Genotype data from 21 microsatellites were used to evaluate genetic diversity and relationships of Gatacko cattle and Busha breeds. A total of 100 animals included in the study were divided into three groups: Gatacko cattle (GC), Busha from eastern Herzegovina (BEH) and Busha from western Herzegovina (BWH). Hardy-Weinberg equilibrium was observed for all loci in all groups, except for single locus (INRA35) in group of Busha from western Herzegovina. A total number of 166 alleles were detected in a total sample. The average number of alleles per locus was 7.9. The highest observed mean heterozygosity (0.688) was found in the Gatacko cattle and the lowest (0.634) in Busha from western Herzegovina. A pairwise population matrix of Fst values among analyzed groups showed significantly high genetic differentiation among all three groups: Gatacko cattle and Busha from western Herzegovina, Gatacko cattle and Busha from eastern Herzegovina and Busha from western and Busha from eastern Herzegovina. This report represents the first genetic characterization of Gatacko cattle and Busha cattle, traditional cattle breed from Bosnia and Herzegovina. The obtained results are valuable for the future development of conservation and management strategies for these cattle breeds.

The variability of 21 microsatellite loci was analyzed in order to estimate the genetic diversity of the Busa cattle breed. A total of 50 animals involved in the study were divided into two groups: Busa from eastern Herzegovina and Busa from western Herzegovina. The mean number of alleles per locus was 6.6. The average expected heterozygosities were 0.6885 and 0.6212 in the eastern and western populations, respectively. The observed heterozygosity values were 0.6579 and 0.6336, in eastern and western population, respectively. The degree of population differentiation (FST) ranged from 0.008 (for ILSTS006 locus) to 0.242 (for BM1818 locus), with a mean of 0.112. Cluster analysis showed that the two populations were clearly clustered into two distinct clades. This report represents the first genetic characterization of pure Busa cattle from Bosnia and Herzegovina. The obtained results are important for the future development of conservation and management strategies for this cattle breed.

Abstract Nothofagus alessandrii (Nothofagaceae) is one of the most endangered trees from Chile. Despite its conservation status, few molecular markers are available to study its population genetic, connectivity and to assist reproduction programs. Novel polymorphic microsatellites from the genome of N. alessandrii were isolated and characterized using high-through sequencing. A total of 30 primer pairs were synthesized and 18 microsatellites were amplified correctly. Polymorphism and genetic diversity was evaluated in 58 individuals from three populations of N. alessandrii. Sixteen of them were polymorphic and the number of alleles in the pooled sample ranged from 2 to 14, the mean number of alleles was 4.81. The mean values of observed heterozigosity (HO) and excepted heterozygosity (HE) of the Chanco population were 0.392 and 0.383. The mean values of HO and HE in the population of Empedrado (0.397 and 0.360, respectively) and Fundo el Desprecio (0.397 and 0.395, respectively) were similar. Linkage disequilibrium was found in a few pairs of loci suggesting that most of the markers can be considered as independent. Significant deviations from Hardy- Weinberg equilibrium (P < 0.05) were found in several loci probably due to low sampling size. Transferability to the congeneric N. pumilio was successful in only four out of the sixteen polymorphic markers. The microsatellite markers developed in this study will be useful to study the genetic diversity and structure and to develop integrated managements plans for the conservation on this endangered species.

Cryptosporidiumfelis is an important cause of feline and human cryptosporidiosis. However, the transmission of this pathogen between humans and cats remains controversial, partially due to a lack of genetic characterization of isolates from cats. The present study was conducted to examine the genetic diversity of C. felis in cats in China and to assess their potential zoonotic transmission. A newly developed subtyping tool based on a sequence analysis of the 60-kDa glycoprotein (gp60) gene was employed to identify the subtypes of 30 cat-derived C. felis isolates from Guangdong and Shanghai. Altogether, 20 C. felis isolates were successfully subtyped. The results of the sequence alignment showed a high genetic diversity, with 13 novel subtypes and 2 known subtypes of the XIXa subtype family being identified. The known subtypes were previously detected in humans, while some of the subtypes formed well-supported subclusters with human-derived subtypes from other countries in a phylogenetic analysis of the gp60 sequences. The results of this study confirmed the high genetic diversity of the XIXa subtype family of C. felis. The common occurrence of this subtype family in both humans and cats suggests that there could be cross-species transmission of C. felis.

Background: Duck farming plays a significant role, next to chicken in the socio-economic uplift men of the rural farmers of North-East India. Pati duck is the most common duck breed in the Brahmaputra valley of Assam and the other common variety reared in North-East India is Chara-Chambeli, however it originatein Kerala. Genetic characterization plays a significant role for formulation of breeding strategies for improvement of any breed. Microsatellites are codominant in nature and are highly polymorphic. High level of allelic variation, co-dominant mode of inheritance and potential for automated analysis make them an excellent tool for genotyping, mapping and genetic characterization. Pati duck is the most common duck breed in the Brahmaputra valley of Assam and the other common variety reared in North-East India is Chara-Chambeli however its origin in Kerela. Therefore, the present investigation was carried out to characterize these two duck breeds using microsatellite markers to evaluate the genetic diversity in these two duck population. Methods: For the present study, Blood sample were collected from 50 Patiducks and 50 Chara-Chambeli ducks from different parts of North-East India. Assessment of genetic characterization of duck breeds of north-east region were carried out using 16 microsatellite markers and population genetics analysis were done by POPGENE software. Result: In the present study, all the studied loci were highly polymorphic. Analysis generated a total of 41 microsatellite alleles. The number of observed alleles (Na) with an overall mean of 1.93±0.258. However, the effective number of alleles (Ne) with a mean of 1.6933±0.2712. The Shannon’s information index was found to a mean value of 0.5685±0.1693. The overall means for observed (HO) and expected (He) heterozygosities were 0.2889±0.2477 and 0.5289±0.0853, respectively. The chi-square (χ2) test for Hardy-Weinberg equilibrium revealed that all the loci are in within Hardy-Weinberg equilibrium.

Random amplified polymorphic DNA (RAPD) markers were used to study genetic diversity in dog samples belonging to populations of German Shepherd and Japanese Spitz. A total of twelve samples were typed using eight RAPD primers. Out of eight primers, three primers gave result in six individuals of dogs. The phylogenetic tree constructed by the neighbor joining method based on Nei. Original measures revealed highest genetic identity found in German Shepherd as 0.9444 and highest genetic distance as 1.2809. The analysis predicts the number of polymorphic loci as 15 and the percentage of polymorphic loci as 83.3. The Cleveland Bay (CB) is the United Kingdom’s oldest established horse breed. In this study we analyzed the genetic variability in CB horses and investigated its genetic relationships with other horse breeds. We examined the genetic variability among 90 CB horses sampled in the USA compared to a total of 3447 horses from 59 other breeds. Analysis of the genetic diversity and population structure was carried out using 15 microsatellite loci. We found that genetic diversity in CB horses was less than that for the majority of other tested breeds. The genetic similarity measures showed no direct relationship between the CB and Thoroughbred but suggested the Turkman horses (likely in the lineage of ancestors of the Thoroughbred) as a possible ancestor. Our findings reveal the genetic uniqueness of the CB breed and indicate its need to be preserved as a genetic resource.

Abstract Schima superba is a common dominant tree species in evergreen broad-leaved forest in subtropical China. Despite its multiple usages in wood industry, reforestation and traditional Chinese medicine, its genetic diversity is poorly studied. To help studying its genetic diversity and structure in the future, after microsatellite enrichment and screening, we identified 16 microsatellites in S. superba. These markers showed polymorphism in three populations. The number of alleles per locus ranged from 3 to 32 with a mean of 14. Within populations, the observed and unbiased expected heterozygosities ranged from 0.048 to 0.926 and from 0.048 to 0.949, respectively. The newly developed 16 microsatellites will be useful for investigating the genetic diversity and structure from large scale patterns to fine-scale structures in this species.

The Neotropical freshwater fishIchthyoelephas longirostris(Characiformes: prochilodontidae) is a short-distance migratory species endemic to Colombia. This study developed for the first time a set of 24 polymorphic microsatellitelociby using next-generation sequencing to explore the population genetics of this commercially exploited species. Nineteen of theselociwere used to assess the genetic diversity and structure of 193I. longirostrisin three Colombian rivers of the Magdalena basin. Results showed that a single genetic stock circulates in the Cauca River, whereas other single different genetic stock is present in the rivers Samaná Norte and San Bartolomé-Magdalena. Additionally,I. longirostriswas genetically different among and across rivers. This first insight about the population genetic structure ofI. longirostrisis crucial for monitoring the genetic diversity, the management and conservation of its populations, and complement the genetic studies in Prochilodontidae.
