Purpose and objectives. To reveal new possibilities for the expression of cannabis genotypes to obtain starting material with increased contents of non-psychotropic cannabinoids, without tetrahydrocannabinol and stabilized biological traits. Material and methods. In our study, we took Ukrainian hemp cultivars, the populations of which were distinguished by high stability of cannabinoid contents in comparison with foreign cultivars, as a basis. The study was carried out using breeding, biochemical and statistical methods: physical mutagenesis, inbreeding, individual selection, thin-layer and gas-liquid chromatography to evaluate plants for cannabinoids (HP 6890 Series Hewlett Packard chromatograph using an internal standard). Results and discussion. It was found that gamma rays at a dose of 200 Gy disrupted the genotypic relationships between cannabinoids, with accumulation of their minimum or maximum amounts. Lowering the irradiation dose to 150 Gy does not destabilize the population in terms of the cannabinoid contents. Inbreeding can be also used in breeding for both to create inbred lines that are homozygous for the ‘no cannabinoids’ trait and to identify lines with increased contents of cannabidiol, cannabigerol or cannabichromen. A lot of early-bred and modern cultivars (French, Polish, Hungarian, German) show a high population variability of the cannabinoid contents, proving that no systemic breeding was conducted to stabilize these traits in the populations of these cultivars. High homogeneity of populations in terms of the formation and accumulation of cannabinoids is only intrinsic to monoecious hemp cultivars bred by the Institute of Bast Crops of NAAS. The homozygosity of a population primarily depends on the control of targeted cross-pollination between cannabinoid-free plants using an express testing plants for these compounds prior to anthesis onset and subsequent analysis of the selected elite plants by thin-layer chromatography. Given the relevance of creating hemp varieties with therapeutic properties, breeding methods have been developed to shift the balance of ‘no cannabinoid at all’ towards a genetically determined increase in one of the cannabinoids (cannabidiol, cannabigerol, cannabichromene) combined with eliminated psychotropic activity (the THC content does not exceed 0.08%). Studies of cannabinoid accumulation revealed differences in their formation during the growing period. Based on the fact that the maximum accumulation of cannabinoids occurs during the seed ripening onset, and, in particular, their synthesis is the most intensive in perianths and small leaves, it is possible to evaluate the prospects for creating hemp cultivars with a branched diamond-shaped inflorescence, which will provide, in addition to a high content of one or another cannabinoid, an increased yield of plant biomass. Conclusions. Physical mutagenesis, inbreeding and selection are promising breeding methods for reducing psychotropic tetrahydrocannabinol, stabilizing cannabis cultivars in terms of the absence of cannabinoids, increasing the contents of cannabidiol, cannabigerol and cannabichromene gaining advantage from valuable collection accessions

The molecular specificity in the biosynthesis of diacylglycerols by rat intestinal mucosa was examined by means of radioactive markers, thin-layer chromatography with silver nitrate and gas-liquid chromatography with radioactivity monitoring. Bile salt micelles of alternately labeled monoacylglycerols and free fatty acids were incubated with everted sacs of intestinal mucosa for various periods of time and the diacylglycerols were isolated by solvent extraction and thin-layer chromatography. Stereospecific analyses of the X-1,2-diacylglycerols labeled from 2-monoacylgiycerols showed that the sn-1,2-isomers (45–55%) were slightly in excess of the sn-2,3-isomers (34–45%) with the X-1,3-diacylglycerols accounting for the rest of the radioactivity (5–10%). This suggests that racemic diacylglycerols may be intermediates in the resynthesis of dietary fat in rat intestinal mucosa. Detailed analyses of the molecular species of the sn-1,2-diacylglycerols labeled from free fatty acids revealed that 10–45% of the total did not contain the acid present in the 2-monoacylglycerol supplied, and therefore had originated from the phosphatidic acid pathway. These findings are at variance with those obtained in isolated microsomes, which have suggested an inhibition of the phosphatidic acid pathway by monoacylglycerols as well as have given evidence of an exclusive synthesis of sn-1,2-diacylglycerols from 2-monoacylglycerols.

The occurrence of abscisic acid (ABA) in the dormant shoots of Douglas-fir (Pseudotsuga menziesii (Mirb.) Franco) was confirmed by bioassay, thin-layer chromatography, gas–liquid chromatography, and gas–liquid chromatography – mass spectrometry. Seasonal variation of ABA in the buds, leaves, and stems was then determined using 2-trans-ABA as an internal standard. Concentrations of ABA were highest in the autumn for buds (2.1 μg/g) and needles (0.79 μg/g) and highest in January for stems (0.34 μg/g). The lowest concentrations for all tissues were in February and March, before bud burst. Close correlation of levels of ABA with previously measured physiological evidence of growth and metabolic activity suggests a possible role in the dormancy cycle of Douglas-fir.

The article shows the importance of the study of feed for broiler chickens on the content of organochlorine pesticides. These substances in significant concentrations can disrupt the development of birds and cause mass poisoning. The study of formula feed for broiler chickens «prestart» and «start» by two methodswas conducted: a certified method for the determination of organochlorine pesticides in feed and formula feed (thin layer chromatography) and a method for detecting pesticides not certified for feed (gas-liquid chromatography with mass spectrometer detector). During the experiment, the presence of organochlorine pesticides in several formulafeed samples was revealed.

Abstract Gas-liquid chromatography (GLC) following conventional isolation procedures has been used to clean up pesticide residues for confirmation by thin layer chromatography (TLC). This procedure is more rapid and efficient than present cleanup procedures and permits the determination of pesticide residues that do not survive these rigorous acid or alkali treatments. The method also permits TLC confirmation of pesticide residues that have similar Rf values but different GLC retention times. Recoveries ranged from 85 to 105% for 25 of 28 pesticides tested

Abstract The Onley-Yip procedure for determining ethylenethiourea (ETU) in milk and crops was modified to reduce interferences by the ethylenebisdithiocarbamates (EBDCs). A 20 g cropmethanol extract is cleaned up by adsorbing the sample onto Gas-Chrom S, desorbing ETU, and eluting ETU from aluminum oxide with chloroform containing ethanol. ETU is converted to the S-butyl derivative for gas-liquid chromatography (GLC) and flame photometric detection (sulfur mode). For liquid chromatography (LC), ETU is cleaned up on another aluminum oxide column and injected directly. LC and GLC results are confirmed by thin layer chromatography. A cooking procedure based on conversion of EBDCs to ETU is included for surveying crops for possible EBDC content. Recoveries from 8 crops and milk fortified at 0.05 ppm ETU ranged from 73 to 100%.

Abstract Thin-layer chromatography (TLC)–densitometry was used to separate, identify, and quantitate clotrimazole, miconazole, and ketokonazole (alone or combined with other drugs) in various pharmacopoeial or proprietary creams and ointments. Clotrimazole was extracted from the cream or ointment with ethyl alcohol, and miconazole and ketokonazole were extracted with a mixture of equal volumes of chloroform and isopropyl alcohol. Active ingredients were separated from excipients and other drugs by TLC on a precoated silica gel F254 plate with a solvent system of n-hexane–chloroform–methanol–diethylamine (50 + 40 + 10 + 1, v/v). The 3 azoles were well separated and easily identified in this chromatographic system. The separated azoles were visualized under short-wave UV light and quantitated by scanning densitometry at 220 nm by comparing the integrated areas of samples with those of standard (one azole was used as internal standard for the other). Recoveries from samples spiked with known amounts of azoles were excellent. The method was validated further by comparison with official liquid chromatographic methods.
