Ensuring stringent temperature control parameters and efficient cooling mechanisms are maintained at a dairy plant are critical to downstream processing capabilities and product quality. This study examined the temperature of incoming raw milk across three production months (June, July, August 2018), to capture peak season processing at one dairy manufacturing plant. ABSTRACT: Transport of cooled raw milk in bulk has greatly improved the quality of the raw material collected by dairy plants as it reduces the proliferation of mesophilic microorganisms that cause milk acidity and hinder its processing. However, refrigeration has favored the growth of psychrotrophic microorganisms which are able to grow at low temperatures (below 10ºC) and that produce heat resistant enzymes which degrade some milk components, reducing milk shelf life and causing organoleptic changes. The aim of this paper was to evaluate the microbiological quality of raw milk in dairy farms and after its transport to the processing dairy plant, through plate counting of mesophilic and psychrotrophic microorganisms. Fourteen milk samples were collected from tanks of the dairy-farming properties, and one sample was collected from their milk transport tanker at the entrance of the processing plant. Our results showed that the mean number of mesophilic microorganisms was higher in samples collected straight from the dairy farm tanks than in the samples collected from the transportation tankers at the entrance of the plant. Of the 14 sampled tanks, 64.3% were non-compliant with legislation. The sample collected from the milk transportation tanker containing milk from all properties showed a higher mean number of psychrotrophic microorganisms than the dairy farm samples. We conclude that the milk from dairy properties showed a higher amount of mesophilic microorganisms, and after transportation, at the entrance of the processing plant, there is a higher amount of psychrotrophic microorganisms.

From February 2004 to March 2005, 140 samples of food handlers - hands and nostrils - (92), raw milk (24), and minas frescal cheese (24) were analyzed for the presence of Escherichia coli in a dairy processing plant of Goiás State. Forty-seven E. coli strains were obtained and compared by DNA macrorestriction patterns obtained from pulsed-field gel electrophoresis following XbaI restriction in order to investigate the possible sources of cheese contaminations. Based on PFGE genotyping, one strain isolated from food the hands of a handler and five strains isolated from raw milk were identical or closely related to six strains from cheese suggesting, in these cases, the probable source of E. coli contamination in cheeses. No strain isolated from the nostrils was related to those found in cheeses or milk strains. The results showed high diversity among the strains, demonstrating a lack of predominance of an endemic clone in the dairy plant. This paper highlights the usefulness of PFGE as an epidemiological tool for determining the source of E. coli contamination in the food industry.

ABSTRACTThe present study aimed to investigate the presence, distribution, and persistence ofArcobacterspp. in an artisanal dairy plant and to test the isolates to determine their different genotypes in the processing plant and in foods. Samples were collected in an artisanal cheese factory on four occasions between October and December 2012. Food samples (raw milk, ricotta cheese, mozzarella cheese, and conditioning liquid), water samples, and environmental samples were analyzed by the culture method; isolates were identified by multiplex PCR and genotyped by pulsed-field gel electrophoresis (PFGE) analysis.Arcobacter butzleriwas isolated from 29 out of 59 samples (46.6%), 22 of which were from environmental samples and 7 of which were from food samples. Cluster analysis divided the strains into 47 PFGE patterns: 14 PFGE clusters and 33 unique types. Our findings indicate that the plant harbored numerousA. butzleripulsotypes and that the manual cleaning and sanitation in the studied dairy plant do not effectively removeArcobacter. The recurrent isolation ofA. butzlerisuggests that the environmental conditions in the dairy plant constitute a good ecological niche for the colonization of this microorganism. In some cases, the presence of indistinguishable strains isolated from the same facilities on different sampling days showed that these strains were persistent in the processing environment.

A target flowmeter, used to measure raw milk flow, was examined for sanitary conditions in a university dairy plant 10 times over a period of eight weeks. The flowmeter connection was swabbed at four different locations along the dairy plant connection at four different times during the work day: i) after chlorine sanitization, before product; ii) after product, before cleaning in place (CIP); iii) after CIP, before acid sanitization; and iv) after acid sanitization, at end of day. Samples were plated in duplicate on standard plate count agar and on violet red bile agar. After routine CIP cleaning and sanitization procedures, bacterial counts were low. Additionally, no finished product contamination problems were detected over the 7 months of flowmeter use as shown by routine quality control tests on pasteurized milk which had flowed past the in-line meter as raw milk. These results indicate that normal cleaning and sanitization procedures were adequate for the in-line flowmeter.

This study investigates the individual and combined impacts of El Niño and the positive Indian Ocean Dipole (IOD) on the Southeast Asia (SEA) rainfall variability. Using composite and partial correlation techniques, it is shown that both inter-annual events have individually distinct impacts on the SEA rainfall anomaly distribution. The results showed that the impacts of the co-occurrence of El Niño and IOD events are significant compared to the individual effects of pure El Niño or pure IOD. During June-July-August and September-October-November, the individual impacts of the pure El Niño and IOD events are similar but less significant. Both events caused negative impacts over the southern part of SEA during June-July-August (JJA) and propagated northeastward/eastward during September-October-November (SON). Thus, there are significant negative impacts over the southern part of SEA during the co-occurrence of both events. The differential impacts on the anomalous rainfall patterns are due to the changes in the sea surface temperature (SST) surrounding the region. Additionally, the differences are also related to the anomalous regional atmospheric circulations that interact with the regional SST. The anomalous Walker circulation that connects the Indian Ocean and tropical Pacific Ocean also plays a significant role in determining the regional anomalous rainfall patterns.

With increases in average temperature and rainfall predicted, more households are expected to be at risk of flooding in the UK by 2050. Data and technologies are increasingly playing a critical role across public-, private- and third-sector organisations. However, barriers and constraints exist across organisations and industries that limit the sharing of data. We examine the international context for data sharing and variations between data-rich and data-sparse countries. We find that local politics and organisational structures influence data sharing. We focus on the case study of the UK, and on geospatial and flood resilience data in particular. We use a series of semi-structured interviews to evaluate data sharing limitations, with particular reference to geospatial and flood resilience data. We identify barriers and constraints when sharing data between organisations. We find technological, security, privacy, cultural and commercial barriers across different use cases and data points. Finally, we provide three long-term recommendations to improve the overall accessibility to flood data and enhance outcomes for organisations and communities.

Psychrotolerant endospore-forming bacteria Bacillus and Paenibacillus spp. are important spoilage organisms in fluid milk. A recently developed rpoB subtyping method was applied to characterize the diversity and phylogenetic relationships among Bacillus and related sporeformers associated with milk processing systems. Milk samples representing the processing continuum from raw milk to pasteurized products were collected from two fluid milk processing plants, held at 6°C uptothe code date that had been established by each processing plant (i.e., either 18 or 21 days), and plated for bacterial enumeration throughout storage. Bacterial colonies selected to represent the visible diversity in colony morphology on enumeration plates were examined further. Among 385 bacterial isolates characterized, 35% were Bacillus spp., and 65% were Paenibacillus spp. A total of 92 rpoB allelic types were identified among these isolates, indicating considerable diversity among endospore-forming spoilage organisms present in fluid milk systems. Of the 92 allelic types identified, 19 were isolated from samples collected from both processing plants. The same rpoB allelic types were frequently identified in paired raw milk and packaged product samples, indicating that Bacillus and Paenibacillus spp. can enter dairy processing systems through raw milk. Certain subtypes were found exclusively in pasteurized samples, including those that were temporally independent, suggesting the possibility of in-plant sources for these spoilage organisms, including through the persistence of selected subtypes in processing plants. Development of effective control strategies for the diverse array of psychrotolerant endospore-forming organisms that currently limit the shelf lives of high-temperature short-time fluid milk products will require comprehensive, integrated efforts along the entire milk processing continuum.

Objective To test the hypothesis that there is seasonal variation in the rates of gestational diabetes (GDM) diagnosed using a 2 hour oral glucose tolerance test. Design Monthly assessment of the percentage of women screened from 1st April 2016 to the 31st December 2020 who were diagnosed as having gestational diabetes Setting London Teaching Hospital Population 28,128 women receiving antenatal care between April 1st 2016 and 31 December 2020. Methods Retrospective study of prospectively collected data. Main Outcome Measures Proportion of women screened diagnosed as having gestational diabetes. Results The mean (SD) percentage of women diagnosed with GDM was 14.78 (2.24) in summer (June, July, August) compared with 11.23 (1.62) in winter (p < 0.001), 12.13 (1.94) in spring (p = 0.002), and 11.88 (2.67) in autumn (p = 0.003). There was a highly significant positive correlation of the percentage testing positive for GDM with the mean maximum monthly temperature (R2 = 0.248, p < 0.001). There was a statistically significant 33.8% increase in the proportion of GDM diagnoses from June 2020 onwards, possibly related to a reduction in exercise secondary to the Covid-19 pandemic. Conclusions There is a 23.3% higher rate of GDM diagnoses in the warmer summer months. There has been a 33.8% rise in GDM diagnoses associated with the Covid-19 pandemic.

Recent global developments concerning food quality and food safety have influenced and stimulated food legislation in Turkey in accordance with internal and international trade and agreements. In this study, the way in which the dairy industry conforms to this legislation was analyzed through a case study of İzmir province, which generally has all the structural characteristics of the dairy sector in Turkey. A survey in which dairy plant managers responded to a special questionnaire was used to collect data from 86 dairy plants chosen on the basis of proportional sampling. According to the results of this study, (i) there are many dairy processors in the region, (ii) most managers have a limited education concerning their positions, (iii) most firms handle small volumes of milk and have little control over the raw milk supply, (iv) resources are too limited in these firms, limiting their ability to adopt most regulations, and (v) few processors apply the regulatory practices imposed by governmental agencies. Thus, food legislation is not enough to ensure food safety in the dairy industry in Turkey. Technical and educational support should be given to farmers and the staff of dairy firms by the Ministry of Agriculture to form an appropriate food safety infrastructure in Turkey for the milk and processed dairy products industry.
