Two strains of lactic acid bacteria were selected for the study (Lactobacillus sakei Lsk-45andLactobacillus sakeiDSM 20017). Bacterial media, based on rice and rice flour, were tested as an alternative to media based on whey. A comparison of the different types of media showed that there was better growth of the selected strains on themedium based on rice flour.Statistical analyses, including factorial experiments and response surface analyses, were used to optimizethe composition of the bacterial medium for Lactobacillus sakei propagation. Bacteriological peptone and rice flour were found to be good growth factors for Lactobacillus sakei.For Lactobacillus sakei Lsk-45,better growth was obtained with the use of 7.75-10 g/L of peptone and 57.5-75 g/L of rice flour. For Lactobacillus sakei DSM 20017, better growth was obtained with the use of 7-10 g/L of peptone and 40-75 g/L of rice flour. Lactic acid bacteria (LAB) has been applied as probiotic mainly to prevent pathogens. This group of bacteria can be found in marine fish products including fermented, shrimp product, cincaluk. The objective of this research was to isolate LAB from cincaluk, and examined the activity against pathogen V. alginolyticus and A. hydrophila. The research was conducted on March-Juny, total of 12 isolates were obtained from cincaluk survey method. The result of research showed that all isolates round, creamy white, smooth-edged, raised surface, and the size is 1.0 to 3.0 mm. The physiological and biochemical test showed that get all isolates had a Gram-positive, catalase positive and are indole negative, 10 isolates were motile, as many as five isolates have citric positive, 10 isolates is Methyl Red (+), all isolates ferment sucrose and lactose, while only 9 isolates ferment glucose, and as much as 6 isolates that produce sulfide. All isolates were found to have inhibitory effects against bacteria V. alginolyticus and A. hydrophila. The highest antimicrobial activity is indicated by B isolates with an average diameter of 12.3 mm and inhibition zone antivibrio activity of 6.3 units. Isolates B with an average diameter of inhibition zone of 9.7 mm and 3.7 units antiaeromonas activity.

Lactic acid bacteria (LAB) are widely used in the food industry due to their probiotic properties and fermentation activities. Traditional fermented fish products are dominated by a diverse variety of lactic acid bacteria with significant probiotic characteristics. Several in vitro and in vivo studies on lactic acid bacteria from fermented fish products have confirmed LAB strains to possess characteristics to be considered as probiotics that contribute to positive health benefits to the host and are generally regarded as safe (GRAS). This paper presents a review of the characteristics of the LAB strain that is considered as a probiotic. It also presents an overview of the probiotics mechanism of action and specifically highlights the LAB species with potential probiotic characteristics isolated from traditional fermented fish products.

Isolation of lactic acid bacteria from fermented fish products, peda, jambal roti and bekasam were carried out. Antibacterial activity of lactic acid bacteria was tested using well diffusion method against Escherichia coli, Bacillus cereus, Pseudomonas aeruginosa and Staphylococcus aureus. Twelve isolates of lactic acid bacteria had been isolated. Results showed that isolate A2 from peda was able to inhibit the growth of the 4 testing bacteria with inhibition zones of 6.7 mm for E. coli; 7.3 mm for P. aeruginosa; 8.3 mm for S. aureus and 10.0 mm for B. cereus. Isolate B5 from jambal roti had inhibition zones 11.6 mm for E. coli; 6.0 mm for P. aeruginosa ; 7.3 mm for S. aureus and 13.3 mm for B. cereus ; while isolate C6 from bekasam had inhibitor zones 7.7 mm for E. coli; 6.0 for P. aeruginosa; 8.0 mm for S. aureus and 9.3 mm for B. cereus.
