Abstract Inonotus obliquus is a parasite on the birch and other trees and is also a well-known medicinal mushroom. Its sterile conk is highly sought for its bioactive compounds such as phenols, polysaccharides, triterpenoids, and steroids. It was traditionally used to treat various gastrointestinal diseases, viral and parasitic infections, to counteract the progression of cancers, and to stimulate the immune system. We used acute gamma irradiation, followed by short-term submerged cultivation, as an oxidative stress inducer to enhance the synthesis of mycelial metabolites. The 300 Gy and 400 Gy doses showed the best results across the whole experimental design. Each assayed criterion had a different corresponding optimal stimulation dose. In one experiment, sublethal doses of irradiation triggered the dry weight of the cultured mycelium to increase by 19.764%. The free radical scavenging potential of the mycelium extracts increased by 79.83%. The total phenolic content of mycelium extracts and culture broth increased by 55.7% and 62.987%, respectively. The total flavonoid and sinapinic acid content of the broth increased by 934.678% and 590.395%, respectively. As such, gamma irradiation pre-treatment of the mycelial inoculum proved an interesting, economically and environmentally effective tool for stimulating secondary metabolite synthesis in submerged mycelium cultures.

Food irradiation technologies are used to reduce the risk of food borne diseases by eliminating pathogenic microorganisms, prolonging shelf life and reducing storage losses by delaying ripening, germination or sprouting. However, application of irradiation in food technology can negatively influence the biologically active compounds in foods. In this research, the effect of gamma-irradiation on the antioxidant activity of Bulgarian teas was investigated. The aim of the study was to evaluate the total phenolic and tannin content and antioxidant activity of ethanolic extracts of Bulgarian herbal teas before and after gamma-irradiation. Mursalski tea ( Sideritis scardica), Mashterka tea ( Thymus serpyllum), Good Night tea (tea mix), Staroplaninski tea (Balkan tea mix), Trakia tea (tea mix), and Mountain tea (Planinski tea mix) were selected for this study. Gamma-irradiation was applied at the absorbed dose of 5 kGy. Antioxidant activity of non-irradiated and irradiated teas was determined by measuring antiradical activity against DPPH• and ABTS•+ and the ability to reduce ferrous ions. The highest total phenolic content was found in Mursalski tea (268 mg/g), and the highest tannin content in Good Night tea (168 mg/g). FRAP, TEAC and DPPH assays revealed that the most active samples were Staroplaninski (2.78 mmol Fe (II)/g), Planinski (0.87 mmol Trolox/g) and Planinski (0.032 mg/mL), respectively. The radical scavenging activity of irradiated tea samples was maintained after gamma-irradiation. The most interesting extract from irradiated tea studied was Staroplaninski, which demonstrated a higher antioxidant potential in the irradiated sample compared with the non-irradiated sample.

Chaga (Inonotus obliquus) is a parasitic fungus, which has been used in traditional medicine in Russia and other northern European countries in the treatment of cancer, gastrointestinal andliver diseases. It has been a subject of intensive researchrecently, confirming many of its health-beneficial effects. In order to obtain a product that would allow modified and prolonged release of the Chaga?s active metabolites, hot water Chaga extract was immobilized using calcium - alginate. The extract, which was predominantly composed of carbohydrates (57 %), also contained a relatively high amount of antioxidants/phenolic compounds (130 mg gallic acid equivalents per g of dry extract) and exhibited pronounced radical scavenging activity. It showed significant antibacterial activity as well, inhibiting growth of tested bacterial strains at concentrations of 1.25-20 mg/mL. Entrapment efficiency was about 80 %, and the extract-alginate system showed pH-dependant extract release; there was negligible release at pH 1.75 (gastric pH), and the release gradually increased with the increase in pH, reaching ~43 % of immobilized extract at pH 8.5 after 90 min. Such a product could be used as a dietary supplement, adjuvant in therapy of gastrointestinal diseases or as a food additive.

Objective : Gamma radiation induces free radicals with a corresponding alteration in the cell’s antioxidant defense system. The present study thus aimed at determining the role of gamma irradiation in improving the cosmeceutical potential of CEK in terms of antioxidants.  Methods : C.emersonii KJ725233 was subjected to low (100 Gy) and high (1000 Gy) gamma irradiation doses and its effect on the chlorophyll content was evaluated. The quantitative alterations in the antioxidant content of CEK were evaluated by phosphomolybdenum assay (TAC), ferric reducing antioxidant potential (FRAP), 2, 2 - Dipheny-1-picryl hydrazyl radical scavenging assay (DPPH), total phenolic (TPC) and total flavonoid content (TFC). Also, the corresponding qualitative alterations in the bioactives of CEK were determined by GC-HRMS analysis.   Results : A 179.57±2.55% increase in the total chlorophyll content alongwith a 71.76±2.96%, 32.08±2.16%, 11.67±0.89%, 42.85±8.0% and 31.37±3.18% increase was observed in the TAC, FRAP, DPPH radical scavenging, TPC and TFC was observed in CEK irradiated at 1000 Gy. GC-HRMS analysis revealed the induction of Vitamin E on irradiation at both the doses with a corresponding decrease in the phytol content whereas 100 Gy stimulated the induction of phytosterols. Conclusion : The potent intrinsic antioxidant activity of cosmeceutically significant CEK can be elevated with the induction of the most sought after antioxidant in cosmetology – Vitamin E on irradiation.

Abstract Phytochemical and antioxidant activity of quinoa flour was evaluated after subjected to gamma irradiation processes at dose 3 and 6 kGy. Both non-irradiated and irradiated quinoa samples were subjected to successive extractions in ethanol solvent. The antioxidant activity after gamma irradiation treatment was investigated via Ferric reducing antioxidant power (FRAP) and radical-scavenging activity (RSA) using 2,2-diphenyl-1-picrylhydrazyl (DPPH).Total phenolic and flavonoid content were analyzed using Folin–Ciocalteu micro-method, aluminium chloride (AlCl3) method and High Performance Liquid Chromatography (HPLC). As well as, effect of irradiation treatment on saponin was also evaluated. Irradiation treatment showed slight differences in the saponin content after exposure to 3 and 6 kGy. Irradiation process enhanced both total phenolic content (TPC) and Total flavonoid content (TFC), TPC were 34.52 and 30.92 mg Gallic Acid Equivalent (GAE)/100g compared to 26.25 mg GAE/100g in non-irradiated quinoa. TFC were 67.44 and 62.89 mg Quercetin Equivalents (QE)/100g compared to 53.15 mg QE/100g. Irradiation dose 3 kGy significantly (p> 0.05) decreased the IC50 as DPPH-RSA and increased the FRAP.

Kaempferia parviflora rhizomes exposed to gamma irradiation at doses of 0, 5, and 7.5 kGy were evaluated every 3 months for their microbial loads, contents of 5,7-dimethoxyflavone (DMF), total phenolic contents, and antioxidant activities (DPPH and FRAP assays) during 12 months of storage at 25 °C. After gamma irradiation, microbial loads were reduced below acceptable limits. Phytochemical contents in irradiated samples were significantly increased as compared to those of the control (0 kGy) while DPPH radical scavenging activities were significantly decreased. However, gamma irradiation had no effect on FRAP values. During storage of irradiated samples, phytochemical contents and antioxidant activities were gradually decreased whereas microbial loads were progressively increased. Even so, microbial quality of irradiated samples was less than acceptable limits through 12 months' storage. Gamma irradiation at doses of 5 and 7.5 kGy was sufficient to control microbial growth and T. castaneum infestation of K. parviflora rhizomes within acceptable limits for at least 12 months at 25 °C. Additionally, 5 and 7.5 kGy were effective doses to retain DMF and phenolics in K. parviflora rhizomes.

Allium tuberosum, commonly known as the Chinese chive (CC) is often used as a traditional medicine in East Asia for its health benefits. To explore the potential of CC as a functional feed additive, antibacterial and antioxidant assays, untargeted metabolomics, and a 2 × 3 × 3 fractional factorial design (FFD) were conducted. In the present study, CC displayed stable DPPH radical scavenging activity with constant total phenolic content, however, the total flavonoid contents and the antibacterial activities were attenuated following heat treatment. The FFD results identified the solid content (SBM) as the main determinant of the antibacterial activity and moisture content of the CC products along with two other factors: drying time and temperature. Two CC products manufactured with 30% (w/v) SBM with 3 h drying at 80 °C and 20% (w/v) SBM with 8 h drying at 60 °C obtained the maximum antibacterial activity and least moisture content (<5%). Liquid chromatography-tandem mass spectrometry based multivariate analysis revealed 14 changed compounds in the non-heated and heated CC including flavonols, sinapinic acid, and lysophospholipids, which might affect the functionality. In conclusion, we propose an empirical approach to the pre-processing of CC juice that is suitable for blending in feed and simultaneously retaining its bioactivities.

Objective: Myristica fragrans Houtt commonly used as traditional medicine for alleviating of various disorders. The purpose of our study was to map out the in vitro antioxidant property and in vivo anti-hyperglycemic and analgesic effect of the methanolic extract of Myristica fragrans Houtt. (Seed and mace) (Myristicaceae) on Swiss albino mice. Methods: The processed powder of Myristica fragrans Houtt (seed and mace) were subjected to methanolic extraction by soxhlet filtration methods, and the desiccated extract was used for screening of antioxidant by DPPH free radical scavenging assessment as well as total phenolic content by using folin-ciocalteu reagent.Anti-hyperglycemic effect and analgesic action tested through alloxan induced antidiabetics test and acetic acid-tempted writhing test on mice. Results: In DPPH free radical scavenging assessment, free radicals neutralization expressed as % of inhibition 49.69±0.06% also by IC50 values as 68.43 µg/ml surmise middle level of antioxidant property. The total phenolic content expressed as 186.25 mg/g equivalent of gallic acid indicates, active phenolic content. Oral administration of 200 and 400 mg/kg of extract dose and reference drug vildagliptin (50 mg/kg) for the duration of the 4-day study period, and initiated % of inhibition the blood glucose level measured as 22.48%, 44.78% and 62.02% regard as the significant anti-hyperglycemic properties. The analgesic activity was investigated by using the acetic acid-induced writhing test in mice, at the dose of 200 mg/kg body and 400mg/kg weight, and resulting 50.4% and 68.10% correspondingly, which was considerably significant with a standard drug. Conclusion: The present study suggests that methanolic extract of seed and mace of Myristica fragrans Houtt can manage moderate oxidative stress as well as perform the painkilling action. Besides, prolong medication may enhance the new dimension of anti-hyperglycemic activity.

Nyctanthes arbour-tristis is a shrub belongs to the family Oleaceae. The flowers of this plant are fragrant since the presence of flavonol glycosides. It has also been reported for the presence of β-sitosterol, iridoid glycosides, tannins etc., and known to have immunostimulant, hepatoprotective, antiviral and antifungal activities. In the present study an attempt is made to identify antioxidant capacity and anthelminthic potential of methanolic flower extract of Nyctanthes arbour-tristis. Antioxidant activity was evaluated by total phenolic content assay, total flavonoids content assay, free radical scavenging activity and reducing ability methods. Anthelmintic activity was evaluated on Perithima posthuma using Piperazine citrate as standard drug. The results obtained for the above activities reveals that Nyctanthes arbour-tristis shows considerable antioxidant activity for all the methods and anthelminthic potential at 300 mg/ml. The current investigation is intended to evaluate the content of phytochemical constituents and antioxidant potential of hydroalcoholic extracts of stem and root of Grewia serrulata DC (HAESGS & HAERGS) and leaf and bark of Grewia Nervosa (Lour.) panigrahi (HAELGN & HAEBGN). Initially, all the extracts at different concentrations were estimated for their total phenolic content and total flavonoid content. The study was further extended for their antioxidant potential evaluation using various in vitro methods such as 1, 1-diphenyl-2-picrylhydrazyl (DPPH) radical, hydroxyl radical and superoxide radical scavenging assays. The total phenolic content (mg gallic acid equivalent per gram of extract) was high in HAELGN (170.82±0.19) and HAERGS (123.00±0.48) than HAESGS (111.2±0.26) and HAEBGN (119.60±0.23). The total flavonoid content (mg quercetin equivalent per gram) is greater in HAERGS (71.24±0.50) and HAESGS (65.68±0.27) than HAELGN (55.82±0.35) and HAEBGN (62.38±0.45). The IC50 values (µg/ml) of different plant extracts inferred that DPPH radical scavenging activity is greater in HAELGN (42.91±0.88) and HAEBGN (53.87±0.35) than HAESGS (126.73±1.20) and HAERGS (88.87±1.25). However, hydroxyl and superoxide radical scavenging activity is more in HAERGS (135.41±1.19 & 88.00±1.42) and HAELGN (172.28±1.91 & 108.163±1.09) than HAESGS (237.3±1.65 & 110.074±0.87) and HAEBGN (204.7±1.04 & 125.54±1.07). The results of present comprehensive analysis demonstrated that both the plants Grewia serrulata DC and Grewia Nervosa (Lour.) panigrahi possess high phenolic, flavonoid contents and potential antioxidant activity, and could be used as a valid source of natural antioxidants and might be utilized for pharmacological screening of various therapeutic activities.
