Objective: Curcuma longa is a known natural medicine for inflammation from ancient times. It has a low absorption rate and poor solubility. Hence, it is used for the green synthesis of nanoparticles. Zinc oxide nanoparticle (ZnO NPs) is famous nanoparticles which are economical, less toxic, and brilliantly biocompatible. They have potential biomedical properties, mainly anticancer, antidiabetic, and antimicrobial.Methods: The present study was designed to investigate in vitro analysis of the antimicrobial activity against pathogenic bacteria and fungi and its ability to scavenge reactive oxygen radicals of ZnO NPs.Results and Conclusion: The results indicated that ZnO NPs produced from C. longa had higher antimicrobial activity against Escherichia coli, Staphylococcus aureus, Streptococcus pyogenes, and Candida albicans. Therefore, we suggest that ZnO NPs can be used as the antimicrobial agent. It is a good scavenger of superoxide radical, nitric oxide, and hydrogen peroxide and has reducing power, which is greater than ascorbic acid at a higher concentration.

Curcumin turmeric (C. longa) has been found capable of inhibiting p300/CBP associated factor (PCAF) histone asetyltransferase (HAT) as a cancer biomarker. However, the molecular mechanism of inhibition is unkown. This study is aimed to analyse the molecular inhibition mechanisms by in silico method. The analysis showed that curcumin’s Gibbs free energy (ΔG) was stable and capable of binding amino acid residues believed to be active site of PCAF HAT. Curcumin is predicted to be a PCAF HAT inhibitor, and further in vitro analysis is required.

In this study we focused on four taxa of the genus Calendula (C. maritima, C. suffruticosa subsp. fulgida, C. arvensis, and the hybrid between the first two ones), collected in Mediterranean area (Sicily). Six extracts for each species were obtained using solvents with increasing polarity (hexane, ethanol 80%, acetone 70%, and water) and through extraction by supercritical fluids (SFE). It has been observed that the solvent with the highest extraction efficiency was ethanol 80% for all species. However, SFE extracts showed high antioxidant activity comparable to the ethanol 80% extract (polyphenol, DPPH, and reducing power method). These findings were confirmed by in vitro analysis (MTT assay) where it was observed that the tested concentration (24 µg/mL), obtained from ethanol 80% and SFE extracts, showed a protective effect comparable to that induced by a synthetic antioxidant. Extraction with SFE ensured a great selectivity by avoiding the use of toxic organic solvents and thus consisted of a promising technique for sustainable production of Calendula extracts.
