Antioxidant peptides derived from natural products have superior performance and broader application prospects. In this study, five novel antioxidant peptides were prepared from Paeonia ostii (P. ostii) seed meal, moreover the bioactive and the relationship between structure and properties of antioxidant peptides were elucidated by quantum chemical calculations. The free radical-scavenging activities were used as indexes to purify and concentrate the antioxidant peptides through five proteases and separation techniques. FSAP (Phe-Ser-Ala-Pro), PVETVR (Pro-Val-Glu-Thr-Val-Arg), QEPLLR (Gln-Glu-Pro-Leu-Leu-Arg), EAAY (Glu-Ala-Ala-Tyr) and VLRPPLS (Val-Leu-Arg-Pro-Pro-Leu-Ser) were identified by nano liquid chromatography–tandem mass spectrometry (LC-MS/MS). In vitro antioxidant activity test, EAAY exhibited the highest 2, 2’-azino-bis (ABTS) and hydroxyl radical-scavenging activity of 98.5% ± 1.1% and 61.9% ± 1.3%, respectively (p < 0.01), at 0.5 mg/mL. In silico calculations were carried out using the density functional theory (DFT) with the B3LYP/6-31G* basis set. According to natural bond orbital (NBO) analysis, the bioactivity of free-radical scavenging of the peptides was presumed. Moreover, the antioxidant peptides demonstrated no obvious cytotoxicity to L929 fibroblast cells. Therefore, the peptides from P. ostii seed by-products might potentially have excellent uses in functional foods, nutraceuticals and pharmacological products.

<p><em>Centella asiatica</em> also known as <em>mandukparni </em>or Indian pennywort or <em>jalbrahmi</em>, which has been used as a medicine in the Ayurveda from ancient times and mentioned in many classical texts of Ayurveda. <em>Centella asiatica</em> has long been used to improve memory and cognitive function.</p><p>The study aimed to identify the phytochemicals present in different solvent extracts of <em>Centella asiatica </em>(i.e. PECA- Petroleum ether extract of <em>C. asiatica, </em>CCA- Chloroform extract of <em>C. asiatica, </em>EACA- Ethyl acetate extract of <em>C. asiatica,</em> ECA- Ethanolic extract of <em>C. asiatica, </em>HACA- Hydro-alcoholic extract of <em>C. asiatica</em>)<em> </em>and evaluate the respective in-vitro antioxidant potentials. <em></em></p><p>The phytochemical screening of extracts was done with standardized procedures and the antioxidant potential of different solvent extracts of <em>Centella asiatica</em> was assessed by its free radical scavenging activity 2, 2-diphenyl -1- picrylhydrazyl (DPPH) as well as hydrogen peroxide scavenging assay respectively for reducing capability.</p><p>In all different solvent extracts of <em>C. asiatica</em> revealed excellent free radical scavenging activity as revealed by 2-2- diphenyl-1-picryl-hydrazyl (DPPH) assay with  EC<sub>50</sub> values for ECA=128.752±1.85 μg/ml, HACA=274.884±1.21 μg/ml and hydrogen peroxide assay against the standard (Butylated hydroxytoluene) BHT, with the EC<sub>50</sub> values ECA=429.69±0.92 μg/ml HACA=458.08±0.58 μg/ml while rest solvent extracts shown very less antioxidant activity.</p><p> The present study indicates that the <em>Centella asiatica</em> extracts have good antioxidant activity which can be used in stress and anxiety and also a good source to be used as natural drugs.</p>

AbstractIn this study, an aqueous extract of leaves from Melothria maderaspatana was tested for in vitro antioxidant activity. Free radical scavenging assays, such as hydroxyl radical, hydrogen peroxide, superoxide anion radical and 2,2-diphenyl-1-picryl hydrazyl (DPPH), 2,2’-azinobis-(3-ethyl-enzothiazoline-6-sulfonic acid) (ABTS) radical scavenging, and reducing power assay, were studied. The extract effectively scavenged hydroxyl radical, hydrogen peroxide and superoxide anion radicals. It also scavenged DPPH and ABTS radicals. Furthermore, it was found to have reducing power. All concentrations of leaf extract exhibited free radical scavenging and antioxidant power, and the preventive effects were in a dose-dependent manner. The antioxidant activities of the above were compared to standard antioxidants such as butylated hydroxytoluene (BHT), ascorbic acid, and α-tocopherol. The results obtained in the present study indicate that the M. maderaspatana extract could be considered a potential source of natural antioxidant.

Pterygota alata is a large deciduous tree of Malvaceae family. The present study was designed to evaluate in vitro antioxidant activity of stem bark of the plant. Ferric reducing power (FRP) test, 1, 1- diphenyl-2-picryl hydrazyl (DPPH) free radical scavenging test and total antioxidant capacity (TAC) assay were used to detect the antioxidant activity. Total phenolic and flavonoid contents were also determined as they are well known phytochemicals with antioxidant property. The bark of the plant showed efficient reducing power as well as free radical scavenging property (IC50 values 52.25 - 172.05). The bark also found rich in total phenolic and flavonoid content. The highest amount of total phenolic content was found in chloroform soluble fraction (29.898 µg/mL) followed by ethyl acetate soluble fraction (15.88 µg/mL). The highest content of total flavonoid also detected in chloroform soluble fraction (107.56 µg/mL) but followed by crude ethanol extract (98.66 µg/mL). Overall, the bark of the plant possesses significant antioxidant activity, therefore can be used as a good natural source of antioxidant. © 2014 JSR Publications. Background: Pyrazole is an important class of heterocyclic compound, has been shown to exhibit diverse biological and pharmacological activities such as anti-inflammatory, anti-cancer, antioxidant, etc. Methods: In this study, a series of novel 3,5-dimethyl-1H-pyrazole derivatives containing hydrazine 4a-l have been synthesized via the reaction of the 2-(3,5-dimethyl-1H-pyrazol-1-yl)acetohydrazide. All synthesized compounds have been tested for their in vitro antioxidant activities via utilization of 1,1-biphenyl-2-picrylhydrazyl (DPPH) as a free radical scavenging reagent. Results: The data reported herein indicates that compound 4k showed potential radical scavenging capacity and compounds 4f and 4g exhibited best activity for the iron binding while comparing with positive controls. Conclusion: Good activity was noted for some compounds. In particular, compound 4k showed the highest antioxidant activity with IC50 values of 22.79 ± 3.64 and 1.35 ± 0.66 μg/mL in the DPPH and ABTS tests, respectively.

Plant polysaccharides have been increasingly employed in the pharmaceutical, industrial, and food environments due to their versatile functional properties. In the present investigation, a heteropolysaccharide galactomannan (GAP) was extracted from Adenanthera pavonina L. seeds and characterized by physicochemical analyses to determine its thermal properties, photostability, antioxidant activity, and acute toxicity. GAP was characterized by FTIR, DSC, and TG. The photostability of GAP submitted to artificial UV irradiation was analyzed. Antioxidant activity was evaluated by the DPPH (2,2-diphenyl-1-1-picrylhydrazyl) free radical-scavenging method, while a bioassay method was carried out to study acute toxicity in Artemia salina L. Physical-chemical and functional characteristics of GAP support its potential role in the food and pharmaceutical industries. GAP was photostable under UV irradiation. In vitro GAP antioxidant evaluation showed that it bears free radical-scavenging activity for DPPH radicals. The median lethal concentration (LC50) of GAP was 239.4 mg∙mL−1, indicating that this biopolymer is nontoxic. Such results indicate that this biopolymer presents characteristics of neutrality, photostability, and nontoxicity that are commercially attractive.

Objective: Four different extracts of Araucaria columnaris (bark peel) and Cosmos sulphureus (flowers) were screened for their phytochemical composition, and free radical scavenging activities.Methods: DPPH method was used to test the antioxidant activity for extracts.Results: Among the different extracts tested, the methanol extract of both the plant species showed significant radical scavenging activities. Phytochemical analysis of the extracts revealed that the radical scavenging activities might be due to the presence of flavonoids, tannins and phenolic compounds.Conclusion: The results obtained suggest that Araucaria columnaris (bark peel) and Cosmos sulphureus(flowers) could be exploited in the treatment of various diseases like cancer, cardiovascular diseases and infection diseases.

A series of quinazolines encompassed with thiazolidinone and azetidinone have been synthesized and evaluated for their antioxidant, anticancer and DNA binding studies. All the synthesized compounds were characterized by IR, 1H & 13C NMR and mass spectra. Antioxidant activity was carried out using % free radical scavenging by DPPH assay. Compounds 4b, 5b and 5d have shown better antioxidant activity (60, 67 and 66%, respectively) among the tested compounds. Compounds having % free radical scavenging activity more than 55% were evaluated for anticancer activity by MTT assay towards cell lines A-549 (lung carcinoma) and MDA-231 (human breast cancer). Results revealed that the tested compounds exhibited moderate to low anticancer activity. Further, DNA binding activity was studied by absorption titration method for all the synthesized compounds, and compound 5b showed a good binding constant of 70.05 and % hyperchromicity of 82.93%.

Ruta graveolens L. is a medicinal plant used in traditional systems of medicine for treatment of psoriasis, vitiligo, leucoderma, and lymphomas with well-known anti-inflammatory and anticancer properties. Therefore antioxidant potential of R. graveolens (in planta and in vitro) was investigated. As antioxidants present in plant extracts are multifunctional, their activity and mechanism depends on the composition and conditions of the test system. Therefore, the total antioxidant capacity was evaluated using assays that detect different antioxidants: free radical scavenging (DPPH and ABTS), transition metal ion reduction (phosphomolybdenum assay), reducing power, and nitric oxide reduction. Content of furanocoumarin-bergapten in the extracts showed good corelation with free radical scavenging, transition metal reduction and reducing power, while total phenolic content showed good corelation with nitric oxide reduction potential. Antioxidant activity of in vitro cultures was significantly higher compared to in vivo plant material. The present study is the first report on comprehensive study of antioxidant activity of R. graveolens and its in vitro cultures.
