Four types of traditional Thai rice—polished, black fragrant, glutinous and black glutinous rice—were separately used as raw material for vinegar production. During alcohol fermentation, using enriched baker’s dried yeast (S. cerevisiae) as a starter culture gave the highest ethanol content over 7 days of fermentation. The conversion of ethanol to acetic acid for vinegar production by Acetobacter pasteurianus TISTR 102 was performed for 25 days. The highest amount of acetic acid was detected with glutinous rice fermentation (6.68% w/v). The biological properties of Thai rice vinegars were determined, including the total phenolic content, antioxidant activity, antibacterial activity and cytotoxicity. Black glutinous rice vinegar exhibited the maximum total phenolic content of 133.68 mg GAE/100 mL. This result was related to the antioxidative activity findings, for which black glutinous rice vinegar exhibited the strongest activity against both ABTS•+ and DPPH• radicals. Cytotoxicity against the human colon cancer cell line (HT-29) provided an IC50 value of 74.02 μg/mL and weak activity in a mouse fibroblast normal cell line (L929) with an IC50 value of 171.06 μg/mL. Glutinous rice vinegar was the most effective vinegar for inhibiting pathogenic bacterial growth of both Gram-negative and Gram-positive bacteria. These results suggested that the value of total phenolic content corresponded to the anticancer activity and antioxidant activity results, while antibacterial activity depended on the acidity of rice vinegar.

In vitro antibacterial activity of 21 plant methanolic extracts were investigated by disc diffusion method against gram-negative bacteria Escherichia coli. Amongst the extracts tested, the Datura stramonium L. and Ocimum basilicum L. showed significant antibacterial activity against the bacterial pathogen. D. stramonium showed the highest antibacterial activity followed by O. basilicum extract. Phytochemical analysis of extracts showed the presence of alkaloids, tannins, saponins, flavonoids, and phenols. The 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging and ABTS analysis of potent extract D. stramonium showed appreciable antioxidant activity. For DPPH and ABTS assay the highest antioxidant activity was observed at 60 mg ml-1 concentration with a percent inhibition of 71.66 and 77.19 with IC50 value 9.71 and 22.16 μg ml-1 respectively. Moreover, the radical scavenging activity of the extract was lower than that observed for the synthetic antioxidants butylated hydroxyanisole (BHA )and butylated hydroxytoluene (BHT). The total phenolic content of D. stramonium extract was found to be 0.592 mg GAE/gm which was assayed using Folin ciocalteau reagent. The results provide evidence that the extract of D. stramonium and O. basilicum can be further recommended in the treatment of the infections caused by E. coli; further D. stramonium is also a potential source of natural antioxidants.

The aim of this study is to investigate antioxidant activity and total phenolic content of ethyl acetate fraction of Sembung leaves. Sembung (Blumea balsamifera L.) is a plant that has the potential as an antioxidant because it contains secondary metabolites of alkaloids, steroids, flavonoids, saponins, and phenolic compounds. In this study, antioxidant activity was tested by using 1,1-diphenyl-2-picrylhydrazyl (DPPH) for free radical scavenging activity assay. Evaluation of total phenolic content was tested by using the Folin-Ciocalteau method. The results showed that the ethyl acetate fraction of Sembung leaves possess showed medium antioxidant activity with IC50 valueof 221.821 compare with reference standard ascorbic acid with IC50 value 68.25 ppm. The ethyl acetate fraction of Sembung leaves has a total phenolic content of 161.101 mg GAE/g. This study provided that Blumea balsamifera leaves possess antioxidant.

This study aims to determine antioxidant activity and total phenolic content of mangosteen leaves in several different solvents. The antioxidant activity test of the extract was determined by the DPPH method and the total phenolic content was determined by the Folin-Ciocalteu method. The phytochemical test of mangosteen leaves showed the presence of phenolic compounds, saponins, triterpenoids and alkaloids. The extraction results increase with increasing water content in methanol, ethanol and acetone. The highest amount of extract was obtained using 50% acetone solvent (34.98 ± 0.17%). Extraction using 100% ethanol showed the highest total phenolic content (614.25 ± 5.58 mg GAE / g extract) and the lowest IC50 value (5.81 mg / L). The results of this study indicate that the use of 50% acetone solvent can optimize the amount of extract and 100% ethanol can optimize the content of total phenolic compounds and antioxidant activity of mangosteen leaves.

Olax scandens Roxb. (family Olacaceae) available in throughout tropical India. The current study, aerial parts of different concentrates of Olax scandens was evaluated for its in-vitro antioxidant potential by FRAP assay taking ascorbic acid as the standard and estimation of total phenolic content and flavonoids content. The IC50 value was originated that methanolic concentrates of Olax scandens are more efficient in antioxidant activity by FRAP methods compared EA & PE concentrates. The methanolic concentrates of Olax scandens & ascorbate exhibited antioxidant potential possessing IC50 207µg/ml & 50µg/ml by Ferric reducing ability Power assay. The methanolic and EA concentrates of Olax scandens showed the total phenolic content (14.426 ± 0.032, 4.128 ± 0.025) respectively, and ৒lavonoids content (11.526 ± 0.054, 3.682 ± 0.042) respectively. Invitro antioxidant studies show methanolic concentrates of Olax scandens have better antioxidant activity as well as a higher content of total phenolic and flavonoids content. These results indicate that aerial parts of methanolic concentrates Olax scandens could serve as a natural antioxidant, which may be useful in preventing free radical-induced diseases.

PENENTUAN TOTAL FENOLIK DAN AKTIVITAS ANTIOKSIDAN FRAKSI ETIL ASETAT DARI TEPUNG PELEPAH AREN (Arenga pinnata) ABSTRAK Tujuan penelitian ini untuk menentukan total fenolik dan aktivitas antioksidan fraksi etil asetat dari tepung pelepah aren (Arenga pinnata). Tepung pelepah aren diekstraksi maserasi menggunakan etanol hasil redestilasi cap tikus kemudian di partisi menggunakan pelarut petroleum eter, etil asetat dan aquades. Penentuan kandungan total fenolik menggunakan metode Folin-Ciocalteu dan aktivitas antioksidan menggunakan metode serapan radikal 1,1-difenil-2-pikrilhidrazil (DPPH). Didapati kandungan total fenolik fraksi etil asetat sebesar 172.857 ± 4.762 mg/kg dan aktivitas penangkal radikal bebas fraksi etil asetat sebesar 90.573 µg/mL. Hasil ini menunjukan bahwa fraksi etil asetat berpotensi sebagai antioksidan alami. Aren’s midrib powder extracted with maceration method with cap tikus redistillated ethanol and then partitioned with solvents such as petroleum ether, ethyl acetate and distillate water. The determination of phenolic content was using Folin-Ciocalteu method and the antioxidant activity was done using diphenylpycrylhydrazyl (DPPH) radical scavenging. Total phenolic value of ethyl acetate fraction is 172.857 ± 4.762 and IC50 value of ethyl acetate fraction is 90.573 µg/mL. This result shows ethyl acetate fraction as natural antioxidant potential. Worldwide, marine algae are known to produce a wide variety of bioactive secondary metabolites and several compounds have been derived from them for the prospective development of novel drugs by the pharmaceutical industry. In this context, Ulva lactuca, Enteromorpha intestinales, and Cladophora vagabunda, isolated from Black Sea coastal waters of Romania, were evaluated for their potential for bioactivity. Extracts of the algae selected for the study were prepared using ethanol. The study presents original results concerning the total phenolic content (TPC), determined by the spectrometric method (Folin-Ciocalteu method) and the individual phenols from each species of seaweed studied were identified by HPLC-DAD techniques. Total antioxidant activity (ACL) was studied by photochemiluminescence method, and antibacterial activity was studied on two strains of bacteria, Escherichia coli ATCC 10536 - gram-negative and Staphylococcus aureus ATCC 6538P - gram-positive. Results indicated that the highest total phenolic content was obtained for Cladophora vagabunda (325.8 mg/100 g f.w.), followed by Ulva lactuca (285.5 mg/100 g f.w.) and the lowest rate was registered by Enteromorpha intestinalis (268.8 mg/100 g f.w.). Through HPLC-DAD techniques, 6 individual phenolic compounds were identified in all analyzed seaweeds. In addition, two other phenolic compounds were identified in lower concentrations in Ulva lactuca, but only one phenolic compound in Enteromorpha intestinalis. The results of this study show that there is a strong correlation between antioxidant activity (ACL), antibacterial activity and total phenolic content (TPC). Antioxidant activity shows the same order as total phenolic content (TPC): Cladophora vagabunda (64.15 nmol/g sample) followed by Ulva lactuca (22.8 nmol/g sample) and Enteromorpha intestinalis (10.4 nmol/g sample). The antibacterial activity recorded in all green seaweeds shows that the inhibition sites are higher on Escherichia coli compared to Staphylococcus aureus.

Eucalyptus (Eucalyptus camaldulensis) and Myrrh (Commiphora Myrrha) essential oils (EOs) stand out for their benefits in terms of health and functionality. Buffalo set yogurt enriched with different concentrations of EOs (0.3, 0.6, and 0.9%) were investigated. The effects of addition on sensory, syneresis, antibacterial activity, and bioactive properties (total phenol content and antioxidant activity) of yogurt were studied. The most acceptable organoleptic properties of treated yogurt were those samples treated with Eucalyptus oil. The levels of syneresis were decreased by increasing the concentration of EOs. Moreover, the antioxidant activity, antibacterial activity, and total phenolic content were enhanced by increasing the concentration of EOs. Yogurt with 0.9% Eucalyptus oil showed the highest antioxidant activity and total phenolic content. The same concentration of Eucalyptus oil showed the highest antibacterial activity against S. typhimurium (the inhibition zone was 20.63 mm) then E. coli (the inhibition zone was 19.43 mm). On the other hand, the highest antibacterial effect against L. monocytogene was for Myrrh oil-enriched yogurt by 0.9% and the inhibition zone was 19.21 mm. The obtained results showed that Eucalyptus and Myrrh oils can be applied to yogurt to improve its beneficial properties in terms of physical characteristics and for human health due to their antioxidant activity and phenolic materials.

Leaves and drupes of the olive varieties "Koroneiki", "Lianolia Kerkyras", "Mastoidis", "Arbequina", "Adramytini", "Megaritiki", "Gaidourelia", "Kalamata", "Konservolia" and "Chalkidiki", were collected at different seasons during the years 2008 & 2009 and investigated for their total phenolic content and antioxidant activity. Total phenolic content and antioxidant activity were determined according to Folin-Ciocalteau and DPPH assays, respectively. Different varieties presented differences in the total phenolic content and total antioxidant activity per tissue. For both years, most varieties showed higher total phenolic content in new season leaves than in green and black drupes. New season leaves on April showed the highest total phenolic content than the leaves of September and December of the same year, with a few exceptions. There was a positive correlation between the antioxidant activity and the total phenolic content. Highest phenolic content was accompanied by a lower IC50 value and as a result higher antioxidant activity.

This study intended to improve physiological characteristics of Magnolia officinalis bark (MOB) extracts by Aspergillus niger fermentation. M. officinalis bark was extracted using distilled water, 95% ethanol, and methanol, and it was then fermented by A. niger. The physiological characteristics of the fermented extracts, namely, tyrosinase inhibitory activity, antioxidant activity, antibacterial activity, and anti-skin-aging activity, were evaluated and compared with those of unfermented extracts. To determine the safety of the fermented extracts, their cytotoxicity was analyzed by measuring the cell viability of CCD-966SK and human epidermal melanocytes (HEMn) after exposure. The fermented methanol extract exhibited the highest antityrosinase activity, total phenolic content, and antioxidant activity. The total phenolic content of the extracts fermented by A. niger was 3.52 times greater than that of the unfermented extracts. The optimal IC50 values for tyrosinase inhibition and 2,2-diphenyl-1-picrylhydrazyl (DPPH) removal by the A. niger-fermented extracts were 30 and 12 μg/mL, respectively. The fermented methanol extracts inhibited skin-aging-related enzymes such as collagenase, elastase, MMP-1, and MMP-2. Compared with the unfermented extracts, the fermented extracts also contained greater antibacterial activity against tested stains including MRSA. These results could be attributed to an increase in the concentration of original active compounds and the biosynthesis of new compounds during fermentation. In cytotoxicity assays, the A. niger-fermented extracts were nontoxic to CCD-966SK cells, even at 500 μg/mL. Hence, in general, methanol-extracted M. officinalis fermented by A. niger for 72 h has the most active antioxidant, skincare, or antiaging compounds for healthy food or cosmetics applications.

This study was aimed at the extraction of total phenolic compounds, as well as, the antioxidant activity of black glutinous rice (Oryza sativa L.). Black glutinous rice of three varieties consists of Leum Pua, Doi Saket, and Phayao were extracted using citric acid solvent. The influences of temperature, time and pH value on total phenolic content and antioxidant activity (percentage of inhibition of DPPH free radical) were investigated. A central composite design was employed to determine the optimum extraction condition to obtain the highest total phenolic content and antioxidant activity. The regression models of total phenolic content were significant (p0.05) and determination coefficients values were 0.8028, 0.8089, and 0.8603 of Leum Pua, Doi Saket, and Phayao, respectively. The regression models of DPPH inhibition were significant (p0.05) and determination coefficients values were 0.8084, 0.8928, and 0.8243 of Leum Pua, Doi Saket, and Phayao, respectively. The selected optimum conditions were 72°C, 91 mins at pH 1.44 for Leum Pua, 72.5°C, 83 mins at pH 1.39 for Doi Saket, and 66.3°C, 75 mins at pH 1.32 for Phayao. Under the optimum condition, total phenolic content and percentage of inhibition of DPPH free radical of extract from Leum Pua, Doi Saket and Phayao were 0.3814 mg GAE g-1 DW with 89.36%, 0.2607 mg GAE g-1 DW with 73.33%, and 0.2617 mg GAE g1 DW with 75.27%, respectively. The results showed that the total phenolic content of the extract indicated a highly positive correlation with antioxidant activity.
