 30 days after sowing. Testing was done by infesting 3 female BPH into a plastic cage with filter paper (Whatman No.40 9 cm in diameter) placed on the bottom, which had been sprayed with a solution of Ninhydrin 0.01 mg/ml in acetone. The treatment consisted of 3 rice varieties (Ciherang, IR-64 dan Pelita) on each ovened and non-ovened filter paper, with three replicates so that a total of 18 treatments were used by infesting each with the 3 female BPH nymphs. Whereas, 3 treatments with non-ovened papers, with 3 replicates, were infested with 3 female BPH imago each. So that the treatments involved a total of 27 feeding tubes. Honeydews released by BPH that ate on the test varieties for 24 hours were collected on filter papers and formed blue/purple spots. The extent of the honeydew spots formed was measured and analyzed. The results showed that the average value of phloem consumption in Ciherang, IR-64, and Pelita rice varieties showed that these three varieties were very susceptible to BPH sucking pests. The more food nutrients sucked by BPH pests from rice plants, the greater the number of honeydew spots and the wider the honeydew spots that were produced. The areas of honeydew spots of Pelita variety was 33.78 m2, IR-64 28.26 mm2, and Ciherang 22.44 mm2, respectively. Pelita rice varieties had a high susceptibility to BPH pests when compared to Ciherang and IR-64 because Pelita does not have resistance genes to BPH pests. Keywords: Ciherang, IR-64, Pelita, planthopper, rice, honeydew   ABSTRAK Wereng batang coklat (WBC) merupakan salah satu serangga hama penting yang menyerang tanaman padi, yang secara tidak langsung menjadi vektor bagi penyebaran penyakit kerdil rumput dan kerdil hampa oleh virus, serta dapat mengkonsumsi makanan yang banyak dalam waktu singkat sehingga dapat menimbulkan ledakan kerusakan dan kerugian yang besar. Penelitian ini bertujuan untuk membandingkan respon makan WBC pada varietas padi tahan dan rentan melalui pengukuran embun madu yang disekresikan. Penelitian dilaksanakan di Laboratorium Toksikologi, Departemen Proteksi Tanaman Fakultas Pertanian, Institut Pertanian Bogor. Bibit padi untuk pengujian embun madu digunakan saat berumur 30 hari setelah semai. Pengujian dengan menginfestasikan WBC betina ke dalam kurungan plastik yang di bagian bawahnya diletakkan kertas saring (Whatman No. 40 berdiameter 9 cm) yang telah disemprot dengan larutan Ninhidrin 0,01 mg/mL aseton. Perlakuan yang terdiri dari 3 varietas padi (Ciherang, IR-64 dan Pelita) pada masing-masing kertas saring oven dan tidak oven dan dengan tiga ulangan sehingga total terdapat 18 perlakuan dengan menginfestasikan masing-masing 3 ekor nimfa betina WBC. Sedangkan pada 3 perlakuan kertas saring tanpa oven, dengan tiga ulangan, yang diinfestasikan masing-masing berupa 3 ekor imago betina WBC. Sehingga total perlakuan seluruhnya meliputi sebanyak 27 tabung makan. Embun madu yang dikeluarkan oleh WBC yang makan pada varietas uji selama 24 jam tertampung pada kertas saring dan membentuk bercak berwarna biru/ungu. Luas bercak embun madu yang terbentuk diukur dan dianalisis. Hasil penelitian menunjukkan bahwa nilai rata-rata tingkat konsumsi floem pada varietas padi Ciherang, IR-64 dan Pelita, menunjukkan bahwa ketiga varietas ini sangat rentan terhadap hama pengisap WBC. Semakin banyak nutrisi makanan yang disedot oleh hama WBC dari tanaman padi, maka semakin banyak jumlah spot bercak honeydew dan semakin luas spot bercak honeydew yang dihasilkan. Luas spot untuk varietas Pelita adalah 33,78 m2, IR-64 28,26 mm2, dan Ciherang 22,44 mm2. Varietas padi Pelita memiliki kerentanan yang tinggi terhadap hama WBC, bila dibandingkan dengan varietas Ciherang dan IR-64, karena varietas Pelita ini tidak memiliki gen ketahanan terhadap hama WBC. AbstractThe brown planthopper Nilaparvata lugens is a serious phloem-feeding pest of rice in China. The current study focuses on a saccharopine dehydrogenase (SDH) that catalyzes the penultimate reaction in biosynthesis of the amino acid lysine (Lys), which plays a role in insect growth and carnitine production (as a substrate). The protein, provisionally designated as NlylsSDH [a SDH derived from yeast-like symbiont (YLS) in N. lugens], had a higher transcript level in abdomens, compared with heads, wings, legs and thoraces, which agrees with YLS distribution in N. lugens. Ingestion of Nlylssdh targeted double-stranded RNA (dsNlylssdh) for 5, 10 and 15 days decreased the mRNA abundance in the hoppers by 47, 70 and 31%, respectively, comparing with those ingesting normal or dsegfp diets. Nlylssdh knockdown slightly decreased the body weights, significantly delayed the development of females, and killed approximately 30% of the nymphs. Moreover, some surviving adults showed two apparent phenotypic defects: wing deformation and nymphal cuticles remained on tips of the legs and abdomens. The brachypterours/macropterours and sex ratios (female/male) of the adults on the dsRNA diet were lowered compared with the adults on diets without dsRNA. These results suggest that Nlylssdh encodes a functional SDH protein. The adverse effect of Nlylssdh knockdown on N. lugens implies the importance of Lys in hopper development. This study provides a proof of concept example that Nlylssdh could serve as a possible dsRNA-based pesticide for planthopper control.
