Objective: This study evaluated the Chaetomium cupreum extracts as biocontrol agents against four plant pathogenic fungi (Cladosporium cladosporioides, Fusarium oxysporum, Phomopsis azadirachtae, and Rhizoctonia solani).Method: The antifungal activity of n-butanol and ethyl acetate extracts of C. cupreum was evaluated against plant pathogenic fungi using food poison method.Result: In n-butanol extract, the percentage inhibition of mycelial growth against C. cladosporoides was 88.3±0.1, F. oxysporum was 59.4±0.2, R. solani was 56.2±0.9, and P. azadirachtae was 52.0±0.1at 0.25 mg/ml, respectively. In ethyl acetate extract, the percentage inhibition of mycelial growth against C. cladosporoides was 86.0±0.5, F. oxysporum was 66.4±0.1, P. azadirachtae was 55.2±0.9, and R. solani was 52.0±0.1 at 0.25 mg/ml, respectively.Conclusion: It was found that n-butanol extract is more effective than ethyl acetate extract of C. cupreum. Future studies will focus on the purification and characterization of compounds of C. cupreum and their biocontrol capacity with the mechanism for plant pathological applications.

Background: In recent years, the study of the structure and biological activity of medicinal plants has a particularly important to search for diabetes medicine. Ruellia tuberosa is used to treat various diseases such as diabetes by inhibiting the activity of α-glucosidase. Objective: In this study, experiment was designed to isolated isolate and identified identify α-glucosidase inhibitory extracts and compounds from Ruellia tuberosa L. through bio-assay guided isolation. Method: Dry powder of Ruellia tuberosa L. was extracted with 70% ethanol, then liquid-liquid partition with n-hexane, ethyl acetate and butanol, respectively. The extracts were evaluated with α-glucosidase inhibition. The potential extracts were isolated and identified compounds. The effects of these compounds on the α-glucosidase inhibitory were evaluated. Results: The a-glucosidase inhibitory activities showed that the n-hexane, ethyl acetate and the butanol extract had the α-glucosidase inhibition with an IC50 of 46.2 0.9, 6.6 2.9 and 8.9  μg/mL, respectively. From the n-hexane and ethyl acetate extracts, the structures of four compounds were elucidated by NMR spectroscopic method, including lupeol (1), syringaresinol (2), apigenin (3), verbascoside (4). The a-glucosidase inhibitory activities showed that all isolated compounds were more active than the positive control - acarbose with an IC50 of 37.5  0.4; 19.5  0.2; 20.1  0.3; 69.3  0.2 µg/mL, respectively. Conclusion: The ethyl acetate extract showed strong activity about 19 times more than positive control - acarbose. The compound syringaresinol (2) was the most powerful α-glucosidase inhibitor. Therefore, Ruellia tuberosa L. contains many compounds that can inhibit α-glucosidase activity.

Purpose: To investigate the effect of different extracts of Cremastra appendiculata (D. Don) Makino onapoptosis of A549 cells, and the underlying mechanism.Methods: The contents of colchicine in ethyl acetate and n-butanol extracts of Cremastra appendiculata(D. Don) Makino were determined using high performance liquid chromatography (HPLC). Lung cancerA549 cells cultured in vitro were divided into blank control, standard colchicine and Cremastra appendiculata (D. Don) Makino extract groups. The effect of different extract concentrations on proliferation of the cells was determined using methyl thiazolyl diphenyl-tetrazolium (MTT) assay, while apoptosis of A549 cells induced by the extracts was evaluated by flow cytometry (FC).Results: Compared with the standard colchicine group, there was no colchicine in the n-butanol and ethyl acetate extracts of Cremastra appendiculata. Results from MTT assay showed that the extract inhibited the proliferation of A549 cells (p < 0.05). Flow cytometry results showed that ethyl acetate extract significantly enhanced apoptosis in A549 cells (p < 0.05). However, n-butanol extract had no significant effect on the apoptosis of A549 cells (p < 0.05).Conclusion: The ethyl acetate extract of Cremastra appendiculata (D. Don) Makino induces apoptosis in lung cancer A549 cells. Therefore, there is a need for further research and development of antitumor drugs from the extract of Cremastra appendiculata (D. Allamanda leaf extracts were made by three organic solvents hexane, methanol and ethyl acetate having different polarities. Thin layer chromatography (TLC) of the refluxing extracts showed each of them contained considerable number of different compounds. By observing Rf value of these extracts it was confirmed that the compounds present in different extracts are not same and two of them (methanol and ethyl acetate) extracts were distilled to remove solvent for bioactivity test. In growth inhibition test, methanol and ethyl acetate extracts of allamanda inhibited mycelial growth of Phomopsis vexans. Ethyl acetate extract at 0.2% and 0.3% concentration inhibited 100% mycelial growth of Phomopsis vexans while methanol extract was not effective in suppressing growth rather it arrested temporarily the growth of Phomopsis vexans. Eggplant seeds treated with ethyl acetate extract in blotter produced higher percentage of seed germination (85.00%) and healthy seedlings (88.36%) and lower percentage of dead seed (15.00%), and rotten seed (5.89%) than those treated with methanol extract. In most cases, seed quality was improved with the increasing of concentration of ethyl acetate extract. It may be summarized that higher amount of antifungal compounds were present in ethyl acetate extract in purified form where methanol also have some compounds that inhibited partially growth of Phomopsis vexans.DOI: http://dx.doi.org/10.3329/jesnr.v5i2.14814 J. This research aimed to evaluate the secondary effects of secondary metabolites produced by streptomycetes on spore germination and mycelial growth of the phytopathogenic fungi Cladosporium fulvum Cooke and Fusarium oxysporium f. sp. lycopersici from tomato plants. Metabolites produced by streptomycete isolates codified as AC-147 and AC-92 caused 94.1% inhibition of C. fulvum while AC-95 isolate caused 33.9% inhibition. AC-92 was the most efficient for F. oxysporum f. sp. lycopersici, causing 94.2% inhibition of spore germination. For mycelial growth, AC-26 and AC-92 were the most efficient in inhibiting C. fulvum growth by 46.6% and F. oxysporum f. sp. lycopersici by 29.9%. These streptomycetes are potential agents for biocontrol development methods of these tomato plant pathogenic fungi.

A chitinolytic enzyme from Bacillus thuringiensis subsp. aizawai has been purified and its molecular mass was estimated ca. 66 kDa by sodium dodecyl sulfate – polyacryamide gel electrophoresis (SDS–PAGE). The enzyme was able to hydrolyze chitin to chitobiosides but not carboxymethylcellulose, cellulose, pullulan, and laminarin. Optimal pH and temperature were detected at 6 and 50 °C, respectively. Stability, in the absence of substrate, was observed at temperatures less than 60 °C and pH between 5 and 8. Enzyme activity was significantly inhibited by K+ and EDTA and completely inhibited by Hg2+. Purified chitinase showed lytic activity against cell walls from six phytopathogenic fungi and inhibited the mycelial growth of both Fusarium sp. and Sclerotium rolfsii. The biocontrol efficacy of the enzyme was tested in the protection of bean seeds infested with six phytopathogenic fungi.Key words: chitinase, Bacillus thuringiensis, purification, phytopathogenic fungi.

The objective of this study was to assess the ethyl acetate extracts of Gastrodia elata Blume (GEB) on vascular tone and the mechanisms involved. GEB was extracted with 95% EtOH followed by a further extraction with ethyl acetate. The effects of GEB and its ingredients on the isometric tensions of the aortic rings from rats were measured. The ethyl acetate extract of GEB induced a vasodilatory effect on rat aorta, which was partially dependent on endothelium. Four chemical compounds isolated from GEB were identified as 3,4-dihydroxybenzaldehyde (DB), 4-hydroxybenzaldehyde (HB), 4-methoxybenzyl alcohol (MA), and 4,4′-dihydroxydiphenyl methane (DM), respectively. All of these compounds induced vasodilatations, which were dependent on the endothelium to different degrees. After pretreatment with Nω-nitro-l-arginine methyl ester, indomethacin, or methylene blue, the vasodilatations induced by DB, HB, and MA were significantly decreased. In addition, the contractions of the rat aortic rings due to Ca2+ influx and intracellular Ca2+ release were also inhibited by DM. Furthermore, the administration of DB significantly enhanced the productions of nitric oxide (NO) and the activities of the endothelial NO synthase in aorta and in endothelial cells. Thus, GEB may play an important role in the amelioration of hypertension by modulating vascular tones.

Characterization of antioxidant compounds from the seeds of basil (Ocimum basilicum L.) has been done. Extraction is done by maceration method using ethanol solvent, fractionation by TLC and column chromatography, antioxidants test using DPPH method, and characterization of antioxidant compound using GCMS. Ethanol extract and results of fractionation ethanol extract of basil seeds using n-hexane, ethyl acetate, n-butanol and methanol-water extracts show that n-butanol extract has the highest antioxidant activity with IC50 values ​​of 41.90 ppm. Results of column chromatography n-butanol extract using n-hexane : ethyl acetate (1:9) as mobile phase yielded 5 fractions with fraction 4 (F4 isolate) has dominant stain of active antioxidants after being sprayed DPPH reagent, it had IC50 values ​​of 39,70 ppm and total phenolic content of 0,003 mg/g. Isolate F4 suspected contains two active compounds as antioxidant which is terpenoid and phenolic compound group, namely squalene and 1,4-di-tert-buthyl-phenol identified by GCMS.DOI :http://dx.doi.org/10.15408/jkv.v0i0.3598
