Protein Digestibility of Kapok Seeds in Vitro for Fish Feed        Kapok seeds is a by-product of agricultural industry having potential to be used as raw material for fish feed as a source of protein and essential fats. The content of the protein in the kapok seed  flour is 28 – 34 % that is an overwhelming amount to be a great source of protein for fish feed. Feasibility studies are needed, however, the use of kapok seed based on the digestibility of the protein. Pepsin with concentrations of 0.02; 0.2; and 2 % (in 0.075 N HCl solution) kapok seed flour added in three repetitions to test the digestibility of kapok seed invitro. Undigested protein was then analyzed using the kjeldahl method. Determination optimal concentration of pepsin was calculated based on the remaining undigested proteins (pepsin indigest) and compared the amount of protein digestibility of proteins obtained before (pepsin digest). The research results revealed that to digestibility of protein on concentration of 0.02; 0,20 and 2,0 % was 64,43; 68,42; 65,33%. Statistical test Anova revealed any significant differences of treatment respectively of  protein digestibility. Test of Least Square Difference (LSD) stated that each  treatment significantly different. Concentration optimum of enzyme that givethe best digestibility value was 0.20 % digestibility values 68,43% in the level of  error 0.05.Keywords : Kapok seed flour, protein, pepsin, invitro, optimum digestibility, proximate analysis ABSTRAK          Biji kapuk merupakan hasil samping industri pertanian yang berpotensi untuk dijadikan bahan baku pakan ikan sebagai sumber protein dan sumber lemak esensial. Komposisi protein pada tepung biji kapuk sebesar 28‑34% adalah jumlah yang sangat potensial untuk dijadikan sumber protein bagi pakan ikan. Akan tetapi diperlukan kajian kelayakan penggunaan biji kapuk berdasarkan kecernaan protein.Pepsin dengan konsentrasi 0,02; 0,2; dan  2%(larutan dalam HCl 0,075 N) ditambahkan pada tepung biji kapuk dengan tiga kali pengulangan untuk menguji kecernaan biji kapuk secara invitro. Protein yang tercerna kemudian dianalisis menggunakan metode kjeldahl. Penentukan konsentrasi  pepsin optimal dihitung berdasarkan sisa protein yang tidak tercerna(pepsin indigest) dan dibandingkan jumlah protein awal sehingga didapatkan kecernaan protein (pepsin digest).Hasil penelitian menyatakan kecernaan protein pada  konsentrasi 0,02;0,2%;dan 2%  berturut turut 64,43; 68,42; 65,33%. Uji statistik Anova menyatakan setiap perlakuan memberikan perbedaan yang signifikan terhadap kecernaan protein. Uji Least Square Difference (LSD) menyatakan setiap perbandingan perlakuan berbeda. Konsentrasi enzim optimum yang memberikan nilai kecernaan terbaik adalah 0,20% (68,43%) pada tingkat kesalahan 0,05.Kata kunci: Tepung biji kapuk, protein, pepsin, invitro, kecernaan optimum, proksimat

Feed is still one of the problems in aquaculture because it takes about 70% of operational costs. High prices is caused by imported raw materials for feed production. One possible effort to overcome this problem is to find alternative local raw materials to replace imported fish meal. The purpose of this study was to examine the proximate content of shrimp shell wastes from processing shrimp crackers (Paneus sp.) to be used as shrimp flour. Samples were colledted three times in a month, at the beginning, middle and end of the month. The waste of the heads, shells and tails of the shrimp were turned into flour for laboratory testing through proximate analysis. The data were tabulated using the Microsoft Excel softwareand then analyzed descriptively by comparing the nutrient content of shrimp waste flour with the feed requirements of mariculture. The results showed that shrimp shell flour contained protein, fat, carbohydrates and ash content of around 27.4%, 2.07%, 14.84% and 47.27%. while the resulting randemen ranged from 81.75%. Therefore, shrimp shell flour can be used as an alternative raw material for preparing marine fish feed.

The objective of this work was to evaluate the Amino Acid profile/score and In-vitro protein digestibility of composite biscuits produced from blends of Wheat flour (WHF), African breadfruit flour (ABF)and Moringa seed flour(MSF) at the following ratios (Sample A: WHF 100%: ABF 0; MSF 0, B= WHF 77.5%:ABF 20%: MSF 2.5%, C=WHF 75%: ABF 20%: MSF 5.0%, D= WHF 72.5%: ABF 20%: MSF 7.5%. E = WHF 70%: ABF 20%: MSF 10%, F = WHF 90%: ABF 0: MSF 10%, G = WHF 80%: ABF 20%: MSF 0). The most predominant Amino Acid in ABF was glutamic (12.27 g/100 g) followed by Aspartic and lysine, with values of 8.96 g/100 g and 6.55 g/100 g, respectively. Glutamic Acid content of the biscuits ranged from 10.96 g/100 g – 12.96 g/100 g, with sample B giving significantly higher value. Substitution with MSF resulted in decreasing glutamic acid content levels in the formulated biscuits, while lysine, phenylalanine and Isoleucine improved with the addition of 10% Moringa seed flour. Amino acid Scores of the biscuits using Hen egg as standard showed that whole egg had a higher amino acid score except glycine (1.04 – 1.25). Percentage In-vitro protein digestibility ranged from 10.64% - 47.33%, showing that addition of moringa seed flour and African breadfruit flour improved digestibility values from 10.64% to 47.33% for sample E with the control sample (wheat flour biscuit) being significantly lower. Substitution with ABF and MSF improved protein digestibility of the produced biscuits. Also, the Amino acid scores of the formulated biscuits were higher than the FAO recommended daily dietary requirements for Amino acids.

The anti- microbial activity tests of the seed oil, NaOH and ash soaps of the seed oil, of C.albidum were carried out to determine their effectiveness against gram+, gram– bacteria and some fungi strains. The proximate analysis of C.albidum seeds was performed to determine its nutritional benefits to mankind. Agar diffusion tests using Disc diffusion for the pure oils and soaps while the cup plate test method for dilutions was used in the determination of in-vitro activities. Standard methods as described by AOAC, (1990); AOAC, (1984) and Kjeldahl method described by Chang (2003) were used for proximate determinations. The results show the activity of the oil against Klebsiella pneumonia at a concentration as low as 10 mg/ml. The ash-soap of the leaves shows the highest antimicrobial activities because its MIC is the closest to the standard, Streptomycin. It possesses activity against Escherichia coli and Klebsiella pneumonia at a concentration as low as 5mg/ml. The % proximate analysis composition shows Moisture content, Ash content, Crude Fibre, fat/Oil, Protein, Carbohydrates as follows (14.98, 1.90, 2.09, 2.38, 12.14 and 67.65)%. The study concluded by unleashing the great potential of Chrysophyllum albidum seeds as a useful raw material in food, cosmetics and pharmaceutical industries rather than being regarded as waste in the environment

Osteopenia is a sign of osteoporosis that usually occurs in adolescent aged 18-24 years. One of the efforts to manage osteopenia is by giving a high calcium diet. Meatballs substituted by tempeh gembus can be used as an alternative to local food-based snacks as source of calcium. To analyze the effect of the substitution of tempeh gembus on the protein content, calcium, digestibility of the protein, and the acceptability of meatballs. The study was conducted in two stages, there are preliminary study and main study, and using randomized single factor experimental, tempeh gembus levels (25%, 50%, 75%, 100%) and control (0% tempeh gembus). The analysis of the content of protein was conducted using Kjeldahl method, calcium contents using Uv-Vis spectrophotometer, and protein digestibility using in-vitro method. Data was analyzed with mann-whitney. Organoleptic tests were carried out by hedonic testing of 30 semi-trained panelists. Substitution of tempeh gembus had significant effect on the protein contents and protein digestibility, but not significant effect on the calcium contents. Formulation with 25% tempeh gembus substitution was found to be a best formulation of meatballs containing protein content of 8.03%, calcium contents of 351.19 mg / 100 g, and protein digestibility of 53.22%. Meatball with 25% tempeh gembus with 78 mg per serving is sufficient for nutrition label reference in general category, 13% energy; protein by 11%; calcium by 25%.

The cost of fish feed is a significant constraint to freshwater fish farming in Asia. In the aquaculture value chain, feed is a responsible factor and accounts for nearly 75% of the total cost of fish production in many countries. Therefore, this study attempts to focus the feasibility of using pig manure fermentation to formulate feed pellets to culture the tilapia fish in the cement pond, along with the cultured using ready-made feed containing the residue from the manure fermentation at different ratios. Accordingly, the experimental studies were approached by two types of experiments, namely, experiment 1 (focused on the nutrients in pig manure collected from an anaerobic system farm), and experiment 2 (studied the appropriate level of pig manure to be used as a raw material for fish feeding). The five months’ fermentation of pig manure contains 17.09% protein, which is the highest amount of nutrition source. Digestate manure was estimated; other primary macronutrients, including nitrogen, phosphorus, and potassium, were 2.73, 0.55 and 1.22 %, respectively. Also, a considerable amount of essential micronutrients, for example, copper, manganese and zinc, were found in the digestate. Nile tilapia were grown at the cement ponds for 120 days. The maximum verified dry matter and protein digestibility in tilapia were attained for diets containing 10 % percent pig manure at 89.28 ± 4.09 and 56.64 ± 1.43 (p> 0.05), respectively. In this study experimental results indicated that fish meal consisting of 15% manure was found to be the most acceptable substitute source of protein for tilapia production. This implies that the cost of fish meal production using fermented pig manure can be significantly lowered, gain higher profit and provide significant economic benefits to farmers.

Biscuits were produced from wheat, African walnut and Moringa seed flour blends and chemical, physical, in-vitro protein digestibility, ant-nutrient and sensory properties of the products were evaluated. Chemical compositions of the biscuit samples revealed that sample E was significantly higher (p < 0.05) in ether extract (30.16%) and ash (4.20%) while sample F had a significantly higher protein content of 11.41%. The weight of the biscuit samples increased with increasing substitution levels with Moringa seed flour from 9.73–12.08 g. The heights of the samples were not affected by substitution levels. In-vitro protein digestibility values of the biscuits showed remarkable improvement from 11.03% for the control sample to 69.03% for sample E. Anti-nutrient content of the formulated biscuits showed that oxalate had values ranging from 18.68–35.71mg/100g, phytate 0.61–9.21mg/100g, saponin 0.46–8.41%, trypsin inhibitor 2.31–6.80mg/100g, tannin 18.68–35.71 mg/100g and cyanide 0.02–0.44mg/100g. Sensory evaluation scores showed decreasing values in flavour and overall acceptability with increased levels of substitution with Moringa seed flour (7.5–10%) in the biscuit formulation. Although, these substitution levels led to an improvement in protein content, fibre and protein digestibility of the biscuit samples.

Mealworm larvae (Tenebrio molitor) are edible insects consumed in feed and food. In the current study, the optimal harvesting time of mealworm larvae for use as aquafeed was investigated during the ages of 30–90 days after hatching (DAH). Development of digestive enzymes, proximate composition, and in vitro protein digestibility using digestive enzymes from African catfish (Clarias gariepinus) and Nile tilapia (Oreochromis niloticus), were used as criteria. The specific activities of pepsin and trypsin significantly decreased with age (p < 0.05) from the first harvesting time until 50 and 45 DAH, respectively, while steadiness in these enzyme activities was observed onwards. Chymotrypsin specific activity appeared constant across all harvesting times. The specific activity of amylase significantly decreased in the later stages of development, while cellulase exhibited a different pattern suggesting it has a major role in dietary fiber utilization relative to starch. Regarding proximate compositions of the mealworm larvae, the moisture and ash contents decreased significantly with age, while the protein content exhibited the opposite trend with the highest contents from 60 to 90 DAH. Crude lipid was generally fairly constant, but its lowest value was observed in the earliest stage. In vitro protein digestibility was not significantly different across all harvesting times for both fish species, except for the significantly decreased digestibility value at 65 DAH relative to 30 and 35 DAH for Nile tilapia. However, based on the economic benefits of time for growth increment and proximate chemical composition, approximately 60 DAH is proposed as suitable for harvesting mealworm larvae to be used in fish feed.
