Evaluation of Anti-diabetic and Anti-hyperlipidemic Activity of Rhododendron arboreum bark extract against Streptozocin Induced Diabetes in Rats

The anti-diabetic and anti-hyperlipidemic activity of R. arboreum in bark extract against streptozocin-induced diabetic rats. R. arboreum bark extract in ethanolic solution was prepared by soxhlatation method and stored in refrigerator for further use. The plant extract was tested for its hypoglycaemic property in glucose overloaded albino rats. The effect of extract was evaluated for its anti-diabetic action in STZ induced diabetic rats on 7, 14, 21 and 28 days respectively.  Treatment with the ethanolic extract of Rhododendron arboreum significantly decreased blood glucose in diabetic rats and glucose overloaded rats, where as the extract also improved the others biochemical parameters associated with diabetes like total cholesterol, triglycerides and LDL were reduced and HDL level was increased .The cardioprotective and nephroprotective qualities of the extract was determined by measuring biomarker LDH in diabetic rats. The histopathology of pancreas was studied to support the experimental results. The histology of pancreatic islet cells was normal in non-diabetic animals, whereas in diabetic treated rats. The significant percentage reduction of glucose level was found to be 44.78% at the dose of 200 mg/kg of aqueous extract of Rhododendron arboreum at 28th day of the treatment.

Objective: The objective of the present study was to evaluate the thyroid hormone stimulating efficacy of Acacia senegal (Gum Arabic) bark extract in 6-n-propyl-2-thiouracil (PTU)-induced hypothyroid albino rats.Methods: Ethanolic extract of A. senegal bark was given to PTU-induced hypothyroid albino rats at the dose of 500 mg/kg body weight. The animals were divided into control, PTU, and A. senegal bark extract treated and standard drug Eltroxin-treated groups for 60-day experimentation. The serum thyroxin levels, organ’s weight, and serum biochemistry were carried out.Results: Oral administration of A. senegal bark extract caused a highly significant increase (p≤0.001) in serum triiodothyronine (T3) and tetraiodothyronine (T4), and significant changes were also observed in organ’s weight with biochemical parameters as compared to hypothyroid albino rats and results were approximate to the standard drug Eltroxin.Conclusion: These results evaluate that ethanolic extract of A. senegal bark helps to compensate or increases the serum thyroxin level in management of hypothyroidism.

Diabetes mellitus develops either due to insufficient insulin secretion or lack of insulin resulting from damaged pancreas beta cells. Dyslipidaemia is an acute complication in diabetes mellitus. The aim of this study was to investigate the extenuating effects of T. cattapa leaves and P. americana seed aqueous extracts on streptozotocin-induced lipids profile perturbation and pancreatic damage in experimental rats. Thirty male albino rats were randomly divided into six groups (n = 5). Group 1: (negative control), group 2: 80 mg/kgbwt streptozotocin (positive control), group 3: (80 mg/kgbwt streptozotocin + 200 mg/kgbwt T. catappa leave extract), group 4: (80 mg/kgbwt streptozotocin + 200 mg/kgbwt Persea americana seed extract), group 5: (80 mg/kgbwt streptozotocin + 200 mg/kgbwt extracts-mixture), group 6: (80 mg/kgbwt streptozotocin + 5 mg/kgbwt glibenclamide (standard drug). A single dose of streptozotocin was administered to the rats intraperitoneally; the extracts and glibenclamide were administered orally for 21 days, after which their pancreas was excised for histology. Thereafter, extracts of T. catappa leaves and P. americana seed on serum lipid levels imbalance in streptozotocin-induced diabetic rats were investigated. Histological studies showed a degenerative effect of the pancreatic islet cells of strotozotocin-induced groups. Results showed improvement in the regulation of lipid metabolism as well as the regeneration of the beta cells of pancreas on treatment with T. catappa leaves and P. americana seed. The extracts both equally exert significant antidyslipidemic effects in diabetic rats, in view of the extenuating effects of the extracts on pancreatic islet cells.

Background: Diabetes an epidemic that gives rise to an increased macro vascular complication mortality rate of 1.5 million annually, increasing by 2035. Treatment of diabetes is complicated and associated with a severe side effect.</p> <p>Objective: To evaluate the antihyperglycemic property of the ethanolic extract of the back of MYCA and its toxicological effect on the hematological parameters with histopathological analysis of selected organs of induced diabetic albino rats.</p> <p>Method: The ethanoic extract (1000 mg/kg bw and 500mg/kg bw) and glibenclamide (0.5mg/kg bw) were administered to diabetic induced rats.  The effect of the extract on the hematological was studied in the diabetic induced rats. Histopathological changes were observed in the liver of induced diabetic rats after the administration of the MYCA extract.</p> <p>Results: The anti-hyperglycaemic effect was observed at two different levels, for two different doses (1000mg/kg and 500mg/kg). The results also showed significant suppression of blood glucose level in glucose fed hyperglycaemic albino rats but showed no significantly suppressed glucose level in overnight fasted normoglycemic rats.</p> <p>Conclusion: The findings revealed that the ethanolic extract of MITRAGYNA CILIATA (MYCA) possess antihyperglycemic property. Besides, the extract can prevent the various complication of diabetes and improve some hematological parameters.

This study aimed to understand Strychnosnuxvomica and Holarrhena pubescens Stem bark extract action towards M3 receptor in controlling blood glucose levels. Strychnos nux vomica  and Holarrhena pubescens are both alkaloidal drugs can help in controlling Hyperglycemic level. This will be useful in the formulation of a new herbal drug molecule for treating diabetes. Chloroform and ethanolic extracts of selected alkaloidal plants were extracted using the soxhlet apparatus and obtained quotes were tested for acute toxicity studies and carried out anti-diabetic action on Wister albino rats for 21 days. Results obtained from Blood glucose levels and histopathological study of test groups are compared with blood glucose levels of standard group, and highly significant action was identified by the chloroform extract of Strychnos nux vomica and Holarrhena pubescens group. Moderate anti-diabetic action was observed remaining two groups of ethanolic extracts. Strychnos nux vomica and Holarrhena pubescens ethanolic extract groups are acting on M3 receptors and controlling Hyperglycemic levels.

Background: The antipyretic, analgesic and anti-inflammatory activities of two concentrations (100 and 200 mg/kg) of ethanolic extract of leaf, bark, flower and fruit pulp of C. fistula were determined in male wistar albino rats. Methods: Antipyretic activity was assessed by E. coli endotoxin induced pyrexia. Analgesic activity was assessed by hot plate, tail immersion and acetic acid induced writhing test. Anti-inflammatory activity was evaluated by carrageenan-induced rat paw edema assay. Result: Significant (p less than 0.05) antipyretic activity was exhibited from 2h onwards by bark extract @ 200 mg/kg and from 3h onwards by bark extract @100 mg/kg and leaves extract @ 200mg/kg as compared to control group. Significant (p less than 0.05) analgesic activity was shown by extract of bark @ 200 mg/kg as it is evident by increase in reflex time in hot plate (90,120,180 min), tail immersion test (120,180 min) and inhibition of writhing (32.12%). Significant (p less than 0.05) anti-inflammatory activity was exhibited from 3h post administration by bark @ 200 and leaves @ 100 and 200 mg/kg.

Background: Appreciable number of medicinal plants are used for the treatment of diabetes in Nigeria. Aim of the Study: The present study aimed to investigate the antidiabetic activity of Zingiber officinale extracts and its potential mechanisms in streptozotocin-induced diabetic rats. Study Duration: The period of the study was done on 30th September, 2018 at the Department of Biochemistry, Faculty of Basic Medical Sciences, university of Jos, Nigeria. Methodology: Albino rats of Wistar strain weighing between 130 g to 160 g were induced with single freshly prepared streptozotocin (55 mg/kg body weight). Diabetes was confirmed after forty eight hours in streptozotocin -induced rats showing fasting blood glucose levels > 10 mmol/l. The rats were randomly divided into four (4) experimental groups (n = 4). A (Control diabetic group fed with normal feed), Group B (Normal control fed with normal feed), Group Group C (Diabetic rats treated with 400 mg/Kg body weight extract of ginger and Group D, (Diabetic rats are treated with 400 mg/Kg body weight of metformin). After 8 days the animals were sacrificed and blood samples were collected for biochemical and hematological analysis. Changes in the animal body weights were also measured within the period. Results: From the results, it was observed that treatment of rats with extract of ginger compensates for the reduction of body weight, and caused an increase in the body weight of the treated rats (+11.5%) in contrast to 24.8% reduction observed in diabetic control. In the same order, serum glucose significantly decreased (p<0.05) after the 8-day treatment compared to diabetic control. The extent of reversal of hyperglycemia in the ginger extract treated animals compared well with the metformin treated group. The results, therefore, showed that ginger extract has a significant (p<0.05) hypoglycemic effect in diabetic rats and moreover, elevations in the measured biochemical parameters were significantly (p<0.05) attenuated in rats treated with ginger extract. Conclusion: Zingiber officinale extracts has a significant effect on some biochemical parameters and hematological assays. These provide scientific evidence to confirm the traditional use of Z. officinale in the treatment of diabetes mellitus.

Background:Effective β-cell regeneration is a recognized therapeutic strategy in the treatment of type 1 diabetes mellitus. Regeneration of β-cells could be achieved via exogenous natural sources as medicinal plant extracts. Medicinal plants selected for the investigation were Spondias pinnata (Linn. f.) Kurz, Coccinia grandis (L.) Voigt and Gmelina arborea Roxb. The objective was to determine the β-cell regenerative potential of these plant extracts in alloxan-induced diabetic rats. Alloxan monohydrate was used to induce diabetes (150 mg/kg, ip).Methods:Wistar albino rats were divided into six groups (n=6); healthy untreated rats (healthy control), alloxan-induced diabetic untreated rats (diabetic control), diabetic rats received the extracts (treatment groups) of S. pinnata (1.0 g/kg), C. grandis (0.75 g/kg), G. arobrea (1.00 g/kg) and diabetic rats received glibenclamide (0.5 mg/kg; positive control). The above treatment was continued for 30 days. On the 30th day, the rats were sacrificed and biochemical parameters were determined. In addition, histopathology and immunohistochemistry on the pancreatic tissue were done on the 30th day.Results:According to the results obtained for biochemical parameters, there was a significant increase in the concentrations of serum insulin and C-peptide in plant extracts treated diabetic rats (p < 0.05). The extract of C. grandis produced the highest degree of β-cell regeneration demonstrated through an increase in the number of islets and percentage of the insulin-secreting β-cells (75%) in the pancreas of diabetic rats (p < 0.05) based on the histopathology and immunohistochemistry findings.Conclusion:The results revealed that the selected extracts of C. grandis (0.75 g/kg), G. arborea (1.00 g/kg) and S. pinnata (1.00 g/kg) exerted β-cell regenerative potential in diabetic rats. The three plant extracts would be valued as natural agents of prompting the β-cell regeneration in vivo.
