The cardiovascular response to sepsis includes an early, hyperdynamic phase followed by a late, hypodynamic phase. Although administration of pentoxifylline (PTX) produces beneficial effects in sepsis, it remains unknown whether this agent prevents the transition from the hyperdynamic to the hypodynamic response during the progression of sepsis. To study this, male adult rats were subjected to polymicrobial sepsis by cecal ligation and puncture (CLP). At 1 h after CLP, PTX (50 mg/kg body wt) or vehicle was infused intravenously over 30 min. At 20 h after CLP (i.e., the late stage of sepsis), cardiac output and organ blood flow were measured by radioactive microspheres. Systemic and regional (i.e., hepatic, intestinal, and renal) oxygen delivery (Do 2) and oxygen consumption (V˙o 2) were determined. Moreover, plasma levels of lactate and alanine aminotransferase (ALT) were measured, and histological examinations were performed. In additional animals, the necrotic cecum was excised at 20 h after CLP, and mortality was monitored for 10 days thereafter. The results indicate that cardiac output, organ blood flow, and systemic and regional Do 2decreased by 36–65% ( P < 0.05) at 20 h after CLP. Administration of PTX early after the onset of sepsis, however, prevented reduction in measured hemodynamic parameters and increased systemic and regional Do 2 andV˙o 2 by 50–264% ( P < 0.05). The elevated levels of lactate (by 173%, P < 0.05) and ALT (by 718%, P < 0.05), as well as the morphological alterations in the liver, small intestine, and kidneys during sepsis were attenuated by PTX treatment. In addition, PTX treatment decreased the mortality rate from 50 to 0% ( P < 0.05) after CLP and cecal excision. Because PTX prevents the occurrence of hypodynamic sepsis, this agent appears to be a useful adjunct for maintaining hemodynamic stability and preventing lethality from sepsis.

AbstractBackground and Aims: Various types of infertility are associated with uncontrolled hyperglycemia and diabetes. Development of oxidative stress is one the most important factors in the alteration of spermatogenesis in diabetic conditions. Consequently, the reduction of oxidative stress with antioxidant compounds can be effective in the reduction of tissue alterations. The aim of this study was to evaluate the efficacy of coenzyme Q10 in improvement of spermatogenesis in adult diabetic rats. Material and Methods: 32 adult rats were divided into four groups of control and treatment. Coenzyme Q10 (10 mg/kg body weight - b.w.) was administrated to one control and one diabetic (intraperitoneal injection of 45 mg/kg b.w. of Streptozotocin) groups. Blood concentrations of FSH, LH and Testosterone were measured. Histology of testicular tissue and sperm analysis were considered for evaluation of spermatogenesis. Results: Administration of Coenzyme Q10 led to increase of pituitary gonadotropins levels in diabetic rats. Testosterone levels were not changed significantly. Testicular morphology, spermatogenic indices and sperm analysis were improved in treated diabetic rats. Conclusions: The results of this study suggest that the use of Coenzyme Q10 has positive effects in reduction of spermatogenic alterations following induction of experimental diabetes in rats.

Abstract: Infertility is an inability of couples to reproduce after carrying out sexual intercourses for at least a year without using any contraceptives. There are 10-15% couples worldwide who experience infertility problems and almost one half of them is on men. Spermatozoa qualities including concentration, motility, and morphology are used as an indicator for men’s fertility. The utilization of herbal medicine (i.e. Eurycoma longifolia Jack extracts) is now being an alternative way to improve fertility among the community. This study was carried out to find the effects of Eurycoma longifolia Jack on spermatozoa qualities. This experimental study was conducted to nine male wistar rats (Rattus norvegicus) weighing from 200-250 grams, aging from 12-15 months. These nine adult rats were divided into 3 groups of 3 rats each. 400 and 600 mg/kg/day of Eurycoma longifolia Jack extracts were orally administrated to group 1 and 2 respectively, while group 3 were treated as control group. After 50 days, the animals of group 1, 2 and 3 were sacrificed. As a result, there is an improvement in qualities of spermatozoa which are statistically significant (p<0,05) in group 2. Thus, provide the beneficial effects of 600mg/kg Eurycoma longifolia Jack extracts on the qualities of spermatozoa.Keywords: pasak bumi, spermatozoaAbstrak: Infertilitas adalah ketidakmampuan pasangan untuk dapat hamil setelah satu tahun berhubungan intim tanpa menggunakan kontrasepsi. Penggunaan tanaman herbal atau yang lebih dikenal jamu telah menjadi pengobatan alternatif di masyarakat. Tanaman herbal yang dimaksud adalah pasak bumi. Penelitian ini bertujuan untuk mengetahui efek dari esktrak pasak bumi (Eurycoma longifolia Jack) terhadap kualitas spermatozoa. Penelitian ini menggunakan 9 ekor tikus wistar jantan (Rattus norvegicus) dengan berat badan 200-250 g berumur 12-15 bulan. Sembilan ekor wistar dibagi menjadi tiga kelompok, dan satu kelompok terdiri dari tiga ekor tikus wistar. Penelitian ini dilaksankan selama 50 hari. Esktrak pasak bumi (Eurycoma longifolia Jack) diberikan dengan dosis 400mg/kgBB per hari pada perlakuan 1 dan dosis 600 mg/kgBB per hari pada perlakuan 2. Setelah 50 hari hewan coba pada kelompok kontrol, perlakuan 1 dan perlakuan 2 diterminasi. Hasil penelitian memperlihatkan terjadi peningkatan kualitas spermatozoa yang signifikan secara statistik (p<0,05) pada perlakuan 2 terhadap kelompok kontrol. Hasil tersebut menunjukan bahwa Esktrak pasak bumi (Eurycoma longifolia Jack) pada dosis 600 mg/kgBB dapat meningkatkan kualitas spermatozoa.Kata kunci: pasak bumi, spermatozoa

We investigated the osteogenic responses to jump training and subsequent detraining in young and adult male rats to test the following hypotheses: 1) jump training has skeletal benefits; 2) these skeletal benefits are preserved with subsequent detraining throughout bone morphometric changes; and 3) there are no differences between young and adult rats during detraining in terms of the maintenance of exercise-induced changes. Twelve-week-old (young) and 44-wk-old (adult) rats were divided into the following four groups: young-sedentary, young-exercised, adult-sedentary, and adult-exercised. The exercised groups performed jump training (height = 40 cm, 10 jumps/day, 5 days/wk) for 8 wk followed by 24 wk of being sedentary. Tibial bone mineral content and bone mineral density in vivo significantly increased with jump training, and the effects were maintained after detraining in both the young and adult exercised groups, although the benefits of training became somewhat diminished. After 24 wk of detraining, the beneficial effects of training on bone mass and strength were preserved and associated with morphometric changes, such as periosteal perimeter, cortical area, and moment of inertia. There were no significant age-exercise interactions in such parameters, except for the periosteal perimeter. These results suggest that there are few differences in bone accommodation and maintenance by training and detraining between young and adult rats.

There has been increasing evidence suggesting that a severe caloric restriction (SCR) (above 40%) has beneficial effects on the hearts of rats. However, most of the reports have focused on the effects of SCR that started in adulthood. We investigated the consequences of SCR on the hearts of rats subjected to SCR since birth (CR50). From birth to the age of 3 months, CR50 rats were fed 50% of the food that the ad libitum group (AL) was fed. Thereafter, a maximal aerobic test was performed to indirectly evaluate global cardiovascular function. Indices of contractility (+dT/dt) and relaxation (−dT/dt) were analyzed in isolated heart preparation, and cardiomyocyte diameter, number, density, and myocardium collagen content were obtained through histologic analysis. Ventricular myocytes were isolated, using standard methods to evaluate phosphorylated AKT levels, and Ca2+ handling was evaluated with a combination of Western blot analysis, intracellular Ca2+ imaging, and confocal microscopy. CR50 rats exhibited increased aerobic performance and cardiac function, as shown by the increase in ±dT/dt. Despite the smaller cardiomyocyte diameter, CR50 rats had an increased heart−body weight ratio, increased cardiomyocyte density and number, and similar levels of myocardium collagen content, compared with AL rats. AKT was hyperphosphorylated in cardiomyocytes from CR50 rats, and there were no significant differences in Ca2+ transient and SERCA2 levels in cardiomyocytes between CR50 and AL rats. Collectively, these observations reveal the beneficial effects of a 50% caloric restriction on the hearts of adult rats restricted since birth, which might involve cardiomyocyte AKT signaling.
