Studies of such quality indicators as immunogenic and antigenic activity of the vaccine against colibacteriosis of animals have been carried out in experiments with laboratory animals. Immunogenic activity was determined by vaccination of white mice with different doses of the vaccine, followed by infection with virulent strains of Escherichia , producers of adhesive antigens, and determination of ID The antigenic activity of the vaccine was studied in experiment on rabbits by determining the level of antibodies to adhesive antigens in agglutination reaction in blood serum of vaccinated animals. The relationship between immunogenic and antigenic activity of the vaccine was detected by studying preventive activity of sera obtained from vaccinated rabbits in the experiment on white mice. The analysis of the obtained research results was given. The relationship between the level of antibodies to adhesive antigens in the blood serum of vaccinated animals and the immunogenic activity of the vaccine in the experiment on laboratory animals was established.

Abstract A fully automated method has been developed for the determination of total cholesterol in blood serum, using the AutoAnalyzer system. According to the new method, based on the cholesterol determination of Abell et al. (1), the serum sample is saponified and subsequently extracted with carbon tetrachloride. After treatment with Liebermann-Burchard reagent, the amount of cholesterol is determined at 630 nm. The method can be used for free and esterified cholesterol because saponification is carried out. The relationship between the new method and that of Abell et al. is linear. The correlation coefficient is 0.98 and the standard error 1.5%. According to the new method, 30 samples can be analyzed per hour.

Determined the key quality indicators, which allow to control the resource of the liquid dielectric. For the first time, “The method of experimental determination of the liquid dielectric resource and measures for its restoration” was developed.

An integral part of preclinical pharmacokinetic studies is the development of a bioanalytical method for determination of the drug in a biological fluid.The aim of the research was to assess the suitability of the test system based on enzyme-linked immunosorbent assay (ELISA) for quantitative determination of rituximab in the blood serum of laboratory animals after intravenous administration of rituximab at a dose corresponding to the therapeutic dose in humans. Th test system was developed by the Scientific and Production Center Probiotech.Materials and methods: the determination of rituximab in biological samples was carried out using a two-stage sandwich-type ELISA, followed by detection based on horseradish peroxidase. The ELISA results were recorded using a microplate photometer at a wavelength of 450 nm.Results: the experiments helped to establish the detection limit (0.24 ng/mL) and the lower limit of quantitation (1.00 ng/mL) of rituximab in rabbit blood serum, they also demonstrated high selectivity of analyte determination in a multicomponent biological matrix. The mean rituximab concentration was within 14 % of the nominal value in the entire working range of the method. The within-run and between-run precision of the assay did not exceed 7.4 %, the total error of the method did not exceed 20.1 %. The linearity of dilution makes it possible to use the assay for the analysis of biological samples with a wide range of rituximab concentrations. The stability of the analyte in the rabbit blood serum was confirmed by storing samples for 6 hours at room temperature, for 50 days at —35 °C, and after 3 freeze-thaw cycles. The validated immunoassay was successfully used to determine the rituximab concentration in biological samples obtained in the rituximab pharmacokinetic trial in rabbits. The accuracy of the results was confirmed for the entire range of the determined concentrations; parallelism was demonstrated between the calibration curve and the results of analysis of serially diluted rabbit serum samples with the maximum concentration of rituximab.Conclusions: the proposed enzyme immunoassay test system can be used for quantitative determination of rituximab in the blood serum of laboratory animals, as it meets acceptance criteria for all validation parameters described in the international guidelines on validation of bioanalytical methods.

Currently, live vaccines are mainly used to prevent brucellosis in animals. The effectiveness of such vaccines is a time-tested fact. However, live vaccines have many disadvantages, they can cause abortions in vaccinated animals, some vaccine strains migrate to unvaccinated animals, and pose an epidemiological danger. In addition, there is a problem of postvaccinal antibodies, which complicates the differential diagnosis of immunized and sick animals. When receiving a weakly agglutinogenic vaccine, it is important to choose a specific protective antigen, since the antigenic set of any bacterium, including brucella, is very large, and not all antigens are of equal importance in the formation of immunity. Studies were conducted on rabbits and guinea pigs that react negatively to brucellosis. Before and after vaccination, the antigenic activity of the test vaccine and the level of antigen-binding lymphocytes with a receptor for the protective brucella antigen were determined in dynamics. To detect the antigenic activity of LAV, a red blood cell diagnostic was prepared, which was obtained by sorption of the protective brucella antigen on tanized red blood cells. Based on the results of serological and bacteriological studies, it was found that the LAV at a dose of 0.1 mg of antigen is optimal. A dynamic increase in serological conversion in vaccinated rabbits and guinea pigs was observed with the introduction of each dose of the vaccine. Depending on the dose range, the highest peak of seroconversion was observed in guinea pigs on day 30 and in rabbits on day 15 of the study. Thus, the optimal vaccination dose of brucellosis weakly agglutinogenic vaccine with a single administration should be considered: for guinea pigs - 0.1 mg, and for rabbits-0.2 mg. Consequently, the administered dose of the test vaccine affected the induction of agglutinins and depended on the injected dose of the drug, which was reflected in the results of seroconversion. Maximum seroconversion was observed in guinea pigs on day 30, in rabbits on day 15 after immunization.

SummaryWhen the International Normalised Ratio (INR) is used for control of oral anticoagulant therapy the same result should be obtained irrespective of the laboratory reagent used. However, in the UK National External Quality Assessment Scheme (NEQAS) for Blood Coagulation INRs determined using different reagents have been significantly different.For 18 NEQAS samples Manchester Reagent (MR) was associated with significantly lower INRs than those obtained using Diagen Activated (DA, p = 0.0004) or Instrumentation Laboratory PT-Fib HS (IL, p = 0.0001). Mean INRs for this group were 3.15, 3.61, and 3.65 for MR, DA, and IL respectively. For 61 fresh samples from warfarin-ised patients with INRs of greater than 3.0 the relationship between thromboplastins in respect of INR was similar to that observed for NEQAS data. Thus INRs obtained with MR were significantly lower than with DA or IL (p <0.0001). Mean INRs for this group were 4.01, 4.40, and 4.59 for MR, DA, and IL respectively.We conclude that the differences between INRs measured with the thromboplastins studied here are sufficiently great to influence patient management through warfarin dosage schedules, particularly in the upper therapeutic range of INR. There is clearly a need to address the issues responsible for the observed discrepancies.

SummaryIn order to determine the correlation between different doses of F. VIII and their clinical effect,. 70 children with severe hemophilia A were studied after treatment with single doses of cryoprecipitate. The relationship between plasma F. VIII levels or doses calculated in u/ kg of body weight and clinical results followed an exponential curve. Plasma F. VIII levels of 0.35 and 0.53 u/ml corresponded to 95 and 99% satisfactory treatment, respectively. Similar clinical results were obtained with 20 and 31 u/kg. When the in vivo recovery of F. VIII after lyophilized cryoprecipitate was 0.015 u/ml for each u/kg injected, plasma F. VIII levels of 0.30 and 0.47 u/ml respectively were achieved. Since home treatment is largely based on single infusions of F. VIII, it is suggested that moderate and severe hemorrhages be treated with a dose which will provide a plasma F. VIII level of 0.5 u/ml.

Objectively and reliably determined indicators of rheological properties of the dough were identified using the alveograph device to create a system of classifications of wheat and flour from it for the intended purpose in the future. The analysis of the relationship of standardized quality indicators, as well as newly developed indicators for identifying them, differentiating the quality of wheat flour for the intended purpose, i.e. for finished products. To do this, we use mathematical statistics methods.

The objective: to еxplore the relationship between the activity of endometrial proliferation and the state of the local immune response in the uterus in the conditions berprestasi process. Patients and methods. Examined 228 women of reproductive and perimenopausal age with endometrial pathology using ultrasound and then performing hysteroresectoscopy. Determination of the concentrations of the cytokines IL-1, IL-2, IL-6 and TNF was performed by solid phase ELISA. Results. Found a trend that confirms the loss of sensitivity to hormones at the stage of malignancy of the endometrium and can be used as diagnostic determinants in determining the nature of intrauterine pathology and criterion of the effectiveness of conservative therapy. Conclusion. Improving etiopatogenetice approach to the therapy of hyperplastic proce.sses of endometrium with determination of receptor phenotype of the endometrium is a research direction in modern gynecology, which will help to improve the results of treatment and prevention of intrauterine pathology. Vertical axis wind turbine technology has been applied last years, very long after horizontal axis wind turbine technology. Aerodynamic problems of vertical axis wind machines are discussible. An important problem is the determination of the incidence law in the interaction between wind and rotor blades. The focus of the work is to establish equations of the incidence depending on the blade azimuth, and to solve them. From these results, aerodynamic torques and power can be calculated. The incidence angle is a parameter of velocity triangle, and both the factors depend not only on the blade azimuth but also on the ratio of rotational speed and horizontal speed. The built computational program allows theoretically selecting the relationship of geometric parameters of wind turbine in accordance with requirements on power, wind speed and installation conditions.
