Summary Background In the presence of conflicting advice about the relationship between selenium-type II diabetes-oxidative stress trio, this study aimed to assess the consequences of selenium supplementation on fasting plasma glucose (FPG) level, antioxidant activities of selenodependent and non-selenodependent enzymes, and other markers of oxidative stress studied for the first time during gestational diabetes mellitus (GDM). Methods This research was carried out among 180 pregnant Algerian women, 60 of whom were in good health, 60 women with GDM did not take supplements, and 60 women with GDM took selenium orally (50 μg/d) for 12 weeks starting from their second trimester of pregnancy. Blood samples were taken in order to assay FPG level and oxidative stress markers. Results Selenium supplementation during GDM has demonstrated its hypoglycemic power in the significant decline of FPG level, and its antioxidant properties in the significant reinforcement of antioxidant activities of erythrocyte selenodependent enzymes (glutathione peroxidase and glutathione reductase), the significant increase in erythrocyte catalase and superoxide dismutase activities simultaneously with the highest decrease in erythrocyte and plasma malondialdehyde levels. This decrease was only significant for plasma carbonyl proteins, which was not the case for erythrocyte carbonyl proteins. Conclusions The recourse to selenium supplementation by seleno-deficient pregnant women with GDM is beneficial for maternal health. This micronutrient exploits its antioxidant and insulin-mimetic properties in the maintenance of blood glucose homeostasis and the fight against oxidative stress, and consequently, its supplementation delays the occurrence of GDM complications.

Background: One of the most common complications of pregnant women is gestational diabetes mellitus (GDM). Oxidative stress can play an important role in GDM. Objective: The aim of this study was to evaluate salivary antioxidants and oxidative stress markers in GDM. Method: Twenty pregnant women with GDM and 20 healthy pregnant women with normal blood glucose test participated in this study. Five mL of unstimulated saliva samples were collected. Spectrophotometric assay was carried out for sialochemical analysis. Stata software was used for data analysis. Results: The GDM group exhibited no significant difference in salivary total antioxidant capacity and malondialdehyde compared to the healthy control group. All of antioxidants markers, the uric acid, total antioxidant, peroxidase and catalase, decreased in GDM group that the difference of peroxidase and catalase was statistically significant. All of oxidative stress markers, the salivary malondyaldehid, total oxidative stress and total thiol, increased in GDM group. GDM group exhibited significantly higher salivary total oxidative stress levels. Conclusion: Catalase level was significantly lower and total oxidative stress was significantly higher. These two markers might have significant importance and might exhibit early changes compared to other factors in GDM. . Some of salivary antioxidants might have diagnostic, prognostic or therapeutic implications in GDM. Other studies with large sample size on salivary and blood samples need to be done to confirm this properties and salivary samples using instead of blood samples in GDM biomarkers changes.

Objective.To determine plasma markers of oxidative stress during the second and third trimester of pregnancy in patients with gestational diabetes mellitus (GDM).Study Design.We conducted a prospective nested case-control study involving 400 pregnant women, 22 of whom developed GDM. As control group, 30 normal pregnant women were chosen randomly. Plasma samples were analyzed for 8-iso-prostaglandin F2α(8-iso-PGF2α), advanced oxidative protein products (AOPPs), protein carbonyl (PCO), glutathione peroxidase-3 (GPX-3), and paraoxonase-1 (PON1) at 16–20 weeks, 24–28 weeks, and 32–36 weeks of gestation.Results.Compared to control subjects, the plasma levels of PCO, AOPPs, and 8-iso-PGF2αwere elevated at 16–20 weeks’ and 32–36 weeks’ gestation in GDM. There was no significant difference in PCO and 8-iso-PGF2αat 24–28 weeks in GDM. GPX-3 was statistically significantly increased at 16–20 weeks and 32–36 weeks in GDM. PON1 reduced in patients with GDM. No significant differences were found at 24–28 and 32–36 weeks between the GDM and control groups. In GDM, PCO, AOPPs, and 8-iso-PGF2αlevels were higher and GPX-3 and PON1 levels were lower in the second than the third trimester.Conclusion.Oxidation status increased in GDM, especially protein oxidation, which may contribute to the pathogenesis of GDM.

Objective: Aims of the study is to specify the significance of thiol/disulfide homeostasis in the aspect of gestational diabetes mellitus (GDM) and GDM-related complications. Material and Methods: This study is a prospective review of the data of 61 healthy and non-pregnant women, 58 healthy pregnant women, and 62 pregnant women with GDM. Results: The patients with gestational diabetes mellitus had significantly higher disulfide/native thiol and disulfide/total thiol concentrations than non-pregnant patients (p<0.001 for both) and healthy pregnant patients (p: 0.015 and p: 0.018, respectively). Besides, in GDM group had significantly lower native thiol/total thiol concentrations than non-pregnant patients and healthy pregnant patients (p<0.001 and p: 0.016, respectively). There were positive and significant correlations between disulfide levels and HbA1c concentrations (r=0.26, p: 0.042), and between disulfide and oral glucose tolerance test first hour concentrations (r=0.26, p: 0.039). The receiver operating characteristic curve analyses for native thiol, total thiol, and disulfide were unable to predict adverse perinatal outcomes in this cohort. Conclusion: The significantly higher concentrations of disulfide/native thiol and disulfide/total thiol in women with GDM could be considered as the presence of increased oxidative stress.  However, these markers failed to predict adverse perinatal outcomes.
