This research aimed to evaluate the in vitro effect of buffer extract obtained from leaves and pseudobulbs (modified shoots) of Dendrobium parishii Rchb. f. on the 2-thiobarbituric acid reactive substances (TBARS) as lipid peroxidation biomarker, aldehydic and ketonic derivatives of oxidatively modified proteins, and total antioxidant capacity (TAC) in the muscle tissue of the rainbow trout (Oncorhynchus mykiss Walbaum). The shoots (pseudobulbs) with leaves of Dendrobium parishii cultivated under glasshouse conditions were sampled at M.M. Gryshko National Botanic Garden (NBG) (Kyiv, Ukraine). Since 1999, the whole collection of tropical and subtropical plants (including orchids) has had the status of a National Heritage Collection of Ukraine and is supported through State funding. Besides, NBG’s collection of tropical orchids was registered at the Administrative Organ of CITES in Ukraine (Ministry of Environment Protection, registration No. 6939/19/1-10 of 23 June 2004). The collected pseudobulbs and leaves were brought into the laboratory for biochemical studies. Freshly collected leaves were washed, weighed, crushed, and homogenized in 0.1M phosphate buffer (pH 7.4) (in proportion 1:19, w/w) at room temperature. The extract was then filtered and investigated for its antioxidant capacity. The extract was stored at -20°C until use. The increase in TBARS level in the muscle tissue exposed to extracts derived from leaves and pseudobulbs of D. parishii was insignificant. The level of ketonic derivatives of oxidatively modified proteins was non-significantly decreased both for leaf and pseudobulb extracts compared to the untreated samples. The extracts obtained from leaves and pseudobulbs of D. parishii significantly increased the TAC level in muscle tissue due to inhibited the Fe2+/ascorbate-induced oxidation of Tween 80. Overall, these findings demonstrate that aqueous extracts of vegetative organs of Dendrobium parishii can enhance the total antioxidant capacity in the muscle tissue of the rainbow trout. Moreover, this antioxidant effect was more intensive for pseudobulb extracts.

AbstractIntroduction: The goal of this study was to assess the influence of vaccination against enteric redmouth disease on oxidative stress biomarkers and antioxidant defence in the muscle tissue of rainbow trout (Oncorhynchus mykiss Walbaum) vaccinated against Yersinia ruckeri in the first and second month after immunisation. Material and Methods: Healthy fish were vaccinated orally with inactivated whole cells of a virulent strain of Y. ruckeri. One and two months after immunisation the muscle samples were collected. Results: No significant difference was noted in lipid peroxidation level in either the first or second month after vaccination, while aldehydic and ketonic derivatives of oxidatively modified proteins (OMB) in the vaccinated group were significantly lower in the second month compared to those in the first month after vaccination (P < 0.05). The content of ketonic derivatives of OMB in muscles in the first month after immunisation was higher compared to untreated control. All these culminated in a depletion of glutathione peroxidase (GPx) activity and low level of total antioxidant capacity (TAC). Conclusion: Correlations between catalase activity and lipid peroxidation and TAC confirmed the pivotal role of catalase in antioxidant defence during immunisation. From a broader perspective, it is suggested that immunisation of fish with Yersinia vaccine is associated with induced free radical formation and oxidative stress. Free radicals would therefore be at least partially responsible for the induction of both humoral and cellular elements of the immunity and increased protective immunity against Y. ruckeri infection.

Background: Based on the structure of RC-121 (D-Phe-c (Cys-Tyr-D-Trp-Lys-Val-Cys)-Thr-NH2, - synthetic derivatives of somatostatin), some analogs were synthesized and tested for in vitro cytotoxic and antioxidant activity. Objectives: The new analogs were modifyed at position 5 with Dap (diaminopropanoic acid), Dab (diaminobutanoic acid) and Orn and at position 6 with the unnatural amino acids Tle (t-leucine). Methods: The in vitro cytotoxic effects of the substances were investigated against a panel of human tumor cell lines HT-29 (Human Colorectal Cancer Cell Line), MDA-MB-23 (Human Breast Cancer Cell Line), Hep G-2 (Human Hepatocellular Carcinoma Cell Line) and HeLa (cervical cancer cell line). The antioxidant capacities were tested by ORAC (Oxygen Radical Antioxidant Capacity) and HORAC (Hydroxyl Radical Averting Capacity) methods. Results: All substances expressed significantly higher antioxidant capacity by comparison with galic acid and Trolox. All substances showed considerable antioxidant capacity as well. Compound 2T (D-Phe-c(Cys-Tyr-DTrp- Dap-Tle-Cys)-Thr-NH2)had the highest antioxidant effect. The compound 4T (D-Phe-c(Cys-Tyr-D-Trp- Orn-Tle-Cys)-Thr-NH2) displayed antiproliferative effect on HeLa cells with IC50 30 µM. The peptide analog 3T (D-Phe-c(Cys-Tyr-D-Trp-Lys-Tle-Cys)-Thr-NH2) exerted the most pronounced inhibition on the cell vitality up to 53%, 56% and 65% resp. against MDA-MB-23, Hep G-2, HeLa in the higher tested concentration. Conclusion: The somatostatin analogs showed moderate influence on the vitality of different tumor cells and could be used in changing their pathology.
